Sambrookら,Molecular cloning:A laboratory manual;Cold Spring Harbor Laboratory Press,Cold Spring Harbor,New York,1989に記載されているように、標準的な方法を使用してDNAを操作した。分子生物学的試薬を製造者の指示書に従って使用した。ヒト免疫グロブリン軽鎖および重鎖のヌクレオチド配列に関する一般的な情報は、Kabat,E.A.ら,(1991)Sequences of Proteins of Immunological Interest,第5版,NIH Publication No91-3242に示されている。
DNA配列は、二本鎖シークエンシングにより決定された。
所望の遺伝子セグメントは、適切なテンプレートを用いるPCRによって生成されたか、または遺伝子自動合成による合成オリゴヌクレオチドおよびPCR産物からGeneart社(ドイツ、レーゲンスブルク)によって合成された。単一の制限エンドヌクレアーゼ切断部位に隣接する遺伝子セグメントを、標準的なクローニング/シークエンシングベクターにクローニングした。形質転換した細菌からプラスミドDNAを精製し、UV分光法により濃度を決定した。サブクローニングされた遺伝子断片のDNA配列を、DNAシークエンシングによって確認した。遺伝子セグメントは、それぞれの発現ベクターへのサブクローニングを可能にする適切な制限部位を用いて設計された。全てのコンストラクトは、真核細胞における分泌のためのタンパク質を標的とするリーダーペプチドをコードする、5’末端DNA配列を含むように設計された。
標準的なプロトコールを参照して、ろ過された細胞培養上清からタンパク質を精製した。簡潔には、抗体を、プロテインA-セファロースカラム(GEヘルスケア)に適用し、PBSで洗浄した。抗体の溶出はpH2.8で達成され、その直後に試料を中和した。凝集したタンパク質を、PBS中または20mMヒスチジン、150mM NaCl(pH6.0)中でのサイズ排除クロマトグラフィー(Superdex200,GEヘルスケア)により、単量体抗体から分離させた。単量体抗体画分をプールし、(必要な場合)例えばMILLIPORE Amicon Ultra(30MWCO)遠心濃縮機を使用して濃縮し、-20℃または-80℃で凍結保存した。これらの試料の一部が、例えばSDS-PAGEおよびサイズ排除クロマトグラフィー(SEC)によるその後のタンパク質解析および分析的特徴付けのために提供された。
NuPAGE(登録商標)Pre-Castゲルシステム(Invitrogen)を製造者の指示書に従って使用した。特に、10%もしくは4-12%のNuPAGE(登録商標)Novex(登録商標)Bis-TRIS Pre-Castゲル(pH6.4)およびNuPAGE(登録商標)MES(還元ゲル、NuPAGE(登録商標)抗酸化剤ランニングバッファー添加剤を含む)またはMOPS(非還元ゲル)ランニングバッファーを使用した。
抗体の凝集およびオリゴマー状態の決定のためのサイズ排除クロマトグラフィー(SEC)を、HPLCクロマトグラフィーによって実施した。簡潔には、プロテインA精製抗体を、Agilent HPLC 1100システムでの300mM NaCl、50mM KH2PO4/K2HPO4(pH7.5)のTosoh TSKgel G3000SWカラムに、またはDionex HPLC-システムでの2xPBSのSuperdex200カラム(GEヘルスケア)に適用した。UV吸光度とピーク面積の積分により、溶出したタンパク質を定量化した。バイオ・ラッドのゲルろ過スタンダード151-1901が標準としての役割を果たした。
それぞれの抗体は、ポリエチレンイミンを使用して、HEK293-EBNA細胞を哺乳動物発現ベクターでコトランスフェクトすることによって産生され
た。細胞に、重鎖と軽鎖に1:1の割合で対応する発現ベクターをトランスフェクトした。
レンチウイルスベクターを作製するために、構成的に活性なヒトサイトメガロウイルス最初期プロモーター(CMV)下で、CARの正確なアセンブリのためのそれぞれのDNA配列を、インフレームでレンチウイルスポリヌクレオチドベクターにクローニングした。レトロウイルスベクターは、ウッドチャック肝炎ウイルス転写後調節エレメント(WPRE)、中心ポリプリン路(cPPT)エレメント、pUC複製起点、および細菌における増殖と選択を促進する抗生物質耐性をコードする遺伝子を含んでいた。
抗CD20抗体GA101をジゴキシゲニル化し、ジゴキシゲニン(DIG)分子組み込みをウエスタンブロット解析で検証した。抗体とジゴキシゲニンのカップリング反応のために、20mM His 140mM NaCl(pH6)に溶解した抗体をまず脱塩し、ZebaTMスピン脱塩カラム(ThermoFisher カタログ#89889)を用いてバッファーを0.1M重炭酸ナトリウム(pH8)バッファーに交換した。抗体とジゴキシゲニン-3-O-メチルカルボニル-e-アミノカプロン酸-N-ヒドロキシスクシンイミドエステル(Sigma Aldrich カタログ#11333054001)の等モル以上(1:3の割合)の量を、シェーカーで室温で1時間、300rpmでインキュベートした。抗体-ジゴキシゲニンコンジュゲートを再び脱塩し、バッファーを20mM His 140mM NaCl(pH6)に交換した。非コンジュゲートDig‐NHSを同じステップで除去した(カットオフ7kDa)。
Jurkat NFATレポーターCAR‐T細胞における抗ジゴキシゲニン‐ds‐scFv‐CD28ATD‐CD28DCD3zSSDの発現とジゴキシゲニン‐Cy5のCARへの結合をFACSにより確認した。抗ジゴキシゲニン-ds-scFv-CD28ATD-CD28DCD3zSSD導入Jurkat NFATレポーターCAR-T細胞を300gで3分間室温でペレット化し、新鮮なRPMI-1640+10%FCS+1%Glutamax(増殖培地)に適量再懸濁させた。その後、3x105個の細胞を96ウェルプレートの各ウェルに添加し、一度300gで5分間スピンダウンし、2%FCSを含むPBS中の100μlに再懸濁させた。Dig‐Cy5を最終濃度20nMになるまで加え、氷上で45分間インキュベートした。その後、細胞をペレット化し、氷冷PBSに再懸濁させた。洗浄ステップをさらに2回繰り返した。その後、フローサイトメトリーにより細胞のCy5シグナル(APCチャネル)を解析した(図8)。陰性対照として、非形質導入Jurkat NFAT細胞を同様に処理し、解析した。
ここに記載されているのは、CD20を発現するSUDHDL4腫瘍細胞を標的細胞として、および抗ジゴキシゲニン-ds-scFv-CD28ATD-CD28CSDCD3zSSDを発現するJurkat NFATレポーターCAR-T細胞のソート済みのプールをレポーター細胞として用いる、レポーターCAR-T細胞アッセイである(図9)。ジゴキシゲニル化(Digoxygeninylated)GA101IgG(抗体:Dig-NHS比1:10)をIgGとして使用したが、これは、一方では腫瘍抗原を認識し、他方では形質導入されたJurkat NFATレポーターCAR-T細胞によって認識される。陽性対照として、96ウェルプレート(Cellstar Greiner-bio-one、カタログ#655185)に、CD3抗体(Biolegend(登録商標)から)を10μg/ml含むリン酸緩衝生理食塩水(PBS)を一晩4℃で、または37℃で少なくとも1時間コーティングした。CD3でコーティングされたウェルをPBSで2回洗浄し、最後の洗浄ステップの後、PBSを完全に除去した。レポーター細胞またはJurkat NFAT野生型細胞を計数し、Cedex HiResを用いてその生存率をチェックした。細胞数は、1x106生細胞/mlに調整した。したがって、細胞懸濁液の適切なアリコートを210gで5分間、室温(RT)でペレット化し、新鮮なRPMI-1640+10%FCS+1%Glutamax(増殖培地)に再懸濁させた。目的の抗原を発現する標的細胞を計数し、それらの生存率についてもチェックした。細胞数は、増殖培地中、1x106生細胞/mlに調整した。標的細胞とレポーター(エフェクター)細胞を、96ウェル懸濁培養プレート(Greiner-bio one)に、E(エフェクター細胞):T(標的細胞)比5:1で3連で蒔いた(ウェル当たり合計110.000細胞)。次のステップとして、目的の抗原を標的とするジゴキシゲニニル化GA101抗体の段階希釈液を、2mlのディープウェルプレート(Axygen(登録商標))を用いて増殖培地で調製した。ウェル当たり200μlの最終体積で1μg/ml‐0.01pg/mlの範囲の最終濃度を得るために、50μlアリコートの異なる希釈物をそれぞれのウェルにピペットで移した。96ウェルプレートを190gおよび室温で2分間遠心分離した。Parafilm(登録商標)で密封し、湿度雰囲気中、37℃および5%CO2でプレートをインキュベートした。20時間のインキュベーション後、マルチチャンネルピペットを用いて10回上下にピペッティングすることより、各ウェルの内容物を混合した。100μl細胞懸濁液を新しい白色フラットクリアボトム96ウェルプレート(Greiner‐bio‐one)に移し、100μlのONE‐GloTMルシフェラーゼアッセイ(Promega)を加えた。ロータリーシェーカーで300rpm、室温暗所で15分インキュベートした後、Tecan(登録商標)Spark10Mプレートリーダーを用いてウェル当たり1秒の検出時間で発光を測定した。標的細胞とレポーター細胞を5:1の比率で20時間共培養すると(灰色の丸)、ジゴキシゲニル化GA101 IgGを抗体として用いた場合、グラフは抗ジゴキシゲニン-ds-scFv-CD28ATD-CD28CSDCD3zSSDを発現するJurkat NFATレポーターCAR-T細胞の用量依存的な活性化を示す(図9)。ジゴキシゲニニル化を伴わないGA101 IgGを用いた場合(図9、灰色の四角形で描画)、形質導入されたJurkat NFATレポーターCAR-T細胞の活性化は検出できなかった。1μg/mlのジゴキシゲニニル化GA101とインキュベートしたが標的細胞なしでインキュベートしたさらなるJurkat NFAT野生型細胞は、いかなる活性化も示さなかった(図9黒い四角形)。対照的に、抗ジゴキシゲニン-ds-scFv-CD28ATD-CD28CSDCD3zSSDを発現するJurkat NFATレポーターCAR-T細胞を、標的細胞なしで、1μg/mlのジゴキシゲニニル化GA101 IgGとインキュベートしたところ、活性化を示した(図9黒い三角)。

ここに記載されているのは、接着性FAPを発現するNIH/3T3-huFAP cl 19腫瘍細胞を標的細胞として用いて実施した、Jurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図13A)または抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図13C)のソート済みのプールを使用した。P329G LALA変異を伴うFAP 4B9 IgGをIgGとして使用したが、これは、一方では腫瘍抗原を認識し、他方ではJurkat NFATレポーターCAR-T細胞によって認識される。アイソタイプ対照として、P329G LALA変異を伴うIgG DP47/vk3を含めた。陽性対照として、96ウェルプレート(Cellstar Greiner-bio-one、カタログ#655185)のウェルに、CD3抗体(Biolegend(登録商標)から)10μg/mlを含むリン酸緩衝生理食塩水(PBS)を37℃で少なくとも1時間コーティングした。CD3でコーティングされたウェルをPBSで2回洗浄し、最後の洗浄ステップの後、PBSを完全に除去した。接着性NIH/3T3‐huFAP cl 19標的細胞をPBSで1回洗浄し、トリプシンで剥離させた。剥離した細胞をDMEM+4.5g LD-グルコース+L-グルタミン+25mM HEPES+10%FCSおよび1%Glutamaxに再懸濁させた。レポーター細胞またはJurkat NFAT野生型細胞を計数し、Cedex HiResを用いてその生存率をチェックした。細胞数は、1x106生細胞/mlに調整した。したがって、細胞懸濁液の適切なアリコートを210gで5分間、室温(RT)でペレット化し、新鮮なRPMI-1640+10%FCS+1%Glutamax(増殖培地)に再懸濁させた。目的の抗原を発現する標的細胞を計数し、それらの生存率についてもチェックした。細胞数は、レポーター細胞について記載したのと同様に、増殖培地中、1x106生細胞/mlに調整した。標的細胞とレポーター細胞を、96ウェル懸濁培養プレート(Greiner-bio one)にE:T比5:1で3連で蒔いた(ウェル当たり合計110.000細胞)。次のステップとして、目的の抗原を標的とするP329G LALA変異を伴うGA101の段階希釈液を、2mlのディープウェルプレート(Axygen(登録商標))を用いて増殖培地で調製した。ウェル当たり200μlの最終体積で1μg/ml‐0.0001μg/mlの範囲の最終濃度を得るために、50μlアリコートの異なる希釈物をそれぞれのウェルにピペットで移した。96ウェルプレートを190gおよび室温で2分間遠心分離した。Parafilm(登録商標)で密封し、湿度雰囲気中、37℃および5%CO2でプレートをインキュベートした。20時間のインキュベーション後、マルチチャンネルピペットを用いて各ウェルの内容物を10回上下にピペッティングすることより混合した。100μl細胞懸濁液を新しい白色フラットクリアボトム96ウェルプレート(Greiner‐bio‐one)に移し、100μlのONE‐GloTMルシフェラーゼアッセイ(Promega)を加えた。ロータリーシェーカーで300rpm、室温暗所で15分インキュベートした後、Tecan(登録商標)Spark10Mプレートリーダーを用いてウェル当たり1秒の検出時間で発光を測定した。
ここに記載されているのは、接着性CEAを発現するMKN45腫瘍細胞を標的細胞として用いるJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図14A)または抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図14C)のソート済みのプールを使用した。P329G LALA変異を伴うCEA A5B7 IgGまたはP329G LALA変異を伴うCEA T84 LCHA IgGのいずれかが使用された。さらに、アイソタイプ対照として、P329G LALA変異を伴うIgG DP47/vk3を含めた。
ここに記載されているのは、接着性TNCを発現するCT26TNC cl 19腫瘍細胞を標的細胞として用いるJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図15C)または抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞(図15A))のソート済みのプールを使用した。IgGとして、P329G LALA変異を伴うTNCA2B10を使用した。さらに、アイソタイプ対照として、P329G LALA変異を伴うIgG DP47/vk3を含めた。

ここに記載されるのは、HLA-A2/WT1-ペプチドバインダー33F05(配列番号139および140)、11D06(配列番号141および142)、33H09(配列番号143および144)および5E11(配列番号145および146)の特異性を評価するために、ペプチドパルスT2細胞を標的細胞として使用したJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞の選別プールを使用した。IgGとして、P329G LALA変異を伴うHLA-A2/WT1-ペプチドバインダーを用いた。HLA-A2/WT1-ペプチド結合抗体およびレポーター細胞とのインキュベーションの前に、T2細胞を、それぞれ10-5Mのペプチドを37℃で2時間パルスするか、またはパルスせずに放置した。標的細胞とレポーター細胞を、5:1のE:T比(ウェル当たり2000標的細胞当たり10.000エフェクター細胞)で、384ウェル白色フラットクリアボトムプレート(Greiner-bio one)に蒔いた。次のステップとして、問題のIgGの段階希釈液を増殖培地で調製した。レポーター細胞、T2細胞およびIgGを37℃で16時間インキュベートした後、ウェル当たり6μlのONE-GloTMルシフェラーゼ基質(Promega)を添加し、TECAN社製インフィニットM1000Proプレートリーダーを使用して発光を直接測定した。
ここに記載されるのは、レポーター細胞として抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFAT CAR-T細胞のソート済みのプールを用いたJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞は、P329G LALA変異を伴うIgGフォーマットのHLA-A2/WT1-ペプチドバインダーに結合し、これにより、試験したHLA-A2/WT1ペプチド(それぞれRMFまたはVLD)を異なる程度に認識する。
ここに記載されるのは、RMF-ペプチドまたはVLD-ペプチドでパルスされたT2細胞を用いたフローサイトメトリーによるHLA-A2/WT1-ペプチドバインダー5E11(配列番号102および103)および33H09(配列番号100および101)の特異性の評価である。HLA-A2/WT1-ペプチド結合抗体とのインキュベーションの前に、T2細胞を、それぞれの10-5Mのペプチドを37℃で2時間パルスするか、またはパルスせずに放置した。問題の抗体の異なる濃度の100000細胞の細胞アリコートへのそれぞれのIgGの結合を氷上で1時間行い、続いてPBSで2回洗浄し、Fortesssaアナライザー(BD Biosciences)でのフローサイトメトリーで90nMの濃度で抗huFc検出(Jackonson ImmunoResicationからの抗ヒトF(ab)2_AF647)によって評価した。バインダー5E11および33H09の両方とも、RMF-ペプチドパルスT2細胞上には明確な濃度依存性結合シグナルを与えるが、VLD-ペプチドパルスT2細胞上には与えない(図20Aおよび図20B)。このフローサイトメトリーに基づく評価によれば、両抗体候補は、RMF-ペプチドパルスT2細胞には特異的に結合するが、VLD-ペプチドパルスT2細胞には結合しないようである。
ここに記載されるのは、患者由来の腫瘍の一次試料(結腸癌の肺転移)を使用するJurkat NFATレポーターCAR-T細胞アッセイである。腫瘍試料を、CEAおよびFAP抗原レベルの発現を示す蛍光活性化細胞ソーティング(FACS)によって特徴付けた。FACS分析は、CD45陽性細胞の約80%がCEAを発現し、約16%が表面にFAPを発現することを示した(図22A)。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞のソート済みのプール(図22B)または抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSDを発現するJurkat NFATレポーターCAR-T細胞のソート済みのプール(図22C)を使用した。P329G LALA変異を伴う、ヒトIgG1抗CEA T84 LCHAまたは抗FAP(4B9)抗体をIgGとして使用した。腫瘍細胞とレポーター細胞を、5:1のE:T比(ウェル当たり200.000標的細胞当たり20.000エフェクター細胞)で、96ウェル白色フラットクリアボトムプレートに2連で蒔いた。次のステップとして、問題のIgGの段階希釈液1μg/mlを増殖培地で調製した。レポーター細胞、標的細胞およびIgGを37℃で8時間インキュベートした後、ウェル当たり50μlのONE-GloTMルシフェラーゼ基質(Promega)を添加し、TECAN社製インフィニットM1000Proプレートリーダーを使用して発光を直接測定した。
Claims (9)
- 試料中の腫瘍細胞の存在を判定するためのアッセイであって、
a)試料を、抗原結合ドメインと認識ドメインとを含む抗原結合分子と接触させるステップであって、抗原結合ドメインが、腫瘍細胞に特異的に結合可能な標的抗原結合部分を含む、接触させるステップ;
b)試料を、キメラ抗原受容体(CAR)発現レポーターT(CAR-T)細胞と接触させるステップであって、レポーターCAR-T細胞が、
i.応答エレメントに作動可能に結合されており、認識ドメインに特異的に結合可能なCAR;
ii.応答エレメントの制御下にあるレポーター遺伝子;
を含む、接触させるステップ:および
c)レポーター遺伝子の発現を測定して腫瘍細胞の存在を確認することにより、T細胞の活性化を判定するステップ
を含み、
認識ドメインが変異Fcドメインであり、変異Fcドメインが、非変異親Fcドメインと比較して少なくとも1つのアミノ酸置換を含み、CARが、変異Fcドメインに特異的に結合することはできるが、非変異親Fcドメインには特異的に結合することができない、
アッセイ。 - 抗原結合ドメインがFab断片であり、認識ドメインがFcドメインである、請求項1に記載のアッセイ。
- 抗原結合分子が、IgGクラス抗体、特にIgG1またはIgG4アイソタイプ抗体である、請求項1または2に記載のアッセイ。
- 変異体Fcドメインは、ヒトIgG1 Fc(配列番号132)の残基117、118、136、180、193、212、214および318からなる群から選択される位置にアミノ酸置換を含み、特に、変異体ヒトIgG1 Fcは、ヒトIgG1 Fc(配列番号132)の残基117にロイシンからアラニン、残基118にロイシンからアラニン、残基136にイソロイシンからアラニン、残基180にアスパラギンからアラニン、残基193にヒスチジンからアラニン、残基212にプロリンからグリシン、残基214にプロリンからグリシン、および/または残基318にヒスチジンからアラニンへのアミノ酸置換を含む、請求項1から3のいずれか一項に記載のアッセイ。
- 変異体Fcドメインは、ヒトIgG1 Fc(配列番号132)の残基212の位置にアミノ酸置換を含み、特に、変異体Fcドメインは、ヒトIgG1 Fc(配列番号132)の残基212にプロリンからグリシンへのアミノ酸置換を含む、請求項4に記載のアッセイ。
- CARが、少なくとも1つの細胞内刺激シグナル伝達および/または補助刺激シグナル伝達ドメインを含む、請求項1から5のいずれか一項に記載のアッセイ。
- 細胞内シグナル伝達および/または補助シグナル伝達ドメインの活性化が応答エレメントの活性化をもたらす、請求項6に記載のアッセイ。
- 応答エレメントの活性化がレポーター遺伝子の発現をもたらす、請求項1から7のいずれか一項に記載のアッセイ。
- 試料が、疾患に罹患している個体に由来する患者試料であり、特に疾患ががんである、請求項1から8のいずれか一項に記載のアッセイ。
