Description
テロメラーゼは、RNA鋳型と、テロメラーゼ活性の主要決定因である「テロメラーゼ逆転写酵素」(TERT)と呼ばれる逆転写酵素を含むタンパク質成分とからなる。特に記載がない限り、本明細書においては、用語「テロメラーゼ」はTERTを指す。
本明細書において、(i)テロメラーゼ触媒活性を失ったイヌテロメラーゼ逆転写酵素(TERT)、または(ii)その断片をコードする配列を含む核酸が提供される。
好ましくは、核酸は、本明細書で定義されるポリヌクレオチド配列、および宿主細胞もしくは宿主生物におけるタンパク質産物の発現(例えば転写および翻訳)を可能にする調節配列(例えば適切な1種以上のプロモーター、1種以上のエンハンサー、1種以上のターミネーター等)を含む遺伝子コンストラクトである。
前記の核酸または免疫原性組成物は、イヌにおける腫瘍の予防または処置のための方法で有用である。
全てのコンストラクトにおいて、TERT配列の上流には、ヒトユビキチンをコードするDNA配列がある。ユビキチンの存在は、TERTタンパク質のプロテアソームへのアドレッシングを高め、かつ得られたTERTペプチドのクラスI提示経路を増加させることとなる。TERT配列の下流には、例えばウェスタンブロットまたは組織化学による融合タンパク質の将来的な精製または検出を容易にするために、インフルエンザタンパク質V5の配列がある。TERTタンパク質をコードするDNA配列は、核小体移入シグナルをコードするN末端領域における47のアミノ酸を欠失している。更に、タンパク質酵素活性を阻害するために、TERTの触媒部位における3つのアミノ酸(VDD)が除去されている。
2.1.材料および方法
細胞培養
トランスフェクションアッセイおよび免疫蛍光実験のために使用されたヒト293T細胞系統は、サイモン・ウェイン−ホブソン教授(パスツール研究所)の好意により提供された。TRAPアッセイのために使用されたCrFK(Crandall−Reeseネコ腎臓)細胞は、J.リチャードソン教授(エコール・ヴェテリネール・ドゥ・メゾン・アルフォール)の好意により提供された。細胞は、37℃、5%CO2で、ダルベッコ変性イーグル培地(DMEM)に10%の熱不活性化されたウシ胎児血清(FCS)、1%のピルビン酸ナトリウム、1%のペニシリン−ストレプトマイシンピルベート、および0.1%のβ−メルカプトエタノールを補充した培地において増殖させた。培養培地の全ての成分は、Life technologies SAS(フランス、サン・トーバン)から購入した。
293T細胞のトランスフェクションは、pCDTまたはpDUV5プラスミドのいずれかでJetPRIME(登録商標)トランスフェクションキット(Polyplus−transfection SA、フランス、イルキルシュ)を使用して製造元の指示に従って実施した。6ウェルプレートにおいて、1ウェル当たり400000個のHeLa細胞または293T細胞を2mLのDMEM培養培地中に撒き、トランスフェクション前に37℃、5%のCO2で24時間培養した。それぞれのウェルにつき、2μgのそれぞれのプラスミドを200μLのjetPRIME(登録商標)バッファー中で希釈したもの、またはそれぞれ4μLのjetPRIME(登録商標)剤だけしか有さない200μLのjetPRIME(登録商標)バッファーを、細胞上に滴下した。トランスフェクション媒体は、4時間後に除去し、そして2mLのDMEM培養培地によって置き換えた。細胞は37℃、5%CO2に置き、そして分析のために24時間後に回収した。
トランスフェクトさせた293T細胞を、プロテアーゼ阻害剤カクテル(完全にEDTA不含、ロシュ・ダイアグノスティックス、米国、インディアナポリス)を含有する放射免疫沈降アッセイ(RIPA)溶解バッファー(RIPAバッファー、Sigma Aldrich chimie SARL、フランス、サン・カンタン・ファラヴィエ)を用いて氷上で10〜20分にわたり溶解させた。次いで、懸濁液を、細胞砕片の除去のために、14000rpmで4℃で15分間遠心分離した。上清を収集し、ブラッドフォード法を使用してタンパク質濃度を測定した。タンパク質試料を、95℃で5分間変性させ、Nu−PAGE(登録商標)Novex 4〜12%Bis−Trisゲル(インビトロジェン、米国、カールスバッド)で分離し、PVDF膜(iBlot(登録商標)transfer stack、インビトロジェン、米国、カールスバッド)へとiBlot(登録商標)装置(インビトロジェン、米国、カールスバッド)を使用して移した。膜は約60kDaのカットであった。まず、上方部の膜を抗V5抗体(インビトロジェン、米国、カールスバッド)でプロービングし、その一方で他の部分は、抗βアクチン抗体(Sigma Aldrich chimie SARL、フランス、サン・カンタン・ファラヴィエ)でプロービングし、次いで試料を、ECL(強化された化学発光)の抗マウスセイヨウワサビペルオキシダーゼ(HRP)結合抗体(GEヘルスケア、フランス、ヴェリジー)によって曝露した。イムノブロットシグナルは、両者ともGEヘルスケア(英国、バッキンガムシア)から購入した製品である18×24のフィルムと相応するカセットを使用して明らかにした。
ヒト293T細胞を、8ウェルのLab−Tek(登録商標)チャンバースライド(Sigma Aldrich chimie SARL、フランス、サン・カンタン・ファラヴィエ)上に200μLの培養培地中で20・103細胞/ウェルで撒き、37℃で一晩インキュベートした。翌日に、培養培地を破棄した。1μgのpCDTまたはPUF2プラスミドのいずれか、50μLのOptiMEM(Life technologies SAS、フランス、サン・トーバン)および2.5μLのFugene HD(Promega France、フランス、シャルボニエール・レ・バン)を含有する10μLの混合溶液を、相応するチャンバーに添加した。コントロールとして、20・103個のHeLa細胞を、プラスミドを含まない同じ混合物10μLと一緒にインキュベートした。チャンバースライドを、インキュベーター中で24時間放置した。トランスフェクトさせた293T細胞を、1×PBSで慎重に洗浄し、そして200μLの2%PFAを各ウェルへと+4℃で10分にわたり加えて、細胞を固定し、そして透過性にさせた。次いでウェルを、0.05%Tween(登録商標)20の1×PBSで2回洗浄し、そして293T細胞を室温で200μLのブロッキング溶液(0.5%TritonX100、3%BSA、10%ヤギ血清)と一緒に30分間インキュベートした。最後に、ウェルを室温で一次マウス抗V5抗体(Life technologies SAS、フランス、サン・トーバン)をブロッキング溶液中で1/200で希釈したものと一緒に僅かに撹拌しつつ1.5時間にわたりインキュベートした。0.05%Tween(登録商標)20の1×PBS中で3回洗浄した後に、二次ヤギ抗マウスAlexa Fluor 488(登録商標)抗体(Life technologies SAS、フランス、サン・トーバン)をブロッキング溶液で希釈(1/500)したものを、それらのウェルの中に入れ、光を避けて僅かに撹拌しつつ室温で45分間置いた。ウェルを、0.05%Tween(登録商標)20の1×PBSで3回洗浄し、そしてウェルにDAPIを含有するVectashield(登録商標)マウンティング培地(Vector laboratories、英国、ピーターバラ)をマウントした。スライドを、画像処理および解析システム(Axiovision、Carl Zeis MicroImaging GmbH、ドイツ、イェーナ)を備えた蛍光顕微鏡(Axio observer Z1、Carl Zeis MicroImaging GmbH、ドイツ、イェーナ)を用いて解析した。
テロメラーゼ活性を、テロメラーゼ活性の定量的測定のための測光酵素イムノアッセイによって、テロメリックリピート増幅プロトコール(TRAP)(Yang et al, 2002)を利用して測定した。CrFK(Crandell Reesネコ腎臓)テロメラーゼ陰性細胞(Yazawa et al., 2003)に、pDUV5またはpCDT TERTコンストラクトをコードするプラスミドをトランスフェクトさせた。ポジティブコントロールとして、CrFK細胞に、野生型ヒトTERT(全活性)をコードするプラスミドをトランスフェクトさせた。簡潔には、トランスフェクションの24時間後に、CrFK細胞を機械的スクレイピングにより採取し、次いで1mLのPBSで2回洗浄し、そして3000gで4℃における5分間の遠心分離によってペレット化させた。テロメラーゼ活性は、TRAP−ELISAアッセイによってTeloTAGGGテロメラーゼPCR ELISAPLUSキット(ロシュ・ダイアグノスティックス、ドイツ)を使用して製造元の指示に従って評価した。細胞抽出物におけるタンパク質濃度は、ブラッドフォード法(バイオ・ラッド・ラボラトリーズ)によって測定した。3マイクロリットルの細胞抽出物(2.1μg、0.21μg、0.021μgに相当)を、キットで提供されたポリメラーゼ連鎖反応(PCR)混合物中でインキュベートした。サイクルプログラムは、25℃で30分のプライマー伸長で行い、次いでその混合物を、94℃で30秒の変性、50℃で30秒のアニーリング、72℃で90秒の重合、および72℃で10分の最終伸長からなる30サイクルのPCRにかけた。2.5μlの増幅産物を、ELISAのために製造元の指示に従って使用した。それぞれのウェルの450nmでの吸光度(690nmの参照波長で)を、Dynex MRX RevelationおよびDynex MRX Revelation TC 96 Well Microplate Readerを使用して測定した。
TERTをコードする新たなプラスミドは、トランスフェクション後にインビトロで機能性を示す
新たなプラスミドコンストラクトの機能性は、pCDTまたはpDUV5でインビトロでトランスフェクトされた細胞の全タンパク質ライセート中のプラスミドでコードされたTERTタンパク質の存在によって示される。本発明者らは、pCDTまたはpDUV5でトランスフェクトされた293T細胞プラスミド(トランスフェクションの24時間後)の全タンパク質ライセートに対するウェスタンブロット分析を行った。それぞれのプラスミドによってコードされたTERTタンパク質配列はV5タンパク質配列でタグ付けされているので、当該融合タンパク質の存在を明らかにするために、セイヨウワサビペルオキシダーゼ(HRP)と結合された抗V5抗体を使用した。
酵素活性の不在を試験するために、TRAPezeアッセイを行った。図3によって概説されるように、pDUV5またはpCDTでトランスフェクトされた細胞からのタンパク質ライセートは、いかなるテロメラーゼ活性も示さない。ポジティブコントロールとして、ネイティブなヒトTERT(hTERT)でトランスフェクトされたCrFK細胞からのタンパク質抽出物を使用した。このように、本発明者らは、pCDTまたはpDUV5プラスミドのいずれかによってコードされるTERTタンパク質が、インビトロでのトランスフェクション後にいかなる機能的な酵素活性も示さないことを実証した。
3.1.材料および方法
マウス
雌のBalb/cByマウスおよびC57BL/6Jマウス(6〜8週齢)をJanvier laboratories(フランス、サン・ベルトヴァン)から購入した。動物を、パスツール研究所の特定病原不在(Specific Pathogen Free)動物施設に収容した。マウスを皮内(ID)または筋内(IM)での免疫化の前に、1×リン酸緩衝生理食塩水(1×PBS、Life technologies SAS、フランス、サン・トーバン)中の2%キシラジン(Rompun、バイエル・サンテ、フランス、ロス)および8%のケタミン(Imalgen 1000、メリアル、フランス、リオン)の混合溶液で、個々の動物の体重および麻酔期間に応じて麻酔をかけた(腹腔内経路)。全ての動物を、良好な動物診療に厳密に従って取り扱い、パリのパスツール研究所の地元の動物実験倫理委員会のガイドラインに従った。
マウスの研究(IFNγ ELIspot)で使用されるTERTペプチドを、マウスのクラスIのMHC、H2Kb、H2DbまたはマウスのクラスIIのH2−IAdに結合するためにオンラインで利用できる4つのアルゴリズム:Syfpeithi(http://www.syfpeithi.de/)、Bimas(http://www−bimas.cit.nih.gov/)、NetMHCpanおよびSMM(http://tools.immuneepitope.org/main/)を使用してインシリコでのエピトープ予測によって予測した。
皮内(ID)での免疫化は、脇腹の下方部でインスリン用ニードル(U−100、29GX1/2’’−0.33×12mm、ベルギー、テルモ)を用いて剃毛後に行った。剃毛後に、免疫化処置の間およびその後に、紅斑は観察されなかった。筋内免疫化(IM)を、前脛骨筋中で、またインスリン用ニードルU−100を用いて実施した。それぞれの動物に、100μgのDNAに相当するpCDTまたはpDUV5のいずれかのプライミング用量を、ワクチン経路とは無関係に投与した。全ての動物を、プライミング14日後に同じ量のプラスミドおよび同じ免疫化経路を使用して追加免疫した。皮内的ワクチン接種直後に、侵襲性ニードル電極(6×4×2、47−0050、BTX、米国)を、注射部位が2つのニードル列(2つのニードル列は0.4cm離れている)の間に位置するように皮膚中に挿入した。異なる電圧の2つのパルスをかけた(HV−LV):HV=1125V/cm(2パルス、50μs−0.2μsパルス間隔)およびLV=250V/cm(8パルス、100V−10ms−20msパルス間隔)。筋内的免疫化の直後に、筋肉注射箇所を超音波ゲル(Labo FH、blue contact gel、NM Medical、フランス)で覆い、ピンセット電極(0.5cm間隔、tweezertrode 7mm、BTXI45−0488、米国)によって取り囲み、そして皮膚エレクトロポレーションと同じパラメータを使用して電圧を印加した。Agilepulse(登録商標)in vivo systemというエレクトロポレーターを全ての実験のために使用した(BTX、米国)。それぞれの免疫化経路(筋内的、皮内的)について、コントロールマウスを、同じ手順で同じ容量の1×PBSを使用して処理した。
簡潔には、PVDFマイクロプレート(IFN−γ Elispot kit、Diaclone、Abcyss、フランス、10×96試験、ref.862.031.010P)を、捕捉抗体(抗マウスIFN−γ)で一晩被覆し、PBS2%ミルクでブロッキングした。pDNA免疫化されたマウスからの脾臓をすりつぶし、細胞懸濁液を70mmナイロンメッシュ(Cell Strainer、BD Biosciences、フランス)を通して濾過した。Ficollで精製された脾臓細胞(Lymphocyte Separation Medium、Eurobio、フランス)を、Cellometer(登録商標)Auto T4 Plusカウンター(Ozyme、フランス)を使用して計数し、そしてプレートに3部で2×105または4×105細胞/ウェルで添加し、そして5μg/mlのdTERTまたはhyTERT関連ペプチドまたはコンカナバリンA(10μg/ml)で刺激するか、または血清不含の培養培地で模擬刺激を行った。19時間後に、スポットを、ビオチンにコンジュゲートされた検出抗体に曝露し、引き続きAP標識ストレプトアビジンおよびBCIP/NBTの基質溶液に曝露した。スポットを、Immunospot ELIspotカウンターおよびソフトウェア(CTL、ドイツ)を使用して計数した。

簡潔には、標的細胞調製のために、ナイーブなC57/Bl6マウス由来の脾臓細胞を、高濃度(5μM)、中濃度(1μM)または低濃度(0.2μM)のCFSEを含有する1×PBS中で標識した(Vybrant CFDA−SE cell−tracer kit;Life technologies SAS、フランス、サン・トーバン)。5μMおよび1μMのCFSEで標識した脾臓細胞を、2種の異なるH2ペプチドで5μg/mlにおいて1時間30分にわたり室温でパルスした。ペプチド987および621を、CFSEで高濃度標識されたナイーブな脾臓細胞とCFSEで中濃度標識されたナイーブな脾臓細胞のそれぞれのパルスのために使用した。CFSEで低濃度標識された脾臓細胞はパルスさせずにおいた。pCDTまたはpDUV5で事前に免役したそれぞれのマウスに、追加免疫注射の10日後に、それぞれの分画からの同数の細胞を含む107個のCFSE標識された細胞混合物を眼窩後方の静脈を通じて投与した。15〜18時間後に、脾臓由来の単細胞懸濁液を、フローサイトメトリーによってMACSQUANT(登録商標)サイトメーター(Miltenyii、ドイツ)で分析した。

Prism−5ソフトウェアを、データ処理、解析および図形表示のために使用した。データは、平均±標準偏差として表される。EliSPOTアッセイの統計分析のために、本発明者らは、マンホイットニーのノンパラメトリック検定を使用し、そしてインビボでの細胞傷害性アッセイのために、ダンの多重比較試験を用いたクラスカル−ウォリス分析を使用した。有意差は、p値<0.05に定めた。
pDUV5は、マウスにおける皮内的もしくは筋内的免疫化およびエレクトロポレーションの後に、強力な細胞傷害性のCD8T細胞応答を誘導する
本発明者らは、pDUV5プラスミドというDNAプラスミドがマウスにおいて効果的な細胞免疫応答(CD8)を顕現できるかどうかを評価した。この目的のために、9〜10匹のC57−Bl/6マウスの異なるグループに、pDUV5を皮内的にまたは筋内的に注射した直後にエレクトロポレーションをした。2週間後に、マウスに同じプロトコールで追加免疫注射を投与した。ブーストの10日後に、マウス脾臓を採取し、そして誘導された免疫応答を、表1に記載されるH2拘束性ペプチドを使用したIFN−γ ELISPOTアッセイを介してモニタリングした。マウスMHCクラスIに拘束性のイヌTERTペプチドを、材料と方法の節に記載したようにインシリコで予測した。図4に示されるように、dTERT特異的IFN−γ分泌性CD8T細胞の度数におけるコントロールマウスと比較しての大きな増加は、皮内的におよび筋内的にワクチン接種された動物の脾臓において観察された。これは、3つのペプチドのうち2つ(p621およびp987)について観察された(p<0.05)。2つの投与経路の間に有意差は認められなかった。
抗腫瘍免疫応答における細胞傷害性CD8T細胞の重要性に鑑みて、本発明者らは、プラスミドpCDTがそのような免疫応答をインビボで促進できるかどうかを評価した。こうして、9〜10匹のC57−Bl/6マウスの異なるグループを、pCDTによって該プラスミドの皮内的または筋内的注射によって免役した直後にエレクトロポレーションを行った。2週間後に、マウスに同じプロトコールで追加免疫注射を投与した。追加免疫の10日後に、マウス脾臓を採取し、そして誘導された免疫応答を、表1に記載されるH2拘束性ペプチドを使用したIFN−γ ELISPOTアッセイを介してモニタリングした。
4.1.材料および方法
イヌTERTペプチドライブラリー
凍結乾燥されたdTERTペプチド(純度>90%)を、JPT Peptide Technologies(ドイツ、ベルリン)から購入した。それぞれのペプチドを、供給元の推奨に従って、使用前に、蒸留したH2O、5%DMSO中で2mg/mLで再懸濁し、使用前に−20℃で凍らせたままにした。イヌTERTペプチドの三分の一(アミノ酸281〜571)を使用して、11アミノ酸が重複していて、かつ図3に示されるようにイヌTERTのこの配列を回復する15アミノ酸のペプチド70個を合成した。4つのペプチドプールを、インビトロ実験とイヌにおけるELIspotアッセイのために使用した。
イヌ血液試料は、Bourgelat研究所(Marcy l’Etoile、フランス)から購入した。その血液試料は、その施設の倫理ガイドラインに従って収容し、給餌し、かつ世話をした健康な4年齢のビーグル犬から採取した。ヘパリン化された血液試料を、1×PBS(Life technologies SAS、フランス、サン・トーバン)中に4倍希釈した。希釈された試料を、次いでLymphocyte Separation Medium(Eurobio、フランス、クールタブフ)上に成層し、そして2200rpmで(室温で)30分にわたり中断なく遠心分離した。イヌPBMCを採集し、そして10%DMSO(Sigma Aldrich chimie SARL、フランス、サン・カンタン・ファラヴィエ)を加えたウシ胎児血清(FCS、PAA Laboratories GmbH、オーストリア、パッシング)中で使用前に液体窒素中で貯蔵した。
0日目に、イヌの凍らせたPBMCを戻し、Cellometer(登録商標)Auto T4 Plusカウンター(Ozyme、フランス)を使用して計数し、そして2部または3部で106細胞/mLで48ウェルの平底プレート(BD、フランス)中でAIM−V培地(Invitrogen)に100ng/mLのcaGM−CSFおよび5ng/mLのcaIL−4(R&DSystems)または50ng/mLのヒトFlT3L(Immunotools)を補充した培地においてプレーティングした。細胞は、37℃、5%CO2でインキュベーター中で培養した。
本発明のワクチン技術の妥当性を強調するために、本発明者らは、目的の種、すなわちイヌにおける事前に存在していたイヌTERT特異的T細胞のレパートリーの存在を裏付けることを望んだ。
6匹のナイーブなビーグル犬に、ワクチン接種および100〜400μg/kgの筋内的な後ワクチン接種の15〜20分前に、2.5mg/kg(静脈内)imalgeneおよび20〜80μg/kg(静脈内)dorbeneの局所麻酔を受けさせた。それらの犬に、400μgのpDUV5 DNAを皮内注射した後にエレクトロポレーションを行った。pDU5 DNAは、0日目、29日目、57日目および142日目にエレクトロポレーションを行った。末梢血を抜き取り、そして単核細胞を、プール6またはプール19のいずれかに属するイヌテロメラーゼ特異的ペプチドについて、Martinuzzi et al., 2011の方法に従って試験した。
pDUV5 DNAエレクトロポレーションが新形成を伴う動物において特異的dTERT T細胞応答を誘導し得ることを示すために、5匹の新形成を伴う飼い犬とコントロールとしての3匹の飼い犬を使用した。それらの病気の動物は広範に異なる腫瘍を引き起こしていた。以下の表2を参照のこと。
