[発明の背景]
米国および世界の全住民は、5000万人を超える人々が死亡した1918年のスペイン(H1N1)の大流行と類似した汎発性インフルエンザ大流行の危険にさらされ続けている。同様に、兵器化されたインフルエンザウイルスは細菌戦争による大きな脅威であり続けている。さらに、抗原連続変異により、インフルエンザに対する予防をしようとしている個体は、毎年ワクチン接種することを必要とし、最近の研究により、季節性ワクチン製剤は、ワクチン接種株と循環株との間にミスマッチが存在する場合、わずかな効果しかないことが示されている。
本明細書に使用される「約」および「およそ」という用語は、数値に関して使用される場合、述べられている値の+または−20%以内を意味する。
一般に「インフル(flu)」として知られているインフルエンザは、インフルエンザウイルスである、オルトミクソウイルス科のRNAウイルスによって引き起こされるトリおよび哺乳動物の感染疾患である。インフルエンザは季節的流行として世界中に広がり、1年に約300万〜500万人の重症疾患の症例および1年に約250,000〜500,000人の死、いくつかの大流行の年には数百万人に増加する死をもたらす。20世紀において、3回のインフルエンザ大流行が発生し、各々はヒトにおけるウイルスの新規株の出現によって引き起こされ、何千万人もの人々を殺した。多くの場合、既存のインフルウイルスが別の動物種からヒトに伝播する場合、または既存のヒト株が、通常トリまたはブタに感染するウイルスから新規遺伝子を取得する場合に新規インフルエンザ株が出現する。
いくつかの態様において、本発明は、操作されたインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体、そのようなポリペプチド、タンパク質および/またはタンパク質複合体を含む組成物、ならびにそのようなポリペプチド、タンパク質および/またはタンパク質複合体を使用する方法を提供する。そのようなタンパク質は、出発点として任意の適切なインフルエンザウイルスHAタンパク質を使用して作製できる。たとえば、本発明のタンパク質は、出発点として任意の適切なインフルエンザ型(たとえばA、BまたはC)、インフルエンザウイルスの亜型(限定されないが、H1N1、H1N2およびH3N2亜型を含む)または株(たとえばH1N1 A/プエルトリコ/8/1934(「PR8」)株(配列番号1))由来のインフルエンザHAタンパク質を使用して作製できる。ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体の重要な性質の1つは、それらが、非常に変わりやすい頭部ドメインと異なるHAタンパク質の三量体ストークドメインを含むことであり、そのHAタンパク質の三量体ストークドメインは、インフルエンザ型、亜型および株の間でより保存されている。したがって、インフルエンザウイルスの同種の型、亜型および株に対する(すなわち、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体を作製するための出発点として使用される同じ型、亜型および/または株のインフルエンザウイルスに対する)ワクチン免疫原として有用であることに加えて、本発明のHAポリペプチド、タンパク質および/またはタンパク質複合体はまた、インフルエンザウイルスの異種の型、亜型および株に対する(すなわち、操作されたHAポリペプチド、タンパク質および/またはタンパク質複合体を作製するための出発点として使用されるものと異なる型、亜型および/または株のインフルエンザウイルスに対する)ワクチン免疫原として有用であってもよい。
ここで記載されている特定のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体を提供することに加えて、本発明はまた、このようなインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸、ならびにこのような核酸を含む組成物およびベクターを提供する。このような核酸は、当該分野において公知の任意の適切な方法を使用して得ることができるか、または作製できる。たとえば、インフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸分子は、クローンDNAから得ることができるか、または化学合成によって作製できる。いくつかの態様において、核酸は、当業者に公知の方法、たとえばランダム−またはポリA−プライム逆転写のいずれかによって調製されたRNAを逆転写することによって得ることができる。起源が何であれ、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸分子は、任意の適切なベクター、たとえば、核酸分子の増殖に使用されるベクターまたは核酸分子の発現に使用されるベクター内にクローニングされていてもよい。核酸は、必要な場合、種々の制限酵素を使用して特定の部位で切断されていてもよい。発現を必要とする態様において、核酸は、所望の細胞種類、たとえば哺乳動物細胞または昆虫細胞における発現を誘導するのに適したプロモーターに作動可能に連結されていてもよく、任意の適切な発現ベクター、たとえば哺乳動物または昆虫発現ベクター内に組み込まれていてもよい。本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸分子は、その核酸分子から発現されるタンパク質の発現レベルを向上させるために当該分野において公知の方法によって最適化された。たとえば、コドン最適化は種間のコドン使用頻度の変化を最小化または排除するために使用されていてもよい。いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、ヒト(たとえば、配列番号63および図48を参照のこと)、Cricetulus griseus(たとえば、配列番号64および図49を参照のこと)、Nicotiana benthamiana(たとえば、配列番号65および図50を参照のこと)、Pichia pastoris(たとえば、配列番号66および図51を参照のこと)、Saccharomyces cerevisiae(たとえば、配列番号67および図52を参照のこと)またはSpodoptera frugiperda(たとえば、配列番号68および図53を参照のこと)における発現のためにコドン最適化されている核酸分子に由来する。
いくつかの態様において、本発明のインフルエンザHAポリペプチドおよび/またはタンパク質は、所望の立体構造的構造、たとえばストークドメインの天然の三量体立体構造を有するタンパク質複合体に構築され、その立体構造を安定化させるために架橋される。架橋され得るインフルエンザHAタンパク質、およびこのような架橋結合を容易にするように設計される特定のインフルエンザHA変異体の特定の領域の詳細は本出願の他の項に記載されている。いくつかの態様において、架橋はインフルエンザHAタンパク質の三次および/または四次構造を安定化させるために使用されていてもよい。いくつかの態様において、架橋結合は分子内および/または分子間架橋結合であってもよい。いくつかの態様において、使用される架橋は標的架橋である。いくつかの態様において、使用される架橋は生理的条件下で安定である。いくつかの態様において、使用される架橋は、たとえば発現の間および/または貯蔵(たとえば高濃度のインフルエンザHAタンパク質を含む組成物の貯蔵)の間、インフルエンザHAタンパク質の凝集体形成を導かない。いくつかの態様において、このような架橋の導入は、免疫原、たとえばワクチン免疫原としての本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質の有効性を向上させることができる。いくつかの態様において、このような架橋の導入は、インフルエンザHAタンパク質内のエピトープ、たとえばストークドメイン内のエピトープを安定化させることができ、それによりエピトープが特定の抗体によって認識されることができ、抗体の産生を誘発でき、および/または抗体が結合するとB細胞受容体を活性化できる。
いくつかの態様において、本発明は、ジ−チロシン(DT)架橋を含むインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体、ならびにこのようなDT架橋したインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体を作製する方法を提供する。
本発明のいくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体(および/またはそれらの合成における中間体)は、たとえば、頭部ドメインのタンパク質分解による除去を容易にするために使用され得る1つ以上のプロテアーゼ認識モチーフを含む。当該分野において公知である任意の適切なプロテアーゼ認識モチーフが使用されていてもよい。このような操作されたプロテアーゼ認識部位は、それらが、頭部ドメインの破壊および/または除去に有用であるが、好ましくはストークドメイン(および/またはストークドメインにおける中和エピトープの立体構造)の天然の三量体立体構造を破壊しないインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体における任意の適切な場所に配置されていてもよい。このような場所は、限定されないが、機能的アッセイ、抗体結合アッセイ、抗原アッセイ、構造アッセイなどにおいて操作されたプロテアーゼ認識部位を導入している効果を試験することを含む、当該分野において公知である方法を使用して決定されていてもよい。いくつかの態様において、このような操作されたプロテアーゼ認識モチーフは可変ループ領域内に位置していてもよい。なぜならこのような領域はインフルエンザHAタンパク質の構造および/または機能を顕著に変化させずにアミノ酸配列の変化を許容することが知られているからである。本発明のインフルエンザHAタンパク質は、インフルエンザHAタンパク質配列内にプロテアーゼ認識配列を生成するために、たとえば、プロテアーゼ認識部位(たとえば、配列番号18、19、21、23および25を参照のこと)を含むか、その一部を含む1つ以上のアミノ酸を挿入することによって、またはインフルエンザHAタンパク質由来の1つ以上のアミノ酸を、プロテアーゼ認識部位(たとえば、配列番号24を参照のこと)を含むか、もしくはその一部を含む異なるアミノ酸と置換することによって、またはアミノ酸(たとえば、配列番号20および22を参照のこと)の挿入および置換の組合せを実施することによって1つ以上のプロテアーゼ認識配列を導入するように操作されていてもよい。操作されたプロテアーゼ認識部位は典型的に約20までのアミノ酸残基からなる。いくつかの態様において、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、以下の一番目の頭部を除去した部位:アミノ酸残基53〜67、アミノ酸残基60〜76、アミノ酸残基269〜277、およびアミノ酸残基277〜290のうちの1つ以上において操作されたプロテアーゼ認識モチーフを含み、また、以下の二番目の頭部を除去した部位:アミノ酸残基142〜146、およびアミノ酸残基155〜164のうちの1つ以上において操作されたプロテアーゼ認識モチーフを任意に含んでいてもよい。いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、インフルエンザHAタンパク質の以下の領域:アミノ酸残基40〜68、アミノ酸残基60〜76、アミノ酸残基77〜114、アミノ酸残基120〜141、アミノ酸残基142〜146、アミノ酸残基148〜178、アミノ酸残基182〜188、アミノ酸残基195〜201、アミノ酸残基209〜242、アミノ酸残基250〜255、アミノ酸残基260〜285、アミノ酸残基277〜290、およびアミノ酸残基286〜320のうちの1つにおいてアミノ酸残基位置において開始するプロテアーゼ認識配列を含む。いくつかの態様において、このようなプロテアーゼ認識モチーフは、インフルエンザHA頭部ドメインにおけるSa、Ca、SbおよびCb抗原部位のうちの1つ以上におけるタンパク質分解による切断を可能にすることができる。いくつかの態様において、プロテアーゼ認識モチーフは、位置48、63、278、282、286または291におけるアミノ酸の直後でインフルエンザHAタンパク質内に挿入される。いくつかの態様において、プロテアーゼ認識モチーフは、インフルエンザHAタンパク質の以下の領域:アミノ酸残基38〜58、アミノ酸残基53〜73、アミノ酸残基268〜288、アミノ酸残基272〜292、アミノ酸残基276〜296およびアミノ酸残基281〜301のうちの1つ以上においてインフルエンザHAタンパク質内に挿入される。いくつかの態様において、プロテアーゼ認識モチーフは、PreScissionプロテアーゼ認識配列(たとえば、LEVLFQGP(配列番号69))もしくはTEV認識配列(たとえば、ENLYFQG(配列番号70)またはENLYFQS(配列番号71))、またはそれらのいずれかの組合せを含んでいてもよい。このようなプロテアーゼ認識部位をコードするヌクレオチド配列は、当該分野において公知である標準的な分子生物学的手法を使用して本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸内に操作されていてもよい。
いくつかの態様において、本発明は、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体を作製する方法を提供する。本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、当該分野において公知である任意の適切な手段によって作製できる。いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、組換え手段によって作製できる。いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質および/もしくはタンパク質複合体、またはそれらのいずれかの部分は、化学合成手段によって作製できる。たとえば、ここで記載されているタンパク質またはタンパク質複合体の一部に対応するペプチドは、ペプチドシンセサイザーの使用によって合成できる。
本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体が組換え手段によって作製される態様において、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体をコードする核酸は、限定されないが、哺乳動物細胞、トリ細胞(たとえばEB66アヒル細胞)および昆虫細胞(たとえばバキュロウイルス発現系を使用したSF9またはHi5細胞)を含む、任意の適切な細胞種類において発現できる。核酸分子からポリペプチドおよびタンパク質を発現する方法は慣用であり、当該分野において周知であり、当該分野において公知である任意の適切な方法、ベクター、系および細胞種類を使用できる。たとえば、典型的に、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体をコードする核酸配列は、適切なプロモーターを含有する適切な発現構築物内に置かれ、次いでそれは発現のために細胞に送達される。
いくつかの態様において、キメラタンパク質/融合タンパク質を産生するため、たとえば、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体の分析および/または単離および/または精製を容易にするために、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体にキメラドメインを付加することが好適であってもよい。いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、リーダー配列、前駆体ポリペプチド配列、分泌シグナル、局在化シグナル、エピトープタグ、プロテアーゼ切断部位などを含んでいてもよい。使用され得るエピトープタグには、限定されないが、FLAGタグ、グルタチオンS−トランスフェラーゼ(GST)タグ、緑色蛍光タンパク質(GFP)タグ、ヘマグルチニンA(HA)タグ、ヒスチジン(His)タグ、ルシフェラーゼタグ、マルトース結合タンパク質(MBP)タグ、c−Mycタグ、プロテインAタグ、プロテインGタグ、ストレプトアビジン(strep)タグなどが含まれる。
いくつかの態様において、ここで記載されているインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、翻訳後修飾を付加もしくは除去することによって、化学修飾もしくは付加物を付加もしくは除去することによって、および/または当業者に公知である任意の他の修飾を導入することによって変化されていてもよい。翻訳または合成の間または後に、たとえば、グリコシル化(または脱グリコシル化)、アセチル化(または脱アセチル化)、リン酸化(または脱リン酸化)、アミド化(または脱アミド化)、ペグ化、公知の保護/遮断基による誘導体化、タンパク質分解による切断によって、または当該分野において公知である任意の他の手段によって修飾されるインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は本発明の範囲内に含まれる。たとえば、いくつかの態様において、インフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、臭化シアン、トリプシン、キモトリプシン、パパイン、V8プロテアーゼ、NaBH4、アセチル化、ホルミル化、酸化、還元による化学的切断、ツニカマイシンの存在下での代謝合成などに供されていてもよい。いくつかの態様において、このような翻訳後修飾は、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体を、より免疫原性、より安定、ならびに/または中和および広域中和抗体とより結合できるか、もしくはその産生をより誘発できるようにするために使用されていてもよい。

いくつかの態様において、本発明は、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体のいずれかを含む組成物を提供する。いくつかの態様において、このような組成物は、免疫原性組成物、ワクチン組成物および/または治療組成物であってもよい。いくつかの態様において、このような組成物は対象に投与されていてもよい。いくつかの態様において、ここで記載されているインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体は、ウイルス様粒子または「VLP」に存在していてもよい。
いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、研究手段として、診断手段として、治療剤として、抗体試薬もしくは治療抗体の産生のための標的として、および/またはワクチン組成物のワクチンもしくは成分として有用であってもよい。たとえば、いくつかの態様において、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、動物対象、たとえばヒトを含む哺乳動物対象におけるワクチン免疫原として有用である。本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体のこれらおよび他の使用はより完全に以下に記載されている。当業者は、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体が、様々な他の適用にも有用であってもよく、全てのこのような適用および使用は本発明の範囲内に含まれることを意図することを理解する。
一態様において、本発明のインフルエンザポリペプチド、タンパク質およびタンパク質複合体は、たとえばELISAアッセイ、Biacore/SPR結合アッセイ、および/または当該分野において公知である抗体結合についての任意の他のアッセイにおいて、抗HA抗体の結合および/または力価をアッセイおよび/または測定するための検体として有用であってもよい。たとえば、本発明のインフルエンザHAポリペプチド、タンパク質およびタンパク質複合体は、抗HA抗体の有効性を分析および/または比較するために使用されていてもよい。

いくつかの態様において、本発明は、本発明のインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体(またはこのようなインフルエンザHAポリペプチド、タンパク質および/またはタンパク質複合体を含む組成物)を対象に投与することを含む方法を提供する。このような方法は、インフルエンザウイルスを有する個体を処置するための方法(すなわち治療的方法)および/または将来のインフルエンザウイルス感染に対して個体を防御するための方法(すなわち予防的方法)を含んでいてもよい。
本発明は、本発明のインフルエンザHAポリペプチド、タンパク質もしくはタンパク質複合体、またはこのようなポリペプチド、タンパク質もしくはタンパク質複合体を含有する組成物を含むキットをさらに提供する。本発明の方法および組成物の使用を容易にするために、ここで記載されている成分および/または組成物のいずれか、ならびに実験もしくは治療もしくはワクチン目的に有用な追加の構成成分がキットの形態で充填されていてもよい。典型的に、キットは、上記構成成分に加えて、たとえば、構成成分、パッケージング材料、容器および/または送達装置を使用するための指示書を含んでいてもよい追加材料を含有する。

米国および世界の全住民は汎発性インフルエンザ発生の危険にさらされ続けており、兵器化されたインフルエンザウイルスは細菌戦争/テロリズムによる大きな脅威であり続けている[23、24]。HAの免疫優勢頭部の代わりに、保存されたストークQNEに対してAb反応を誘発できるインフルエンザウイルスHAに基づいたワクチン免疫原は、同種(H1N1)、ならびに同種ドリフトバリアント、群1異種(H5N1)、および群2異種チャレンジ(H3N2)から防御できる広域中和抗体を生じさせることが予想される。したがって、単一のユニバーサル免疫原は、季節性、汎発性および兵器化されたインフルエンザウイルスに対する防御免疫反応を誘発できる。インフルエンザウイルスが全住民に対して与える商業的および公衆衛生への影響により、今日の米国における生命保険会社が1918年のスペイン風邪大流行の再発の可能性に対して資本を保持する必要があるという事実が強調される(Oliver,Wyman,& CO,2012 & [25])。ここで記載されているアプローチは、大量のワクチン産物を貯蔵でき、各々の新たな脅威に対処するために必要とされる製造の際の時間の増加に由来する現実の脅威を取り除く広域防御インフルエンザワクチンを提供する可能性を有する。
天然の可溶性および組換えHA三量体を生成し、三量体のストークにおける三量体形成相互作用を共有結合により架橋するために標的ジチロシン(DT)「ステープル」を適用することによって、DT安定化HA三量体が、抗原プロファイルを完全に保存した状態で操作される。HAストーク領域における三量体の共有結合安定化はストークの四次構造を安定にするように操作され、これにより、ストークのQNEを保存しながら、後の頭部のタンパク質分解による除去が可能となる。DT結合は、タンパク質/複合体が完全に折り畳まれた後、複合体を安定化させるために導入され、それにより、タンパク質の構造的機能的完全性を維持しながら、天然構造を固定する[16〜18]。これらの安全で不可逆的なゼロ長の架橋は、非常に近い構造的近接性でTyr残基の間のみに形成し、タンパク質の構造を歪めない。それらはどれもジスルフィド結合で観察されるような非特異的凝集体形成を引き起こさない[17、19−22]。標的DT架橋結合手法は可溶性HA三量体をその正確に折り畳まれた構造に共有結合により安定化させるために適用されていてもよく、次いでそれが実際に鍵となるQNEを提示しているかどうかを決定できる。その後、免疫優勢頭部は、アミノ酸変化に対する可変ループ許容度およびHAのトランスポゾンに基づいた突然変異分析から集めた情報を使用して、配列特異的プロテアーゼ切断部位を導入することによって除去できる。頭部のないHA上でのQNEの提示は、ドリフトバリアントおよび異種ウイルスを用いた致死チャレンジ研究における防御の広さにより改良すると予想される。HIVにおける本発明者らの以前の研究は、高度にグリコシル化された多量体(たとえばHIV Env)が、関連する四次構造および抗原性を維持しながら、切断されたEnv三量体内の種々の位置におけるDT架橋によって一緒に効果的に固定され得ることを示している。
HIVエンベロープスパイクは、その基部における十分に特徴付けられた相互作用およびスパイクの先端における相互作用を介して三量体形成される[33、34]。スパイクの先端における三量体形成相互作用を安定化させるために、チロシン置換基を導入し、タンパク質を発現させ、精製し、DT架橋した。蛍光により、7つのバリアントは、任意の最適化前に、DT特異的励起(320nm)および発光(405nm)波長を使用して定量して平均80%+の効率で分子間三量体形成架橋を形成することを特定した。立体構造的および三量体特異的bnAbに結合するこれらの構築物の能力をアッセイした。DT架橋結合は、プロトマーと三量体の両方に結合する、抗CD4結合部位bnAb b12、および優先的に三量体と結合するが、単量体にも結合する抗V2 bnAb PG9の結合を十分に保存する。さらに、立体構造的固定はまた、ELISAアッセイにおいて非中和mAb、たとえばb6およびb13との結合を顕著に減少させる。DT結合の位置はDT−Env三量体のトリプシン断片のMS/MSによって確認した。より重要なことには、立体構造的に固定したHIV Env三量体は、WTプロトマーと比較して、非常に広域に中和し、有効な抗HIV Env bnAb、PG16のうちの1つと顕著に良好に結合することが見出され;PG16エピトープは天然/機能的HIVエンベロープ三量体上にのみ存在する[28]。改良されたPG16結合は、β−シートとしてPG16が結合する重複しているエピトープのα−ヘリカル配座異性体と結合する、不十分な中和抗V2mAb、CH58との結合の際の顕著な減少と相関する。このDT固定された可溶性HIV Env三量体を用いた次の工程は、それを動物免疫原性実験において試験することである。
DT固定したHA三量体からのHA頭部ドメインのタンパク質分解による除去は、基質特異的プロテアーゼ(たとえばTEV)のためのHA1頭部ドメイン内に認識モチーフを操作することを必要とする。トランスポゾンに基づいた突然変異誘発スクリーニングを使用して、PR8 HA1球状頭部内の4つの領域は、操作されたTEVプロテアーゼ部位とほぼ同じ大きさの外来配列の挿入を許容することが特定されている。さらなる最適化を用いずに、これらの領域のうちの2つ(アミノ酸残基128および223に位置する)は、PR8球状頭部における4つの主要な抗原部位のうちの3つ(Sa、CaおよびSb部位)のタンパク質分解による切断を可能にする[36]。残りのCb部位も除去される。HA1におけるこれらの部位に挿入を有するウイルスは融合可能なままであるので、HA複合体は機能的にインタクトなままである。次いでタンパク質分解反応が実施される。
実験設計。WT HAおよび上記のバリアントの可溶性形態(たとえば、膜貫通ドメインを欠き、T4フォルドン三量体形成モチーフを保有している)は、分泌タンパク質としてSF9またはHi5細胞において発現され、十分に確立された方法によって精製される[37〜38]。Tyrへの置換およびDT架橋の抗原効果は、抗HAストーク広域中和mAbのパネル(たとえば6F12、C179、CR6261、F10、A66およびD8)を使用してELISAにおいて決定される。なぜなら、Tyrへの置換によって引き起こされる構造変化は、これらの抗体のいくつかとの結合を減少または増加させることができるからである。方法:完全曲線結合アッセイは、WT HAを非架橋および架橋HAバリアントと比較する。結合の変化を、結合曲線の非線形回帰分析(Graphpadソフトウェア)を使用して決定して、各mAbを有する各構築物についてEC50値を算出し、比較する。分子間結合形成は、還元SDS−PAGEにおけるゲルシフトにより確認され(ウェスタンブロット/クマシー;DT結合は減少しない);DT架橋結合は上記のように蛍光分光分析によって定量される。このような方法は、分子間DT結合を形成し、操作されたインフルエンザ免疫原を架橋結合した後、野生型PR8 HAと同等の鍵となる抗ストーク四次bnAbとの結合を保持するHAバリアントを産生するために使用できる。
PreScissionプロテアーゼ認識配列(LEVLFQGP(配列番号69)(QとG残基との間の切断)および/またはTEV認識配列(ENLYFQG(配列番号70)(QとG残基との間の切断)およびENLYFQS(配列番号71)(GとS残基との間の切断))を規定された(たとえばアミノ酸残基128および223)またはさらなる位置に挿入して、発現され、精製され、完全に折り畳まれ、DT安定化された可溶性HA前駆体であるバキュロウイルスからHAの球状頭部の大部分を除去できる。抗原確認後、アミノ酸分析および質量分析を実施して架橋分子を生化学的に特徴付けることができる。
PR8 HAバリアントは、mg定量で発現させ、DT架橋し、タンパク質分解し、精製し、抗原的に特徴付けることができる。PR8、NL09およびVN04 HALO/PR8_6+2変異ウイルス調製物を作製できる。チャレンジウイルスの各々についてLD50を確立するために、各ウイルスについて公表されたLD50付近で各群について示されるウイルスの10倍希釈物を使用して、各ウイルスについて4C57BL/6マウス(雌、6〜8週齢(Charles River Laboratories)の4つの群に接種できる。5XのLD50用量の同種(PR8)チャレンジから動物の80%+を防御する精製したDT固定した頭部のないHA三量体免疫原の最適用量を確立するために、5C57BL/6マウス(雌、6〜8週齢(Charles River Laboratories)の4つの群を、一定量のPoly I/Cアジュバント(10μg)と共に様々な量の精製したDT−頭部のないHA免疫原の連続注射からなるプライムブースト戦略により免疫化できる。簡潔に述べると、各群は、アジュバントとしてPoly I/Cと共に製剤化した0μg、2.5μg、5μgおよび10μgのDT固定した頭部のない三量体により免疫化できる。3週間後、各々等量のアジュバント免疫原によりマウスを追加免疫できる。追加免疫の3週間後、それらを、5XLD50用量の同種(PR8)インフルエンザウイルスにより鼻腔内でチャレンジできる。マウスは、適切な時間、たとえば14日、罹患率および死亡率についてモニターでき、評価できる。それらの初期体重の25%超を損失したマウスは屠殺し、死亡とスコア付けできる。生存は25%未満の体重損失として定義できる。ドリフトバリアントおよび群1異種チャレンジに対する防御について免疫化マウスを試験するために、C57BL/6マウスの3つの群は10μgのPoly I/Cアジュバントのみ(「アジュバントのみ」対照群)により免疫化でき、残りの3つの群は上記に特定した最適用量のアジュバントDT−頭部のないHA免疫原により上記のスケジュールに従って免疫化できる(「DT固定した頭部のない三量体」群)。

頭部のないHAの操作における従来の試みは、球状頭部領域が組換えにより切り出されたHAタンパク質を発現させることを含んでいた。このような従来の頭部のないHA構築物は当該分野においてかなりの反響を引き起こした。なぜならそれらは改良された交差反応性Ab反応を誘発したからである。しかしながら、これらのAbは交差防御せず、同種チャレンジに対して防御しただけであった。これらの従来の組換え頭部のない構築物は交差防御の立体構造的ストークAbの現在のレパートリーと結合せず、このことは、インタクトな球状頭部の非存在下で少なくともある程度のストークミスフォールディングを示唆している。これらの従来の観察は、最も広域に交差反応する抗ストークAbの1つであるC179を使用して、免疫蛍光分析によって確認された(図6を参照のこと)。DTに基づいた立体構造的固定の適用は、頭部のタンパク質分解による除去の前にストーク三量体をその天然の立体構造に一緒に保持することによってこの欠点を回避し、それにより決定的なストークQNEに対するAb反応に焦点を当てるDT固定された頭部のないHA免疫原を生じる。
DT架橋をPR8 HAストークドメイン内に導入すると、DT架橋HA三量体は天然の抗原性を維持した。cr6261 bnAb(pdbファイル:3GBN)との複合体における1918 H1N1 HA三量体、およびPR8 HA(pdbファイル:1RU7)の結晶構造に基づいて、DT架橋形成を可能にするためにチロシンへの置換をHAストークドメイン内に首尾良く操作した。そのDT架橋形成は、球状頭部がタンパク質分解により除去されると、四次抗原を維持するはずである。続いて293T細胞にチロシンへの変異体の分泌バリアントをトランスフェクトし、トランスフェクトした細胞上清中で405nm蛍光にて測定してDT結合の形成を決定した。405nm蛍光(DT結合に非常に特異的である)の大きな増加により、いくつかのチロシンへの変異体において強固な架橋結合が実証される(図7A)。陽性対照(インスリン)およびDT標準との比較に基づいて、70%超の架橋結合効率を、任意の最適化前に、これらの構築物のうちの4つについて確認した。図7Bに示されるように、C179 Ab結合は架橋結合反応の前後で変化していない。これらのデータにより、PR8 HAストークが架橋できること、および最も広域の交差反応性立体構造的mAbの1つである、C179(2)が結合する鍵となる四次ストークペプチドが維持されることが示される。
導入:立体構造的に固定された頭部のないHAを設計する際に、1918 HA:cr6261複合体(PDB:3GBN)およびPR8 HA(PDB:1RU7)の原子構造を分析して、1)ストークbnAb結合を維持するためにcr6261エピトープから十分な距離でストークにおいてジチロシン(DT)架橋を可能にし、2)頭部を除去するために使用できるプロテアーゼ切断部位の挿入を可能にする位置を特定した。
いくつかのチロシン変異を、三量体が高い効率でそれらの天然の融合前状態に固定できるようにPR8 HAのストーク内に操作した。図85Aは、DT架橋結合後の三量体状態への明確なシフトを実証しており(還元SDS−PAGE);図85Bは、ジ−チロシン結合が405nmにおける特異的な蛍光によって形成していることを確認している。架橋種の濃度測定により、三量体状態への80%超の変換が実証される。最も重要なことには、8D4、ストーク特異的bnAbの結合が完全に維持される(図85C)。結晶学的分析により、8D4がcr6261と同じエピトープに結合することが示されている。これらのデータにより、PR8 HAが、鍵となるVH1−69四次ストークエピトープの天然の立体構造を維持しながら、そのストークにおいて架橋され得ることが確認される。
HAの機能を破壊せずにTEVプロテアーゼ切断部位が挿入され得るPR8 HAの頭部内の領域を特定した。構造に基づいた設計に関して、PR8 HAおよびTEVプロテアーゼ認識部位の構造データを合わせ、切断部位挿入を以下の基準に基づいてHAの頭部の領域内に特異的に標的化した:i)免疫優勢頭部の除去を最大化するためにストーク先端への近接性;ii)構造摂動を最小化するためにHAの二次構造とTEV切断部位との間の類似性;iii)トランスポゾンに基づいた突然変異誘発スクリーニングからのデータによるトランスポゾン変異誘発スクリーニングに基づいた挿入に対する許容度として特定される領域(Heaton and Palese PNAS Vol.110,No.50;pp.20248−53)。

配列番号1のアミノ酸残基229〜519と少なくとも65%の配列同一性を有するアミノ酸配列から本質的になるインフルエンザヘマグルチニン(HA)ポリペプチドであって、前記アミノ酸配列が、アミノ酸位置403、406、411、422、429、432、433および435またはそれらに対応するアミノ酸残基のうちの1つ以上においてチロシンへの点変異を含む、インフルエンザヘマグルチニン(HA)ポリペプチド。

前記ポリペプチド、タンパク質またはタンパク質複合体が、三量体ストーク立体構造に折り畳まれ、少なくとも1つのジ−チロシン架橋を含み、前記少なくとも1つのジ−チロシン架橋の一方または両方のチロシンがチロシンへの点変異から生じている、[1]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

前記ポリペプチドが、三量体ストーク立体構造に折り畳まれるインフルエンザHAタンパク質複合体により構成され、前記インフルエンザHAタンパク質複合体が少なくとも1つのジ−チロシン架橋を含み、前記少なくとも1つのジ−チロシン架橋の一方または両方のチロシンがチロシンへの点変異から生じている、[3]に記載のインフルエンザHAポリペプチド。

前記架橋が1つ以上の対合したチロシン残基の間に位置し、前記対合したチロシン残基が、残基403および433;411および422、403および429、403および432、433および435、ならびに406および433からなる群から選択される、[4]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

前記架橋が1つ以上の対合したチロシン残基の間に位置し、前記対合したチロシン残基が、残基403および433;411および422、403および429、403および432、433および435、ならびに406および433からなる群から選択される、[5]に記載のインフルエンザHAタンパク質複合体。

前記インフルエンザHAポリペプチド、タンパク質またはタンパク質複合体が、配列番号3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、86、87、88、89、90、91、92、93、96、97、98、99、100、101、102、103、104、105、106、107、108、109、110もしくは117のアミノ酸配列、またはこれらの配列のいずれかと65%以上の配列同一性を有するアミノ酸配列を有する、[1]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

前記ポリペプチド、タンパク質またはタンパク質複合体が、HA−ストーク特異的抗体と結合できる、[1]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

前記ポリペプチド、タンパク質またはタンパク質複合体が、抗体C179と結合できる、[9]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

前記ポリペプチド、タンパク質またはタンパク質複合体が、三量体ストーク立体構造に折り畳むことができる、[1]に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。

Claims (15)
- 配列番号1のアミノ酸残基292〜519と少なくとも70%の配列同一性を有するアミノ酸配列を含むインフルエンザヘマグルチニン(HA)ポリペプチド、タンパク質またはタンパク質複合体であって、前記アミノ酸配列がチロシンへの少なくとも2つの点変異を含み、前記2つの点変異は、アミノ酸位置が(a)403および433、(b)411および422、(c)403および429、(d)403および432、(e)433および435、(f)406および429、または(g)406および433であるか、または配列番号1とのアラインメントによりそれらに対応するアミノ酸残基であり、かつ、前記ポリペプチド、タンパク質またはタンパク質複合体が、インフルエンザHA三量体ストークの一部を形成し、および/または、インフルエンザHA三量体ストークを有する、または、形成できるタンパク質複合体に折り畳まれる、インフルエンザヘマグルチニン(HA)ポリペプチド、タンパク質またはタンパク質複合体。
- 配列番号1のアミノ酸残基292〜519と少なくとも70%の配列同一性を有するアミノ酸配列から本質的になる頭部のないインフルエンザヘマグルチニン(HA)ポリペプチドであって、前記アミノ酸配列がチロシンへの少なくとも2つの点変異を含み、前記2つの点変異は、アミノ酸位置が(a)403および433、(b)411および422、(c)403および429、(d)403および432、(e)433および435、(f)406および429、または(g)406および433であるか、または配列番号1とのアラインメントによりそれらに対応するアミノ酸残基であり、かつ、前記ポリペプチドが、インフルエンザHA三量体ストークの一部を形成し、および/または、インフルエンザHA三量体ストークを有する、または、形成できるタンパク質複合体に折り畳まれる、頭部のないインフルエンザヘマグルチニン(HA)ポリペプチド。
- 前記ポリペプチド、タンパク質またはタンパク質複合体が、少なくとも1つのジ−チロシン架橋を含むインフルエンザHA三量体ストークの中にあるか、もしくはそれを含む、請求項1に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。
- 請求項2に記載の頭部のないインフルエンザHAポリペプチドであって、前記ポリペプチドが、少なくとも1つのジ−チロシン架橋を含むインフルエンザHA三量体ストークの中に含まれる、頭部のないインフルエンザHAポリペプチド。
- 前記少なくとも1つの架橋が、(a)残基403および433;(b)411および422、(c)403および429、(d)403および432、(e)433および435、(f)406および429、ならびに(g)406および433からなる群から選択される対合したチロシン残基の間に位置する、請求項3に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体、または請求項4に記載の頭部のないインフルエンザHAポリペプチド。
- 前記ポリペプチド、タンパク質またはタンパク質複合体が、HA−ストーク特異的抗体と結合できる、請求項1に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体、または請求項2に記載の頭部のないインフルエンザヘマグルチニン(HA)ポリペプチド。
- 前記ポリペプチド、タンパク質またはタンパク質複合体が、抗体C179と結合できる、請求項6に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。
- 前記ポリペプチド、タンパク質またはタンパク質複合体が三量体形成ドメインをさらに含み、任意に前記三量体形成ドメインがフォルドンドメインである、請求項1に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体、または請求項2に記載の頭部のないインフルエンザヘマグルチニン(HA)ポリペプチド。
- 配列番号1のアミノ酸残基292〜519と少なくとも80%の配列同一性を有するアミノ酸配列を含む、請求項1に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体、または請求項2に記載の頭部のないインフルエンザヘマグルチニン(HA)ポリペプチド。
- 前記ポリペプチド、タンパク質またはタンパク質複合体が、配列番号3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、86、87、88、89、90、91、92、93、96、97、98、99、100、101、102、103、104、105、106、107、108、109、または110のアミノ酸配列を含む、請求項1に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。
- 請求項2に記載の頭部のないインフルエンザHAポリペプチドであって、前記ポリペプチドは、配列番号96、97、98、99、100、101、102、103、104、105、106、107、108、109、もしくは110のアミノ酸配列を含む、頭部のないインフルエンザHAポリペプチド。
- 配列番号96、97、98、99、100、101、102、103、104、105、106、107、108、109、および110からなる群から選択されるアミノ酸配列を含む、インフルエンザHAポリペプチド、タンパク質またはタンパク質複合体であって、前記ポリペプチド、タンパク質またはタンパク質複合体が、インフルエンザHA三量体ストークの一部を形成し、および/または、インフルエンザHA三量体ストークを有する、または、形成できるタンパク質複合体に折り畳まれる、インフルエンザHAポリペプチド、タンパク質またはタンパク質複合体。
- 請求項1から12のいずれか1項に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体をコードする核酸分子。
- 請求項1から12のいずれか1項に記載のインフルエンザHAポリペプチド、タンパク質またはタンパク質複合体を含む組成物。
- 前記組成物が、アジュバント、担体、免疫刺激剤またはそれらのいずれかの組合せをさらに含む、請求項14に記載の組成物。
