本発明は、一態様では、被検者に1つ以上の食事介入を適用することによって達成できる減量の程度を予測する方法に関する。特定の実施形態では、本方法を用いて、被検者の減量する可能性について情報に基づいた予測を行ない、それに応じて1つ以上の食事介入を選択又は調整しても良い。例えば、食事介入が低カロリー食である場合、本方法を用いて、被検者にとって適切な食事を選択すること、又は減量の程度に影響する毎日のカロリー摂取又は特定の食事の期間を調整すること、又は被検者に対する現実的な期待を設定することによって低カロリー食に対する順守を増加させることが可能になる。また本方法を用いて、被検者の生活習慣を変更することを支援しても良い。
好ましくは被検者は哺乳動物、好ましくはヒトである。被検者は代替的に、非ヒトの哺乳動物(例えば、馬、牛、羊、又は豚など)であっても良い。一実施形態では、被検者はコンパニオンアニマル例えば犬又は猫であっても良い。
本発明には、被検者から得られる1つ以上のサンプル中の1つ以上のバイオマーカーの濃度を判定するステップが含まれている。
用語「食事介入」が意味するのは、被検者の食事の変化をもたらす被検者に適用される外部因子である。一実施形態では、食事介入は低カロリー食である。
一実施形態では、1つ以上のバイオマーカーの濃度を食事介入の前に判定する。別の実施形態では、1つ以上のバイオマーカーの濃度を食事介入の前後で判定する。バイオマーカー濃度を、食事介入の全体に渡って所定の時間に判定しても良い。これらの所定の時間は、食事介入の全体に渡って周期的(例えば、毎日又は3日ごと)であっても良いし、又は試験中の被検者、分析中のサンプルのタイプ、及び/又は達成すると予測される減量の程度に依存しても良い。
フルクトサミンは、糖と第1球アミンとの間の糖化反応からもたらされる化合物である。生物学的に、フルクトサミンはフルクトサミン−3−キナーゼによって認識される。
第VII因子は血液凝固タンパク質である。これは、抗血友病因子(AHF)としても当該技術分野で知られている。
アディポネクチンは、ヒトの中でADIPOQ遺伝子によってコード化されるタンパク質である。それは当該技術分野において、GBP−28、apM1、AdipoQ、及びAcrp30とも言われる。
インスリンは、膵臓のベータ細胞によって生成されるペプチドホルモンである。
個々のバイオマーカーは、本発明の方法において予測値を有している場合があるが、本方法の品質及び/又は予測能力は、複数のバイオマーカーからの値を組み合わせることによって改善される場合がある。
本方法には、試験サンプル中の個体バイオマーカーの濃度を1つ以上の基準値又は対照値と比較するステップが更に含まれていても良い。基準値を、食事介入に従って体重を減らす被検者の所定の能力に対応付けても良い。いくつかの実施形態では、基準値は、ある食事介入に従う被検者又は被検者のグループに対して、以前に得た値である。基準値は、食事介入に従う被検者のグループからの平均濃度(例えば、平均又は中央値濃度)に基づいても良い。
一実施形態では、本方法には更に、1つ以上のバイオマーカーの濃度を被検者の1つ以上の身体測定及び/又は生活習慣特性と組み合わせることが含まれる。この情報を組み合わせることによって、改善された予測モデルが被検者が達成できる減量の程度に対して提供される。

本発明の方法によって予測される減量の程度を、1つ以上の所定の閾値と比較しても良い。このような閾値を用いて、被検者を、予測される減量の程度(例えば、低い、中間の、高い及び/又は非常に高い予測される減量の程度)を示すカテゴリに階層化しても良い。閾値から逸脱する程度は、どの被検者が特定の介入から最も利益を得るのかを判定するのに有用である。このようにして、食事介入及び生活習慣の変更を最適化することができ、被検者によって達成される減量の現実的な期待を設定することができる。
更なる態様では、本発明によって、被検者の生活習慣を変更するための方法が提供される。被検者における生活習慣の変更は、本明細書で説明する任意の変更であっても良く、例えば、食事の変更、もっと運動すること、異なる作業及び/又は生活環境などである。
一態様では、本発明によって、減量用の低カロリー食の一部として用いるダイエット食品が提供される。ダイエット食品は、本明細書で説明する方法によってある程度の減量を達成すると予測される被検者に投与される。
更なる態様では、本発明によって、被検者に1つ以上の食事介入を適用することによって達成できる減量の程度を予測するためのキットが提供される。
本明細書で説明する方法を、汎用ハードウェア(例えば1つ以上のコンピュータプロセッサ)上で実行されるコンピュータプログラムとして実装しても良い。いくつかの実施形態では、本明細書で説明する機能を、デバイス例えばスマートフォン、タブレット末端又はパーソナルコンピュータによって実装しても良い。
被検者はDiogenes研究の参加者であった。この研究は、汎ヨーロッパのランダム化比較食事介入研究であり、8つのヨーロッパセンタにおける肥満及び過体重家族における減量及び体重維持に対する食事タンパク質及びグリセミック指数の効果を詳しく調べている(Larsenら,Obesity reviews(2009)、11、76〜91)。

実施例2には、実施例1と同じ被検者が含まれているが、BMI2(低カロリー介入後のBMI)を予測する代わりに、可能性被検者が「減量敏感」又は「減量抵抗」である可能性を予測することに焦点を当てている。
Claims (26)
- 被検者に1つ以上の食事介入を適用することによって達成できる減量の程度を予測するための方法であって、
前記被検者から得られる1つ以上のサンプル中の1つ以上のバイオマーカーの濃度を判定するステップであって、前記バイオマーカーは、フルクトサミン及び第VII因子から選択される、ステップを含む、方法。 - 1つ以上のサンプル中のアディポネクチンの濃度を判定するステップを更に含む、請求項1に記載の方法。
- 1つ以上のサンプル中のインスリンの濃度を判定するステップを更に含む、請求項1又は2に記載の方法。
- フルクトサミン、第VII因子、アディポネクチン及びインスリンのそれぞれの濃度は、判定され、前記サンプル中のフルクトサミン及びインスリンの濃度の低下及び第VII因子及びアディポネクチンの濃度の増加は、前記被検者の減量の程度の増加を示す、請求項1〜3のいずれか一項に記載の方法。
- 前記1つ以上のサンプルは、血液由来である、請求項1〜4のいずれか一項に記載の方法。
- 前記フルクトサミンの濃度は、糖化アルブミンを測定するステップによって判定される、請求項1〜5のいずれか一項に記載の方法。
- 前記食事介入は、低カロリー食である、請求項1〜6のいずれか一項に記載の方法。
- 前記低カロリー食は、約600〜約1200kcal/日のカロリー摂取量を含む、請求項7に記載の方法。
- 前記低カロリー食は、少なくとも1種のダイエット食品の投与を含む、請求項7又は8に記載の方法。
- 前記低カロリー食は、6〜12週間の継続期間を有する、請求項7〜9のいずれか一項に記載の方法。
- 前記1つ以上のバイオマーカーの濃度と、前記被検者の1つ以上の身体測定及び/又は生活習慣特性とを組み合わせるステップを更に含む、請求項1〜10のいずれか一項に記載の方法。
- 前記身体測定は、性別、体重、身長、年齢、及び体格指数からなる群から選択され、前記生活習慣特性は、前記被検者が喫煙者であるか又は非喫煙者であるかである、請求項11に記載の方法。
- 前記減量の程度は、被検者が食事介入の適用によって達成すると予測される体格指数によって表される請求項1〜12のいずれか一項に記載の方法。
- 被検者に対する1つ以上の食事介入を最適化する方法であって、
前記被検者が達成できる減量の程度を、請求項1〜13のいずれか一項に記載の方法により予測するステップと、
前記被検者に前記食事介入を適用するステップとを含む、方法。 - 被検者が食事介入により達成するであろうと期待される体格指数(BMI2)を予測するための方法であって、
a.前記被検者から得られる1つ以上のサンプル中のフルクトサミン、第VII因子、アディポネクチン及びインスリンの濃度を判定するステップと、
b.式(1):

を用いてBMI2を予測するステップと、を含み、

- ユーザから1つ以上のバイオマーカーの濃度が与えられたときに、減量の程度をプログラム可能なコンピュータに予測させるためのコンピュータ実施可能な命令を含むコンピュータプログラム製品であって、前記バイオマーカーは、フルクトサミン及び第VII因子から選択される、コンピュータプログラム製品。
- 前記コンピュータプログラム製品に、前記ユーザから身体測定及び/又は生活習慣特性が更に与えられ、前記身体測定は、年齢、体重、身長、性別、及び体格指数からなる群から選択され、前記生活習慣特性は、前記ユーザが喫煙者であるか又は非喫煙者であるかである、請求項21に記載の製品。
- 前記バイオマーカーには、アディポネクチン及び/又はインスリンが更に含まれる、請求項21又は22に記載のコンピュータプログラム製品。
- 食事介入の後に被検者が達成できる減量の程度を予測するためのキットであって、
a.第VII因子に特異的な抗体と、
