Description
受動液滴融合
幾何学的に介在された受動液滴融合は、体積拡張部における連続相の排除に基づく(図1)。この方法は、液滴間の汚染が少ないという利点を特徴とするが、追加的に設計が複雑になる流路を必要とし、能動誘導法よりもスループットが低い。
能動液滴融合法は、その多くが液滴を融合させるために、電極の製造及び電気信号の正確なタイミングを必要とするため、多くの受動液滴融合法よりも複雑である。これらの方法は、液滴の合体に必要な表面張力の不安定性を生じさせるために電気を用いる。最も有利な設計の1つは、図4に示されたピコインジェクターである。液滴がピコインジェクターの近くを通過する際に、電極が活性化され、試薬を液滴に注入することができる水/油界面の薄膜不安定性を発生させる。高いスループット(数キロヘルツ)及びピコリットル以下の注入精度があると主張された。本方法の主な不便な点は、通過する液滴との融合の最中に、注入溶液リザーバーの汚染の可能性があることである。電気の使用は、DNAやたんぱく質などの生物学的分子への電気信号の生物学的適合性が問題となる。
特許文献1は、連続相と、反応体液滴と、スペーサーとを含む3相マイクロ流体システムを開示する。スペーサーは、反応体液滴の合体を防ぐ不混和性液体または疎水性粒子によって形成される。この出願はまた、T字型接合マイクロ流体デバイスを用いた注入によって試薬液滴を追加することを開示しているが、どのように注入を行うかについては具体的に説明していない。特に、この出願は、合体前に試薬液滴を分けることについて教示も示唆もしていない。
(a)マイクロ流体またはミリ流体システムの流路内で、連続相と、少なくとも2つの反応体液滴と、2つの反応体液滴AnとAn+1との間の少なくとも1つのスペーサープラグと、を含み、nが0以上の整数である一連の3相マイクロ/ミリ流体を発生させる段階と、
(b)インジェクターと呼ぶ側面インレットによって、一連の流体内に、非濡れ性の単一の液滴B内に試薬を注入する段階であって、液滴Bが反応体液滴An+1に直接先行するスペーサープラグの注入点を通過する間にその場で生成され、反応体液滴An+1が注入点に到達する前に、液滴Bが注入点から分離してインジェクター端部において試薬流体のメニスカスが形成され、反応体液滴An+1が注入点にあるときに試薬流体メニスカスが側面インレット内に収縮する、試薬を注入する段階と、
(c)液滴Bを注入点から離れた位置で反応体液滴An+1と融合または接触させる段階と、を含み、
(d)n>1の場合に、段階(b)及び(c)を反復する。
・連続相は、反応体液滴の流体及び試薬液滴の流体と実質的に混合しない。
・スペーサー流体は、連続相、反応体液滴の流体及び試薬液滴の流体と実質的に混合しない。
i)その場にとどまっている、すなわち試薬リザーバーと依然として接続され、制御された体積を有する試薬液滴の発生(段階b)
ii)注入点からの試薬液滴の離脱(段階b)

例1:一連の3相ミリ流体の生成
一連の3相は、連続相(フッ化物オイル Novec HFE7500+0.5%v/v パーフルオロオクタノール界面活性剤)と、反応体液滴(ペプトン−グルコース−塩媒体、pH4.6)と、スペーサー流体(空気または鉱物油)と、からなる。一連の流体は、T接続を使用して、750umのID FEPテフロン(登録商標)管内に生成され、液体の流速は、シリンジポンプ及び圧力制御部による気体の圧力で制御される。一連の空気のスペーサーでは、フッ化物オイル及び媒体の流速はそれぞれ6.5及び7.5ml/hであり、空気の圧力は1Barである。一連の鉱物油スペーサーについては、フッ化物オイル、媒体及び鉱物油の流速はそれぞれ、5、5、及び10ml/hである。

2. 反応対象物液滴の注入(メチレンブルー水溶液またはアセテートバッファ(pH4.5)溶液+BodipyFL) 本方法の要素は図8に示されている。
段階:
a.反応体液滴の蛍光信号検出。
b.(管内に穿孔された穴を通した)反応体液滴の前方への反応対象物液滴の注入。注入は、アクリル系マニホールド(Lee社製マニホールド取付けキット)によって、穿孔された穴に整列されたソレノイドバルブ(Lee社製VHS型)によって行われる。画像取得による評価(GuppyProファイアーワイヤーカメラ及びNikon10倍対物レンズ)。図6a。
c.反応対象物液滴の穿孔された穴への付着。図6a。
d.同期:穿孔された穴から連続相毛細管ブリッジ(可動受動融合チャンバー)内への試薬液滴の離脱。画像取得による評価。図6b。
e.可動受動融合チャンバー上における、試薬液滴及び反応体液滴の接触。画像取得による評価。図6c。
f.可動受動融合チャンバーの連続相の排出。画像取得による評価。図6c。
g.液滴融合。画像取得及び蛍光信号モニタリングによる評価。図6d。
スペーサー流体として空気(図6)及び鉱物油を有する一連のミリ流体での試薬液滴注入試験は、自己同期形式の4つの典型的な段階、試薬液滴の発生、試薬液滴のノズルからの離脱、液滴の接触及び連続相の排出、並びに液滴の融合を示した。
Aspergillus nigerの変異株の胞子が、0.000375のODを得るための抽出後に、PGS媒体内で希釈される。この濃度により、PGS媒体反応体液滴内への単一の胞子の閉じ込めが可能となる。空気スペーサーを有する、3105個の反応体液滴の一連のミリ流体が、30mの長さのFEPテフロン(登録商標)管(750um ID)内で生成された。一連の流体の生成について、(0.5% v/v パーフルオロオクタノール界面活性剤を有する)Novec7500フッ化物オイル及びPGS媒体の流速は、それぞれ6.5及び7.5ml/hであり、空気の圧力は1Barである。一連の流体は、一定の圧力(250mBar)を印加することによって前後に循環する。一連の流体の全てがレーザー液滴検出器を通過した後、一連の液滴の流動方向は自動的に切り替えられる。30℃における菌の成長の30h後の酵素活性を決定するために、5nLのBodipy FL液滴が、本発明のモジュールを用いて、一連の流体の循環のために250mBarの圧力、試薬液滴追加のために300mBarの圧力を印加し、50μmの半径の穿孔された穴について、ソレノイドバルブ解放のための3msパルスを発生させて、一連のミリ流体の各液滴内に注入される。液滴追加と蛍光検出との間の45秒の遅延は、追加モジュールと蛍光モジュールとが30cmだけ離れて位置することによって得られる。この遅延により、多数の先行試験によって示されるように、酵素活性曲線の線形区間内に各液滴の単一のデータ点を確実に集めることができる。追加された一連の流体の蛍光信号が、図9b(拡大図)に示されている。この曲線において、ベースラインは、空の液滴の信号に対応し、ピークは菌で占められた558個の液滴のアミラーゼ活性による蛍光に対応する。
蛍光により、追加された液滴の体積を決定するために、蛍光色素濃縮液滴が、スペーサーとして気泡を有する希釈された蛍光色素液滴で形成された一連のミリ流体内に追加される。一連のミリ流体発生条件は、前述の例1で示されたものと同一である。ソレノイドバルブの作動は、蛍光による液滴検出に基づく。FITC蛍光色素 1E−04M液滴が、本発明のモジュール(穿孔された穴の半径:50μm)を用い、ソレノイドバルブ(Lee社製VHS型)の開放の10msパルス、一連の流体の循環のための150mBarの圧力、及び試薬液滴追加のための150から300mBarの可変圧力を印加して、一連のミリ流体(FITC蛍光色素 1E−06M液滴)の各液滴内に注入される。一連のミリ流体への注入の完了後、流動は追加された液滴の蛍光信号を記録するために反転される。注入された一連の流体の蛍光信号は図13に示されており、各ステップは試薬液滴追加の異なる圧力に対応する。圧力の増加とともに試薬液滴の体積の増加が図14において見られ、これは取得されたビデオのスナップショットに対応する。質量バランスは、ターゲット液滴体積の試薬液滴体積に対する比(VT/VR)の定量化を可能にし、VT/VR=(CF−CT)/(CR−CF)で表され、CF、CT及びCRは、追加された液滴、ターゲット液滴及び試薬液滴の濃度である。蛍光色素の濃度と蛍光ピークの高さとの間の比が与えられると、CF及びCTは、追加された液滴及びターゲット液滴についての蛍光ピークの高さに対応する。CR/CT=100では、各ステップについての平均ピーク高さの値を用いて、(VT/VR)についての計算値は、図15に印加された圧力の試薬液滴追加の関数としてプロットされる。98から16のVT/VRの比が、実験した圧力範囲で観察され、幅広い体積の範囲に渡って、試薬液滴を生成するための開発されたモジュールの柔軟性を明らかにした。この特徴は、マイクロ及びミリ流体システム内の濃度勾配の生成についての潜在的な用途を有している。
注入システムにキャパシタを導入することによるメニスカス収縮の増加を行うために、気泡が試薬リザーバー内に導入される。試薬液滴形成の際の離脱後、メニスカスの位置がビデオ取得によって記録された。図16は、2.5ml/hで流れるフッ化物オイルNovec7500+0.5% v/v パーフルオロオクタノール界面活性剤の流動における、50μmの半径の穴を通過する、連続的な試薬流動下における最大メニスカス収縮(水、印加圧力100mBar)を示している。同一の動作条件を維持して、気泡がない状態で行った平行試験(図16、左)と比較すると、メニスカス収縮は気泡が存在する場合の方が大きい(図16、右)。この効果は、試薬液滴が分離した後の気泡の圧縮性によって圧力の変化が大きくなることに起因している。

Claims (15)
- マイクロ流体またはミリ流体デバイスにおける、反応体液滴と試薬液滴とを融合または接触させるための方法であって、
(a)前記マイクロ流体またはミリ流体システムの流路内において、連続相、少なくとも2つの反応体液滴A n 及びA n+1 、並びにnが0以上の整数である2つの前記反応体液滴AnとAn+1との間の少なくとも1つのスペーサープラグを含む一連の3相マイクロ/ミリ流体を発生させる段階と、
(b)インジェクターと呼ぶ側面インレットによって、一連の流体内に、非濡れ性の単一の試薬液滴Bとして試薬流体を注入する段階であって、前記試薬液滴Bが、前記反応体液滴An+1に直接先行する前記スペーサープラグの注入点の通過の際にその場で生成され、前記反応体液滴An+1が前記注入点に到達する前に、前記インジェクター端部に試薬流体のメニスカスを形成しつつ前記試薬液滴Bが前記注入点から離脱し、前記反応体液滴An+1が前記注入点にあるときに前記試薬流体のメニスカスがインジェクターと呼ぶ前記側面インレット内に引き込まれる、段階と、
(c)前記試薬液滴Bが、前記注入点から離れて、前記反応体液滴An+1と融合または接触する段階と、
(d)n>1のときに、段階(b)及び(c)を繰り返す段階と、
を含む、方法。 - 前記試薬液滴Bが、前記注入点に付着した状態を保ち、前記反応体液滴An+1の前方の毛細管ブリッジが前記試薬液滴Bに到達する際に分離する、請求項1に記載の方法。
- rが前記試薬液滴の半径であり、Rが円筒形の幾何形状を有する前記流路の半径であるか、または平面幾何形状を有する流路の幅の半分もしくは高さの半分である場合に、比r/Rが0.1から1の範囲である、請求項1または2に記載の方法。
- 段階(b)において注入される前記試薬液滴Bの体積が、1pLよりも大きい、請求項1から3のいずれか一項に記載の方法。
- dが前記側面インレットの直径であり、Dが円筒形の幾何形状を有する前記流路の直径であるか、またはDが平面幾何形状を有する前記流路の高さもしくは幅である場合に、比d/Dが0.05から0.5の範囲である、請求項1から4のいずれか一項に記載の方法。
- 前記注入が不連続である、請求項1から5のいずれか一項に記載の方法。
- 前記試薬液滴Bがバルブ、特にソレノイドバルブによって作動される、請求項6に記載の方法。
- 段階(b)の前に、前記反応体液滴An+1が、光学的手段、電気的手段、磁気的手段、放射線検出手段、音響手段からなる群から選択された検出手段によって検出される、請求項1から7のいずれか一項に記載の方法。
- 段階(b)がさらに、前記反応体液滴An+1を検出する段階の後であって、段階(c)の前に、検出された前記反応体液滴An+1を選択し、それによって、ヌル、すなわち前記試薬液滴Bが注入されないか、または前記試薬液滴Bの体積が1pLより大きいように、前記試薬液滴Bの体積を調整する段階を含む、請求項1から8のいずれか一項に記載の方法。
- 前記スペーサーが、前記連続相、反応体相、及び試薬相と非混合性である気体または液体である、請求項1から9のいずれか一項に記載の方法。
- 前記スペーサーが、鉱物油などの非混合性液体である、請求項10に記載の方法。
- 前記デバイスがミリ流体デバイスである、請求項1から11のいずれか一項に記載の方法。
- コンビナトリアル化学、化学生物学スクリーニング、遺伝子シーケンシングにおいて使用される、請求項1から12のいずれか一項に記載の方法。
- 連続相、少なくとも2つの反応体液滴、nが0以上の整数である2つの反応体液滴AnとAn+1との間の少なくとも1つのスペーサープラグを含む一連の3相マイクロ/ミリ流体で満たされたミリ/マイクロ流体流路管と、
前記流路管上に開口する開口部であって、前記開口部が液体相の試薬を含むリザーバーと接続された、開口部と、
前記リザーバー及び前記開口部に結合するバルブであって、試薬液滴Bを前記流路管に放出するように構成された、バルブと、
前記流路管上であって、前記開口部の上流側に配置された検出器であって、前記反応体液滴A n+1 が前記開口部に到達する前に、前記試薬液滴Bが前記流路管内に注入されるように、前記検出器が、前記反応体液滴A n+1 を検出し、前記バルブの開放を前記反応体液滴A n+1 の到達と同期させるように構成された、検出器と、を含む、

2つの抵抗R1及びR2が直列接続され、
前記2つの抵抗の間の接続部が、抵抗R3と直列に素子ICに接続され、

R1及びR1が注入点の両側における一連の液滴の流動抵抗に対応し、
R3が試薬リザーバーと前記バルブとの間の流動抵抗に対応し、
R4が試薬流体が注入チャンバーを通過する流動に起因する抵抗に対応し、
C1が注入チャンバー内部のキャパシタンスに対応する、
