Description

本発明は、HLA−B*27重鎖のアルファ−1、アルファ−2、および/またはアルファ−3のIgドメイン内に見出される1つまたはそれ以上のアミノ酸と相互作用する抗HLA−B*27抗体を包含する。HLA−B*2705(配列番号385)およびHLA−B*2709(配列番号386)の場合、アルファ−1のIgドメインは、対応する完全な重鎖アミノ酸配列のアミノ酸25〜114からなり、アルファ−2のIgドメインは、対応する完全な重鎖アミノ酸配列のアミノ酸115〜206からなり、アルファ−3のIgドメインは、対応する完全な重鎖アミノ酸配列のアミノ酸207〜298からなる。抗体が結合するエピトープは、HLA−B*27重鎖の1つまたはそれ以上のIgドメイン内に位置する3つまたはそれより多くの(例えば、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20またはそれより多くの)アミノ酸の単一の連続した配列からなっていてもよい。あるいは、このようなエピトープは、HLA−B*27重鎖の1つまたはそれ以上のIgドメイン内に位置する複数の連続していないアミノ酸(またはアミノ酸配列)からなっていてもよい。また本発明の抗体は、β2m配列(配列番号388)内の1つまたはそれ以上のアミノ酸とも相互作用する可能性がある。
完全ヒトモノクローナル抗体等のモノクローナル抗体の作成方法は当技術分野で公知である。そのような公知の任意の方法を本発明と関連して用いてヒトHLA−B*27に特異的に結合するヒト抗体を作ることができる。
本発明の抗HLA−B*27抗体および抗体フラグメントは、説明した抗体の配列とは異なるがヒトHLA−B*27に結合する能力を保持するアミノ酸配列を有するタンパク質を包含する。そのような変異抗体および抗体フラグメントは、親配列と比較した場合にアミノ酸の1個またはそれ以上の付加、欠失または置換を含むが、説明した抗体の生物化学的活性と本質的に同等である生物学的活性を示す。同様に、本発明の抗HLA−B*27抗体をコードするDNA配列は、開示した配列と比較した場合にヌクレオチドの1個またはそれ以上の付加、欠失または置換を含むが本発明の抗HLA−B*27抗体または抗体フラグメントと本質的に生物学的に同等である抗HLA−B*27抗体または抗体フラグメントをコードする配列を包含する。そのような変異アミノ酸およびDNA配列の例は上記で論じられている。
本発明は、細胞毒素、化学療法薬、免疫抑制剤または放射性同位体等の治療部分に結合した抗HLA−B*27モノクローナル抗体(「免疫結合体」)を包含する。細胞毒性剤として、細胞に有害である任意の薬剤が挙げられる。免疫結合体の形成に適した細胞毒性剤および化学療法剤の例は当技術分野で公知である(例えばWO05/103081参照)。
本発明の抗体は、単一特異性、二重特異性または多重特異性であり得る。多重特異性抗体は、1つの標的ポリペプチドの異なるエピトープに特異的であってもよく、または1超の標的ポリペプチドに特異的な抗原結合ドメインを含有してもよい。例えばTutt他、J.Immunol.147:60〜69ページ(1991);Kufer他、Trends Biotechnol.22:238〜244ページ(2004)参照。本発明の抗HLA−B*27抗体を別の機能性分子(例えば別のペプチドまたはタンパク質)に連結させることができ、または機能性分子と共発現させることができる。例えば、抗体またはそのフラグメントを別の抗体または抗体フラグメント等の1つまたはそれ以上の別の分子的実在物に(例えば化学的カップリング、遺伝子融合、非共有結合性会合または別の方法によって)機能的に連結させることにより、第2の結合特異性を備えた二重特異性抗体または多重特異性抗体を産生することができる。例えば、本発明は、免疫グロブリンの一方のアームがHLA−B*27またはそのフラグメントに特異的であり、免疫グロブリンの他方のアームが第2の治療標的に特異的であるかまたは治療部分に結合されている二重特異性抗体を含む。
本発明は、本発明の抗HLA−B*27抗体またはその抗原結合フラグメントを含む医薬組成物を提供する。本発明の医薬組成物は、適切な担体、賦形剤および移送、送達、耐性等を改善する別の薬剤と共に製剤化される。全ての薬剤師に公知の処方集:Remington’s Pharmaceutical Sciences、Mack Publishing Company、Easton、PA中に多数の適切な製剤を見出すことができる。これらの製剤として、例えば粉末、ペースト、軟膏、ゼリー、ワックス、油、脂質、脂質(カチオン性またはアニオン性)含有ベシクル(LIPOFECTIN(商標)等)、DNA接合体、無水吸収ペースト、水中油型のおよび油中水型のエマルション、エマルション・カーボワックス(様々な分子量のポリエチレングリコール)、半固体ゲルならびにカーボワックスを含有する半固体混合物が挙げられる。Powell他、「Compendium of excipients for parenteral formulations」PDA(1998)J Pharm Sci Technol 52:238〜311ページも参照。
本発明の抗体(およびそれらを含む医薬組成物)は、HLA−B*27活性に関連する、もしくはHLA−B*27活性が介在するあらゆる疾患または障害、またはHLA−B*27が介在する抗原提示を遮断もしくは弱化することによって処置可能なあらゆる疾患または障害の処置、予防、および/または改善に特に有用である。本発明の抗HLA−B*27抗体を用いて処置が可能な例示的な疾患および障害としては、例えば、強直性脊椎炎、反応性関節炎(ライター症候群)、急性前部ブドウ膜炎、筋実質炎、乾癬性関節炎、潰瘍性大腸炎等などの脊椎関節症が挙げられる。本発明の抗HLA−B*27抗体を用いて予防、処置および/または改善が可能な追加の疾患および障害としては、固形臓器移植の拒絶、および移植片対宿主病(GVHD)が挙げられる。
本発明の特定の実施形態によれば、決められた時間経過で抗HLA−B*27抗体の複数回の用量を対象に投与してもよい。本発明のこの形態に係る方法は、抗HLA−B*27抗体の複数回の用量を対象に連続投与することを含む。「連続投与」は、本明細書で用いられる場合、異なる時点で、例えば予め決められた間隔(例えば、数時間、数日、数週間または数カ月)で隔てられた異なる日に、抗HLA−B*27抗体の各用量を対象に投与することを意味する。本発明は、1回の初回用量の抗HLA−B*27抗体、それに続いて1回またはそれ以上の第二の用量の抗HLA−B*27抗体、場合によりそれに続いて1回またはそれ以上の第三の用量の抗HLA−B*27抗体を患者に連続投与することを含む方法を包含する。
本発明は、少なくとも1種の追加の治療活性を有する成分と組み合わせて、本発明の抗HLA−B*27抗体を投与することを含む治療用投与レジメンを包含する。そのような追加の治療活性を有する成分の非限定的な例としては、他のHLA−B*27遮断剤、例えば、第2の抗HLA−B*27抗体などが挙げられる。本発明の抗HLA−B*27抗体と組み合わせた投与が有益な可能性がある他の物質としては、サイトカイン阻害剤(例えば、小分子サイトカイン阻害剤、および例えばIL−1、IL−2、IL−3、IL−4、IL−5、IL−6、IL−8、IL−9、IL−11、IL−12、IL−13、IL−17、IL−18などのサイトカインまたはそれぞれ個々の受容体に結合する抗体等)、抗ウイルス剤、抗生物質、鎮痛薬、コルチコステロイド、ならびに/またはNSAIDが挙げられる。
また本発明の抗HLA−B*27抗体を使用して、例えば診断目的で、サンプル中のHLA−B*27またはHLA−B*27発現細胞を検出および/または測定できる。例えば、抗HLA−B*27抗体、またはそのフラグメントを使用して、HLA−B*27対立遺伝子サブ変異体(例えば、HLA−B*2705またはHLA−B*2709)の存在を特徴とする状態または疾患を診断できる。HLA−B*27に関する例示的な診断アッセイは、例えば患者から得たサンプルを本発明の抗HLA−B*27抗体と接触させることを含むことができ、抗HLA−B*27抗体は検出可能な標識またはレポーター分子で標識されている。あるいは、それ自体が検出可能に標識されている二次抗体と組み合わせて、標識されていない抗HLA−B*27抗体を診断用途で用いることができる。検出可能な標識またはレポーター分子は、3H、14C、32P、35Sもしくは125I等の放射性同位体;フルオロセイン・イソチオシアネートもしくはローダミン等の蛍光部分もしくは化学発光部分;またはアルカリ・ホスファターゼ、ベータ−ガラクトシターゼ、西洋ワサビ・ペルオキシダーゼもしくはルシフェラーゼ等の酵素であることができる。サンプル中のHLA−B*27を検出するためにまたは測定するために用いることができる特定の例示的なアッセイとして、酵素結合免疫吸着アッセイ(ELISA)、ラジオイムノアッセイ(RIA)および蛍光活性化細胞選別(FACS)が挙げられる。
ヒトHLA−B*27に対するヒト抗体の作成
抗HLA−B*27抗体を作成するために、可溶性HLA−B*27融合タンパク質(HLA制限ペプチドに融合したHLA−B*2705重鎖細胞外ドメインおよびβ2m配列を含む)、またはHLA−B*27を発現する組織/細胞のいずれかを用いた、加えてその組み合わせを用いた標準的な方法に従ってVELOCIMMUNE(登録商標)マウス(例えば米国特許第6,596,541号を参照)を免疫化した。HLA−B*27特異的なイムノアッセイによって抗体免疫反応をモニターした。所望の免疫反応が達成されたら、脾細胞を回収してマウス骨髄腫細胞と融合させ、その生存率を保ち、ハイブリドーマ細胞株を形成した。ハイブリドーマ細胞株をスクリーニングして選択することにより、HLA−B*27特異的抗体を産生する細胞株を同定した。この技術を使用して、数種の抗HLA−B*27キメラ抗体(すなわち、ヒト可変ドメインとマウス定常ドメインとを有する抗体)を得た;この方式で作成された代表的な抗体を以下のように名付けた:2477N、2490N、2482N、2477N2、2490N2、2482N2、2480N、2497N、2491N、2499N、および2718N。
重鎖および軽鎖可変領域アミノ酸配列
表1に、選択された抗HLA−B*27抗体の重鎖および軽鎖可変領域(HCVRおよびLCVR)ならびに相補性決定領域(CDR)に関するアミノ酸配列の識別子(配列番号)を記載する。
表面プラズモン共鳴によって得られたヒトモノクローナル抗HLA−B*27抗体の結合親和性および速度定数
リアルタイム表面プラズモン共鳴バイオセンサーアッセイ(Biacore T200、3000、またはT100−2)を使用して、精製した抗HLA−B*27抗体に対する抗原結合に関する会合速度定数および解離速度定数(それぞれkaおよびkd)と計算された平衡解離定数および解離半減期(それぞれKDおよびt1/2)を決定した。以下の構成要素:N末端のHLA制限ペプチド配列(SRYWAIRTR、配列番号387のアミノ酸1〜9);続いて第1のリンカー配列(配列番号387のアミノ酸10〜19);続いてヒトベータ−2ミクログロブリン(β2m)配列(配列番号387のアミノ酸20〜118);続いて第2のリンカー配列(配列番号387のアミノ酸119〜133);続いてHLA−B*2705重鎖の細胞外領域(配列番号387のアミノ酸134〜409);およびmyc−myc−ヘキサヒスチジンC末端タグを有する終端(配列番号387のアミノ酸410〜437)からなる加工されたHLA−B*27細胞外ドメインを有するインライン融合タンパク質(「scHLA−B27−mmH」、配列番号387)への結合に関して抗体を試験した。
インビトロで抗HLA−B*27抗体はHLA−B*27によって誘導されるT細胞活性化を遮断する
2種の哺乳動物細胞株:HLA−AおよびHLA−B分子を低い内因性レベルで発現するかまたはそれらの内因性レベルがゼロであることがこれまでに示されているBリンパ芽球様細胞株であるC1R−neo(ATCC、番号CRL−2369);およびJurkatヒトCD4+T細胞株から得られた混合培養物を利用することによりバイオアッセイを行って、HLA−B*27−ペプチド複合体と対応するT細胞受容体(TCR)との相互作用により誘導されたT細胞活性化を測定した。
HLA−B*27発現細胞への抗HLA−B*27抗体の結合を確認するためのフローサイトメトリー分析
フローサイトメトリー分析を利用して、HLA−B*27+細胞への抗HLA−B*27抗体の結合を確認した。試験した細胞は以下の通りである:(a)C1R−neo−B*2705細胞株;(b)HLA−B*27トランスジェニックマウスからの脾細胞;および(c)HLA−B*27+個体からの一次ヒト末梢血単核細胞(PBMC)。C1R−neo−B*2705細胞株は、HLA−B*2705対立遺伝子の重鎖およびヒトベータ2ミクログロブリンを過剰発現するように安定してトランスフェクトされた親C1R−neo細胞株の派生株である(実施例4を参照)。トランスフェクトされていないC1R−neo親細胞を陰性対照として使用した。HLA−B*2705の重鎖およびヒトβ2ミクログロブリンのトランスジェニックヘテロ接合型発現によりトランスジェニックマウス株[C57BL/6J−Tg(HLA−B27)30−4Trgマウス株;Jackson Laboratory、ストック番号003440]を作成した。野生型同腹子からの脾細胞を、ヘテロ接合型HLA−B*27+脾細胞の陰性対照として使用した。HLA−B*27陽性として一次ヒトPBMCを遺伝子型解析し、市販の抗HLA−B*27抗体(HLA ABC Ab;Millipore番号MAB1285F)で染色することによってHLA−B*27対立遺伝子の発現を確認した。HLA−B*27-ドナーからのPBMCを陰性対照として使用した。
末梢血単核細胞(PBMC)へのHLA−B*27抗体の結合
HLA−B*27以外の対立遺伝子を発現することが示されているPBMCを使用して追加のフローサイトメトリー実験を行った。4人のドナーからのヒト血液をK2−EDTA抗凝血薬チューブ(BD、番号366643)に収集した。QIAamp DNA血液ミニキット(Qiagen、番号51304)を使用して、全血サンプルからゲノムDNAを製造元の説明書の通りに精製した。次いでゲノムDNAサンプルを増幅し、次のHLA遺伝子座特異的シーケンス反応を、SeCoreキット(Invitrogen、遺伝子座Aは番号5300925、遺伝子座Bは番号5311925D、遺伝子座Cは番号5320925)を使用して実行した。ABI3730xL DNA解析装置を使用してDNA配列を決定し、uType SBT HLA配列決定ソフトウェア(Invitrogen、番号53999−1)を使用してHLAタイプを判断した。
表面HLA*BへのHLA−B*27抗体の結合
フローサイトメトリー分析をさらに利用して、様々なHLA*B対立遺伝子を過剰発現するように加工された細胞株に対する抗HLA抗体の結合特異性を決定した。様々なHLA対立遺伝子を発現する細胞株を作成するために、親K562(不死化ヒト骨髄性白血病)細胞をDNAプラスミドでエレクトロポレーションして、23種の異なるHLA対立遺伝子(表11に示される)と、ネオマイシンまたはハイグロマイシンのいずれかに対する抗生物質耐性遺伝子との安定な統合を促した。次いで、抗生物質による選択条件下にエレクトロポレーションした細胞株を2.5週間から5週間置き、特異的なHLA対立遺伝子を発現する細胞を選択した。フィコエリトリン(Biolegend、番号311406)にコンジュゲートしたHLA−A、B、C pan特異的抗体(クローンW6/32)を使用して、得られたK562/HLA細胞株をFACSでソートして、最大レベルの表面HLA(発現するもののうち上位10%)を示す集団を得た。
HLA−B*27対立遺伝子サブ変異体に対する抗体の優先結合
電気化学発光検出技術を使用して、細胞表面のヒトHLA−B*27対立遺伝子サブ変異体(すなわち、HLA−B*2705およびHLA−B*2709)に結合する抗HLA−B*27抗体の能力を、そのHLA−B*07対立遺伝子変異体への結合能力と比較して測定した。これらの研究のために、HLA−B*07重鎖(CR1−B7、ATCC番号CRL−2371)、HLA−B*2705重鎖(配列番号385のアミノ酸25〜362)、またはHLA−B*2709重鎖(配列番号386のアミノ酸25〜362)のいずれかを発現するように安定してトランスフェクトされたC1R−neo細胞を使用した。それぞれのHLA−B*発現細胞株はさらに、HLA重鎖と非共有結合により複合体を形成するヒトベータ2ミクログロブリン(β2m;配列番号388のアミノ酸21〜119)も発現した。
インビボで決定された抗HLA−B*27抗体の活性
インビボでの抗HLA−B*27抗体の活性を調査した。DNAの流体力学的送達(HDD)によりJackson Laboratoryから得られたC57BL/6野生型(WT)マウスの肝臓で、HLA−B*27(UniProt受託番号P03989のアミノ酸25〜362)およびヒトベータ2ミクログロブリン(β2m;GenBank受託番号NP_004039のアミノ酸21〜119)を過剰発現させた。HDD実験のために、WTマウスをコホート1つあたりマウス2〜5匹のグループに分け、各マウスに、HLA−B*27を発現するプラスミド50μgおよびβ2mを発現するプラスミド50μg、または空の対照プラスミド50μgのいずれかを注射した。HDD注射から3日目および10日目に、4つのコホートのそれぞれのマウスに、30mg/kgの抗HLA抗体H4H2542P、H4H2499NもしくはH4H2482P、またはアイソタイプ対照抗体を腹腔内注射した。HDDから14日目にマウスを処分し、コラゲナーゼ処置とパーコールでの遠心分離により肝臓に浸潤する細胞を単離した。ACK溶解緩衝液(Gibco、番号A1492−01)を使用して肝臓の赤血球を溶解させた。浸潤免疫細胞を、生細胞/死細胞色素(Invitrogen、番号L34957)と抗CD45抗体(eBioscience、番号12−0451−82)とで染色して浸潤白血球を検出し、さらに抗CD3(BD Biosciences、番号552774)、抗CD4(Biolegend、番号100414)、および抗CD8(Biolegend、番号100734)抗体で染色して、様々なT細胞群を同定した。FACS Cantoでフローサイトメトリーを行い、データをFlowJoソフトウェアを使用して分析した。処分された動物の肝臓への、一般的には免疫細胞、特定にはCD8+およびCD4+細胞の浸潤レベルを、頻度に関して、すなわち(例えば肝細胞とは逆に)コラゲナーゼ処置とパーコールでの遠心分離を用いて単離された肝臓からのインタクト細胞/生細胞(リンパ球、CD8+細胞、またはCD4+細胞)のパーセンテージとして測定した。
それゆえに、抗HLA抗体H4H2499Nは、HDDによりHLA−B*27とβ2mとを過剰発現するマウスの肝臓への炎症細胞の浸潤を遮断できる。
抗HLA−B*27抗体のヒスチジン置換変異体の構築
pH依存性の結合特性(すなわち、中性pHと比較して低いpHにおける結合の低下)を有し、向上したインビボでの有効性(例えば、より長い抗体の血清中半減期、T細胞活性化の持続的な阻害等)を有する抗HLA−B*27抗体変異体を作成する試みにおいて、一連の変異抗体を構築した。特に、各抗体の相補性決定領域(CDR)内の(ヒスチジン以外の)各アミノ酸をそれぞれヒスチジンに突然変異させた親抗体H4H2477N2およびH4H2499Nの突然変異体バージョンを構築した。表1で示されるように、親H4H2477N2抗体の重鎖可変領域(HCVR)は、配列番号50のアミノ酸配列を含み、親H4H2477N2抗体の軽鎖可変領域(LCVR)は、配列番号58のアミノ酸配列を含み、親H4H2499N抗体の重鎖可変領域(HCVR)は、配列番号146のアミノ酸配列を含み、親H4H2499N抗体の軽鎖可変領域(LCVR)は、配列番号154のアミノ酸配列を含み;親H4H2477N2およびH4H2499N抗体のCDR配列は、それぞれ表19Aおよび19Bに表される。
HLA−B*27発現細胞に対する抗HLA−B*27抗体のヒスチジン置換変異体のpH選択的な結合
2つの抗HLA抗体、H4H2477N2およびH4H2499NのCDR領域における親アミノ酸(単数または複数)の単一部位および複数部位にヒスチジン置換を行って、pH選択的結合を増加させた(実施例10を参照)。電気化学発光検出技術を同様に使用して、pH7.2およびpH6.0の両方におけるC1R−neoHLA−B*2705細胞上で発現された細胞表面のヒトHLA−B27への結合に関して、これらのヒスチジン変異体およびそれらの親抗体を試験した。上述したようにして、細胞を回収し、計数し、96ウェルの炭素電極プレートに植え付け、遮断した(実施例8を参照)。pH6.0での結合を決定するために、抗体添加の前に、プレートに結合した細胞をpH6.0の緩衝液[50mMのMES、0.0025MのKCl、0.137MのNaCl、0.0005MのMgCl2×6H2O、0.0009MのCaCl2、0.5%(w/v)BSA;5MのNaOHの添加によりpH6.0に調節]ですすいだ。pH7.2で結合を決定するための中性pH緩衝液[PBS+0.5%(w/v)BSA;pH7.2]中、またはpH6.0での結合を決定するためのpH6.0緩衝液中のいずれかで、抗HLA抗体を50nMから開始して希釈し、12ポイントで3倍の連続希釈液を得て、次いでプレートに結合した細胞に移し、25℃で1時間インキュベートした。AquaMax2000プレートウォッシャー(MDS Analytical Technologies)を使用して未結合の抗体を洗い落とした。SULFO−TAG(商標)とコンジュゲートした抗ヒトCH1特異的抗体(社内で作成)を使用して細胞に結合した抗HLA抗体を検出した。MSD読取り用緩衝液の添加、電気化学刺激、および発光検出も前述したようにして行った。発光強度(RLUとして示される)は、細胞に結合した抗体の量との相関を示した。ヒスチジン変異体に関するC1R−neoB*2705細胞への抗HLA抗体H4H2499NおよびH4H2477N2の結合のEC50値をPrismソフトウェアを使用して計算し、それぞれ表21および22に示した。
Claims (6)
- HLA−B * 27に結合し、HLA−B * 27によって誘導されるT細胞活性化をインビトロで遮断する単離抗体またはその抗原結合フラグメントであって、抗体または抗原結合フラグメントは、それぞれ:配列番号148−150−152−156−158−160のアミノ酸配列を有するHCDR1−HCDR2−HCDR3−LCDR1−LCDR2−LCDR3領域を含む、単離抗体または抗原結合フラグメント。
- 抗体または抗原結合フラグメントは、配列番号146/154のHCVR/LCVRアミノ酸配列対を含む、請求項1に記載の単離抗体または抗原結合フラグメント。
- pH5.75において、pH7.2におけるHLA−B*27への抗体結合のKDよりも少なくとも5倍大きいKDでHLA−B*27と結合し、HLA−B * 27によって誘導されるT細胞活性化をインビトロで遮断する単離抗体またはその抗原結合フラグメントであって、前記抗体またはその抗原結合フラグメントは、配列番号226/234、258/266、290/298、338/346、および370/378からなる群より選択されるHCVR/LCVRアミノ酸配列対を含む、単離抗体または抗原結合フラグメント。
- 請求項1または2に記載の抗体または抗原結合フラグメントと、薬学的に許容される担体または希釈剤とを含む医薬組成物。
- 脊椎関節症を処置する、予防する、または改善する方法において使用するための、請求項4に記載の医薬組成物。
- 脊椎関節症は、強直性脊椎炎、反応性関節炎(ライター症候群)、急性前部ブドウ膜炎、筋実質炎、乾癬性関節炎、および潰瘍性大腸炎からなる群より選択される、請求項5に記載の医薬組成物。
