また、標識ステップS1において特定部位を標識する標識物質は、特定波長λ0において吸収ピークを有する光吸収性標識物質であるのが好適であり、この場合、屈折率分布取得ステップS2において、特定波長λ0に対して短波長側または長波長側にある第1波長λ1および第2波長λ2それぞれにおいて細胞の屈折率分布を取得するのが好適である。また、屈折率分布取得ステップS2において、第1波長λ1および第2波長λ2のうちいずれか一方を特定波長λ0に対して短波長側とし、他方を特定波長λ0に対して長波長側として、第1波長λ1および第2波長λ2のそれぞれにおいて細胞の屈折率分布を取得するのも好適である。
Claims (12)
- 第1波長および第2波長それぞれにおける屈折率が互いに異なる標識物質で細胞の特定部位を標識して、前記第1波長および前記第2波長の双方または何れか一方において標識部位と非標識部位との間で検出可能な屈折率差を生じさせる標識ステップと、
前記標識ステップで前記特定部位が標識された前記細胞の屈折率分布を前記第1波長および前記第2波長それぞれにおいて取得する屈折率分布取得ステップと、
前記第1波長および前記第2波長それぞれにおける屈折率分布を比較することで前記細胞における前記特定部位の分布を評価する解析ステップと、
を備える、細胞評価方法。 - 前記標識ステップにおいて、前記第1波長および前記第2波長のうちの長波長における屈折率が短波長における屈折率より高い標識物質で前記細胞の前記特定部位を標識する、請求項1に記載の細胞評価方法。
- 前記標識ステップにおいて、前記第1波長および前記第2波長のうちの長波長における屈折率が短波長における屈折率より低い標識物質で前記細胞の前記特定部位を標識する、請求項1に記載の細胞評価方法。
- 前記標識ステップにおいて、特定波長において吸収ピークを有する光吸収性標識物質で前記細胞の特定部位を標識し、
前記屈折率分布取得ステップにおいて、前記標識ステップで前記特定部位が標識された前記細胞の屈折率分布を、前記特定波長に対して短波長側または長波長側にある前記第1波長および前記第2波長それぞれにおいて取得する、請求項1~3の何れか1項に記載の細胞評価方法。 - 前記標識ステップにおいて、特定波長において吸収ピークを有する光吸収性標識物質で前記細胞の特定部位を標識し、
前記屈折率分布取得ステップにおいて、前記標識ステップで前記特定部位が標識された前記細胞の屈折率分布を、前記第1波長および前記第2波長のうちいずれか一方を前記特定波長に対して短波長側とし、他方を前記特定波長に対して長波長側として、前記第1波長および前記第2波長のそれぞれにおいて取得する、請求項1~3の何れか1項に記載の細胞評価方法。 - 前記標識ステップにおいて、色素タンパク質をコードするDNA配列が任意タンパク質をコードするDNA配列のN端末またはC端末に接続された細胞内発現用コンストラクトを前記細胞に導入して、この細胞内発現用コンストラクトを前記標識物質として用いて前記特定部位を標識する、請求項1~3の何れか1項に記載の細胞評価方法。
- 前記解析ステップにおいて、前記第1波長および前記第2波長それぞれにおける屈折率分布の比または差に基づいて前記細胞における前記特定部位の分布を評価する、請求項1~6の何れか1項に記載の細胞評価方法。
- 前記解析ステップにおいて、前記第1波長および前記第2波長それぞれにおける屈折率分布の比較の結果を前記細胞の強度画像または蛍光画像に重ねることで、前記細胞の前記特定部位に関する解析を行う、請求項1~7の何れか1項に記載の細胞評価方法。
- 前記標識ステップにおいて、第3波長および第4波長それぞれにおける屈折率が互いに異なる他の標識物質で前記細胞の他の特定部位を標識して、前記第3波長および前記第4波長の双方または何れか一方において標識部位と非標識部位との間で検出可能な屈折率差を生じさせ、
前記屈折率分布取得ステップにおいて、前記標識ステップで標識された前記細胞の屈折率分布を前記第3波長および前記第4波長それぞれにおいて取得し、
前記解析ステップにおいて、前記第3波長および前記第4波長それぞれにおける屈折率分布を比較することで前記細胞における前記他の特定部位の分布を評価する、請求項1~8の何れか1項に記載の細胞評価方法。 - 前記第1波長または前記第2波長は、前記第3波長または前記第4波長と等しい、請求項9に記載の細胞評価方法。
- 第1波長および第2波長それぞれにおける屈折率が互いに異なる標識物質で特定部位が標識されて前記第1波長および前記第2波長の双方または何れか一方において標識部位と非標識部位との間で検出可能な屈折率差が生じた細胞の屈折率分布を前記第1波長および前記第2波長それぞれにおいて取得する屈折率分布取得部と、
前記第1波長および前記第2波長それぞれにおける屈折率分布を比較することで前記細胞における前記特定部位の分布を評価する解析部と、
を備える、細胞評価装置。 - 第1波長および第2波長それぞれにおける屈折率が互いに異なる標識物質で特定部位が標識されて前記第1波長および前記第2波長の双方または何れか一方において標識部位と非標識部位との間で検出可能な屈折率差が生じた細胞の屈折率分布を前記第1波長および前記第2波長それぞれにおいて取得する屈折率分布取得ステップと、
前記第1波長および前記第2波長それぞれにおける屈折率分布を比較することで前記細胞における前記特定部位の分布を評価する解析ステップと、
をコンピュータに実行させるためのプログラム。
