Description
本発明は、yicL、ydiN、ydhK、aaeB、yeeA、rhtC、およびemrDからなる群から選択される1つ以上の遺伝子を過発現することで、L-アミノ酸生産能が向上した大腸菌(Escherichia coli)変異株を提供することを目的とする。
L-アミノ酸の生産において遺伝子yddGが及ぶ影響を確認するために、菌株の遺伝子yddGを欠失させた変異株を製造した。
W0GおよびW1G菌株のL-トリプトファン生産能、MWTR42およびMWTR5菌株のL-フェニルアラニン生産能を確認した。
遺伝子yicL、ydiN、ydhK、aaeB、yeeA、rhtC、emrDがそれぞれ挿入された大腸菌変異株を製造した。
W0GにベクターpTrc99Aのみが挿入されたW0G/pTrc99Aおよび変異株W0G/pTrc99A::yicL、W0G/pTrc99A::ydiN、W0G/pTrc99A::ydhK、W0G/pTrc99A::aaeB、W0G/pTrc99A::yeeA、W0G/pTrc99A::rhtC、W0G/pTrc99A::emrDのL-トリプトファン生産能を確認した。
遺伝子yicL、ydiN、ydhK、aaeB、yeeA、rhtC、emrDがそれぞれ挿入された大腸菌変異株を製造した。
MWTR42にベクターpTrc99Aのみが挿入されたMWTR42/pTrc99Aおよび変異株MWTR42/pTrc99A::yicL、MWTR42/pTrc99A::ydiN、MWTR42/pTrc99A::ydhK、MWTR42/pTrc99A::aaeB、MWTR42/pTrc99A::yeeA、MWTR42/pTrc99A::rhtC、MWTR42/pTrc99A::emrDのL-フェニルアラニン生産能を確認した。
遺伝子yicL、ydiN、ydhK、aaeB、yeeA、rhtC、emrDがそれぞれ挿入されたコリネバクテリウムグルタミカム変異株を製造した。
DW28GにベクターpEKOのみが挿入されたDW28G/pEKOおよび変異株DW28G/pEKO::yicL、DW28G/pEKO::ydiN、DW28G/pEKO::ydhK、DW28G/pEKO::aaeB、DW28G/pEKO::yeeA、DW28G/pEKO::rhtC、DW28G/pEKO::emrDのL-トリプトファン生産能を確認した。
Claims (3)
- yicL、ydiN、ydhK、aaeB、yeeA、およびrhtCからなる群から選択される1つ以上の遺伝子を過発現することでL-アミノ酸生産能が向上した変異株であって、前記変異株が、大腸菌(Escherichia coli)またはコリネバクテリウムグルタミカム(Corynebacterium glutamicum)であり、前記L-アミノ酸は、L-トリプトファンおよびL-フェニルアラニンからなる群から選択される、変異株。
- 前記yicLは配列番号1の塩基配列、ydiNは配列番号2の塩基配列、ydhKは配列番号3または配列番号37の塩基配列、aaeBは配列番号4の塩基配列、yeeAは配列番号5の塩基配列、rhtCは配列番号6の塩基配列からなるものである、請求項1に記載の変異株。
- (a)請求項1に記載の変異株を培地で培養するステップと、
(b)前記変異株または培地からL-アミノ酸を回収するステップと、を含む、L-アミノ酸の生産方法であって、前記L-アミノ酸は、L-トリプトファンおよびL-フェニルアラニンからなる群から選択される、方法。
