Description
i)一般的にnOMV表面に結合した少なくともnOMV糖成分を活性化するステップ、及び
ii)このようにして得られた活性化糖を少なくとも1つの選択された抗原に結合するステップ
を含む、前記コンジュゲートを調製する方法に言及する。

種々の酸化条件を試験した。例えばnOMV ΔtolR 1418の酸化について、範囲5~20mMのNaIO4濃度を試験した。-OAg鎖長への酸化の効果を評価した。鎖長は、酸化が進行するに従って低減した。より高いNaIO4濃度は、OAgサイズを低減させる傾向があった。NaIO4モル濃度を増加させることは、5から45%の高い酸化レベルをもたらすだけでなく(方法に関与する主たる糖としてラムノースを用いて)、-OAg鎖長を低減したことが実証された。同時に小胞サイズ分布に変化はなく、小胞の完全性は維持されていた。
nOMV-Agコンジュゲーションステップ(fVi-ネズミチフス菌nOMV;リンカーを介した間接コンジュゲーション)
fViを無作為(r)又は選択的に(s)のいずれかでADHリンカーとの反応によって改変した。改変fViは、次に、還元的アミノ化を使用して酸化ネズミチフス菌nOMVにコンジュゲートした。fViがADHを用いて無作為に活性化された場合、fVi対nOMVの1:1の重量比をコンジュゲーションにおいて使用し、一方3:1の重量比をfViが末端でADHにより誘導体化された場合に使用した。コンジュゲートは全タンパク質含有量(nOMV回収)を決定するためのマイクロBCA/ローリーアッセイを使用して特徴付けした。高速アニオン交換クロマトグラフィー-パルスドアンペロメトリー検出器(HPAEC-PAD)を全Vi含有量(nOMV干渉無し)を決定するために使用した。高速液体クロマトグラフィーサイズ排除クロマトグラフィー(HPLC-SEC、TSK gel 3000 PWxlカラムを使用)を、示差屈折率(dRI)を使用して遊離Vi%を算出するために使用し、動的光散乱/高速液体クロマトグラフィー-サイズ排除クロマトグラフィー/多角度静的光散乱(DLS/HPLC-SEC MALS)を、サイズを決定するために使用した。本コンジュゲーション方法のコンジュゲーション条件の、nOMVあたりのfVi鎖の最終数への影響を、ナノ粒子トラッキング分析(NTA)によるnOMVの数と併せてHPAEC-PAD回収データを検討して評価した。
Vi-ネズミチフス菌nOMVコンジュゲート(リンカーを介した間接コンジュゲーション)
上の表3によるコンジュゲート1及び5(それぞれVi:nOMVのw/w比0.45及び0.44を有する)をマウスにおいて試験した。比較のために、CRM197も担体として使用し、fVi+nOMVの単純混合物も試験した。
nOMV-Agコンジュゲーションステップ(ETEC CTF1232-ネズミチフス菌ΔtolR 1418又はソンネ菌1790 nOMV;リンカーを用いない直接コンジュゲーション)
CTF1232は、大腸菌抗原である(配列番号14、2種類のnOMV小胞にコンジュゲートされたC-末端ヘキサ-ヒスチジンタグを含む)。ポリペプチドは、小胞における酸化糖への連結のために使用できる5個のリシン残基を含む。
nOMV-Agコンジュゲーションステップ(Na2SO3の存在下でのPfs25-ネズミチフス菌nOMVコンジュゲーション)
マラリア抗原Pfs25を2つの異なる化学によって、すなわちSH-マレイミド又はクリック化学によりタンパク質へ又はNaIO4酸化OAgへにより、ネズミチフス菌nOMV小胞にコンジュゲートした。酸化-OAgへの連結のために、1:1比のnOMV:Pfs25を、PBS中2.6mg/mlのPfs25濃度で、室温での一晩インキュベーションで使用した。コンジュゲート形成は、過剰のNaIO4をNa2SO3を用いてクエンチし(濃度10mMが本実験において、10分間使用された)、続いて同じポットへのPfs25の直接添加(0.2mg/ml最終濃度)を行う場合にも得られた。図4Aは、3種のコンジュゲート;Pfs25単独;又は小胞と物理的に混合したPfs25、に応答した抗Pfs25 IgG力価を示している。全ての構築物は、アルハイドロゲルを用いて製剤化された。マウスを0及び28日目に0.1μg Pfs25/用量で皮下免疫した。抗Pfs25 IgG力価を0、14、28及び42日目に測定した。
RO6C-ネズミチフス菌nOMVコンジュゲート(リサイクルステップ)
マラリア原虫RO6C抗原を還元的アミノ化を使用して酸化ネズミチフス菌nOMV小胞にコンジュゲートした。さらなるコンジュゲートを第1のコンジュゲーションバッチから未反応RO6Cをリサイクルし、それをコンジュゲーションのために再使用することによって作製した。全タンパク質に対するRO6Cの比を競合的ELISAによって測定したところ、非リサイクルコンジュゲートについて7.2%、リサイクルコンジュゲートについて11.1%であった。比較のために、RO6C単独を使用した。全ての構築物は、アルハイドロゲルを用いて製剤化された。
実施例7a
dOMV(ナイセリア・メニンギティディスB群由来)のfHbp v3との反応(反応無し)
本実施例のdOMVを界面活性剤抽出工程によって調製し、ここでデオキシコール酸(deoxhycholate)を選択された界面活性剤として使用する。それにより得られた界面活性剤抽出小胞を本発明の方法により、選択された抗原(fHbp)と反応させた。具体的には、0.96mg/mL濃度のdOMVを、NaIO4を10mMで用いて30分間、室温で暗所にてインキュベートした。過剰のNaIO4を最終濃度20mMのNa2SO3により、15分間、室温でクエンチした。fHbp(dOMVとfHbpとのw/w比1:1、dOMV濃度0.335mg/mL)及びNaBH3CN(3mg)を反応混合物に直接加えた。一晩室温で穏やかに撹拌後、コンジュゲートを超遠心(110000rpm、4℃、1時間)によって精製し、PBS中に再懸濁し、SDS PAGE/ウエスタンブロットによって分析した。
nOMV(ナイセリア・メニンギティディスB群由来)のfHbp v3との反応(本発明のnOMV-fHbpコンジュゲートの形成)
本実施例のnOMVを、Koeberling et a. Vaccine (2014) 32:2688に記載のとおり、いかなる界面活性剤も使用することなく調製した。それにより得られた抽出小胞を、本発明の方法により選択された抗原(fHbp)と反応させた。具体的には、濃度0.96mg/mLのnOMVを5mM NaIO4と30分間、室温で暗所にてインキュベートした。過剰のNaIO4を最終濃度20mMのNa2SO3により、15分間、室温でクエンチした。fHbp(dOMVとfHbpとのw/w比1:1、dOMV濃度0.335mg/mL)及びNaBH3CN(3mg)を反応混合物に直接加えた。一晩室温で穏やかに撹拌後、コンジュゲートを超遠心(110000rpm、4℃、1時間)によって精製し、PBS中に再懸濁し、SDS PAGE/ウエスタンブロットによって分析した。SDS PAGE/抗fHbpウエスタンブロット分析では、nOMVを用いた還元的アミノ化化学によってのみコンジュゲート形成が確認され、dOMVを用いた還元的アミノ化化学では確認されなかった。10%SDS pageゲル。
実施例7bの手順に従ったnOMV(サルモネラ由来)のfHbp v1との反応
実施例7bと同じ実験をネズミチフス菌由来nOMVを使用して実施し、同様の結果が得られ、本発明のnOMV-fHbpコンジュゲートが得られた。
本発明に従った、(NANP)3-SH-Pfs25を使用する多官能基化nOMVの調製
Pfs25タンパク質を次の手順によりEMCSリンカーで誘導体化した。PBS緩衝液中、濃度2.6mg/mLのPfs25をEMCSリンカーに加えた(EMCSリンカーとPfs25 Lys残基とのモル比0.3)。反応物を室温で4時間混合した。得られた誘導体化タンパク質(Pfs25-EMCS)をPD10カラムによって10mM NaH2PO4 pH6に対して精製した。MALDI-TOF MSによる分析は、Pfs25分子あたりEMCSリンカー平均4個が導入されたことを明らかにした。(NANP)3とEMCSリンカーとのモル比3:1、Pfs25濃度0.7mg/mLとなるように(NANP)3をPfs25-EMCSの溶液に加えた。反応物を室温で一晩混合した。この後、Pfs25-(NANP)3誘導体をPBS緩衝液に対しVivaspin 10Kによって精製した。SDS PAGE/ウエスタンブロット及びMALDI-TOF MSによる分析では、産物形成が確認された。100mM NaH2PO4、pH6.5中のネズミチフス菌nOMV(濃度2.1mg/mL)を5mM NaIO4と共に30分間、25℃で暗所にてインキュベートした。過剰のNaIO4を最終濃度10mMのNa2SO3により、10分間、室温でクエンチした。Pfs25-(NANP)3(nOMVとPfs25-(NANP)3とのw/w比1:1、nOMV濃度0.45mg/mL)及びNaBH3CNを反応混合物に直接加えた。一晩室温で穏やかに撹拌後、コンジュゲートを超遠心(110000rpm、4℃、30分間)によって精製し、PBS中に再懸濁し、SDS PAGE/ウエスタンブロットによって分析し、コンジュゲート形成を確認した。
本発明の実施形態に従った、クエンチを用いて又は用いずにネズミチフス菌nOMV粒子をPfs25抗原にコンジュゲートすることによって得られた本発明のコンジュゲートのin-vivoデータ
CD1雌マウスを、クエンチステップを用いて又は用いずにPfs25抗原にコンジュゲートした全タンパク質ネズミチフス菌nOMV粒子2.5μgを用いて0及び28日目に皮下免疫した(実施例5を参照されたい)。両方のコンジュゲートは、競合的ELISAにより20%に近い、全タンパク質に対するPfs25のw/w比を示し、アルハイドロゲル(0.7mg/mL Al3+)に吸着した。抗Pfs5及び抗OAg IgG力価を0、14、27及び42日目に測定した。すべての時点で、2つのコンジュゲートは、図6aに示すとおり、同様の抗Pfs25 IgG応答を誘導し(マンホイットニー検定)。同様に、図10bに示すとおり、コンジュゲートは、同様の抗OAg IgG応答を誘導できた。還元的アミノ化によるコンジュゲーションにおけるクエンチのステップは、マウスにおいて誘導された免疫応答に影響を与えることなく導入することができ、それによりGMMA酸化中間体精製を回避することができる。
nOMV(ナイセリア・メニンギティディスB群由来)とMenCとの反応(本発明のnOMV-MenCコンジュゲートの形成)
MenC多糖を100mM AcONa pH4.5中に濃度40mg/mLで可溶化した。ADHリンカー及びNaBH3CNを、MenC/ADH/NaBH3CNのw/w比1:1.2:1.2でそれぞれ加えた。混合物を30℃で一晩加熱し、次にG10カラムによって脱塩した。TNBS比色分析法及びHPAEC-PADによる特徴付けは、100%誘導体化を示した。
nOMV(ナイセリア・メニンギティディスB群由来)とMenAとの反応(本発明のnOMV-MenAコンジュゲートの形成)
MenA OSを100mM AcONa pH6.5中に濃度40mg/mLで可溶化した。ADHリンカー及びNaBH3CNを、MenA/ADH/NaBH3CNのw/w比1:1.2:1.2でそれぞれ加えた。混合物を30℃に5日間加熱し、次にG10カラムによって脱塩した。TNBS比色分析法及びHPAEC-PADによる特徴付けは、90%誘導体化を示した。fHbpを過剰発現しているMenB GMMAへのコンジュゲーションは、MenC多糖について記載されたとおりに実施した。
MenA及びMenCとのnOMV(ナイセリア・メニンギティディスB群由来)のコンジュゲートの調製
MenA-及びMenC-MenB GMMAコンジュゲートの合成について記載されたのと同じコンジュゲーション条件を同じGMMA粒子上の両方の多糖のコンジュゲーションのために使用した。GMMAを既に記載のとおり酸化し、Na2SO3によるクエンチ後、MenA-ADH及びMenC-ADHを同時にそれぞれMenA/MenC/GMMAのw/w比8:8:1で加えた。

Claims (20)
- 少なくとも抗原に結合された少なくとも表面糖成分を有する、機能性TolRタンパク質を発現しない大腸菌、サルモネラ菌又は赤痢菌の細菌株に由来する未変性の外膜小胞(nOMV)を含む免疫原性コンジュゲート。
- 前記大腸菌、サルモネラ菌又は赤痢菌の細菌株が、msbB、htrB、ddg及び/又はPagP遺伝子の変異をさらに含む、請求項1に記載の免疫原性コンジュゲート。
- 第1の抗原に結合された少なくとも表面糖成分を有するnOMVを含み、前記第1の抗原は、第1の抗原とは異なる第2の抗原に結合されている、請求項1に記載の免疫原性コンジュゲート。
- 第1の抗原に結合された少なくとも表面糖成分及び第1の抗原とは異なる第2の抗原に結合された少なくとも別の表面糖成分を有するnOMVを含む、請求項1に記載の免疫原性コンジュゲート。
- 前記抗原が、免疫原性ポリペプチド又は免疫原性多糖である、請求項1~4のいずれか一項に記載の免疫原性コンジュゲート。
- 前記nOMV及び前記抗原が、同じ又は異なる細菌株由来である、請求項1~5のいずれか一項に記載の免疫原性コンジュゲート。
- 前記ポリペプチドが、大腸菌ポリペプチド抗原;N.メニンギティディスポリペプチド抗原;連鎖球菌ポリペプチド抗原;並びにマラリア原虫ポリペプチド抗原からなる群から選択される、請求項5に記載の免疫原性コンジュゲート。
- 前記大腸菌ポリペプチド抗原が、CTF1232、405及び3526からなる群から選択される、請求項7に記載の免疫原性コンジュゲート。
- 前記N.メニンギティディスポリペプチド抗原が、NHBA、NadA、NsPA、NhhA、App及びfHbpからなる群から選択される、請求項7に記載の免疫原性コンジュゲート。
- 前記連鎖球菌ポリペプチド抗原が、A群連鎖球菌由来のGAS25(Slo)、GAS40(SpyAD)及びGAS57(SpyCEP)抗原からなる群から選択される、請求項7に記載の免疫原性コンジュゲート。
- 前記マラリア原虫ポリペプチド抗原が、熱帯熱マラリア原虫由来のPfs25である、請求項7に記載の免疫原性コンジュゲート。
- 前記多糖が、インフルエンザ菌B型多糖;ナイセリア・メニンギティディス血清群A、C、W135、X及びY多糖;肺炎球菌血清型1、2、3、4、5、6A、6B、7F、8、9N、9V、10A、11A、12F、14、15B、17F、18C、19A、19F、20、22F、23F及び33F多糖;チフス菌(Salmonella enterica serovar Typhi)Viを含むサルモネラ菌多糖、全長又は断片(fViと示される)のいずれか;ストレプトコッカス・アガラクチア血清型Ia、Ib及びIII多糖;化膿連鎖球菌多糖;赤痢菌種多糖;並びにA及びB群連鎖球菌多糖からなる群から選択される細菌性莢膜多糖である、請求項5に記載の免疫原性コンジュゲート。
- 前記nOMVがGMMA小胞である、請求項1~12のいずれか一項に記載の免疫原性コンジュゲート。
- 前記nOMVを、機能性TolRタンパク質を発現しない大腸菌、サルモネラ菌又は赤痢菌の遺伝子改変された細菌株から産生させるステップ、
そのようにして産生されたnOMVの表面上の糖成分を活性化するステップ、及び
活性化糖成分を選択された抗原に直接結合するステップ、又は
活性化糖成分を二価リンカー基に結合して、nOMV-リンカーコンジュゲートを形成し、その後それを選択された抗原に結合するステップ、
を含む、請求項1~13のいずれか一項に記載の免疫原性コンジュゲートを調製する方法。 - 前記nOMVの表面上の糖成分を活性化するステップが、前記nOMV表面糖成分の1つ以上のヒドロキシル基のアルデヒド官能基への酸化を含む、請求項14に記載の方法。
- 前記酸化が、アルカリサルファイトによりクエンチされる、請求項15に記載の方法。
- 前記アルカリサルファイトがNa 2 SO 3 である、請求項16に記載の方法。
- 請求項1~13のいずれか一項に記載の免疫原性コンジュゲート及び少なくとも1つの薬学的に許容される担体又は賦形剤を含む、免疫原性組成物。
