本出願は、その全体が参照により本明細書に組み入れられる、2018年8月23日に提出された米国仮出願第62/721,679号の優先権を主張する。
本開示は、治療有効量のGsx1タンパク質(例えばGsx1-細胞透過性ペプチド融合タンパク質)またはGsx1をコードする核酸分子を投与することにより神経障害を処置することによって、外傷性脊髄損傷もしくは脳損傷等の神経障害またはパーキンソン病等の障害を処置するための方法を提供する。
本発明は、米国教育省によって授与されたP200A150131の下、米国国立衛生研究所によって授与されたT32GM008339の下、および米国教育省によって授与されたP200A150131の下、政府支援を受けてなされた。政府は本発明に一定の権利を有する。
全米脊髄損傷統計センターによると、米国では毎年およそ17,000の新たな脊髄損傷(SCI)の事例が存在する。SCIの主な原因としては、車両衝突、仕事に関連する損傷、転落、暴力行為、およびスポーツが挙げられる。SCIは、乏突起膠細胞、ニューロン、および星細胞の著しい喪失を引き起こし、長期ケアを必要とする病的状態および身体障害の原因となる。広範囲にわたる探究にもかかわらず、SCI後の神経再生および機能回復は非常に困難である。
ゲノムスクリーニングホメオボックス1(Gsx1またはGsh1)は、胚形成期の中枢神経系において高度に発現する神経原性因子である(Gong et al., Nature 425:917-925 (2003))。脊髄の発生の間、Gsx1とその相同体のGsx2とは神経幹/前駆細胞(NSPC)の増殖および分化を調節する。成体の脊髄では、Gsx1発現は低いかまたは検出されない(Chen et al., PLoS One 8, e72567 (2013))。
外側片側切断損傷を有する成体マウス脊髄にGsx1を形質導入するためのレンチウイルス媒介遺伝子発現系の使用が、損傷部位における細胞増殖およびNSPCの活性化を促進するGsx1発現をもたらすことが本明細書に示される。さらに、損傷部位またはその近傍におけるレンチウイルス媒介Gsx1発現(Gsx1処置)は、グルタミン酸作動性およびコリン作動性ニューロンの数を増加させ、GABA作動性介在ニューロンの数を減少させる。Gsx1処置は、損傷したマウスにおいてグリア性瘢痕形成を減弱し、歩行運動機能を劇的に改善した。全ゲノムトランスクリプトーム分析は、Gsx1処置がNSPC活性化、すなわちニューロン分化と相関するNotchシグナル伝達経路を誘導することを明らかにし、Gsx1媒介機能回復に関する分子的洞察を提供する。

限定された神経発生、増加した反応性アストログリオーシス、および瘢痕形成は、SCI後の神経再生および機能回復の主な障壁である。損傷部位またはその近傍におけるGsx1処置がNSPCの活性化および介在ニューロンの特異的亜型(例えばグルタミン酸作動性およびコリン作動性ニューロン)の生成を促進することが本明細書において示される。Gsx1発現はまた、反応性アストログリオーシスおよびグリア性瘢痕形成を阻害し、外側片側切断SCIを有するマウスにおいて劇的な歩行運動機能回復をもたらす。RNA-SeqおよびRT-qPCR分析は、Gsx1処置が細胞増殖、NSPC活性化、神経発生、アストログリオーシス、および瘢痕形成と関連する遺伝子の発現量を変更し、これがSCI後の機能回復と相関することを実証する。

材料および方法
本実施例は、実施例2~7に示す結果を取得するために使用した材料および方法を提供する。
Gsx1とレポーターRFPとをコードするレンチウイルス(レンチ-Gsx1-RFP)および対照レンチウイルス(レポーターRFPのみをコードする、レンチ-Ctrl-RFP)(ABM(登録商標)LV465366およびLV084)を、HEK293T細胞に標的ベクター(レンチ-Gsx1-RFPまたはレンチ-Ctrl-RFP)、エンベローププラスミド(pMD2.G/VSVG、Addgene 12259)、および第3世代パッケージングプラスミド(pMDLg/pRRE、Addgene 12251、およびpRSV-Rev、Addgene 12253)の混合物をトランスフェクトすることによって生成した。HEK293T細胞を、10%ウシ胎児血清(FBS)、1%非必須アミノ酸(MEM NEAA100x Life Technology 11140050)、および1%Glutamax I100X(Life Technology 35050061)を補充した高グルコースダルベッコ変法イーグル培地(DMEM)中で培養した。HEK293T(ヒト胎児腎)細胞のトランスフェクションを、培養が約50~60%の集密度に達する時に実施した。ウイルス含有上清をトランスフェクション後2日目および4日目に収集した。ウイルスを、ウイルス上清をポリエチレングリコール6000(PEG6000)法により沈殿させることによって濃縮した(Kutner et al., Nat Protoc 4, 495-505 (2009))。ウイルス力価を、HEK293T細胞を感染させることによって決定した(Kutner et al., Nat Protoc 4, 495-505 (2009))。
若い成体(8~12週)マウスを動物実験委員会(IACUC)の承認の下で使用した。全ての動物を動物飼養施設に12時間明/暗周期で収容した。各実験条件下のマウスを、可能な場合は等しい数の雄および雌マウスを用いて、無作為に割り当てた。
片側切断SCIおよびレンチウイルス注射のために、マウスを初めに3~4分間の5%イソフルラン吸入により麻酔し、次いで残りの外科的処置の間2.5%イソフルランに維持した。次に、皮膚を、ベタジンスクラブおよび70%エタノールシートを用いて消毒した。椎弓切除術をT9~T10付近において実施して、脊髄を露出した。次に、局所麻酔(0.125%マーカイン)をかけ、背側血管を、焼灼器を使用して焼いた。次いで、外側切断を脊髄の左側に対して実施し、切断は、片側切断SCIのため、脊髄の正中線で終える。損傷直後、約1~2μLのウイルス(1×108TU/ml)を病変中心点に対して約1mm吻側および尾側に注射した。ウイルスを約1μL/分において注射し、針を2~3分間そのままにして、拡散を可能にし、漏出または逆流を予防した。シャム動物に関しては、皮膚および筋肉を切断して、脊髄を露出した。筋肉を縫合し、皮膚をステープルにより元通りに閉じた。外科的処置直後、1mg/kgのメロキシカム、鎮痛薬、および抗生物質として50mg/kgのセファゾリンを皮下投与した。
各動物の歩行運動を、オープンフィールド試験によるバッソマウススケール(BMS)スコアに基づいて評価した(Basso et al., J Neurotrauma 23:635-659 (2006))。BMSスケールは、0(完全に麻痺している)~9(正常)の歩行運動の範囲である。BMSスコア評価は、処置の種類について盲検化されている3名の独立した観察者によって、動物1匹当たり2~3分の観察の後に与えられた。BMS評価は、損傷前、その後は損傷の最大8週間後まで週に2回実施する。
損傷3、7、14、および56日後(DPI)の脊髄組織を、1xリン酸緩衝生理食塩水(PBS)とそれに続く4%(w/v)パラホルムアルデヒド(PFA)とを用いた心臓内灌流後に採取した。次いで、脊髄組織を顕微鏡手術によりで切り分け、ローター上で4%PFAにおいて一晩(18~24時間)固定した。固定した脊髄組織を、1xPBSを用いて30分間3回洗浄し、次いで組織が底に沈むまで30%(w/v)スクロース中に一晩置いた。次に、組織をTissue-Tek(登録商標)最適切断温度(O.C.T.)に包埋することによって凍結保存し、必要となるまで-80℃で保管した。脊髄組織の矢状切片または横断切片(12μm厚)を、クリオスタット(ThermoScientific)を使用して生成した。
免疫染色を、以前に確立されたプロトコールに若干の修正を加えたものに従って実施した(Li et al., Sci Rep 6, 38665 (2016))。簡潔に述べると、切片を、冷メタノールを用いて、室温で10分間、固定および抗原回収のために処理した。全ての抗体を、0.05%トリトンX-100、2%ロバ血清、3%ウシ血清アルブミン(BSA)、およびPBS(1X)を含有するpH7.4のブロッキング溶液中に希釈した。切片を、一次抗体(表1)と共に4℃で一晩インキュベートし、PBSを用いて10分間3回洗浄した。次いで、二次抗体(表1)を室温で1時間インキュベートし、PBSを用いて10分間3回洗浄した。核染色に関しては、4’,6-ジアミジノ-2-フェニルインドール(DAPI、200ng/ml)を添加し、次いで試料を、PBSを用いて3回洗浄し、Cytoseal20(ThermoFisher Scientific 8310-4)を用いて封止する。
各スライド/動物からの少なくとも5つの切片を分析した。画像を、Zeiss LSM800共焦点顕微鏡またはZeiss AxioVision Imager A.1を使用して、条件ごとに同じ露出および閾値、ならびに同じ強度において捕捉した。ImageJにおける自動セルカウンター(Rueden et al., BMC Bioinformatics 18:529 (2017)、Schneider et al., Nat Methods 9:671-675 (2012))を使用して、細胞の総数を計数した。細胞型特異的マーカーとRFPとの同時標識を手動で計数した。試料サイズを、以前の実験から実施された検出力分析に基づいて決定した。データは平均±平均の標準誤差(SEM)として表す。全てのデータを、Microsoft Windows(登録商標)用のGraphPad Prismバージョン5.0ソフトウェアを使用して分析した。2つの条件間の統計的有意性はスチューデントのt検定によって算出し、多群比較は一元配置分散分析とそれに続くテューキー事後検定とを使用して実施する。0.05未満のP値を統計的に有意と考える。
3、14、35DPIの脊髄組織(各時点に関してn≧3、(図3A))を切り出し、損傷/注射セグメント(病変の各側から約2~3mmに及ぶ実質セグメント)を液体窒素中に迅速に急速凍結した。総RNAを、RNeasy脂質組織ミニキット(Qiagen、#74804)を製造業者のプロトコールに従って使用して、脊髄組織から単離した。総RNAの濃度を、Qubit RNA BRアッセイキット(Life Technologies)を使用して決定し、総RNAの品質を、2100バイオアナライザー自動電気泳動システム(Agilent Technologies)においてRNA6000ナノチップを使用して決定した。
ライブラリー調製およびRNA配列決定は、Admera Health(South Plainfield、NJ)によって実施された。総RNAを各試料のライブラリー調製のために使用し、その後、製造業者の使用説明書(Illumina)に従ってバーコードを付して調製した。ライブラリーを、Illumina MiSeqペアエンドキットを使用して調製し、Illumina MiSeqにおいて、ペアエンド、2×150bpとして配列決定した。配列決定ランを製造業者の使用説明書に従って実施し、試料1つ当たり総計4000万のリードを生成した。
FastQC(www.bioinformatics.babraham.ac.uk/projects/fastqc/)を使用した生fastqファイルの品質チェックの後、全ての配列を、STARバージョン2.0(Dobin et al., Bioinformatics 29:15-21 (2013))を用いて、マウス参照ゲノムmm10に対してアラインメントした。生リードカウントを、HTSeq(バージョン0.6.0)(Anders et al., Bioinformatics 31:166-169 (2015))を使用して生成した。R/BioconductorパッケージのDESeq2(Love et al., Genome Biol 15:550 (2014)、Anders & Huber, Genome Biol 11:R106 (2010))を使用して、カウントを正規化し、HTSeqによって生成したカウント行列における変動遺伝子発現量を求めた。Gsx1処置群と対照群との間で発現変動した転写物/遺伝子を、統計的有意性(p値)および生物学的関連性(倍率変化)によって定義した。下流経路分析を、QIAGENの独創性経路分析(IPA、QIAGEN Redwood City、www.qiagenbioinformatics.com/products/ingenuity-pathway-analysis)を使用して実行した。
相補的DNA(cDNA)を、製造業者のプロトコールを使用してSuperScript III第1鎖合成システム(Invitrogen、18080051)を使用し、総RNAから合成した。qPCRを、StepOnePlusリアルタイムPCRシステム(Applied Biosystem)を使用して、パワーSYBR(商標)グリーンPCRマスターミックスおよび遺伝子特異的プライマー(表2)を用いて実施した。GAPDHを参照ハウスキーピング遺伝子として使用する。リバック(Levak)法を使用して、倍率変化を、Shamに対して正規化することによって算出する。
方法の概要
SCI後の成体の脊髄におけるGsx1の異所性発現がNSPC活性化および神経発生を促進するかどうかを決定するために、胸椎(T)10レベルにおいて、脊髄の正中線から左側に対する外側片側切断を実施した。外側片側切断SCIの完全性および無矛盾性は、左後肢における麻痺の観察によって確認した。SCI直後、Gsx1とレポーター赤色蛍光タンパク質(RFP)とをコードするレンチウイルス(レンチ-Gsx1-RFP)1μL/部位(1×108TU/ml)を損傷した脊髄に、損傷部位に対しておよそ1mm吻側および尾側に注射した(図1A)。RFPレポーターのみをコードするレンチウイルス(レンチ-Ctrl-RFP)を対照として使用する。動物を以下の3つの群(3~6匹/群)に無作為に割り当てた:1)シャムマウス(損傷を有しない脊椎を露出、Sham)、2)レンチ-Ctrl-RFPの注射を伴うSCIマウス(SCI+Ctrl)、および3)レンチ-Gsx1-RFP注射を伴うSCIマウス(SCI+Gsx1)。免疫組織化学によって損傷3日後(DPI)および7DPIでの、ならびにRT-qPCRによって3DPIでの脊髄組織におけるレンチウイルス媒介Gsx1発現が初めに確認された(図2A~2E)。対照と比較して、Gsx1処置は、Gsx1発現を有するウイルス形質導入RFP+細胞のパーセンテージを大幅に増加した。
Gsx1処置は損傷した脊髄において細胞増殖を高める
SCIは病変部位において細胞増殖を高める。損傷3日後(DPI)において細胞増殖に対するGsx1処置の効果を決定するために、細胞増殖マーカーKi67の発現を、免疫組織化学とそれに続く共焦点イメージング分析とによって検査した(図1B)。RFP+およびKi67+細胞は注射部位付近に位置付けられることが見出された。以下の3つの対照および実験群:Sham(n=3)、SCI+Ctrl(n=6)、およびSCI+Gsx1(n=6)における損傷/注射区域付近の、DAPI+細胞のうちのKi67+細胞のパーセンテージ(図1C)。Ki67+細胞のパーセンテージの大幅な増加が、Shamマウスと比較して、ウイルス注射を受けた両損傷群において観察され、SCI+Gsx1群において最も大きい増加であった(図1C)。加えて、RFP+細胞のうちのKi67+/RFP+同時標識細胞の大幅に高いパーセンテージが、レンチ-Ctrl-RFP注射を受けたマウスと比較して、レンチ-Gsx1-RFP注射を伴うマウスにおいて見出された(図1D)。さらに、Ki67 mRNA発現量の増加がRT-qPCR分析によって確かめられた。Gsx1処置は、SCI+Gsx1群において、SCI+Ctrl群と比較して大幅に高いレベルのKi67 mRNAレベルを誘導する(約4倍;図1E)。これらの結果は、Gsx1処置が成体の損傷した脊髄において細胞増殖を促進することを示す。
Gsx1処置はSCI後のNSPC活性化を促進する
成体の脊髄において、NSPCは、正常な状態の下では静止状態にて存在するが、損傷後は活性化する。NSPCの活性化に対するGsx1処置の効果を調査するために、免疫組織化学および共焦点イメージング分析を介した、3DPIでの損傷した脊髄におけるNSPCマーカー(NestinおよびNotch1)の発現を検査した。
損傷した脊髄における神経発生の誘導
成体の脊髄において、損傷活性化NSPCは大半が星細胞および乏突起膠細胞を生成する。Gsx1処置がNSPC系統発生における細胞運命決定を変更するか否かを決定するために、14DPIにおける脊髄組織の矢状切片(図6A~6D)および横断切片(図7A~7D)を、SCI+Ctrl(n=6)およびSCI+Gsx1(n=6)群において初期ニューロンのマーカーダブルコルチン(DCX)(図6A)、星細胞マーカーGFAP(図6B)、および乏突起膠細胞前駆体マーカーPDGFRa(図6C)を用いて検査した。DCXは、大半が神経芽細胞および未成熟ニューロンにおいて発現し、成体の神経発生と関連するが、反応性グリオーシスとは関連しない。SCI+Ctrl群と比較して、Gsx1処置は、RFP+細胞のうちの、DCX+/RFP+同時標識細胞のパーセンテージを大幅に増加させ、GFAP+/RFP+およびPDGFRa+/RFP+同時標識細胞のパーセンテージを減少させた(図6D)。DCX+、GFAP+、およびPDGFR+細胞の数における顕著な差は、SCI+CtrlおよびSCI+Gsx1群内の14DPIにおける脊髄の背側領域と腹側領域との間において見出されなかった(図7A~7D)。
Gsx1処置は介在ニューロンの特異的亜型の数を増加させる
Gsx1処置によって誘導された成熟ニューロンの特異的亜型を同定した。56DPIにおける脊髄組織の矢状切片を、成熟ニューロンのマーカーNeuN(図9A)、コリン作動性ニューロンのマーカーChAT(図9B)、グルタミン酸作動性介在ニューロンのマーカーvGlut2(図9C)、およびGABA作動性介在ニューロンのマーカーGABA(図9D)を用いて染色した。SCI+Gsx1群(n=6)におけるRFP+細胞のうちの、NeuN+、ChAT+、およびvGlut2+細胞のパーセンテージの大幅な増加、ならびにGABA+細胞の減少がSCI+Ctrl群(n=6)と比較して観察された(図9E)。さらに、誘導された細胞が成熟に達する35DPIにおけるSham(n=4)、SCI+Ctrl(n=4)、およびSCI+Gsx1(n=4)群からの脊髄組織のRT-qPCR分析は、NeuN(またはHrnbp3)のわずかに増加したmRNA発現量を伴って、vGlut(またはSlc17a6)およびChatの大幅に増加した発現量を示した(図9F)。
Gsx1処置はグリア性瘢痕形成を低減する
SCIは、反応性アストログリオーシスおよびグリア性瘢痕形成の原因となるミクログリアおよび星細胞の活性化を引き起こす。グリア性瘢痕は、大半の場合、反応性星細胞(RA)、非ニューロン細胞(例えば周皮細胞および髄膜細胞)、およびプロテオグリカン豊富細胞外基質(ECM)から構成される。活性化星細胞は、グリア性瘢痕の主な成分を構成するコンドロイチン硫酸プロテオグリカン(CSPG)を分泌する。
Gsx1処置はSCI後の歩行運動機能を改善する
T10レベルにおける外側片側切断SCIを有する全てのマウスは、損傷後、左後肢に麻痺を呈した(図11A)。機能回復に対するGsx1処置の効果を実証するために、歩行運動行動を、確立されたオープンフィールド歩行運動試験およびBMSスコアスケール(Basso et al., J Neurotrauma 23:635-659 (2006))を使用して、損傷前日(-1DPI)から56DPI(SCIの8週間後)まで評価した。各マウスに関して、BMSスコアは3名の観察者によって二重盲検で割り当てられた。BMSスコアは0(完全麻痺および足首の運動がない)~9(正常な歩行)の範囲である。Sham動物は、正常な歩行運動行動を表し、BMSスコアは-1~56DPIまで約9のままであった(図11B)。損傷群(SCI+CtrlおよびSCI+Gsx1)におけるマウスは、片側切断損傷の当日(0DPI)に0のBMSスコアで、左後肢に麻痺を呈し(図11A~11B、およびstemcell.rutgers.edu/Treatment.mov、stemcell.rutgers.edu/Control.mov、stemcell.rutgers.edu/Sham.movにおいて見られる動画)、外側片側切断SCIを誘導することの成功を確認した。SCI+Ctrl群におけるマウス(n≧6)に関しては、BMSスコアは56DPIまでに約3(背側での歩み)まで漸進的に改善した(図11B)。対照的に、SCI+Gsx1群におけるマウス(n≧6)は大幅に改善した歩行運動機能を有し、BMSスコアは30DPIまでに約0から約5まで漸進的に増加した(図11B)。約30DPIから、Gsx1処置動物は、Shamマウスと比較して正常に近い歩行運動(BMSスコアは約6~7に達する)を示した(図11B)。これらの結果は、Gsx1処置がSCI後の歩行運動機能回復を劇的に改善したことを実証する(図11A~11B)。
Claims (22)
- 哺乳動物対象における脊髄損傷を処置する方法における使用のための組成物であって、前記組成物は、配列番号2または4のアミノ酸配列と少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むGsx1タンパク質、または配列番号1または3のヌクレオチド配列と少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含む、Gsx1をコードする核酸分子を含み、前記方法は、前記対象に前記組成物を投与することにより前記脊髄損傷を処置することを含む、組成物。
- 前記Gsx1タンパク質がGsx1-細胞透過性ペプチド(CPP)融合タンパク質を含む、請求項1に記載の組成物。
- 前記Gsx1タンパク質または前記Gsx1-CPP融合タンパク質のGsx1部分が、配列番号2または4を含むか、またはGsx1または前記融合タンパク質の前記Gsx1部分をコードする前記核酸分子が、配列番号2または4を含むタンパク質をコードする、請求項1または2に記載の組成物。
- Gsx1または前記Gsx1-CPP融合タンパク質の前記Gsx1部分をコードする前記核酸分子が、配列番号1または3を含む、請求項1から3のいずれか一項に記載の組成物。
- Gsx1または前記Gsx1-CPP融合タンパク質をコードする前記核酸分子がプラスミドまたはウイルスベクターを含む、請求項1から4のいずれか一項に記載の組成物。
- 前記ウイルスベクターがレンチウイルスベクターまたはアデノ随伴ウイルスベクターである、請求項5に記載の組成物。
- Gsx1または前記Gsx1-CPP融合タンパク質の前記Gsx1部分をコードする前記核酸分子がプロモーターに作動可能に連結する、および/または前記プロモーターが神経特異的エンハンサーに作動可能に連結する、請求項1から6のいずれか一項に記載の組成物。
- 前記プロモーターが構成的プロモーターまたは中枢神経系(CNS)特異的プロモーターである、請求項7に記載の組成物。
- 前記構成的プロモーターがCMVプロモーターである、請求項8に記載の組成物。
- 前記CNS特異的プロモーターが、シナプシン1(Syn1)プロモーター、グリア線維性酸性タンパク質(GFAP)プロモーター、ネスチン(NES)プロモーター、ミエリン関連乏突起膠細胞塩基性タンパク質(MOBP)プロモーター、ミエリン塩基性タンパク質(MBP)プロモーター、チロシンヒドロキシラーゼ(TH)プロモーター、またはフォークヘッドボックスA2(FOXA2)プロモーターである、請求項8に記載の組成物。
- 前記細胞透過性ペプチドが、配列番号61~79のいずれか1つと少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含む、請求項2から10のいずれか一項に記載の組成物。
- 前記脊髄損傷が、車両衝突、転落、暴力行為、スポーツ、または他の物理的外傷によって引き起こされる、請求項1から11のいずれか一項に記載の組成物。
- 前記投与することが注射を含む、請求項1から12のいずれか一項に記載の組成物。
- 前記注射がCNSへの注射を含む、請求項13に記載の組成物。
- 前記哺乳動物対象がヒト対象である、請求項1から14のいずれか一項に記載の組成物。
- 前記Gsx1タンパク質、Gsx1-CPP融合タンパク質、またはGsx1もしくはGsx1-CPP融合タンパク質をコードする核酸分子が医薬組成物中に存在する、請求項1から13のいずれか一項に記載の組成物。
- 前記投与することが、前記Gsx1タンパク質、Gsx1-CPP融合タンパク質、またはGsx1もしくはGsx1-CPP融合タンパク質をコードする核酸分子の少なくとも2回の別個の投与を含む、請求項1から16のいずれか一項に記載の組成物。
- 前記少なくとも2回の別個の投与が、少なくとも1週間、少なくとも2週間、少なくとも3週間、少なくとも4週間、少なくとも1か月、少なくとも2か月、少なくとも3か月、少なくとも6か月、少なくとも9か月、または少なくとも1年空けられる、請求項17に記載の組成物。
- 前記投与することが、前記脊髄損傷の発症の1時間以内、2時間以内、3時間以内、4時間以内、5時間以内、6時間以内、12時間以内、24時間以内、48時間以内、72時間以内、96時間以内、1週間以内、2週間以内、3週間以内、4週間以内、1か月以内、2か月以内、または3か月以内に行われる、請求項1から18のいずれか一項に記載の組成物。
- 前記方法が、前記対象に神経障害治療剤を含む第2の治療剤を投与することをさらに含む、請求項1から19のいずれか一項に記載の組成物。

(1)配列番号2または4と少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含む単離されたGsx1タンパク質、
配列番号2または4と少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むGsx1部分と、配列番号61~79のいずれか1つと少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を必要に応じて含む細胞透過性ペプチド(CPP)部分とを含む単離されたGsx1-CPP融合タンパク質であって、Gsx1部分と前記CPP部分とがリンカーによって必要に応じて接合する、Gsx1-CPP融合タンパク質、
配列番号1または3と少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むGsx1タンパク質をコードするか、または配列番号2または4と少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むGsx1タンパク質をコードする核酸分子、あるいは
Gsx1-CPP融合タンパク質をコードする核酸分子であって、前記Gsx1-CPP融合タンパク質のGsx1部分が、配列番号1または3と少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含む核酸分子によってコードされるか、または配列番号2または4と少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むGsx1タンパク質をコードし、必要に応じて前記Gsx1-CPP融合タンパク質のCPP部分をコードする核酸分子が、配列番号61~79のいずれか1つと少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%の配列同一性を含むタンパク質をコードする、Gsx1-CPP融合タンパク質をコードする核酸分子と、
(2)リポソームと
を含み、前記Gsx1タンパク質、Gsx1-CPP融合タンパク質、Gsx1タンパク質をコードする核酸分子、またはGsx1-CPP融合タンパク質をコードする核酸分子が前記リポソームに封入される、組成物。
