「抗体−薬物複合体」は、細胞傷害性薬または細胞増殖抑制薬に複合体化された抗体を指す。典型的には、抗体−薬物複合体は、細胞表面上の標的抗原(例えば、NTB−A)に結合した後、抗体−薬物複合体を細胞の中に内在化させ、薬物を放出する。
本発明は、とりわけ、NTB−Aに特異的に結合する20F3抗体、ならびにそのキメラ、ベニヤ、及びヒト化形態を提供する。20F3抗体との結合に関して競合するか、または20F3抗体と同じエピトープに結合する抗体も提供される。本抗体は、例えば、様々なNTB−A発現癌の治療及び診断、ならびにNTB−Aの検出(例えば、細胞上のNTB−A発現の検出)に有用である。裸抗体及び複合体化抗体を含む本発明の抗体を使用する、かかる治療、診断、及びNTB−A検出方法も提供される。
別途記載のない限り、NTB−AはヒトNTB−Aを意味する。例示的なヒト配列は、配列番号1(Swiss−ProtQ96DU3)を割り当てられている。4つのスプライス変異体アイソフォームが既知である。成熟細胞外領域は、Q96DU3の残基22〜226によって結合される。アイソフォーム4中のより短い細胞外ドメインは、C2ドメインと2つのITSMモチーフを有し、免疫グロブリンドメインを含む残基18〜121を欠く。
A. 結合特異性及び機能特性
本開示は、とりわけ、名称20F3のマウス抗体、及び20F3抗体のキメラ、ベニヤ、及びヒト化形態、ならびにNTB−Aとの結合に関して20F3抗体と競合する抗体、及び20F3抗体と同じエピトープに結合する抗体に言及する。20F3抗体は、通常型のヒトNTB−A(配列番号1)、ヒトアイソフォーム4、及びカニクイザルNTB−Aに結合する。これらの結合特性により、そのエピトープは、アイソフォーム4に不在であるNTB−Aのセグメントの外側であり(残基128〜226内)、NTB−Aのヒト型とカニクイザル型との間で完全にまたは実質的に保存されるエピトープに存在することが示唆される。

抗NTB−A抗体は、細胞傷害部分または細胞増殖抑制部分と複合体化されて、抗体薬物複合体(ADC)を形成し得る。抗体との複合体化に特定好適な部分は、細胞傷害性薬(例えば、化学療法薬)、プロドラッグ変換酵素、放射性同位体もしくは化合物、または毒素(治療薬と総称されるこれらの部分)である。例えば、抗NTB−A抗体は、化学療法薬などの細胞傷害性薬、または毒素(例えば、細胞増殖抑制もしくは殺細胞薬、例えば、アブリン、リシンA、緑膿菌外毒素、またはジフテリア毒素)と複合体化され得る。細胞傷害性薬の有用なクラスの例は、例えば、DNA小溝結合、DNAアルキル化剤、及びチューブリン阻害剤を含む。例示的な細胞傷害性薬としては、例えば、アウリスタチン、カンプトテシン、カリチアマイシン、デュオカルマイシン、エトポシド、マイタンシノイド(例えば、DM1、DM2、DM3、DM4)、タキサン、ベンゾジアゼピン(例えば、ピロロ[1,4]ベンゾジアゼピン二量体、インドリノベンゾジアゼピン二量体、及びオキサゾリジノベンゾジアゼピン二量体を含む、ピロロ[1,4]ベンゾジアゼピン、インドリノベンゾジアゼピン、及びオキサゾリジノベンゾジアゼピン)、及びビンカアルカロイドが挙げられる。
式IV、IVa、及びIVbのNTB−A標的−薬物複合体を参照すると、添字pは、抗体分子に対する薬物負荷(抗体分子に結合された薬物の分子数)を表し、整数値である。抗体−薬物複合体分子の集団を含む組成物において、平均薬物負荷(例えば、集団における抗体当たりの薬物−リンカー分子の平均数)は、標的細胞に送達され得る薬物の量を決定するため、重要な品質特性である。平均薬物負荷は、整数値または非整数値であってもよいが、典型的には非整数値である。
本明細書に記載の抗NTBA−抗体は、裸抗体または抗体薬物複合体として、NTB−A発現細胞と関連する疾患または障害を治療するための方法において使用され得る。

NTB−A抗体産生マウスの脾臓及びリンパ節から採取したリンパ球を骨髄腫細胞に融合させた。融合させた細胞をハイブリドーマ増殖培地中で一晩回復させた。回復したら細胞を遠沈させ、続いて半固体培地にプレーティングした。ハイブリドーマをインキュベートし、IgG産生ハイブリドーマクローンを選抜した。20F3抗体は、ADCとして強力な細胞傷害を示し、カニクイザルNTB−Aに結合した少数の抗体のうちの1つであった。
ヒト化抗体はマウス20F3抗体に由来した。5つのヒト化重鎖(HA−HE)及び4つのヒト化軽鎖(LA−LD)を、復帰突然変異を異なる位置に組み込んで作製した。場合によっては、復帰突然変異はマウス生殖系列と一致することになるが、他の場合には一致しない(体細胞変異がある場合など)。ヒト化重鎖とヒト化軽鎖とを対にした。配列アラインメントに関する図1及び2、ならびに表1〜4を参照されたい。
EC50を決定するために、3点用量漸増を行った未標識のヒト化抗ヒトNTB−A抗体を、Alexa Fluor 647と複合体化した定濃度(最終5nM)のマウス20F3抗体と混合した。抗原陽性Ramos細胞を、ウェル当たり1×105細胞で、96ウェルV底プレート(Thermo Scientific,Rochester,NY)にプレーティングした。50倍連続希釈の抗体を、FAC緩衝液(PBS+2%ウシ胎仔血清)中で調製し、2回繰り返して細胞に添加した。抗体を、遮光しながら1時間氷上で細胞と共にインキュベートした。細胞を、FAC緩衝液で2回洗浄し、LSRIIフローサイトメーター(BD BioSciences,San Jose,CA)で分析した。EC50値をGraphPad Prismソフトウェア(La Jolla,CA)によって決定した。結果を表5に示す。
補体依存性細胞傷害性(CDC):正常なヒトT細胞(All Cells,Alameda,CA)または癌細胞系(ATCC,Manassas,VA)を5μMのSytox Green(Life Technologies,Grand Island,NY)によって標識標識した後、RPMI1640培地+1020%ヒト血清(Complement Technology,Tyler,TX、またはSolomon Park Research Laboratories,Kirkland,WA)中で、連続用量漸増を行った抗ヒトNTB−A h20F3抗体またはPBD二量体ADC(0.02〜50μg/mL)と結合させた。細胞を、37℃、5%CO2で2時間インキュベートし、溶解した細胞からの蛍光を、Envisionプレートリーダー(Perkin Elmer,Waltham,MA)によって測定した。データは、1%TritonX−100で処理した細胞(Sigma,ST.Louis,MO)の陽性対照を用いて決定した最大細胞溶解率として表す。
多発性骨髄腫細胞系(MM.1RまたはU−266)を飽和濃度のADC(10μg/mL)と結合させ、培地で洗浄し、37℃または4℃でインキュベートした。試料をPE標識ヤギ抗ヒト抗体(Jackson ImmunoResearch,West Grove,PA)によって染色した時点を収集し、1%PFA/PBS中で固定した。全ての時点を収集したら、平均蛍光強度を、FACs Caliburフローサイトメーター(BD BioSciences,San Jose,CA)で測定した。抗NTB−A h20F3ec−1910(2)PBD二量体ADCはMM.1R細胞中に急速に内在化し、ADCの80%が37℃のインキュベーションで4時間以内に内在化した。h20F3ec−1910(2)ADCの20%のみが、細胞を4℃の対照条件下に保った4時間目に内在化した。
ヒト化抗体(HDLD)をPBD及びアウリスタチン抗体薬物複合体として試験した。抗有糸***剤モノメチルアウリスタチンE(MMAE)を、カテプシン切断性バリン−シトルリン(vc)リンカーを介して抗NTB−A mAbに複合体化した。第2のアウリスタチンであるアウリスタチン−2を、ペプチドリンカーを介して抗NTB−A mAbに複合体化した。PBD複合体について、薬物−リンカーSGD−1910を、抗体のIgG1鎖の位置239(付番はEUインデックスに従う)に導入されたシステイン残基のチオール基を介して抗NTBA抗体と複合体化すると、平均薬物負荷は抗体当たり約2薬物となった。位置239にシステインを有する抗体は、ECの表示を伴う。調製は、本明細書に記載の抗NTBA抗体を使用して、WO2011/130613に記載されている通りに行った。ADCを培地に3倍に連続希釈して、10点用量曲線(1,000ng/mL〜0.05081ng/mL)を得て、96ウェルアッセイプレート中で培養された多発性骨髄腫細胞に適用した。細胞系を、4回繰り返して抗NTB−A ADCで処理し、37℃、5%CO2で96時間インキュベートした。細胞の生存性を、Cell Titer Glo発光細胞傷害性アッセイ(Promega)を使用してアッセイし、EnVisionプレートリーダー(Perkin Elmer)を使用してデータ収集した。用量効果曲線及びIC50値を、GraphPad Prismソフトウェアを使用して計算した。結果を表11及び12に示す。
休止ヒトT及びBリンパ球に対する抗NTB−A ADCの細胞傷害作用を評定するため、精製した細胞を黒色の96ウェルアッセイプレートに播種した。次に細胞を、10ug/mLから開始して合計8の希釈点で5倍に漸減させた抗NTB−A ADCで処理した。遊離薬物処理のため、細胞に、それぞれ1,000nM及び100nMから開始して合計8の希釈点で5倍に漸減させたMMAE及び遊離PBD二量体を投与した。プレートを、37℃、5%CO2で96時間インキュベートした後、室温に平衡化させた。等体積のCell−Titer Glo試薬を各ウェルに添加し、プレートを室温で更に30分間インキュベートした。次に、プレートをPerkin Elmer Envisionプレートリーダーで読み取った。用量効果曲線及びIC50値を、GraphPad Prismソフトウェアを使用して計算した。結果を表13に示す。
NSG(NOD scidガンマ;NOD.Cg−Prkdcscid Il2rgtm1Wjl/SzJ)マウスに、動物当たり100万個のMM.1R細胞を静脈内移植し、多発性骨髄腫の播種性モデルを生成した。腫瘍細胞移植5日後に、治療群当たりn=10マウスに、h20F3ec−1910(2)PBD ADCまたは非結合対照hIgGec−1910(2)PBD ADCの単一腹腔内注射を行った。検査したPBD ADC用量レベルは、333μg/kg、111μg/kg、及び37μg/kgであった。進行した腫瘍負荷のマウスは、後肢麻痺、頭蓋腫大、及び/または瀕死の症状を示したときに屠殺した。図3に示されるように、HDLD h20F3ec−1910(2)PBD二量体ADCは、全ての用量レベル(単一用量)で10頭中10頭のマウスにおいて持続的な完全奏功をもたらし、一方、非結合対照PBD ADC投薬マウスは、研究の80日目までに、疾患により全て屠殺した。h20F3ec−1910(2)ADC群(37μg/kg)と対照hIgGec−1910群(2)ADC(333μg/kg)群との間の統計的に有意な差(P<0.0001、Mantel−Cox試験)を得た。
SCID(C.B−17/Sz−Prkdcscid)マウスに、動物当たり500万個のRaji非ホジキンリンパ腫細胞を皮下移植した。平均腫瘍体積が100mm3に達したとき、治療群当たりn=10のマウスに、h20F3ec−1910(2)PBD ADCまたは対照hIgGec−1910(2)PBD ADCの単一腹腔内注射を行った。検査したPBD ADC用量レベルは、100μg/kg、50μg/kg、及び25μg/kgであった。個々のマウスを、皮下Raji腫瘍体積が1,000mm3に達したときに屠殺した。図11に示されるように、HDLD h20F3ec−1910(2)PBD二量体ADCは、100μg/kgの用量レベルで全てのマウスにおいて持続的な完全奏功をもたらした。加えて、h20F3ec−1910(2)PBD ADCは、50ug/kgの用量レベルで6頭中4頭のマウスにおいて持続的な完全奏功をもたらし、一方、腫瘍成長の遅延は、25ug/kgの用量レベルで観察された。hIgGec−1910(2)対照PBD ADCは、100ug/kgで抗腫瘍活性を有さず、6頭中0頭で完全奏功をもたらした。

(a)前記成熟重鎖可変領域が、Kabatによって定義される3つの重鎖相補性決定領域(CDR)であって、配列番号10からなる重鎖CDR1、配列番号11からなる重鎖CDR2及び配列番号12からなる重鎖CDR3を含み、前記成熟軽鎖可変領域が、Kabatによって定義される3つの軽鎖CDRであって、配列番号19からなる軽鎖CDR1、配列番号20からなる軽鎖CDR2及び配列番号21からなる軽鎖CDR3を含む、あるいは
(b)前記成熟重鎖可変領域が、IMGTによって定義される3つの重鎖相補性決定領域(CDR)であって、配列番号22からなる重鎖CDR1、配列番号23からなる重鎖CDR2及び配列番号24からなる重鎖CDR3を含み、前記成熟軽鎖可変領域が、IMGTによって定義される3つの軽鎖CDRであって、配列番号25からなる軽鎖CDR1、アミノ酸配列ATSからなる軽鎖CDR2及び配列番号26からなる軽鎖CDR3を含む、抗体。 - ヒト化抗体、キメラ抗体、またはベニヤ抗体である、請求項1に記載の抗体。
