Description
本発明は、バイオフィルム及び前記バイオフィルム内又は周囲に生存する微生物を衛生化及び/又は実質的に除去するための化合物、組成物、方法、及び使用に関する。特に、本発明は、バイオフィルム及び前記バイオフィルム内及び周囲に生存する病原性微生物を処理する創傷処置、創傷被覆材、医療機器、水処理、食品加工、及び歯科治療に使用することができる金属EDTA化合物に関する。
微生物による、皮膚及び医療機器を含む表面(非生物及び生物)のコロニー形成は、患者への重大な感染リスクを表す。これらの微生物が表面上に付着して増殖する際、それらはバイオフィルムを形成する。バイオフィルム環境内に生存する微生物集団が治癒の遅延及び感染リスクの増加に寄与することがますます認識されている。例えば、慢性創傷は、病原性バイオフィルムが存在する場合には、効果的に治癒しないであろう。そのようなバイオフィルムは、患者の免疫系及び抗菌処置に抵抗し、組織破壊、炎症、及び/又は重篤な感染に導きうる。これらの創傷は、しばしば、多微生物集団によりコロニー形成される。そのような慢性創傷において、バイオフィルム内の微生物は、それらのプランクトン対応物と比較して、抗菌性介入に対して100~1000倍耐性でありうる(Singh and Barbul, Wound Rep Reg. 16:1, 2008)。Jamesらによる最近の試験(Wound Rep Reg. 16: 37-44, 2008)では、それらの試験において分析された慢性創傷の少なくとも60%がバイオフィルムによりコロニー形成されることが示されている。
とりわけ、本明細書又は他所で特定されているかにかかわらず、先行技術の少なくとも1つの欠点に対処する化合物を提供すること、又は既存の抗菌剤、例えば先行技術の金属-EDTA化合物の代替物を提供することが、本発明の1つの目的である。例えば、創傷処置又は医療機器において抗バイオフィルム活性及び/又は抗菌活性及び/又は抗炎症活性を提供する化合物又は組成物を提供することが、本発明の目的でありうる。

a)金属-EDTA錯体を、Ag、Al、Au、Ba、Bi、Tl、Ce、Co、Cu、Fe、Ga、Ir、Mo、Rh、Ru、Ti、及びZnイオンより選択される第1の金属イオンの塩と反応させる工程;及び
b)工程a)において形成された、Ag、Al、Au、Ba、Bi、Tl、Ce、Co、Cu、Fe、Ga、Ir、Mo、Rh、Ru、Ti、及びZnイオンより選択される1つの金属イオンを含むEDTA錯体を、Ag、Al、Au、Ba、Bi、Tl、Ce、Co、Cu、Fe、Ga、Ir、Mo、Rh、Ru、Ti、及びZnイオンより選択される第2の金属イオンの塩と反応させる工程;
を含み、ここで、第1及び第2の金属イオンは異なる。
Na4(EDTA)標準、
塩酸(HCl)ACS試薬、
チオシアン酸塩溶液-100mlの水中のチオシアン酸アンモニウム、
Fe3+溶液-少量のHCl中にアンモニウム硫酸鉄(III)12水和物を溶解し、酢酸ナトリウム緩衝液を加えて、次に水に加えることにより形成する。
1.1gのPUを10mlのジメチルホルムアミド(DMF)中に60℃で一晩溶解させることによりポリウレタン(PU)溶液を調製する。
2.四銀EDTA錯体を調製し、異なる濃度(100mg/ml、50mg/ml、10mg/ml、1mg/ml、及び0mg/ml)でPU溶液中に懸濁する。
3.ガラスカバースリップ及びプラスチックディスク上に四銀EDTAをコーティングしてフィルムを作製する。
4.コーティングしたカバースリップ及びプラスチックディスクを次に、2mlの脱イオン水中に1週間にわたり浸漬する。
5.四銀EDTAの溶出を、標準曲線方法に従って測定する。
合成:4Ag+(aq) + EDTA4-(aq) → Ag4(EDTA) (s)

Claims (16)
- 式Mn(EDTA)の化合物であって、式中:
nが2~4の整数であり;
各々のMが金属イオンであり;そして
Mnが、Ag、Al、Au、Ba、Bi、Tl、Ce、Co、Cu、Fe、Ga、Ir、Mo、Rh、Ru、Ti、及びZnイオンより選択される少なくとも2つの異なる金属イオンを含む化合物。 - n=3又は4であり、Mnが2つのAgイオンを含む、請求項1記載の化合物。
- Mnが少なくとも1つのAgイオン及び1つのZnイオンを含む、請求項1又は請求項2記載の化合物。
- 式Ag2Zn(EDTA)を有する請求項3記載の化合物。
- Mnが少なくとも1つのAgイオン及び1つのCuイオンを含む、請求項1又は請求項2記載の化合物。
- 式Ag2Cu(EDTA)を有する、請求項5記載の化合物。
- 請求項1~6のいずれかに記載の1つ又は複数の化合物を含む組成物。
- 1つ又は複数の化合物を含むヒドロゲルである、請求項7記載の組成物。
- 1つ又は複数の化合物と接触する繊維を含む、請求項7記載の組成物。
- 非金属イオン抗菌剤を含む、請求項7~9のいずれかに記載の組成物。
- 界面活性剤を含む、請求項7~10のいずれかに記載の組成物。
- 請求項1~6のいずれかに記載の化合物又は請求項7~11のいずれかに記載の組成物を含む創傷被覆材。
- 請求項1~6のいずれかに記載の化合物又は請求項7~11のいずれかに記載の組成物を含む医療機器。
- 基質のバイオフィルムを衛生化する及び/又は基質からバイオフィルムを実質的に除去するための、請求項1~6のいずれかに記載の化合物又は請求項7~11のいずれかに記載の組成物の使用であって、ここで基質が、医療機器の一部、あるいは食品調製及び加工装置又は食品の一部である使用。
- 医薬として使用するための、請求項7~11のいずれかに記載の組成物。
- 請求項1~6のいずれかに記載の化合物又は請求項7~11のいずれかに記載の組成物及び医療機器又は創傷被覆材を含むキット。
