薬物組成物を使用する場合、安全かつ有効な量の免疫コンジュゲートを哺乳動物に投与し、安全かつ有効な量は通常少なくとも約10ng/kg体重であり、ほとんどの場合、約50mg/kg体重を超えず、好ましくは、投与量は約50ng/kg体重-約1mg/kg体重である。もちろん、具体的な投与量は、投与経路、患者の健康などの他の要因などの要因も考慮する必要があり、これらはすべて熟練した医師のスキルの範囲内である。

本明細書で使用される用語「CAR-T細胞」、「CAR-T」、「本発明CAR-T細胞」はすべて本発明の第19の態様に記載のCAR-T細胞を指す。
本明細書で使用されるキメラ抗原受容体(Chimeric antigen receptor:CAR)は、細胞外ドメイン、任意のヒンジ領域、膜貫通ドメイン、および細胞内ドメインを含む。細胞外ドメインは、任意のシグナルペプチドと標的特異的結合エレメント(抗原結合ドメインともよばれる)を含まれます。細胞内ドメインは、共刺激分子とζ鎖部分を含む。共刺激シグナル伝達領域とは、共刺激分子を含む細胞内ドメインの一部を指す。共刺激分子は、リンパ球が抗原に効果的に応答するために必要な細胞表面分子であり、抗原受容体またはそれらのリガンドではない。
CARがT細胞で発現すると、細胞外セグメントは特定の抗原を認識することができ、このシグナルは細胞内ドメインを介して伝達され、細胞の活性化と増殖、細胞溶解毒性、IL-2やIFN-γなどのサイトカインの分泌を引き起こし、腫瘍細胞に影響を及ぼし、腫瘍細胞が成長しない、死に至る、または他の方法で影響を受け、患者の腫瘍負荷の軽減または排除をもたらす。抗原結合ドメインは、好ましくは、共刺激分子およびζ鎖の1つまたは複数からの細胞内ドメインと融合する。
本発明はまた、Her2タンパク質を検出する方法に関する。前記方法のステップはおよそ次のとおりです。細胞および/または組織サンプルの獲得、サンプルの媒体への溶解、前記溶解されたサンプルのHer2タンパク質のレベルの検出。
本発明の検出方法において、使用されるサンプルは特に限定されず、代表例は、細胞保存液中に存在する細胞含有サンプルである。
本発明は、本発明の抗体(またはそのフラグメント)または検出プレートを含むキットをさらに提供し、本発明の一好ましい例において、前記キットは、容器、取扱説明書、緩衝液などをさらに含む。
本発明は、Her2レベルを検出するための検出キットをさらに提供し、前記キットは、Her2タンパク質を認識する抗体、サンプルを溶解するためのクラッキング媒体、様々な緩衝液、検出標識、検出基質などの検出に必要な一般的な試薬および緩衝液を含む。前記検出キットは、生体内診断装置であることができる。
本発明は、本発明の免疫コンジュゲートを含むキットをさらに提供し、本発明の一好ましい例において、前記キットは、容器、取扱説明書、同位体トレーサー及び造影剤、フロー検出試薬、細胞免疫蛍光検出試薬、ナノ磁性粒子とイメージング剤からなる群から選択される1つまたは複数の試薬をさらに含む。
好ましくは、本発明のキットは、被験者のHer2発現を非侵入的に検出するための生体内診断キットである。
上述のように、本発明のナノボディは、幅広い生物学的応用価値および臨床的応用価値を有し、その応用は、Her2関連疾患の診断および治療、基礎医学研究、生物学研究などの多くの分野に関する。一好ましい応用は、Her2に対する臨床診断および標的療法である。
(a)本発明のナノボディは、正しい空間構造を有するヒトHer2タンパク質に対して高度に特異的である。
(b)本発明のナノボディの親和性は強い。
(c)本発明のナノボディの製造は簡単である。
(d)本発明のナノボディは、ヒト由来のHer2に特異的に結合でき、Her2高発現腫瘍モデルに効果的に蓄積でき、トラスツズマブまたはペルツズマブと競合しない。本発明のHer2ナノボディは、Her2標的癌診断および治療効果評価、ならびに新世代のHer2の標的インビボ放射線療法に非常に適している。
(1)ヒトHer2のヌクレオチド序列をpCDNA3.1(-)担体に合成し、その後その細胞外フラグメント序列をpFUSE-IgG1担体にサブクローニングし、ここで、hHer2(ECD)のC端にTEV酵素分解ポイントに導入して、FcタグのないhHer2(ECD)タンパク質の調製を容易にする。
(2)Omegaプラスミド大抽出キットで構築されたpFUSE-IgG1-hHer2(ECD)プラスミドを抽出した。
(3)HEK293F細胞をODが2.0×106個/mLになるまで培養した。
(4)プラスミドとトランスフェクション試薬PEIを1:5に均一に混合した後、10分間静置し、その後HEK293F細胞に加え、37℃、6% CO2シェイキングインキュベーターで5-6日間培養した。
(5)細胞上清を回収し、室温でプロテインAビーズと1時間結合させた。
(6)pH7.0のリン酸緩衝液でビーズを洗浄した後、0.1MのpH3.0グリシンでタンパク質を溶出した。
(7)溶出したタンパク質をPBSで限外ろ過し、収量を測定した後、サンプリングしてSDS-PAGE検出(テスト結果は、図1に示した通りである)し、前記抗原の純度は95%を以上であり、後続の免疫反応に使用することができる。
(8)続いて、TEV酵素を使用してタンパク質を酵素分解し、後続抗体スクリーニングのためにタグのない抗原タンパク質Her2(ECD)を獲得した。
(1)1mgのhHer2(ECD)-Fc抗原とフロイントアジュバント(freund’s adjuvant)を等量混合し、新疆バクトリアラクダを週に1回、3回免疫し、B細胞を刺激して抗原特異的ナノボディを発現させた。
(2)3回の免疫後、100mLのラクダ末梢血リンパ球を抽出し、トータルRNAを抽出した。
(3)cDNAを合成し、ネストされたPCRを使用してVHHを増幅した(図2のAは、第1のPCR増幅産物であり、サイズが約700bpのVHH-hinge-CH2-CH3フラグメントをタッピングによって回収し、図2のBは、第2のPCR増幅産物であり、サイズが約400bpのVHHフラグメントである)。
(4)制限酵素Pst IおよびNot Iを使用して、20μgのpMECSファージディスプレイベクター(Biovectorから提供)および10μgのVHHを切断し、2つのフラグメントを連結した。
(5)連結産物をエレクトロコンピテント細胞TG1に形質転換し、Her2ナノボディライブラリを構築し、保存容量を測定した。結果は、図2のCに示した通りであり、構築したライブラリーは勾配希釈した後にプレーティングされ、1/5の濃度で103倍、104倍、105倍、106倍のクローン数を希釈し、モノクローナル抗体の数を計算してライブラリーのサイズが2×109CFUであることを確認した。
(6)同時に、コロニーPCR検出のために24個のクローンをランダムに選択した。図2のDはコロニーPCRの結果を示し、結果は構築したライブラリのVHH挿入率は100%であることを示した。
抗体のスクリーニング
(1)100mMのNaHCO3、pH8.2に溶解した10μgのhHer2(ECD)-Fc抗原(10μg Fc in NaHCO3を対象とする)をNUNCプレートにコンジュゲートし、4℃で一晩放置した。
(2)翌日に100μLの0.1%BSAを添加し、室温で2時間ブロックした。
(3)2時間後、100μLのファージ(2×1011CFU免疫ラクダナノボディファージディスプレイ遺伝子ライブラリー)を加え、室温で1時間反応させた。
(4)0.05%のPBS+Tween-20で5回洗浄して、非特異的ファージを洗い流した。
(5)100mMのトリエタノールアミンでHer2に特異的に結合するファージを分離し、対数期に増殖する大腸菌TG1細胞に感染させ、37℃で1時間培養し、次のスクリーニングのためにファージを生成および精製した。同じスクリーニングプロセスを3回繰り返し、濃縮結果は、図3に示したとおりであり、3回のバイオパニング(bio-panning)プロセスの後、最終的に110倍の濃縮が現れた。
(1)上記2-3回のスクリーニング後、ファージを含む細胞培養皿から600個の個々のコロニーを選択し、100μg/mLのアンピシリン(Ampicillin)を含むTB培地(1LのTB培地には、2.3gのKH2PO4、12.52gのK2HPO4、12gのペプトン、24の酵母エキス、4mLのグリセロールが含まれる)に接種し、対数期に成長した後、1mMのIPTG最終濃度で添加し、28℃で一晩培養した。
(2)浸透法により粗抗体を得て、抗体を抗原被覆ELISAプレートに移し、室温で1時間放置した。
(3)非結合抗体をPBSTで洗浄し、マウス抗HA抗体(北京康為世紀生物科技有限公司から購入)を加え、室温で1時間放置した。
(4)PBSTで非結合抗体を洗浄し、ヤギ抗マウスアルカリホスファターゼ標識抗体を加え、室温で1時間放置した。
(5)非結合抗体をPBSTで洗浄し、アルカリホスファターゼ発色溶液を加え、ELISA装置において405nmの波長で吸収値を読み取った。
(6)サンプルウェルのOD値が対照ウェルのOD値の3倍以上である場合(Ratio+/->3)、陽性クローンウェルと判断される。 PE-ELISAの結果、合計486の陽性クローンを示し、比率(比:+/-)が3~30でしあり、その後、すべての陽性クローンをLA培地に移して培養し、プラスミドを抽出してシーケンスを実行した。シーケンシング結果が多く、シーケンシング結果はほとんど繰り返しシーケンスであるため、表2に示すように、最終的に得られた40ナノボディの対応するELISA結果のみを表示した。
(1)実施例3のシーケンス分析で得られた異なるクローン(7つのナノボディをランダムに選択)について、対応する各プラスミドを大腸菌WK6に電気形質転換し、LA+グルコース、即ちアンピシリンとグルコースを含む培養プレートにコーティングし、37℃で一晩培養した。
(2)単一のコロニーを選び、5mLのアンピシリンを含むLBブロスに接種し、37℃でシェイキング培養した。
(3)1mLの一晩培養した細菌を330mLのTB培地に接種し、37℃でシェイキング培養し、OD値が0.6-1に達したら、IPTGを加え、28℃で一晩シェイキング培養した。
(4)遠心分離して細菌を収集した。
(5)浸透法により抗体粗抽出物を獲得した。
(6)ニッケルカラムイオンアフィニティークロマトグラフィーにより精製されたナノボディを調製した。
精製結果は、図4に示したとおりであり、精製後、抗Her2ナノボディの純度は95%以上に達することができる。
(1)ヒトおよびマウス種の抗原タンパク質HER2をコーティング:ヒトおよびマウス種の抗原タンパク質Her2を添加し、4℃で一晩放置した。
(2)翌日にPBSTで3回洗浄し、1%のBSAを加えて室温で2時間ブロックした。
(3)精製したナノボディを勾配で希釈し、コーティングされたHer2抗原と室温で1時間放置した。
(4)非結合抗体をPBSTで洗浄し、マウス抗HA抗体を加えて、室温で1時間放置した。
(5)非結合抗体をPBSTで洗浄し、ヤギ抗マウスアルカリホスファターゼ標識抗体を加え、室温で1時間放置した。
(6)非結合抗体をPBSTで洗浄し、アルカリホスファターゼ発色溶液を加え、ELISA装置において、405nmの波長で吸収値を読み取り、吸収値に従ってナノボディの特異性を判断した。
検出結果は図5に示したように、本発明のナノボディは、ヒトHer2にのみ結合する。
(1)細胞タイプ:BT474及びMDA-MB-231。PBSで細胞を2回洗浄した。
(2)ナノボディをPBSで0.1ug/uLの濃度に希釈した。
(3)洗浄した細胞をウェルプレートに分注し、各サンプルの細胞数を3×105にした後、希釈したナノボディを各ウェルに加え、均一に混合し、4℃で20分間放置した。
(4)細胞をPBSで2回洗浄し、細胞を100uLのPBSで再懸濁し、マウス抗HA Alexa Fluor488標識抗体を、サンプルあたり1uLに加え、均一に混合し、4℃で20分間放置した。
(5)細胞をPBSで2回洗浄し、300uLのPBSを含むフローチューブに細胞を再懸濁し、光から保護して氷上に置き、機械でテストした。
結果は、Her2発現が高い(BT474)細胞株において、本発明のナノボディの陽性率は>99%であり、Her2発現が低い(MDA-MB-231)細胞株において、ナノボディの陽性率は、6-14%であり、2つの間の差は少なくとも約6倍であったことを示し、これは、本発明のナノボディがHer2に対して非常に良好な特異性を有することをさらに示唆している。いくつかのナノボディの結果を表4に示した。
(1)固定:固定相抗原タンパク質Her2をカルボキシアミノ反応によりCM-5センサーチップの表面に固定した。
(2)結合:ナノボディをHBSバッファーで異なる濃度に希釈し、ナノボディとトラスツズマブまたはペルツズマブまたは抗原のみの結合プロセスを観察した。
(3)チップの再生:次の抗体測定を実行するとき、10mMグリシンですすいで、再生した。
結果は、本発明のナノボディがHer2との親和性がナノモル濃度レベル以上に達し、トラスツズマブまたはペルツズマブと競合してHer2に結合しないことを示した。いくつかのナノボディのデータを表5に示した。
(1)150μLの抗体原液を取り、100μLの0.02mol/L pH7.4リン酸緩衝液と50μLのNa125I溶液を加え、均一に混合し、20μLの5mg/mLクロラミンT溶液を加え、ミキサーで室温で70秒間反応させ、200μLのメタ重亜硫酸ナトリウム溶液(5mg/mL)を加え、5分間作用させた。
(2)PD10カラムで分離し、溶出液は0.02mol/L pH 7.4のリン酸緩衝液であり、チューブあたり10滴を収集し、ペーパークロマトグラフィーを使用してI-125標識ナノボディの純度を確認した。
(3)NOD/SCIDマウスに、細胞接種の1日前に右背中にエストロゲン錠剤を皮下移植し、右***脂肪パッドに1×107Her2高発現(BT474)細胞を接種し、腫瘍が150-200mm3に成長すると、正式な実験研究に使用した。
(4)担癌マウスをイソフルランで麻酔し、I-125標識ナノボディ(~50ug、5MBq)を尾静脈に注射した。
(5)投与の30分後にスキャンし、取得方法は静的15分SPECTおよび中解像度全身CTである。
結果は、本発明の複数のナノボディが、Her2高発現腫瘍モデルの効果的な蓄積に効果的であり、癌の診断および治療に適用されることを示した。同時に、非結合抗体は腎臓と膀胱を介して血液から迅速に除去され、身体の放射線量を減らす。
担癌マウスの一部のナノボディの30分のSPECTスキャン写真と身体分布データを図6と表6に示した。
(1)それぞれ5.5mgのNa2CO3、15.2mgの酒石酸カリウムナトリウム、及び20.5mgのNaBH4を10mLの滅菌ボトルに加え、20分間のCOを通過させ、1mL(35mCi)のNa[99mTcO4]を加え、反応ボトルを密閉し、80℃で30分間加熱した。
(2)50μlの抗体原液を取り、500μlの99mTc(CO)3(H2O)3反応溶液(6.56mCi)を加え、45-50℃で90分間反応させた。
(3)PD10カラムで分離し、溶離液は0.02mol/L pH 7.4リン酸緩衝液であり、薄層クロマトグラフィーにより99mTc標識ナノボディに放射純度を確認した。
(4)NOD/SCIDマウスに、細胞接種の1日前の右背中にエストロゲンの丸薬を皮下移植し、右***脂肪パッドに1×107Her2高発現(BT474)細胞を接種し、腫瘍が150-200mm3に成長すると、正式な実験研究に使用した。
(5)担癌マウスをイソフルランで麻酔し、Tc-99m標識ナノボディ(~10ug、5MBq)を尾静脈に注入するか、20x非標識ナノボディを同時に混合するか、72時間前に20xトラスツズマブまたは20xペルツズマブ尾静脈注射により前処置した。
(6)投与の30分後にスキャンし、取得方法は静的15分SPECTおよび中解像度全身CTである。
結果は、本発明の複数のナノボディが、Her2高発現腫瘍モデルの効果的な蓄積に効果的であり、トラスツズマブまたはペルツズマブと競合しないことを示した。Her2をターゲットにした癌の診断と有効性の評価に使用でき、同時にHer2をターゲットにした新世代の治療法を開発することができる。
担癌マウスの一部のナノボディの30分のSPECTスキャン写真と身体分布データを図7と表7に示した。
Claims (16)
- トラスツズマブまたはペルツズマブと競合せずにHer2に結合する抗Her2ナノボディであって、
Her2エピトープに対するナノボディであり、SEQ ID NO.:1-40のいずれか1つに示されるアミノ酸配列のVHH鎖を有することを特徴とする、前記抗Her2ナノボディ。 - ポリヌクレオチドであって、
請求項1に記載の抗Her2ナノボディのタンパク質をコードすることを特徴とする、前記ポリヌクレオチド。 - SEQ ID NO.:41-80のいずれか1つに示されるヌクレオチド配列を有することを特徴とする
請求項2に記載のポリヌクレオチド。 - 発現担体であって、
請求項2に記載のポリヌクレオチドを含むことを特徴とする、前記発現担体。 - 宿主細胞であって、
請求項4に記載の発現担体を含むか、請求項2に記載のポリヌクレオチドがそのゲノムに組み込まれることを特徴とする、前記宿主細胞。 - 抗Her2ナノボディの生成方法であって、
ナノボディの生成に適した条件の下で、請求項5に記載の宿主細胞を培養して、前記抗Her2ナノボディを含む培養物を獲得するステップ(a)と、及び
前記培養物から前記抗Her2ナノボディを分離または回収するステップ(b)とを含むことを特徴とする、前記抗Her2ナノボディの生成方法。 - 免疫コンジュゲートであって、
(a)請求項1に記載の抗Her2ナノボディと、及び
(b)検出可能なマーカー、薬物、毒素、サイトカイン、放射性核種、酵素、金ナノ粒子/ナノロッド、ナノ磁性粒子、ウイルスコートタンパク質またはVLP、またはそれらの組み合わせからなる群から選択される結合部分を含むことを特徴とする、前記免疫コンジュゲート。 - Tc-99m、Ga-68、F-18、I-123、I-125、I-131、In-111、Ga-67、Cu-64、Zr-89、C-11、Lu-177、Re-188、またはそれらの組み合わせからなる群から選択される(i)診断用同位体と、および/または

(a)Her2分子を検出するための試薬と、(b)腫瘍を治療するための薬物とを調製するために使用されることを特徴とする、前記請求項1に記載の抗Her2ナノボディの使用。 - 薬物組成物であって、
(i)請求項1に記載の抗Her2ナノボディ、または請求項7に記載の免疫コンジュゲートと、及び
(ii)薬理学的に許容される担体とを含むことを特徴とする、前記薬物組成物。 - CAR-T細胞であって、
前記CAR-T細胞は、キメラ抗原受容体CARを発現し、前記CARの抗原結合ドメインは、請求項1に記載の抗Her2ナノボディを有することを特徴とする、前記CAR-T細胞。 - 製剤であって、
請求項15に記載のCAR-T細胞、及び薬理学的に許容される担体、希釈剤または賦形剤を含むことを特徴とする、前記製剤。
