本明細書で使用される時、「報酬関連行動」は、行動を強化するプロセス、すなわち、行動を行った直後またはすぐ後の刺激の送達または出現によって、しばしば快楽的な反応の形態で、特定の行動の率、確率、または強度を増加させる何らかのものである。報酬関連行動には、食物の獲得、性行動、賭博行動、および/または薬物関連の嗜癖行動が含まれる可能性があるが、それらに限定されない。
側坐核(asaccumbens nucleus)または中隔側坐核(nucleus accumbens septi)としても知られる、側坐核(nucleus accumben)(NAc)は、腹側線条体の主要部を形成するニューロンの集合である。それは、報酬、快楽、笑い、嗜癖、攻撃、恐怖、およびプラセボ効果において重要な役割果たすと考えられている。側坐核において見出される主な神経細胞型は、中型有棘ニューロン(MSN)である。これらのニューロンによって産生される神経伝達物質は、中枢神経系の主要な抑制性神経伝達物質のうちの1つである、ガンマ−アミノ酪酸(GABA)である。MSNはまた、側坐核の主要な投射または出力ニューロンである。側坐核におけるニューロンの95%は、中型有棘GABA作動性の投射ニューロンであるが、脳のこの領域における細胞の約1%を構成する、大型無棘コリン作動性介在ニューロン等の、他の神経型もまた見出される。
本明細書では、個体における少なくとも1つの報酬関連行動を妨害するために、個体の側坐核および線条体におけるコリン作動性ニューロンを選択的に過分極化するための、光遺伝学的ベースの組成物および方法が提供される。光遺伝学とは、機能している無処置の生体系とペースを保つために必要とされる時間精度(ミリ秒の時間スケール)で、自由に移動しているほ乳類および他の動物の体内でさえも、生体組織の標的細胞内の具体的な事象を制御するために使用される、遺伝的および光学的方法の組み合わせを指す。光遺伝学は、具体的な標的機構の使用を通じて細胞型分解能を維持しながら、ニューロン膜電位の時間的に正確な操作を可能にする、標的神経細胞の原形質膜への高速光応答性オプシンチャネルまたはポンプタンパク質の導入を必要とする。

本開示は、ほ乳類細胞の原形質膜への輸送を強化する、1つ以上のアミノ酸配列モチーフの追加によって、細胞において発現される光応答性オプシンタンパク質の修飾を提供する。進化的により単純な生物に由来する構成要素を有する、光応答性オプシンタンパク質は、ほ乳類細胞によって発現されない、または耐えられなくてもよく、あるいはほ乳類細胞において高レベルで発現される時に低下した細胞内局在性を示してもよい。その結果として、いくつかの実施形態では、細胞において発現される光応答性オプシンタンパク質は、シグナルペプチド、小胞体(ER)移行シグナル、膜輸送シグナル、および/またはN末端ゴルジ移行シグナルから成る群より選択される、1つ以上のアミノ酸配列モチーフに融合させることができる。ほ乳類細胞の原形質膜への光応答性オプシンタンパク質輸送を強化する、1つ以上のアミノ酸配列モチーフは、光応答性オプシンタンパク質のN末端、C末端、またはNおよびC末端の両方に融合させることができる。随意で、光応答性オプシンタンパク質および1つ以上のアミノ酸配列モチーフは、リンカーによって分離されてもよい。いくつかの実施形態では、光応答性オプシンタンパク質は、細胞原形質膜へのタンパク質の輸送を強化する輸送シグナル(ts)の追加によって修飾することができる。いくつかの実施形態では、輸送シグナルは、ヒト内向き整流カリウムチャネルKir2.1のアミノ酸配列に由来することができる。他の実施形態では、輸送シグナルは、アミノ酸配列KSRITSEGEYIPLDQIDINVを含むことができる。

本明細書で提供される組成物および方法のいくつかの態様では、1つ以上の光応答性プロトンポンプが、側坐核または線条体のコリン作動性介在ニューロンの原形質膜上で発現される。
本明細書で提供される方法のいくつかの態様では、光応答性イオンチャネルのチャネルロドプシンファミリーの1つ以上のメンバーは、側坐核または線条体のコリン作動性介在ニューロンの原形質膜上で発現される。
本開示はまた、本明細書に記載される光応答性オプシンタンパク質をコードするヌクレオチド配列を含む、ポリヌクレオチドも提供する。いくつかの実施形態では、ポリヌクレオチドは、発現カセットを含む。いくつかの実施形態では、ポリヌクレオチドは、上述の核酸を含むベクターである。いくつかの実施形態では、本開示の光応答性オプシンタンパク質をコードする核酸は、プロモータに作動可能に連結される。プロモータは、当該技術分野で周知である。コリン作動性介在ニューロンにおいて機能する任意のプロモータを、本開示の光応答性オプシンタンパク質および/またはその任意の変異形の発現に使用することができる。具体的な動物細胞における光応答性オプシンタンパク質またはその変異形の発現を駆動するのに有用である、開始制御領域またはプロモータは、多数あり、当業者に周知である。これらの核酸を駆動することが可能な事実上いかなるプロモータも使用することができる。いくつかの実施形態では、光応答性タンパク質の発現を駆動するために使用されるプロモータは、コリン作動性介在ニューロンにおける導入遺伝子の強固な発現を駆動することが可能である、コリンアセチルトランスフェラーゼ(ChAT)プロモータであり得る(例えば、Gong et al.,J.Neurosci.,27,9817−9823(2007)を参照されたい)。

ニューロンにおいて発現される光応答性タンパク質を活性化するための波長を有する光を適用することが可能である任意のデバイスが、ニューロンを脱分極化するおよび/または過分極化するために使用されてもよい。例えば、1つ以上のニューロンの膜電圧に影響を及ぼすためにイオンチャネルおよび/またはイオン性ポンプを活性化するための光送達デバイスが使用されてもよい。光送達デバイスは、脳の標的領域に光刺激を提供するように構成することができる。光送達デバイスは、基部、基部に取り付けられたカニューレガイド、およびカニューレガイドを介して基部に取り付けられた1つ以上の光導管を備えてもよい。基部は、光導管から、側坐核または線条体等の標的組織領域に光を送達するように位置付けられた1つ以上の光送達ポートを備えてもよい。光導管は光ファイバーであってもよく、ファイバーの近位端は光学的光源に取り付けられ、遠位端は光送達ポートと連通している。光学的光源は、連続光および/またはパルス光を提供することが可能であり得、所定のパルス周期で光を提供するようにプログラム可能であり得る。光送達デバイスは、所望されるように、例えば、1、2、3、4、5、10、15、20個等の任意の数の光導管を有してもよい。光導管は、各々が、同じまたは異なる波長の光を担持してもよい。送達された光は、黄色、緑色、または青色光等の450nm〜600nmの波長を有してもよい。光送達デバイスは、所望されるように、例えば、1、2、3、4、5、10、15、20個等の任意の数の光送達ポートを有してもよい。いくつかの変形においては、光導管と同じ数の光送達ポートが存在してもよい一方で、他の変形においては、異なる数の光導管および光送達ポートが存在してもよい。例えば、2つ以上の光送達ポートに光を伝達する単一の光導管が存在してもよい。代替的にまたは追加的に、単一の光導管が、単一の光送達ポートに接続されてもよい。カニューレガイドは、光導管を光送達ポートに固定して揃える一助となるように構成されてもよい。いくつかの実施形態では、光送達デバイスは、個体における少なくとも1つの報酬関連行動を妨害するために、光を側坐核または線条体に送達するように構成される。光送達デバイスまた、神経活動を測定するために構成され得る1つ以上の測定電極を備えてもよい。例えば、測定電極は、ニューロンが刺激に応答するときに、膜電位(例えば、活動電位)および/または1つ以上のニューロンの膜を横切る電流における変化を記録してもよい。いくつかの変形においては、測定電極は、1つ以上のニューロンの光刺激に対する電気的応答を測定してもよい。測定電極は、細胞外電極または細胞内電極であってもよい。
動物の側坐核または線条体におけるコリン作動性介在ニューロンの細胞膜上で発現された光応答性オプシンタンパク質を含む非ヒト動物が、本明細書に提供され、ここでタンパク質は、光に応答性であり、介在ニューロンが光で照射される時、介在ニューロンの膜分極状態を変化させることが可能であり、オプシンの照射は、動物の少なくとも1つの報酬関連行動を妨害する。いくつかの実施形態では、光応答性タンパク質は、本明細書に記載されるNpHR、BR、ARおよびGtR3から成る群より選択される。例えば、本明細書に記載されるNpHRタンパク質のうちのいずれかは、標的ニューロンの細胞膜上で発現されてもよい。いくつかの実施形態では、報酬関連行動は、薬物関連嗜癖行動である。薬物は、オピオイド(例えば、アヘンおよびヘロイン)、メタンフェタミン、コカイン、ケタミン、MDMA(3,4−メチレンジオキシメタンフェタミン)、リゼルギン酸ジエチルアミド、カンナビノイド、アルコール、ニコチン、または任意の他の規制物質等であるが、それらに限定されない、任意の嗜癖薬物であり得る。一実施形態では、薬物は、コカインである。別の実施形態では、報酬関連行動は、コカイン嗜癖である。

本開示は、生存動物における強化された行動の制御または特徴付けに有用であると考えられる。本発明の具体的な適用は、生存動物における嗜癖および他の強化された行動の査定を促進する。本明細書で開示される例示的な実施形態の多くの態様は、この分野における以前の開発に関し、またそれらに大いに基づいているため、以下の考察は、かかる以前の開発を要約して、実装形態の詳細および修正が導き出される元となり得る基盤および根底にある教示の確固たる理解を提供する。次の考察は、この文脈に関連して、かつ参照により本明細書に組み込まれる参考文献中のその教示と共に、提供される。本発明は、必ずしもかかる適用に限定されるわけではないが、本発明の種々の態様は、この前後関係を使用して、種々の例の考察を通じて理解されてもよい。
我々は、高い時間的分解能および高い細胞型特異性の両方を用いて、これらの細胞中の活動電位発火を選択的に駆動または遮断することによって、この課題を解決するために、光遺伝学的アプローチ(42〜44)に着手した。微生物オプシン遺伝子をコリン作動性介在ニューロンにおいて特異的に発現させるために、我々は、コリンアセチルトランスフェラーゼ(ChAT)プロモータ下でCreリコンビナーゼを発現するトランスジェニックマウス株を用いた(45)。我々は、NAc中に(図1A)、強化黄色蛍光タンパク質(eYFP)(46、47)に対するコード配列を伴ってフレーム内で融合された、オプシン遺伝子を担持するCre誘発性アデノ随伴ウイルス(AAV)ベクターを定位的に注射した。オプシン遺伝子は、青色光ゲートカチオンチャネルチャネルロドプシン−2(ChR2)(48、49)または黄色光ゲート第3世代クロライドポンプハロロドプシン(eNpHR3.0)(50)のいずれかをコードした。
対象
BACトランスジェニックコリンアセチルトランスフェラーゼ(ChAT)::Creマウスを、GENS AT(原液名:Tg(Chat−cre)24Gsat/Mmcd)(Gong et al.,J.Neurosci 27,9817−9823(2007))から得、Charles RiverからのC57BL6マウスと交配させた。実験的マウスは、Cre(+/−)に対する異型接合、あるいは制御同腹子(−/−)のずれかであった。マウスを、逆行の12時間明/暗サイクルで維持されたコロニーで群飼し、餌および水を随意に与えた。実験プロトコルは、実験動物の管理と使用に関する、国立衛生研究所の指針のガイダンスを満たすために、Stanford University IACUCによって認可された。
前述のように(Tsai et al.,Science 324、1080−1084(2009)、Sohal et al.,Nature 459、698−702(2009))、Cre誘発性組み換えAAVベクターは、オプシン(ChR2(H134R)またはeNpHR3.0のいずれか)が逆配向でlox部位の間に挿入された、2対の不適合なlox部位(loxPおよびlox2722)を担持するDNAカセットに基づいた。この二重フロックス(double−floxed)逆オプシンカセットを、発現を強化するためのpEF−1αプロモータおよびWoodchuck肝炎ウイルス転写後調節要素(WPRE)を担持すするAAV2−MCSベクターのバージョン中にクローン化した。Cre誘発性ChR2 AAV構築物、ならびにeNpHR3.0 導入遺伝子の完全なマップは、www.stanford.edu/group/dlab/optogenetics/sequence_info.htmlにて入手可能である。組み換えAAVベクターを、AAV5コートタンパク質で血清型別し、University of North Carolinaにてウイルスベクターコアによってパッケージングした。最終ウイルス濃度は、ChR2ウイルスについて3×1012粒子/mL、およびNpHR3.0ウイルスについて1.5×1012粒子/mLであった。
マウスを、ケタミン/キシラジンで麻酔し(60μL/マウス、80mg/mLケタミンおよび12mg/mLキシラジンの混合物の使用)、次いで定位ヘッド器具中に配置した。手術を、生理学実験のために4〜6週齢マウスに、および行動実験のために8〜12週齢マウスに行った。眼が乾燥することを予防するように、眼軟膏を塗布した。頭皮の正中切開、続いて開頭術を行い、次いで、ウイルスを、10μLシリンジおよび34ゲージ金属針を用いて注射した。注射量および流速(0.15μL/分で1μL)を注入ポンプによって制御した。各NAcは、2回の注射(注射1:AP1.15mm、ML0.8mm、DV−4.8mm、注射2:AP1.15mm、ML0.8mm、DV−4.2mm)を受容した。ウイルス注射および線維位置を、事実上殻全体が刺激されるように選定した。マウスの殻の小さいサイズを考慮すると、ウイルス拡散および光を内殻に完全に限定することは不可能であり、コアの内側部を含めた(前交連に対して内側)。注射後、針を更に5分間定置に残し、次いで非常にゆっくりと回収した。行動実験のために、マウスに両側性で注射し、次いで1.5mmの中心間距離を有する両側カニューレを、注射部位の上部に配置した(AP 1.15mm、DV 3.8mm)。行動中の神経活動を操作するために、ファイバーがカニューレの末端を超えて200〜300μm突出するようにカニューレを通じて挿入された2つの300μm直径光ファイバー(0.37N.A.)を通じて、光を両側性で送達した。
ChATニューロンにけるオプシン発現の特異性を決定するために、最初にPBSを用いて、次いでリン酸緩衝食塩水(PBS、pH7.4)中に溶解させた4%パラホルムアルデヒド(PFA)を用いて、マウスを、ボタナシア(beuthanasia)で麻酔し、経心的に灌流させた。脳を除去し、4%PFA中に一晩4℃で後固定し、次いでPBS中の30%ショ糖中で平衡化した。40μm厚の冠状片を、凍結ミクロトーム(Leica)上で調製し、凍結保護物質中(PBS中25%グリセロールおよび30%エチレングリコール)4℃で保管した。自由に浮動する薄片をPBS中で洗浄し、次いで30分間、0.3%Triton X−100(Tx100)および3%正常ロバ血清(NDS)中でインキュベートした。切片を、4℃で一晩、3%NDS(ヤギ抗ChAT 1:200、Millipore)中の一次抗体を用いてインキュベートした。薄片を次いでPBSで洗浄し、2時間室温で二次抗体(Cy3またはCy5に共役させたロバ抗ヤギ、Jackson Laboratories)を用いてインキュベートした。切片を次いで洗浄し、DAPI(1:50,000)を用いて20分間インキュベートし、再び洗浄し、PVA−DABCOでスライド上に載置した。共焦点蛍光画像を、レーザー走査型顕微鏡上で、5Xもしくは10X空気対物、または40X油浸対物を使用して得た。
我々は、この生体内標的化戦略の特異性および機能性を検証し、eYFP発現が、ChATを発現したニューロンに対して高度に特異的であることを見出し、更に、ChATを発現したニューロンの大多数はまた、eYFPも発現した(図1B,C)。任意の所与の細胞がChAT介在ニューロンであるという前試験確率は非常に低く、したがって稀有な標的化の漏れでさえも標的化特異性を劇的に減少させたであろうため、観察された特異性は、そのようなまばらな神経集団について特に印象的であった。双方のオプシンは、ChATニューロンの表面膜上で発現され(図1D)、標的ニューロンは、背側線条体におけるコリン作動性介在ニューロンの以前に確立された応答に対応する様態で、電流注射に対して応答した(図1E)(51)。背側線条体生理学と更に一致するように、静止電位および入力抵抗の両方は、ChATニューロン(YFP+ニューロン)について、MSN(YFP−ニューロンとして識別される、表S1、VMについてp<10−4およびRinputについてp=0.004、両側t検定)よりも高かった。最後に、eNpHR3.0は、莫大な過分極を駆動し(図1F;平均±標準誤差:−83.8±11.9mV、n=4)、ChR2は、最大20〜30Hzの発火を確実に駆動したため(図1G、H)、双方のオプシンは、ChAT細胞において強力に機能的であった。

ChAT介在ニューロンは、生体内において持続的に活性であると考えられるが(3〜10Hz(52、53))、まばらなChAT細胞中のこの種の遅い活性が、局所回路活性または行動に影響を及ぼすことに関与し得る様子(または更に関与し得るかどうか)は、謎のままである。我々は、光遺伝学的制御を生かして、薄片電気生理学、生体内電気生理学、および自由移動行動の組み合わせを用いて、この課題に取り組んだ。
急性脳切片生理学
冠状脳切片を、ウイルスを事前(切片作製の2週間よりも前)に注射した成体マウスから、標準的な技法を使用して、Stanford Universityの動物実験委員会によって認可されたプロトコルに厳格に従って調製した。冠状切片250μm厚を、135mM NMDG、1mM KCl、1.2mM KH2PO4、20mMコリンビカルボナート、10mMブドウ糖、1.5mM MgCl2、および0.5mM CaCl2を含有する、氷冷N−メチル−D−グルカミン(NMDG)ベースの切断溶液中、サファイア刃を使用するビブラトームで切断した。切片を、それ以降、119mM NaCl、2.5mM KCl、2.5mM CaCl2、1.3mM MgCl2、1mM NaH2PO4、26.2mM NaHCO3および11mMブドウ糖を含有する、人工脳脊髄液(ACSF)中で維持した。切片を、ACSF中で、30分間37℃で、およびそれ以降、室温で維持した。ACSFに、95%O2/5%CO2で一定して気泡を注入し、全ての実験のために34℃まで加熱した。ニューロンを、eYFPを可視化するためにDIC光学およびフィルターセットの両方を装着した正立顕微鏡(Leica DM−LFSA)上で、40倍水浸漬対物および電荷結合素子(CCD)カメラを使用して可視化した。ニューロンから全細胞記録を、120mMグルコン酸カリウム、20mM HEPES、10mM EGTA、1mM MgCl2、2mM Na−ATP、および0.2mM Na−GTP(pH7.3、290mOsm/L)を含有する電極溶液を使用して行った。IPSCを記録する実験においては、KClを使用してグルコン酸カリウムを置き換えた。ピペット抵抗は3〜5ΜΩであり、記録を、直列抵抗補償を用いずに行った。膜電位は、液界電位からもたらされる誤差に対して補正した。電圧クランプ実験についての保持電位(VM)は、−70mVであった。次の薬剤を示されるように添加した:5μΜ SR−95531、5μΜ 2,3−ジヒドロキシ−6−ニトロ−7−スルファモイル−ベンゾ(F)キノキサリン(NBQX)、5μΜ(R,S)−CPP、メカミラミン(10μΜ)、5μΜコカイン塩酸塩。5μΜのコカイン濃度を、複数の基準によって注意深く選定した。最初に、それは以前の切片作業の選定と一致していた(Thompson et al.,Neuropharmacology 49,185−194(2005))。第2に、有意に高い濃度は、局所麻酔効果をもたらすであろう(Thompson et al.,Neuropharmacology 49,185−194(2005))。最後に、マウスにおけるコカイン薬物動態の研究によれば、行動実験で使用されたレベルに匹敵して、10mg kgの腹腔内注射は、脳内で15分後に4.7μΜのコカインを産出し、20mg/kgは、9.4μΜを産出するであろう(Shah et al,Toxicology and Applied Pharmacology 53,279−284(1980))。光電流を、光学スイッチを使用して、300Wキセノンランプおよび470±20nmまたは580±20nmいずれかのバンドパスフィルターを用いて誘発させた。検体における光パワーは、11.52mWmM−2(470nm)または10.64mWmM−2(580nm)であった。電流を2kHzでフィルターし、50kHzでデジタル化し、pClamp10ソフトウェア(Axon Instruments)を使用してディスクに記録した。データは、平均±平均値の標準誤差として表され、統計的有意性を、適宜、対応のあるまたは対応のないt検定を使用して決定した。MSNにおけるIPSC測定については(図2B〜Eおよび図5A〜B)、光を用いずに10回反復し、その後、光を用いて10回反復前した。各反復は、5秒間の長さであり、5秒間の休息期間によって分離された。細胞切片中のChATのコカイン応答を試験するために(図4A〜C)、全細胞記録を、発火の上昇が図4に要約されるように可変であったが、対照条件における発火の典型的なランダウンと対照をなした、内殻の腹側部分から得た。探査的研究は、コアおよび殻中のその他の場所におけるChAT細胞がコカインに対して応答性がより低かったことを示唆した。
同時光刺激および細胞外電気的記録を、以前に記載されたように行った(Gradinaru et al.,J.Neurosci 27,14231−14238(2007))。オプトロードは、電極の先端がファイバーを超えて300〜500μm突出した、光ファイバー(300μmコア直径、0.37N.A.)に接着されたタングステン電極(1ΜΩ、0.005in、パリレン絶縁)からなっていた。電極を、NAcを通じておよそ100μmの増分で低下させ、光学応答を、各増分につき記録した。光ファイバーは473nmまたは560nmレーザーに連結させた。パワー密度は、双方の波長について、ファイバー先端で約140mW/mm2であり、それは、470nm光について約7〜17mW/mm2および560nm光について約10〜22mW/mm2である、電極の先端における密度に対応した。シグナルを増幅し、バンドパスフィルター(300Hz低域カットオフ、10kHz高域カットオフ)処理した後、デジタル化およびディスクへの記録を行った。各部位につき、5回の刺激反復を提示し、保存した。各刺激エポックは、各反復の開始時間の間で80〜90秒間の回復期間を用いながら10〜15秒間持続し、各反復について50秒のデータをディスクに記録した。
最初に、MSNにおけるシナプス後電流を、急性NAc切片において、図1にあるように標的とされた、ChR2−eYFPを発現するChAT細胞の光遺伝学的光刺激中に監視した(ChR2−eYFP非発現細胞)(図2A)この設定においてChATニューロンを刺激することにより、MSNにおいて記録されたγ−アミノ酪酸A型(GABAA)受容体媒介型抑制性シナプス後電流(IPSC)の周波数は強固に増加された(図2BおよびC)。誘発抑制性電流は、一般に、6msのモダルレイテンシ(modal latency)を伴って光パルスと同期し(図2D)、非同期性IPSCのより小さい強化を伴った(図5A〜C)。全ての記録された細胞にわたって、MSNにおいて観察されたIPSCの平均周波数は、ChATニューロンの光刺激中に525.8±154.3%増加した(n=7、平均±標準誤差、P=0.01、対応のあるt検定)一方で、平均IPSC振幅は、影響を受けなかった(P>0.05、対応のあるt検定、n=7、図2E)。この効果は、ニコチン性アンタゴニストメカミラミンによって減弱された(図7、n=5、P<0.05、対応のあるt検定)。
次に、生体内Cre依存性eNpHR3.0発現を用いて、ChAT介在ニューロンを特異的に抑制することの結果を探索した。
マウス、オプトロード記録、および脳切片を、上記のように調製した。
ChATニューロン興奮で観察されたものとは対照的に、NAcニューロンの発火は、典型的に、ChAT細胞の光遺伝学的抑制によって、MSNにおいて高い可能性で増加した。代表的な細胞は図3Aに示される)。パワースペクトル分析により、これらの生体内記録において、約4Hzの発火パターンにおけるストライキング周波数ピークが明らかとなった(図3B)。要約データは、図3Cに提示される。有意に変調された部位の集団にわたって、ほとんどのニューロンは、ChATニューロンの光遺伝学的抑制によって興奮させられた(n=17)。我々は、稀有な推定ChAT介在ニューロンから単一ユニット記録を得ることができ、それはeNpHR3.0によって完全にシャットダウンされ(図3D)、ChAT介在ニューロン(37)の長い活動電位持続期間特性を示した(この細胞については2.0msであるが、我々の記録におけるMSNについての発火持続期間は1.1〜1.7msの範囲であった)。要約データ(図3E)は、全ての記録された部位についての興奮および抑制の力学を示し、それは興奮の優性パターンを例証している(発火は、光によって興奮させられた部位において130.5+/−17.5%増加した)。まとめると、ChATニューロンの体内光遺伝学的興奮および抑制からの結果は、切片生理学からの我々の知見と一致し、これらの稀有な細胞についての、NAcにおける局所回路活性を制御することにおける驚くほどに強力な役割を指摘している。
この強力なNAc制御機構が、自由に移動しているマウスにおける側坐依存性報酬行動と関連するかどうかを試験することを決定した。急性NAc切片におけるこれらの識別されたChATニューロンの活性に対する、急性投与されたコカインの最初の効果を試験した。次に、eNpHR3.0を使用して、動物が環境とコカインを関連付けることを学習する、コカイン場所嗜好条件付け(CPP)の報酬関連行動における、ChAT細胞のこのコカイン誘発型活性またはベースライン活性のいずれかにおける因果的役割について試験した。
場所嗜好条件付け
全ての行動実験を、動物の暗(活動)サイクル中のウイルス注射の4〜6週間後に行った。場所嗜好条件付け(CPP)プロトコルは、不偏の、平衡のとれた場所嗜好の以前の報告からのものと同様であった(Bardo et al.,Neurosci Biobehav Rev 19,39−51(1995))。CPP器具は、一方の側面のコンパートメントが23cm×26cmの寸法で、黒色壁および格子の床を備え、中央コンパートメントが23cm×11cmの寸法で、透明プレキシガラス壁およびプレキシガラス床を備え、他方の側面のコンパートメントが23cm×26cmの寸法で、白色壁および穴のあいた金属床を備えた、長方形の部屋からなっていた。各試験日のマウス位置を、ビデオトラッキングシステムを使用して監視した。床は、マウスが特定の部屋に対する平均ベースライン偏りを示さないように選択し、ある部屋への強力な初期嗜好を有する(1日目に部屋の中で費やされた差異が5分間を超える)いずれのマウスも排除した。CPP試験は、次のものからなっていた。1日目、各マウスを中央室に配置し、器具全体を20分間自由に探索させた(前試験)。2日目は、条件付けからなっていた。朝に、各マウスを、側面室のうちの一方に閉じ込め、午後に、他方の側面室に同じ期間にわたって閉じ込めた。コカインCPP実験のために、対象に腹腔内コカイン注射を受容させた(別途明記されない限り20mg/kg)後、一方の部屋に配置した一方で、対象に同等量の腹腔内食塩水注射を受容させた後、他方の部屋に配置した。(この濃度のコカインは、対照動物において1日の訓練で強固な条件付けを可能にし、光遺伝学的介入を促進する)。マウスに、それらがコカイン注射と対になった部屋を探索した20分間中、黄色光または青色光のいずれかを受容させた一方で、それらに、他方の部屋を探索している時に光を発光していない「模造の」ファイバーを接続した。青色光(470nm)の強度を、ファイバー先端で140〜200mW/mm2のパワー密度を発生するように選定したが、それはNAcの中央における約4〜7mW/mm2のパワー密度に対応するはずである。黄色光(590nm)の強度を、ファイバー先端で70〜140mW/mm2のパワー密度が存在するように選定したが、それはNAcの中央における約3.5〜7mW/mm2のパワー密度に対応するはずである。3日目、1日目と全く同様に、マウスを中央室に配置し、器具全体を20分間自由に探索させた(後試験)。コカインを伴わなかったCPP実験を同一に行ったが、コカインまたは食塩水の腹腔内注射を省略したことを例外とする。
オープンフィールド試験を、オープンプラスチックアリーナで行った(50cm長さ×50cm幅×40cm深さ)。マウスを、部屋の中心に個々に配置し、3分間自由に探索させた。自動ビデオトラッキングシステム(Viewer II,BiObserve)を使用して、フィールドの中心および周辺両方における活動を測定した。中心における時間は、マウスが、オープンフィールドの中心35×35cm領域で費やした時間を指す。
恐怖条件付け器具は、ショック発生器およびスクランブラに配線された格子床を備え、音響室によって囲まれた、正方形条件付けケージ(18×18×30cm)からなっていた。部屋の最上部を、ファイバーのための開口部を導入することによって、訓練中に光送達を可能にするように修正した。全てのマウスに、訓練中、連続黄色光を受容させたが、翌日の試験中には受容させなかった(590nm、CPP実験の場合と同じパワー密度)。恐怖条件付けを誘発するために、マウスをケージに120秒間配置し、純粋なトーン(2.9kHz)を次いで20秒間再生し、続いて即座に2秒間の足部ショック(0.5mA)を行った。この手順を反復し、第2のショックの30秒後、マウスをそれらのホームケージに戻した。すくみ(完全な不動)を、条件付け日の最初のトーン前に30秒間定量化して、ベースラインすくみを査定し、同様に条件付け日の最後のショック直後に30秒定量化して、即座のすくみを査定した。前後関係上のおよび聴覚合図による恐怖条件付けを、条件付けの翌日に査定した。前後関係上の恐怖条件付けを試験するために、マウスを元の条件付けケージに配置し、すくみを5分間測定した。聴覚合図による恐怖条件付けを試験するために、マウスを、異なる前後関係、すなわちプレキシガラス床を備えるピラミッド形のケージに配置した。新規の環境の影響に対する対照として、すくみをこの新たなケージ中で2.5分間測定し、次いで2.9kHzトーンを2.5分間再生し、その間、条件付けされたすくみを測定した。
腹側内側NAc ChAT細胞中で、コカインは、自発的発火を顕著に増加させることが見出された(代表的なChATニューロンは、図4、AおよびBに示される)。要約データにより、コカインが、ChATニューロンにおいて、10分時点で発火率を0.60±0.41Hzから1.74±0.56Hzまで増加させた一方で(n=7、P<0.005、対応のあるt試験)、ビヒクルのみを受容する細胞の対照群においては、発火率は、同じ時間枠にわたって0.69±0.24Hzから0.09±0.09Hzまで減少したことが明らかとなった(n=6、2つの群を比較してP<0.05、両側t検定)(図4C)。

まとめて、ChATニューロン集団の特異的および急性光遺伝学的制御からのこれらの結果は、自由に移動している哺乳動物において、局所回路活性を制御することおよび報酬関連行動を実施することにおける、これらの稀有かつまばらに分布したニューロンについての強力な役割を指摘する。ChAT介在ニューロンの急性サイレンシングが、薬物の不在下で、場所嗜好に影響を及ぼすことなく薬物関連学習を妨害するという事実は、この超小型回路に対する制御を使用して、一般に欲求性または嫌悪性反応に影響を及ぼすことなく、乱用薬物の嗜癖特性を選択的に妨害することが可能であるという興味深い可能性、つまり莫大な臨床的便益となる可能性を示唆する。興味深いことに、行動的結果は、コリン作動性介在ニューロンの慢性から生じる結論を支持しないが(34)、代わりに、NAcにおけるより速いがより低い細胞標的型の薬理学的変調から生じる解釈とより一致している(23〜26)。コリン作動性介在ニューロンのアブレーションは、NAcにおけるドーパミンの代償性増加等の間接的な効果につながる可能性があり、それが今度はコカイン報酬を強化する可能性がある。実際、急性操作と慢性操作との間の基本的な差異は、ニコチンにおける急性増加(恐らく、コリン作動性受容体に作用する)が、ドーパミンにおける対応する急性増加を引き起こす(54、55)一方で、ドーパミンまたはアセチルコリンレベルにおける慢性変化が、例えば、パーキンソン病のドーパミン枯渇において見られるような(57)、他の神経変調物質のレベルにおける対向する変化を引き起こすことができる(56)という知見等の、脳内のアセチルコリン/ドーパミンバランスの我々の理解における、臨床的に関連する明らかな矛盾を説明し得る。

Claims (5)
- 個体における報酬関連行動を妨害する方法に使用するための組成物であって、光応答性オプシンタンパク質をコードするポリヌクレオチドを含み、光応答性オプシンタンパク質が配列番号1に対して少なくとも95%のアミノ酸配列同一性を有するアミノ酸配列を含み、かつ、膜輸送シグナル及び小胞体(ER)移行シグナルを含み、光応答性オプシンタンパク質が個体の側坐核または線条体におけるコリン作動性介在ニューロンの細胞膜に発現されるように個体の側坐核または線条体に直接投与され、当該タンパク質は光に応答性であり、介在ニューロンが光で照射された時に介在ニューロンの膜過分極を誘発することが可能であり、それによって、光による前記タンパク質の活性化が個体における少なくとも1つの報酬関連行動を妨害する、組成物。
- 個体における報酬関連行動が薬物嗜癖であり、個体が薬物を摂取することをもはや所望しなくなる、請求項1に記載の組成物。
- 薬物嗜癖がコカイン嗜癖、オピオイド嗜癖、又はメタンフェタミン嗜癖である、請求項2に記載の組成物。
- ポリヌクレオチドが、アデノ随伴ウイルスベクター、レトロウイルスベクター、アデノウイルスベクター、単純ヘルペスウイルス(HSV)ベクター、およびレンチウイルスベクターから成る群より選択されるウイルスベクターであってもよいベクターである、請求項1〜3のいずれか1項に記載の組成物。
- 膜輸送シグナルがアミノ酸配列KSRITSEGEYIPLDQIDINVを含み、ER移行シグナルがアミノ酸配列FCYENEVを含む、請求項1に記載の組成物。
