Description
本明細書は、多孔質細胞足場及びその利用に関する。 The present specification relates to a porous cell scaffold and its utilization.
Conventionally, a porous cell scaffold made of a biocompatible polymer or the like has been used for producing a three-dimensional structure using cells. As one of the methods for producing such a porous cell scaffold, there is a method called a porogen leaching method in which pores corresponding to pologens are formed in a polymer matrix using insoluble solid particles (porogens). In the pologen leaching method, particles (porogen) such as sugars and sodium chloride that are insoluble in the non-aqueous solvent are mixed in advance in a solution in which a biocompatible polymer such as polylactic acid is dissolved in a non-aqueous solvent such as dioxane and chloroform. This is a method in which after evaporating a non-aqueous solvent, only chloroform is dissolved in a solvent that dissolves chloroform to form a porous structure in a polymer matrix (Non-Patent Document 1). The polo-gen leaching method has an advantage that it can deal with a complicated shape and can form pores according to the size of the polo-gen.
しかしながら、ポローゲンリーチング法で得られる多孔質体は、細孔がそれぞれ独立した独立気泡であることが多く連通率が低い場合が多かった。かかる多孔質体の表面に細胞を播種して場合、多孔質体の更なる内部方向への細胞の移動や増殖が妨げられやすく、意図した細胞構造体が必ずしも得られるわけではなかった。 However, in the porous body obtained by the pologen leaching method, the pores are often independent cells, and the communication rate is often low. When cells are seeded on the surface of such a porous body, the movement and proliferation of cells in the further internal direction of the porous body are likely to be hindered, and the intended cell structure is not always obtained.
本明細書は、細孔の連結性に優れる多孔質細胞足場基材を提供する。 The present specification provides a porous cell scaffold substrate having excellent pore connectivity.
As a result of various studies on the pologen leaching method, the present inventors have provided cavities derived from a conjugate formed by linking polymers, and further, ice crystals grown from the surface of the polymer or its vicinity. We have succeeded in obtaining a porous material having a polymer matrix having pores derived from. According to such a porous material, a high porosity and pore communication rate are provided because the pores are derived from pologene, the pores are derived from the connecting portion of pologene, and the pores are derived from ice crystals associated with pologene. It was found that the above can be satisfied. The present specification provides the following means based on such findings.

For example, animal cells, plant cells and the like can be mentioned. The animal cell may be a cell constituting an animal tissue or organ including human. It may be a scaffolding for.
以下、本足場、その製造方法等について詳細に説明する。 Hereinafter, the scaffold, its manufacturing method, and the like will be described in detail.
(多孔質細胞足場)
(マトリックス)
本足場は、多孔質構造を規定し、ポリマーを含むマトリックスを備えることができる。以下、適宜図1を参照して説明する。なお、図1は、本足場を例示するものではあるが、本足場は図1に開示されるものに限定されない。
(Porous cell scaffold)
(matrix)
The scaffold can define a porous structure and include a matrix containing a polymer. Hereinafter, description will be made with reference to FIG. 1 as appropriate. Although FIG. 1 illustrates the main scaffold, the main scaffold is not limited to the one disclosed in FIG.
The polymer constituting the
本足場100におけるマトリックス10は、後述する第1の細孔20、第3の細孔40及び第2の細孔60を規定する周辺構造体である。また、マトリックス10は、種々の細孔を包含し規定する、ウォール及び/又はフレームを少なくとも部分的に有する構造体を構成することができる。ウォール及びフレームの厚み及び形状等は、内包する細孔の大きさ、個数、形状等によって、種々の形態を採りうる。
The
また、マトリックス10のウォール及び/又はフレームである構造体は、それ自体多孔質体であってもよい。マトリックス10は、第1のサイズ、第3のサイズ、及び後述する第2のサイズの20%以下、10%以下又は5%以下程度の孔径の微小な孔を多数備える多孔質体などとすることができる。
Further, the structure which is the wall and / or frame of the
(第1の細孔)
マトリックス10は、互いに連通する第1のサイズの第1の細孔20を規定することができる。マトリックス10においては、少なくとも二つの隣接する第1の細孔20が連結してキャビティ22を構成している。典型的には、例えば、図1に示すように、3個以上あるいはさらに多数個の第1の細孔20が連結したキャビティ22を構成することができる。
(First pore)
The
(First size of the first pore)
The first size of the
第1のサイズは、特に限定するものではないが、培養する細胞の大きさ等を考慮して決定することができる。例えば、第1のサイズは、例えば、200μm以上400μm以下とすることができる。かかるサイズを備えていることで、十分な空孔率を確保することができる。第1のサイズは、好ましくは、240μm以上390μm以下である。 The first size is not particularly limited, but can be determined in consideration of the size of the cells to be cultured and the like. For example, the first size can be, for example, 200 μm or more and 400 μm or less. By having such a size, a sufficient porosity can be ensured. The first size is preferably 240 μm or more and 390 μm or less.
第1のサイズは、例えば、走査型電子顕微鏡において、1200μm×850μmの画像30枚につき、第1の孔20として確認できた細孔について、ImageJソフトウェアあるいはそれと精度及び正確性において同等である他の画像解析ソフトウェアを用いて、2点間オートスケール機能を用いて測定することができる。なお、二点間距離は、孔の長軸及び短軸を避けて中間径を測定する。
The first size is, for example, in a scanning electron microscope, for 30 images of 1200 μm × 850 μm, for the pores confirmed as the
( Third pore)
The
( Third size of the third pore)
The third size of the
The third size is not particularly limited, but can be, for example, 90 μm or more. This is because having such a size makes it possible to sufficiently secure the mobility of cells and the like as well as the mobility of cells and the like. From the viewpoint of more efficient culture, for example, it is 100 μm or more, 110 μm or more, and 120 μm or more, for example. The upper limit is not particularly limited, but is, for example, 180 μm or less, 170 μm or less, and 160 μm or less, for example. The range of the third size can be set by appropriately combining these lower and upper limits, and can be, for example, 90 μm or more and 180 μm or less, 100 μm or more and 170 μm or less.

(多孔質基材の作製)
(1)ポリマー濃度が5%(w/w)になるようにPLLA(ポリL−乳酸)と1,4−ジオキサンをバイアル瓶に入れた後、スターラーで6時間撹拌しPLLAを溶解させた。PLLAが溶解した後、超音波処理を30分間行い、マトリックス原液を調製した。
(Preparation of porous substrate)

(空孔率、吸水率及び連結率の評価)
図5A及び図5Bに、実施例の多孔質基材、対照例1及び2の多孔質基材の空孔率、吸水率及び連結率についての評価結果を示す。なお、これらの評価は、以下の方法によって行った。
(Evaluation of vacancy rate, water absorption rate and connection rate)
5A and 5B show the evaluation results of the porosity, water absorption rate and coupling rate of the porous base material of Examples and the porous base materials of Control Examples 1 and 2. These evaluations were carried out by the following methods.
(1) Pore ratio For the pore ratio, the bulk density (ρa) is calculated from the dry weight and the apparent volume of the
(2) Water absorption rate After measuring the weight of each porous base material, the weight was measured after sufficiently absorbing water. The result was applied to the following formula to obtain the water absorption rate. The weight after water absorption was defined as the weight after immersing the porous substrate in 70% ethanol for 15 minutes, sufficiently replacing it with ultrapure water, and allowing it to stand at 37 ° C. for 3 hours.
Water absorption rate (%) = (weight of porous material after water absorption-weight of porous material when dried) / weight of porous wet material when dried x 100
(3) Linkage rate The water absorption rate (theoretical water absorption rate) per unit mass (g) of the

(Evaluation of cell seeding and infiltration)
After absorbing water into the porous substrate of the example and the porous substrate of the control example (diameter 14 mm × thickness 6 mm, respectively ), 1 ml of MC3T3-E1 cell suspension (2 × 10 5 cells / ml) was seeded. .. The next day, the porous substrate was cut and the living cells in the substrate were evaluated by Calcein-AM. The results are shown in FIG. As shown in FIG. 6, according to the porous substrate of the example, it was found that the cells infiltrated into the substrate even within 12 hours.
