本発明の目的、技術構成および利点を明確にするために、以下具体的な実施例を示して、本発明をより詳細に説明する。一般当業者が理解できるように、以下の実施形態は例示を目的とするものであり、本発明の保護範囲は、後述する請求項の範囲に基づくものである。
(1)抗VEGF抗体、変異体の発現ベクターの構築
本文に記載された抗体(その対応する野生型配列は、USP Medicines Compendiumから得られたものであり、具体的には、重鎖配列は配列番号:1に示され、軽鎖配列は配列番号:2に示され;これを基に、本発明の変異体166Cは、kabatナンバリングによる軽鎖166位のアミノ酸をシステインに置換して得られたものであり、コントロール抗体124Cは、軽鎖124位のアミノ酸をシステインに置換して得られたものである)をコードするDNAは、化学合成した(GENEWIZ,Inc. Genomics Suzhou, China)。その後、抗体の重鎖遺伝子をBstBIとPacIで二重消化し、軽鎖遺伝子をHindIIIとEcoRIで二重消化した。重鎖遺伝子は、T4リガーゼにて、BstBIとPacIで処理した真核発現ベクターであるpCGS3(Biovector NTCC Inc.)にライゲーションし、ライゲーションした後、HindIIIとEcoRIで二重消化して、T4リガーゼで軽鎖遺伝子を挿入し、重鎖と軽鎖の両方を含む発現ベクターの構築した。
構築した発現ベクターをDH5αコンピテントエシェリヒア・コリ菌株に形質転換した。形質転換されたDH5α株は、BstBIとPacIの2種の酵素とHindIIIとEcoRIの2種の酵素でそれぞれ二重消化して、シーケンス解析して確認した。ポジティブクローンは選択した。得られたエシェリヒア・コリは、プラスミド抽出キット((CWBIO, Beijing, Chinaから購入)を用いて、溶解してプラスミドを抽出して、重鎖と軽鎖の両方を含む発現ベクターを得た。
精製した発現ベクターは、電気穿孔法でCHO-K1細胞(ATCCから購入)にトランスフェクションし、96ウェルプレートに播種した。15日間培養した後、モノクロナールをピックアップし、ELISAにて細胞の抗体生産量を測定し、ベストの20%の株をピックアップして24ウェルプレートに播種した。7日間培養した後、細胞の抗体生産量を測定し、ベストの5つ~6つの株をピックアップしてローラーボトルに移して培養した。CHO-K1細胞での抗VEGF抗体の発現を達成した。無血清培養が安定した後、7日間の連続培養後に細胞を採取し、精製して後続実験に使用した。

精製した発現ベクターは、電気穿孔法でCHO-K1細胞(ATCCから購入)にトランスフェクションし、96ウェルプレートに播種した。15日間培養した後、モノクロナールをピックアップし、SDS-PAGEにて細胞の抗体生産量を測定し、ベストの20%の株をピックアップして24ウェルプレートに播種した。7日間培養した後、細胞の抗体生産量を測定し、ベストの5つ~6つの株をピックアップしてローラーボトルに移して培養した。CHO-K1細胞での抗抗体の発現を達成した。無血清培養が安定した後、7日間の連続培養後に細胞を採取し、Niアフィニティークロマトグラフィーカラムで精製して後続実験に使用した。
細胞と培地を50 mL遠心チューブに移して、高速凍結遠心機にセットし、8000rで15 min遠心した。上清を回収し、細胞デブリを捨てた。培地を0.22μmのマイクロポアろ過膜で一回ろ過し、ろ液を回収して使用まで予備した。ポンプ、シリンジ及び精製カラムは、カラムボリューム10倍の超純水、PBSで洗浄し、ローディングを待った。ローディング;0.1 M Gly-HClで溶出して、精製した抗体蛋白液をエッペンチューブに引取し、1.5 M Tris-HClでpHを7~8に調整し、2-8℃で保存した。結果を図1に示した。
本発明は、Ellmanの方法(Thermo Fisher Scientific (China) Co, Ltd.)を用いて、抗体製品における遊離スルフヒドリル基の含有率を検出することにより、さらなるコンジュゲーションに適した細胞株を選択した。0.1 M Na2HPO4溶液、DTNB(10 mM)溶液、システイン標準溶液を終濃度0.0059M~0.375Mになるように調製した。同時、濃度5 mg/mLの被検抗体サンプルを準備した。システイン標準溶液または被検サンプルを入れた1.5 mLエッペンチューブに、それぞれ、0.1 M Na2HPO4を250μL 、DTNB溶液を5μL加え、vortexしてよく混ぜた。避光して室温で5 minインキュベートした。波長412 nmのOD値を測定した。OD値をもとに標準曲線を作成し、サンプルのスルフヒドリル基の含有率を算出した。
抗体とコンジュゲートのコンジュゲート反応の条件は、抗体:コンジュゲート(ポリペプチド又はmPEG2000-MAL)=1:30(モル比)、反応温度37℃、反応時間3~4h。反応バッファーはPBS(pH 7.2)であった。抗VEGF抗体とmPEG2000-MAL(化学試薬会社から購入、例えば、SHANGHAI ZZBIO CO . , LTD、製品認識ナンバリングZZP-MPEG-MAL-2K-01))とコンジュゲートした後のSDS-PAGE電気泳動図は、図2に示した。
ポリペプチドは、Ang2中和作用を有する活性ポリペプチドであり、配列はGln-Lys(Ac)-Tyr-Gln-Pro-Leu-Asp-Glu-Lys(Ac)-Asp-Lys-Thr-Leu-Tyr-Asp-Gln-Phe-Met-Leu-Gln-Gln-Gly-CONH2(配列番号:33)(参考文献のHanhua Huang, Jing-Yu Lai, Janet Do, Dingguo Liu, Lingna Li, Joselyn Del Rosario. Specifically Targeting Angiopoietin-2 Inhibits Angiogenesis, Tie2-Expressing Monocyte Infiltration, and Tumor Growth. Clin Cancer Res; 17(5) March 1, 2011,1001-1011参照)であり、リンカーと接続するためのLysは下線で示した。前記リンカーはホルムアミド-PEG4-NHSであり、構造式ば下記式Iに示した:

サンプルは、限外ろ過チューブで前処理して、2回蒸留水(DDW)に溶解した。得られたサンプルは、マススペクトル解析をした。マススペクトルシステムとしては、高性能タンデム質量分析Triple TOF 4600 LC/MS/MSシステム(SCIEX China)、エレクトロスプレーイオン化源を用いて、ポジティブイオンモードで解析した。マススペクトルパラメータ:GS1:55;GS2:55;CUR:35;スキャンレンジ:100-2000 Da;ネブライジング電圧(ISVF):5500 V;ネブライジング温度:350℃;デクラスタリング電圧(DP):150 V;衝突エネルギー(CE):10 eV。結果は図10-13に示した。図10と図11から、DCとコンジュゲートした後、変異体166Cは、メインピークが右に6478.8 Da(両分子のDCの分子量に相当)分シフトしたことがわかる。これは、166Cの両方のスルフヒドリル基はDCにコンジュゲートしたことを証明した。
図12~13から、DCとコンジュゲート反応した後、変異体124Cは、それぞれ、0分子のDCとのコンジュゲート、1分子のDCとのコンジュゲート、2分子のDCとのコンジュゲートのピークが生じ、コンジュゲートの均一性と効率性の両方が166Cより低いことがわかる。
バイオフィルム干渉技術(BLI)、Pro A probe(ForteBio)を用いて、野生型抗体、抗体変異体、抗体ポリペプチドコンジュゲートの、VEGFとのアフィニティーを測定した。抗体サンプルはPBSに溶解して、作業濃度10μg/mLとした。VEGF-165(Sino Biological Inc.)はPBSに溶解して、それぞれ50nM、75 nM、100 nM、150 nM、200 nM濃度にグラジェント希釈した。作業容量はすべて200μLとした。
バイオフィルム干渉技術(BLI)、Pro A probeを用いて、野生型抗体、抗体変異体、抗体ポリペプチドコンジュゲートの、Ang2とのアフィニティーを測定した。抗体サンプルはPBSに溶解して、作業濃度20μg/mLとした。Ang2(Sino Biological Inc.)はPBS(0.02% tween-20,0.1% BSA)に溶解して、それぞれ25nM、50 nM、75 nM、100 nM、 200nM濃度にグラジェント希釈した。作業容量はすべて200μLとした。
OCTETシステムとデータ処理ソフトウェアでデータ図をフィッティングし、ソフトにて抗体などとVEGF又はAng2との分子間力を算出し、KD値で表し、その結果を表5に示した。
抗体によるVEGF165中和作用は、HUVEC細胞(ScienCell Research Laboratories,Inc.)で検出した。96ウェルプレートに、4000細胞/50μL/ウェルの接種密度で細胞を接種した。ブランクコントロールウェルを設定した。コントロールウェルは6つ以上であった。グラジェント希釈方法により、グラジェント濃度の抗体サンプル(0.001μg/mL、0.01μg/mL、0.05μg/mL、0.1μg/mL、0.5μg/mL、1μg/mL、5μg/mL、10μg/mL)を調製した。VEGF165を、試験培地で110 ng/mLの濃度になるように希釈した。調製したVEGF165試薬は、VEGF165の終濃度が10 ng/mLになるように、グラジェント濃度のサンプルと混合した。96ウェルプレートをCO2インキュベータインに入れて、48 hキュベーションした後、CCK-8カウントした。サンプル及びそのコントロールのOD値から、増殖率を算出した。結果は図14に示し、野生型抗体(WT)、変異体(124Cと166C)およびコンジュゲート(124ADCと166ADC)は、内皮細胞であるHUVECの増殖(VEGF刺激による増殖)に対する抑制効果を示した。上記図から、抗体変異体は、選択された部位のシステイン変異により活性が変化しなかったことが分かり、選択された部位が抗体の生物的活性に影響を与えていないことが証明した。同様に、DCにコンジュゲートすることで生じたコンジュゲートも、抗体の細胞増殖に対する抑制活性に影響をを与えなかった。

- 配列番号:1のアミノ酸配列の重鎖と、配列番号:7のアミノ酸配列の軽鎖とを含むか、配列番号:9のアミノ酸配列の重鎖と、配列番号:13のアミノ酸配列の軽鎖とを含むか、又は、配列番号:17のアミノ酸配列の重鎖と、配列番号:21のアミノ酸配列の軽鎖とを含む、抗体又はその抗原結合断片であって、前記抗原結合断片が、Fab断片、Fab’断片又はF(ab’)2断片である、抗体又はその抗原結合断片。
- 前記Fab断片の重鎖アミノ酸配列が配列番号:25の配列であり、前記Fab断片の軽鎖アミノ酸配列が配列番号:29の配列である、請求項1に記載の抗体又はその抗原結合断片。
- 請求項1又は2に記載の抗体又はその抗原結合断片を含む、二重特異性抗体又は融合タンパク質。
- 請求項1又は2に記載の抗体又はその抗原結合断片を含む、コンジュゲート。
- ポリエチレングリコール、細胞障害性薬剤、活性ペプチド、ナノボディ、単一ドメイン抗体、Fab断片、Fab’断片、scFv、低分子薬、化学療法剤又は放射線療法剤からなる群から選ばれる別の機能性分子を、さらに含む、請求項4に記載のコンジュゲート。
- 前記活性ペプチドがAng2中和活性ペプチドである、請求項5に記載のコンジュゲート。
- 前記活性ペプチドが配列番号:33のAng2中和活性ペプチドである、請求項5に記載のコンジュゲート。
- 前記Ang2中和活性ペプチドが、リンカーを介して抗体にコンジュゲートしており、前記抗体が、配列番号:1のアミノ酸配列の重鎖と、配列番号:7のアミノ酸配列の軽鎖とを含む、請求項6又は7に記載のコンジュゲート。
- 前記ポリエチレングリコールが、モノメトキシポリエチレングリコールである、請求項5に記載のコンジュゲート。
- 前記ポリエチレングリコールが、mPEG2000、mPEG5000、又はmPEG10000である、請求項5に記載のコンジュゲート。
- 前記ポリエチレングリコールがさらに薬物分子とコンジュゲートしている、請求項5~11のいずれか1項に記載のコンジュゲート。
- 前記ポリエチレングリコールが直鎖状二官能基化ポリエチレングリコール、直鎖状ヘテロ官能基化ポリエチレングリコール又はMulti-Arm官能基化PEGである、請求項5~12のいずれか1項に記載のコンジュゲート。
- 請求項1又は2に記載の抗体又はその抗原結合断片、請求項3に記載の二重特異性抗体又は融合タンパク質、又は請求項4~13のいずれか1項に記載のコンジュゲートを含み、任意選択的に、薬学的に許容可能な担体を含む、医薬組成物。
- 請求項1又は2に記載の抗体又はその抗原結合断片、請求項3に記載の二重特異性抗体又は融合タンパク質、請求項4~13のいずれか1項に記載のコンジュゲート、又は請求項14に記載の医薬組成物の、癌を治療するための医薬品又はキットの製造における、使用。
- 前記癌が、非小細胞肺癌、結腸直腸癌、乳がん、悪性神経膠腫、又は腎細胞がんである、請求項15に記載の使用。
- 前記癌が、進行期、転移性又は再発性非扁平上皮非小細胞肺がん又は転移性結腸直腸癌である、請求項15に記載の使用。
- 請求項1又は2に記載の抗体又はその抗原結合断片、請求項3に記載の二重特異性抗体又は融合タンパク質、請求項4~13のいずれか1項に記載のコンジュゲート、又は請求項14に記載の医薬組成物を含むキット。
- さらに、前記抗体又はその抗原結合断片と併用する薬剤を含む、請求項18に記載キット。
- 前記薬剤が、カルボプラチン又はシスプラチンである、請求項19に記載キット。
