Description
ファージライブラリより、熱安定性VHH抗体として、配列番号:1および配列番号:12のVHH抗体を単離した。配列番号:1のTmは51.2℃、配列番号:12のTmは60.3℃を示した。より熱安定性の高いVHH抗体を得るため、両者の配列を比較し、配列番号:1のアミノ酸置換を検討した。
配列番号:1の36位のグリシンをアスパラギンに置換したもの(配列番号:3)、50位のアルギニンをロイシンに置換したもの(配列番号:4)、78位のグリシンをアラニンに置換したもの(配列番号:5)、116位のセリンをアルギニンに置換したもの(配列番号:6)をそれぞれ作成した。これらの抗体は、表2に記載のプライマーを用いてポリヌクレオチドを増幅し、これを導入したベクターにより大腸菌を形質転換し、産生させた。なお、表2における、例えばG36Dなる表記は、36位のグリシンをアスパラギン酸に置換したことを示す。これらのVHH抗体の熱安定性を、上記の方法に従って測定した。配列番号3~6のVHH抗体について、配列番号1のVHH抗体のTmとの差(ΔTm)を表3に示す。
配列番号:1の36位のグリシンをアスパラギン、116位のセリンをアルギニンに置換したもの(配列番号:7)、50位のアルギニンをロイシン、78位のグリシンをアラニンに置換したもの(配列番号:8)、36位のグリシンをアスパラギン、50位のアルギニンをロイシン、116位のセリンをアルギニンに置換したもの(配列番号:9)、36位のグリシンをアスパラギン、78位のグリシンをアラニン、116位のセリンをアルギニンに置換したもの(配列番号:10)、36位のグリシンをアスパラギン、50位のアルギニンをロイシン、78位のグリシンをアラニン、116位のセリンをアルギニンに置換したもの(配列番号:11)をそれぞれ作成した。これらのVHH抗体の熱安定性を、上記の方法に従って測定した。結果を表4に示す。
得られたVHH抗体のヒト血清アルブミンに対する結合親和性を表面プラズモン共鳴を用いて測定した。結果を表5に示す。

- 配列番号:10で示されるアミノ酸配列を含む、VHH抗体。
- 配列番号:11で示されるアミノ酸配列を含む、VHH抗体。
- 配列番号:12で示されるアミノ酸配列を含む、VHH抗体。
