Description
本出願は、その内容が参照によりその全体が本明細書に組み込まれる米国仮特許出願第62/397,212号、2016年9月20日出願の優先権を主張する。
本技術は、カンナビス由来カンナビノイド生合成酵素遺伝子の毛状突起特異的プロモーター、毛状突起特異的プロモーターのヌクレオチド配列、および生物におけるカンナビノイド産生を調節するためのまたは生化学物質の他の毛状突起特異的産生を調節するためのプロモーターの使用に一般に関する。本技術は、毛状突起特異的プロモーターを含む、植物細胞および植物を含むトランスジェニック細胞および生物にも関する。

本技術の開示は、毛状突起細胞においてそれに作動可能に連結されているコード核酸配列の転写を制御できる、23個のプロモーターの同定に関する。
一部の実施形態では本技術の開示は、それに作動可能に連結されているコード核酸配列の毛状突起特異的発現を方向付けるために十分である、「カンナビノイドオン」または「CANON」断片と名付けられた核酸分子の同定にも関する。

本技術の核酸構築物は、植物細胞などの宿主細胞を形質転換するために利用され得る。一部の実施形態では、本技術の核酸構築物は、植物の少なくとも一部の細胞を形質転換するために使用される。これらの発現ベクターは、宿主植物細胞に一過性に導入されてもよく、または当業者に周知の種々の方法によってトランスジェニック植物を作出するように宿主植物細胞のゲノム中に安定的に組み込まれてもよい。

カンナビノイドは、カンナビス毛状突起において合成され、蓄積される。オリベトールシンターゼ(OLS)、オリベトール酸環化酵素(OAC)、芳香族プレニルトランスフェラーゼ(PT)、ヘキサノイル-CoA合成酵素(AAE1-1)、CBDAシンターゼ、CBCAシンターゼおよびTHCAシンターゼは、カンナビノイド生合成経路の酵素である。したがって、これらの酵素それぞれのためのプロモーターは、毛状突起細胞においてコード核酸の発現を方向付けることが期待される。本例は、天然ではカンナビノイドを産生しない植物および植物細胞においてカンナビノイド生合成酵素を発現させるための、本技術の毛状突起特異的プロモーターまたはそれらの生物学的に活性な断片の使用を実証する。
ベクター構築物 プロモーター配列(配列番号1~3、5、6、8、9、11~14、16~19、21~26、28、29または31~33)を、Ti プラスミドベクターなどのNicotiana tabacum細胞における発現のために適合されたベクターにおいてGUS-Aマーカーの前に位置付ける。構築物をAgrobacterium tumafaciensに組み込み、当技術分野において周知の方法によりN.tabacumを形質転換するために使用する。構築物を形質転換し、カナマイシン選択下で再生し、初代再生体(T0)を種子に生育させる。
本技術のプロモーターまたはそれらの生物学的に活性な断片を含む発現ベクターを用いて遺伝子操作された植物および植物細胞は、毛状突起特異的転写活性を示す。図2Bに示すとおり、CBDAS 20800:GUSを用いて形質転換したトランスジェニック植物の毛状突起は、明るい青色の毛状突起を示す一方で、植物の毛状突起以外の組織は着色されていない。カンナビノイド生合成経路における他のプロモーターについての毛状突起特異的発現は、定性的に同様である(図3)。図3の結果は、タバコ毛状突起での完全なカンナビノイド生合成経路からのプロモーターの毛状突起特異的発現を実証している。これにより、これらの結果は、本明細書に記される完全なカンナビノイド生合成経路由来のプロモーターが、根、葉、茎または植物の他の組織における発現と比較して、毛状突起組織において作動可能に連結されている遺伝子の発現を優先的に方向付けるために有用であることを実証している。この毛状突起特異的発現は、細胞傷害性カンナビノイドなどの毛状突起特異的生化学化合物の生合成のマニピュレーションのための重要なツールである。加えてこれらのプロモーターは、特定の生化学化合物の管理された産生のためのバイオ工場としてタバコまたはカンナビス毛状突起を使用することを目的とする戦略に重要である。
「カンナビノイドオン」(「CANON」)断片の核酸配列を配列番号31に記載する。CANON断片の核酸配列を推定TATAボックス、5’UTRおよび開始コドンと共に配列番号32に記載する。4×CANON断片合成プロモーターの核酸配列を配列番号33に記載する。コンセンサスCANON断片は、THCAシンターゼ19603、1330、50320、CBCAシンターゼ3498およびCBDAシンターゼ20800からの毛状突起特異的プロモーター由来である。
CANON断片をBLAT検索機能を使用してパープルクッシュゲノム配列を検索することによって同定した。THCAシンターゼ遺伝子を使用するパープルクッシュゲノム配列のBLAT検索は、次の結果をもたらした:

Claims (17)

ヌクレオチド配列が異種性核酸に作動可能に連結されている、合成DNA分子。 - ポリペプチドをコードする1つまたは複数の核酸配列に作動可能に連結されている、請求項1に記載のDNA分子を含む発現ベクター。
- 請求項2に記載の発現ベクターを含む遺伝子操作された宿主細胞。
- Cannabis sativa細胞である、請求項3に記載の遺伝子操作された宿主細胞。
- Nicotiana tabacum細胞である、請求項3に記載の遺伝子操作された宿主細胞。
- 請求項1に記載の合成DNA分子を含むキメラ核酸構築物を含む細胞を含む遺伝子操作された植物。

- キメラ遺伝子がゲノム中に組み込まれている遺伝子操作された植物または植物細胞であって、キメラ遺伝子が、相同性または異種性核酸配列に作動可能に連結されている毛状突起特異的プロモーターを含み、プロモーターが、
(a)配列番号1~3のいずれか1つのヌクレオチド配列;および
(b)配列番号1~3のいずれか1つのヌクレオチド配列と少なくとも90%同一であり、植物毛状突起腺特異的転写活性を有するプロモーターをコードするヌクレオチド配列
からなる群から選択される、遺伝子操作された植物または植物細胞。 - N.tabacum植物である、請求項10に記載の遺伝子操作された植物または植物細胞。

- 植物毛状突起においてポリペプチドを発現させるための方法であって、
(a)宿主細胞中に、
(i)配列番号1~3のいずれか1つに記載されているヌクレオチド配列;および
(ii)配列番号1~3のいずれか1つのヌクレオチド配列と少なくとも90%同一であり、植物毛状突起腺特異的転写活性を有するプロモーターをコードするヌクレオチド配列
からなる群から選択されるヌクレオチド配列を含む発現ベクターを導入するステップであって、(i)または(ii)の核酸配列が、ポリペプチドをコードする1つまたは複数の核酸配列に作動可能に連結されている、ステップ、ならびに
(b)ポリペプチドの発現を可能にする条件下で植物を生育させるステップ
を含む方法。 - 宿主植物毛状突起においてカンナビノイドを増加させる方法であって、
(a)宿主細胞中に、
(i)配列番号1~3のいずれか1つに記載のヌクレオチド配列;および
(ii)配列番号1~3のいずれか1つのヌクレオチド配列と少なくとも90%同一であり植物毛状突起腺特異的転写活性を有するプロモーターをコードするヌクレオチド配列;
からなる群から選択されるヌクレオチド配列を含む発現ベクターを導入するステップであって、(i)または(ii)の核酸配列が、カンナビノイド生合成経路の酵素をコードする1つまたは複数の核酸配列に作動可能に連結されている、ステップ、ならびに
(b)カンナビノイド生合成経路酵素の発現を可能にする条件下で植物を生育させるステップであって、カンナビノイド生合成経路酵素の発現により、植物は、同様の条件下で生育された対照植物と比較してカンナビノイド含有量が増加している、ステップ
を含む方法。 - カンナビノイド生合成経路酵素が、カンナビジオール酸(CBDA)シンターゼ、カンナビクロメン酸(CBCA)シンターゼまたはΔ9テトラヒドロカンナビノール酸(THCA)シンターゼである、請求項14に記載の方法。
- 植物にカンナビゲロール酸(CBGA)を提供することをさらに含む、請求項15に記載の方法。
