Description
前記非ペプチド性重合体は、インスリンβ鎖の25〜29位のいずれかのアミノ酸残基に結合されることがより好ましい。
前記非ペプチド性重合体が結合するインスリンβ鎖のアミノ酸残基は、アミノ基又はチオール基を有することが好ましい。
免疫グロブリンFc領域が非グリコシル化されたものが好ましい。
免疫グロブリンFc領域が、CH1、CH2、CH3及びCH4ドメインからなる群から選択される1つ〜4つのドメインからなることが好ましい。
免疫グロブリンFc領域が、IgG、IgA、IgD、IgE又はIgMに由来するFc領域であることが好ましい。
免疫グロブリンFc領域がIgG4 Fc領域であることが好ましい。
非ペプチド性重合体は、インスリンβ鎖のアミノ酸残基側鎖のアミノ基又はチオール基と、ペプチド(peptide)、ヘミチオアセタール(hemithioacetal)、イミン(imine)又はチオジオキソピロリジニル(thiodioxopyrrolidinyl)結合を形成することが好ましい。
他の態様として、本発明は、前記インスリン結合体を含み、生体内持続性及び安定性が向上したインスリンの持続性製剤を提供する。
さらに他の態様として、本発明のは、(1)非ペプチド性重合体をインスリンβ鎖のN末端を除くアミノ酸残基に共有結合で連結する段階と、(2)前記(1)の反応混合物からインスリンβ鎖のN末端を除くアミノ酸残基に非ペプチド性重合体が共有結合したインスリン連結体を分離する段階と、(3)分離した連結体の非ペプチド性重合体の他方の末端に免疫グロブリンFc領域を共有結合で連結することにより、非ペプチド性重合体の両末端がそれぞれ免疫グロブリンFc領域及びインスリンに結合したインスリン結合体を生成する段階とを含む、インスリン結合体の製造方法を提供する。
前記非ペプチド性重合体の両末端は、それぞれインスリンβ鎖のN末端を除くアミノ酸残基、及び免疫グロブリンFc領域のアミノ基又はチオール基により結合されることが好ましい。
前記段階(1)は、pH9.0±2のアルカリ環境で行われることが好ましい。
前記段階(3)のインスリン連結体と免疫グロブリンFc領域のモル比は1:1〜1:10であることが好ましい。

前記インスリンは、天然のインスリン、天然のインスリンから一部のアミノ酸の置換、付加、欠失及び修飾のいずれかの方法又はそれらの方法の組み合わせにより作製された変異体、インスリン誘導体、インスリンアゴニスト又はそれらのフラグメントであることが好ましい。
インスリン誘導体とは、天然のインスリンと比較して、少なくとも80%のアミノ酸配列相同性を有し、アミノ酸残基の一部の基が化学的に置換(例えば、α−メチル化、α−ヒドロキシル化)、欠失(例えば、脱アミノ化)又は修飾(例えば、N−メチル化、グリコシル化、脱脂酸)された形態であり、体内で血糖を調節する機能を有するペプチドを意味する。
また、インスリンアゴニスト、誘導体、フラグメント及び変異体においてそれぞれ用いる製造方法は、独立して用いてもよく、組み合わせて用いてもよい。例えば、本発明のインスリンペプチドには、天然のインスリンとアミノ酸配列が1つ以上異なり、N末端アミノ酸残基が脱アミノ化(deamination)された、体内で血糖調節機能を有するペプチドも含まれる。
本発明のインスリン結合体は、両末端に反応性官能基を有する非ペプチド性重合体をリンカーとして用いて、重合体の各末端をインスリンβ鎖及び免疫グロブリンFc領域にそれぞれ共有結合させて製造したことを特徴とする。非ペプチド性重合体の両末端が、それぞれ免疫グロブリンFc領域と、インスリンβ鎖アミノ酸残基側鎖のアミノ基又はチオール基に結合されることが好ましい。
本発明は、インスリンの血中安定性を向上させるために、ポリエチレングリコール(PEG)及び免疫グロブリン定常領域(以下、免疫グロブリンFc又はFcともいう)との結合体を製造する上で、インスリンβ鎖上でPEG−Fcの結合位置によってインスリン受容体に対する結合力が変化することを確認し、さらにインスリン結合力を向上させることにより活性を向上させることのできる結合位置を解明したことを特徴とする。例えば、PEG−Fcとの結合により血中安定性を向上させながらも、前記結合がインスリン受容体との結合を阻害して活性を減少させることのない位置を確認した。インスリンα鎖においては、結合体を形成する際に活性が大幅に減少することが知られているので、インスリンのβ鎖上で最適な結合位置を探索した。その結果、非ペプチド性重合体の結合位置は、インスリンβ鎖のN末端を除く、アミノ基又はチオール基を有する任意のアミノ酸残基であることが確認された。
前記非ペプチド性重合体は、インスリンβ鎖のアミノ酸残基側鎖のアミノ基又はチオール基と、ペプチド、ヘミチオアセタール、イミン又はチオジオキソピロリジニル結合を形成することが好ましい。
本発明における「免疫グロブリンFc領域」とは、免疫グロブリンの重鎖と軽鎖の可変領域、重鎖定常領域1(CH1)及び軽鎖定常領域(CL1)を除いたものであり、重鎖定常領域2(CH2)及び重鎖定常領域3(CH3)部分を意味し、重鎖定常領域にヒンジ(hinge)部分を含むこともある。また、本発明の免疫グロブリンFc領域は、天然のものと実質的に同等又は向上した効果を有するものであれば、免疫グロブリンの重鎖と軽鎖の可変領域を除き、一部又は全部の重鎖定常領域1(CH1)及び/又は軽鎖定常領域1(CL1)を含む拡張されたFc領域であってもよい。また、CH2及び/又はCH3に相当する非常に長い一部のアミノ酸配列が欠失された領域であってもよい。すなわち、本発明の免疫グロブリンFc領域は、(1)CH1ドメイン、CH2ドメイン、CH3ドメイン及びCH4ドメイン、(2)CH1ドメイン及びCH2ドメイン、(3)CH1ドメイン及びCH3ドメイン、(4)CH2ドメイン及びCH3ドメイン、(5)1つ又は2つの以上のドメインと免疫グロブリンヒンジ領域(又はヒンジ領域の一部)との組み合わせ、(6)重鎖定常領域各ドメインと軽鎖定常領域の二量体であってもよい。
本発明における免疫グロブリンFc領域と非ペプチド性重合体の結合は、前記インスリンβ鎖と非ペプチド性重合体の結合と同様に、インスリンに結合しない非ペプチド性重合体の他方の末端の反応性官能基と、免疫グロブリンFc領域のアミノ酸残基のアミノ基又はチオール基の共有結合により形成される。よって、非ペプチド性重合体は、免疫グロブリンFc領域のN末端、又は免疫グロブリンFc領域内のリジン残基のアミノ基もしくはシステイン残基のチオール基に結合する。ここで、免疫グロブリンFc領域において非ペプチド性重合体が結合されるアミノ酸残基の位置は限定されない。
本発明における「投与」とは、任意の適切な方法で患者に所定の物質を導入することを意味し、前記結合体の投与経路は、薬物を標的組織に送達できるものであれば、いかなる一般的な経路を介して投与してもよい。腹腔内投与、静脈内投与、筋肉内投与、皮下投与、皮内投与、経口投与、局所投与、鼻腔内投与、肺内投与、直腸内投与などが挙げられるが、これらに限定されるものではない。しかし、経口投与の場合はペプチドが消化されるので、経口用組成物は、活性薬剤をコーティングしたり、胃での分解から保護されるように剤形化することが好ましい。注射剤の形態で投与することが好ましい。また、持続性製剤は、活性物質を標的細胞に送達することのできる任意の装置により投与することができる。
さらに他の態様として、本発明は、(1)非ペプチド性重合体をインスリンβ鎖のN末端を除くアミノ酸残基に共有結合で連結する段階と、(2)前記(1)の反応混合物からインスリンβ鎖のN末端を除くアミノ酸残基に非ペプチド性重合体が共有結合したインスリン連結体を分離する段階と、(3)分離した連結体の非ペプチド性重合体の他方の末端に免疫グロブリンFc領域を共有結合で連結することにより、非ペプチド性重合体の両末端がそれぞれ免疫グロブリンFc領域及びインスリンに結合したインスリン結合体を生成する段階とを含む、インスリン結合体の製造方法を提供する。
インスリン粉末を10mM HClに溶解し、その後3.4K butyraldehyde−PEG−succinimidyl valerate(両末端にそれぞれブチルアルデヒド基とスクシンイミジルバレレートを官能基として有するPEG,Laysan Bio,Inc.米国)によりインスリンβ鎖の29位のアミノ酸残基にペグ化させるために、インスリン:PEGのモル比を1:2とし、インスリン濃度を1.5mg/mlとして、常温で約1時間反応させた。ここで、反応は60.8mMホウ酸ナトリウム(Sodium Borate)pH9.0、45%イソプロパノールにおいて行われ、クエン酸ナトリウム(pH3.0)、45%エタノールを含む緩衝液とKCl濃度勾配を利用したSource Sカラム(GE Healthcare)を用いて、反応液からモノペグ化インスリンを精製した(図1A〜図1B)。
実施例1でペグ化されたインスリンにおける3.4K PEGの結合部位を確認するためにGlu−Cマッピング方法を用いた。1mg/mlの濃度のモノペグ化インスリン50μgに1mg/mlの濃度のペプチド内部加水分解酵素Glu−C 10μgを添加した。反応液は50mM HEPES、pH7.5であり、25℃で8時間反応させた。その後、1NのHCl 50μlを添加して反応を終結させた。マッピングはHPLC逆相クロマトグラフィーを用いて行った。その結果を図2に示す。
インスリン−PEG−免疫グロブリンFcフラグメント結合体を製造するために、実施例1の方法を用いて得られたモノペグ化(mono-PEGylated)インスリンと免疫グロブリンFcフラグメントのモル比が1:1.2となるようにし、全タンパク質濃度を20mg/mlとして25℃で13時間反応させた。ここで、反応液は100mM HEPES、2M塩化ナトリウム(NaCl)、pH8.2であり、還元剤として20mMシアノ水素化ホウ素ナトリウムを添加した。
インスリンのN末端にPEG−Fcが結合したインスリン結合体と、B29にPEG−Fcが結合したインスリン結合体のインスリン受容体に対する結合力を確認するために、表面プラズモン共鳴法(surface Plasmon resonance、SPR、BIACORE 3000)を用いた。インスリン受容体としては、HEK293F細胞で発現させた細胞外ドメイン(extracellular domain、ECD)を精製して用いた。アミンカップリング法を用いて前記インスリン受容体をCM5チップに固定化し、その後1μM〜6.25nMのN末端又はB29インスリン結合体を分注してその結合力を確認した。これらインスリン結合体は結合緩衝液(HBS−EP)で希釈し、インスリン結合体を固定化したチップに4分間結合させて6分間の解離過程を経た。その後、他の濃度のインスリン結合体を結合させるために、インスリン受容体に結合したインスリン結合体に50mMのNaCl/5mMのNaOHを約30秒間分注した。結合力はBIAevaluationプログラムの1:1 Langmuir binding modelを用いて分析した。その結果を図4に示す。
Claims (28)
- インスリン及び免疫グロブリンFc領域が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコール−プロピレングリコール共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、デキストラン、ポリビニルエチルエーテル、生分解性高分子、脂質重合体、キチン類、ヒアルロン酸及びそれらの組み合わせからなる群から選択される非ペプチド性重合体リンカーにより連結されたインスリン結合体であって、前記非ペプチド性重合体は、一方の末端がインスリンβ鎖の27〜29位のいずれかのアミノ酸残基に結合され、他方の末端が免疫グロブリンFc領域に結合されており、前記インスリンが、
(i)配列番号1の第1のペプチド及び配列番号2の第2のペプチドを含む天然のインスリン、
(ii)配列番号1の第1のペプチド及び配列番号2の第2のペプチドの少なくとも一方の1つのアミノ酸残基が、天然のインスリンのアミノ酸配列とは置換によって異なるペプチドを含むインスリン変異体であって、それを投与された対象の体内の血糖を調節する機能を有するインスリン変異体、
(iii)配列番号1の第1のペプチド及び配列番号2の第2のペプチドの少なくとも一方の1つのアミノ酸残基が化学的に修飾されたペプチドを含むインスリン誘導体であって、それを投与された対象の体内の血糖を調節する機能を有するインスリン誘導体、又は
(iv)前記天然インスリン、前記インスリン変異体、又は前記インスリン誘導体のフラグメントであって、それを投与された対象の体内の血糖を調節する機能を有するフラグメント

- スクシンイミド誘導体が、スクシンイミジルカルボキシメチル、スクシンイミジルバレレート、スクシンイミジルメチルブタノエート、スクシンイミジルメチルプロピオネート、スクシンイミジルブタノエート、スクシンイミジルプロピオネート、N−ヒドロキシスクシンイミド又はスクシンイミジルカーボネートである請求項15に記載のインスリン結合体。
- 非ペプチド性重合体が、両末端にそれぞれブチルアルデヒド基とスクシンイミジルバレレート反応性官能基を有するものである請求項15に記載のインスリン結合体。
- 請求項1〜17のいずれか一項に記載のインスリン結合体を含み、生体内持続性及び安定性が向上したインスリンの持続性製剤。
- 糖尿病治療用である請求項18に記載の持続性製剤。
- (1)非ペプチド性重合体をインスリンβ鎖の27〜29位のいずれかのアミノ酸残基に共有結合で連結する段階と、
(2)前記(1)の反応混合物からインスリンβ鎖の27〜29位のいずれかのアミノ酸残基に非ペプチド性重合体が共有結合したインスリン連結体を分離する段階と、
(3)分離した連結体の非ペプチド性重合体の他方の末端に免疫グロブリンFc領域を共有結合で連結することにより、非ペプチド性重合体の両末端がそれぞれ免疫グロブリンFc領域及びインスリンに結合したインスリン結合体を生成する段階とを含む、請求項1のインスリン結合体の製造方法。 - 非ペプチド性重合体は、インスリンβ鎖のアミノ酸残基側鎖のアミノ基又はチオール基と、ペプチド、ヘミチオアセタール、イミン又はチオジオキソピロリジニル結合を形成するものである請求項20に記載の製造方法。
- 非ペプチド性重合体が、両末端にそれぞれ独立してアルデヒド誘導体、マレイミド誘導体又はスクシンイミド誘導体を反応性官能基として有するものである請求項20に記載の製造方法。
- 非ペプチド性重合体の両末端は、それぞれインスリンβ鎖の前記アミノ酸残基、及び免疫グロブリンFc領域のアミノ酸残基側鎖のアミノ基又はチオール基により結合されるものである請求項20に記載の製造方法。
- 非ペプチド性重合体が、両末端にそれぞれブチルアルデヒド基とスクシンイミジルバレレート反応性官能基を有するものである請求項20に記載の製造方法。
- 段階(1)が、pH9.0±2のアルカリ環境で行われる請求項20に記載の製造方法。
- 前記段階(1)のインスリンと非ペプチド性重合体のモル比が1:1.5〜1:10である請求項20〜25のいずれか一項に記載の製造方法。
- 前記段階(3)のインスリン連結体と免疫グロブリンFc領域のモル比が1:1〜1:10である請求項20〜25のいずれか一項に記載の製造方法。
- インスリンが天然のインスリンであるか、又は配列番号1の第1のペプチド及び配列番号2の第2のペプチドを含む変異体であって、配列番号1及び配列番号2の少なくとも一方の1つのアミノ酸残基が、天然のインスリンのアミノ酸配列とは置換によって異なる変異体であり、前記変異体は、それを投与された対象の体内の血糖を調節する機能を有する請求項1に記載のインスリン結合体。
