Description
−lipoplexes(核酸とカチオン脂質の間で形成された複合体)、例えば、リポフェクタミン(登録商標)2000、
−polyplexes(核酸とカチオン性重合体の間で形成された複合体、例えば、ポリエチレンイミンPEI)、
−核酸とデンドリマーの間で形成された複合体、例えば、デンドリマー含有ポリ(アミドアミン)PANAM、
−lipopolyplexes(核酸、リポソームと重合体の間で形成された複合体)、例えば、ポリエチレングリコール(PEG)タイプの重合体によって安定させたリポソームを形成するカチオン脂質と補助剤の混合物に相当するSNALPs(<安定した核酸−脂質粒子>)、
−核酸と高分子ポリ(DL−ラクチド−コグリコリド)PLGAナノ粒子の間で形成された複合体、
の使用が引用され得る。
この目的のため、且つ本発明の最初の課題により、連続水相と少なくとも1つの分散相を含んで成る、ナノエマルション形態の製剤を提供する。
そのため、本発明の第一の目的は、連続水相と少なくとも1つの分散相を含ん成り、且つ以下の:
−少なくとも5モル%の両親媒性脂質;
−以下のものを含んで成る15〜70モル%の少なくとも1つのカチオン性界面活性剤:
−以下の中から選択される少なくとも1つの親油性基:
−12〜24個の炭素原子及びフォスファチジルエタノールアミンを有する脂肪酸のエステル又はアミド、及び
−ポリ(酸化プロピレン)、並びに
−1〜12個の炭素原子を有し、且つ少なくとも1つのカチオン性基によって分断及び/又は置換された直鎖又は分岐アルキル基、及び
−少なくとも1つのカチオン性基を含んで成る親水性重合性基、そして、
−少なくとも25単位のエチレンオキシドを有する少なくとも1つのポリ(エチレンオキシド)鎖を含んで成る、10〜55モル%の補助界面活性剤;
−少なくとも1つの脂肪酸グリセリドを含んで成る可溶性脂質;
−任意にヘルパー脂質;
を含んで成り、両親媒性脂質、カチオン性界面活性剤、及び補助界面活性剤のモルパーセンテージが、全体(両親媒性脂質/カチオン性界面活性剤/補助界面活性剤/任意のヘルパー脂質)に対してである、ナノエマルション形態の製剤を提供することである。
本発明の製剤は、以下のもの含んで成るカチオン性界面活性剤を含んで成る:

一実施形態において、本発明の製剤は、エンドソーム膜を不安定化することによってサイトゾル放出を容易にすることができるヘルパー脂質を含んで成る。好ましくは、この脂質は、ジオレイルホスファチジルエタノールアミン(DOPE)である。
製剤は、製剤の液滴のクラウン部に配置された少なくとも1つの両親媒性脂質を含んで成る。
製剤はまた、ナノエマルションの分散相中、より詳しく述べると、液滴のコア部分の中に含まれる少なくとも1つの脂肪酸グリセリドを含んで成る可溶性脂質を含んで成る。
−脂肪酸及び脂肪アルコールのエステル、例えばパルミチン酸セチルなど;又は
−グリセロールの誘導体、特に脂肪酸でのグリセロールのエステル化によって得られたグリセリド、

分散相はまた、液滴のコアに含まれる他の1若しくは複数の油を含んでいてもよい。
分散相はさらに、好適な量の、例えば色素、安定剤、保存料又は他の有効成分などの他の添加剤を含んでいてもよい。
製剤は、ナノエマルションを安定させることができる補助界面活性剤を含んで成る。
−例えば抗体、ペプチド、サッカリド、アプタマー、オリゴヌクレオチドなどの標的化生物学的リガンド、又は例えば葉酸などの化合物;

一実施形態において、製剤は、細胞内又は患者体内の液滴分配、したがって、RNA干渉の内在性メカニズムを調節することができる核酸配列の分配の視覚化を有利に可能にするイメージング剤を含んで成る。
−ポジトロン放出型断層撮影法(PET)(例えば18F、11Cなどの放射性核種、金属カチオンのキレート68Ga、64Cuを含んで成る化合物であると思われるイメージング剤)、
−単光子放射型コンピュータ断層撮影法(SPECT)(放射性核種、例えば123I又はキレート99mTc若しくは111Inを含んで成る化合物であると思われるイメージング剤)、
−磁気共鳴映像法(MRI)(ガドリニウムのキレート、又は例えば酸化鉄のナノ結晶、鉄−プラチナFePtの酸化マンガンなどの磁気ナノ結晶であると思われるイメージング剤)、
−光学映像法又はX線映像法(親油性フルオロフォア、又は、例えばイオパミドールなどのヨード分子、アミドトリアゾエート、若しくは金ナノ粒子であると思われるイメージング剤)。
本発明の製剤は、治療剤を含んでいてもよい。
一般に、液滴の成分に対する液滴のコア中の成分のモル割合は、10〜80%、特に25〜75%、好ましくは33.35〜73.99%である。言い換えれば、分散相に対する(すなわち、分散相のすべての成分に対する)全体(可溶性脂質/任意の油/任意のイメージング剤/任意の親油性治療剤)のモル割合(mol/mol)は、一般に10〜80%、特に25〜75%、好ましくは33.35〜73.99%である。
本発明に使用されるナノエマルションの水相は、水及び/又はリン酸緩衝液、例えばPBS(「リン酸緩衝液生理的食塩水」)若しくは生理的食塩溶液、特に塩化ナトリウムなどの緩衝液から好ましくは形成される。
一実施形態において、製剤の粘性は、25℃で1ポアズ(0.1Pa.s)より高い。

先に規定された<プレミックス>製剤の液滴には、通常、20〜200nmの直径がある。特に、この直径は、MalvernのZetaSizer装置による動的光散乱によって計測される。
−25〜45モル%の補助界面活性剤(25モル%未満では、製剤が安定性の問題を呈する可能性がある);及び/又は
−15〜50モル%のカチオン性界面活性剤、
を含んで成る製剤が好まれる。
−45〜50モル%の補助界面活性剤(45モル%未満では、製剤は、安定性の問題を呈する可能性がある。50%超では、核酸配列との複合体形成後の<最終的な>製剤のトランスフェクションの有効性が低い);及び/又は

先に規定した<プレミックス>製剤のRNA干渉の内在性メカニズムを調節することができる核酸配列との複合体形成によって、前記配列を送達するのに有効である<最終的な>製剤が得られる。
−低分子(short又はsmall)干渉RNA−siRNA;
−ロックド核酸(locked nucleic acid)−LNA;
−マイクロRNA又はmiRNAとも呼ばれる合成マイクロRNA模倣体。
図1で例示されているように、本発明による製剤の液滴は、コア−リング型で構成されており、ここで:
−コアは、以下のものを含んで成り:
−可溶性脂質、

第二の態様によると、本発明は、先に規定されるような<プレミックス>製剤、及び別々に、200未満の塩基を含んで成る一本鎖核酸配列又は200未満の塩基対を含んで成る二重鎖核酸配列である、少なくとも1つの一本鎖又は二重鎖核酸配列を含んで成るキットは、RNA(リボ核酸)干渉の内在性メカニズムを調節することができる。
−液滴のクラウン中のカチオン性界面活性剤のモル割合、
−液滴のクラウン中の両親媒性脂質のモル割合、
−液滴のクラウン中の補助界面活性剤のモル割合、
−補助界面活性剤が少なくとも25エチレンオキシド単位を含んで成るポリ(エチレンオキシド)鎖を含んで成るという事実、
−液滴のコア中の可溶性脂質の存在、及び/又は
−液滴のコアのモル及び/又は重量の割合、
を用いて先に例示した<プレミックス>製剤は、有利に安定している。<プレミックス>製剤のこの安定性は、<プレミックス>製剤の長期保存を可能にし、そしてそれが、先に定義したキットの保存及び販売の前提条件である。
第三の態様によると、本発明は、上記で規定した製剤の調製方法に関する。
(i)可溶性脂質、両親媒性脂質、カチオン性界面活性剤を含んで成る油相を調製する工程;
(ii)補助界面活性剤を任意に含んで成る水相を調製する工程;
(iii)十分な剪断作用下で、水相中に油相を分散させて、<プレミックスが>製剤を形成する工程;続いて
(iv)RNA干渉の内在性メカニズムを調節することができる核酸配列を、形成した<プレミックス>製剤に加える工程;続いて
(v)こうして形成された製剤を回収する工程、
を含んで成る。
一般に、油相の調製は、製剤の油性成分(可溶性脂質/両親媒性脂質/カチオン性界面活性剤)を混合することを含んで成る。製剤が、エンドソーム膜の不安定化によってサイトゾル放出を促す脂質、及び/又は油、及び/又はイメージング剤、及び/又は治療剤を含んで成る場合、これらは、工程(i)の油相に一般に加えられる。
有利なことには、油相は、液体状態で水相中に分散される。相の1つが周囲温度にて凝固する場合、一方の相、又は好ましくは、両方の相を融解温度以上に加熱することによって、混合をおこなうことが望ましい。
−油滴の平均直径は、通常、10nm超、且つ200nm未満、好ましくは30〜190nmであり;そして、
−好ましくは、製剤の水相が0.15mMのNaCl水溶液であるときに、ゼータ電位は、20mV超、通常25mV〜60mV、好ましくは、40〜55mVである。
次に、工程(iv)は、前記核酸配列を送達するのに使用される製剤の調製を、前記核酸配列を工程(iii)の最後に得られた<プレミックス>製剤と複合体形成することによって、可能にする。
−RNA干渉の内在性メカニズムを調節することができる核酸配列の移動の観察を可能にするアガロースゲル電気泳動法。良好な複合体形成により、RNA干渉の内在性メカニズムを調節することができる核酸配列は、より重くなり、ウェル内に見られる。遊離の少ない複合体形成により、RNA干渉の内在性メカニズムを調節することができる核酸配列が別の位置に移動する。
−動的光散乱法(DLS)、流体力学直径により複合体形成の影響について観察することによる。複合体形成が効果的であるほど、プロフィールがより単一様相の分配に向かう傾向がある。
製剤は、例えば、カラム精製又は透析によって精製できる。
これにより得られたエマルションは、任意の希釈後に、使用できる状態である。
製剤が式(I)の界面活性剤を含んで成る実施形態において、RNA干渉の内在性メカニズムを調節することができる核酸配列との複合体形成に使用されるナノエマルションは、以下の:

−G1は、アルデヒドであり、且つG2は、アミン、次にどのGがG'基を含んで成るかで式(XXI)の基を表しながら形成される式(I)の界面活性剤である。

製剤は、有利なことには、安定していてほとんど毒性がない。それは、形質移入剤として、及び/又はRNA干渉の内在性メカニズムを調節することができる核酸配列を送達するためのシステムとして使用できる。
第四の態様によると、本発明は、真核生物細胞に、RNA干渉の内在性メカニズムを調節することができる核酸配列を挿入する方法であって、本発明の製剤と真核生物細胞とを接触させることを含んで成る方法に関する。この方法は、トランスフェクション方法である。
本発明の製剤は、RNA干渉の内在性メカニズムを調節することができる核酸配列のインビトロ又はインビボ送達のシステムである。
−眼疾患、特に加齢性黄斑変性症(AMD)、緑内障又は先天的爪肥厚症;
−癌、固形腫瘍、転移だけでなく、慢性骨髄性白血病も;
−腎臓病、特に移殖又は急性腎不全後のもの;
−高コレステロール血症;
−特にC型肝炎ウイルス(HCV)、ヒト免疫不全ウィルス(HIV)、及び呼吸系発疹ウイルス(RSV)による感染に関連するウイルス病。
本明細書において、「ナノエマルション」という用語は、少なくとも2つの相、一般に油相及び水相を有する組成物であって、分散相の平均的大きさが、1ミクロン未満、好ましくは10〜500nm、特に20〜100nm、最も好ましくは20〜70nmである組成物を意味する(論文C. Solans, P. Izquierdo, J. Nolla, N. Azemar and M. J. Garcia-Celma, Curr Opin Colloid In, 2005, 10, 102-110を参照)。
−炭素環:不飽和(例えば、シクロヘキセン)又は芳香族(すなわち、フェニル)の炭素原子(好ましい飽和炭素環は、特に例えばシクロペンチル又はシクロヘキシルなどのシクロアルキルである)で構成された飽和クラウン。
−複素環化合物:別段の指示がない限り、5〜6個の原子を含んで成り、且つO、N及び/又はSの中から選択される1若しくは複数のヘテロ原子を含んで成るクラウン基である。前記複素環化合物は、飽和であっても、部分不飽和であってもよく、1若しくは複数の二重結合を含んでいてもよい。この場合、ヘテロシクロアルキル基という用語が使用される。それはまた、別段の指示がない限り、5〜6個の原子を含んで成る芳香族であり、とのとき、ヘテロアリール基を表してもよい。
使用した水相は、PBS 1X緩衝溶液であった。

(i)油相の調製:
ダイズ油、suppocire NC、両親媒性脂質、DOTAP、任意のDOPEを計量し、そして、ジクロロメタンと混合し、その後、60℃まで加熱して、均質の粘性溶液を得る。ジクロロメタンは可溶化を促進する。次に、溶媒を真空内で留去した。
エタノール留去相の間、水相は調製した。5mlのエッペンドルフチューブ中で、補助界面活性剤、グリセロール、及びPBSの水溶液(154mMのNaCl、pH7.4)を混合し、そして、75℃の水浴内で溶かした。
油相が約40℃であり(粘着性形態)、水相が約70℃であった(水浴のまま)。水浴を油相に注ぎ入れた。
2つの相が入ったボトルを、AV505R超音波処理器(Sonics, Newton, USA)の超音波処理チャンバー内に備え付けた。プロトコールでは、40分間にわたる100Wの出力での超音波処理サイクル(30秒ごとに10秒の作動)を伴った。
次に、生じた液滴を、透析(カットオフ閾値:12kDa、154mMのNaClに対して一晩)によって精製して、LNP中に組み込まれていない脂質成分を取り除いた。最終的に、製剤を、セルロース膜による濾過によって滅菌した。
−製剤の組成の影響
表1〜5の結果は、補助界面活性剤(Myrj S40)の割合の低下が液滴の直径の増大につながることを示している。
製剤の液滴サイズ(表6)とゼータ電位(表7)における傾向を、40℃(加速安定性)で計測した。2回の測定の間、製剤を40℃で保存した。
以下の一般手法に従った:
複合体形成は、先に調製した製剤と、バッファー中のsiRNA溶液との単純な混合を伴った。構想している適用に関連するバッファーの選択:インビトロ研究のために、トランスフェクション工程に最適化した媒質は、OptiMEMであった。複合体形成試験のために、バッファーは、5mMのHepesを使用した。
−siRNA移動の観察を可能にしたアガロースゲル電気泳動法。良好な複合体形成があれば、そのときには、複合体形成したsiRNAを含んで成る液滴が、遊離のsiRNAより重くなるので、ウェル内に見られる。複合体形成がそれほどでなければ、遊離のsiRNAが別の位置に向かって移動する。
−尺度;
−遊離siRNA(基準);
次に、siRNAの複合体形成によって得られた移動:
−製剤B1を用いた(比較例)(複合体形成なし、siRNAは遊離);
−8:1の<プレミックス>製剤中のカチオン性界面活性剤による陽電荷の量とsiRNAによって提供される陰電荷の量との間の比により製剤B6を用いた(6カ月前に製剤したプレミックス)(定量的な複合体形成);
−8:1の<プレミックス>製剤中のカチオン性界面活性剤による陽電荷の量とsiRNAによって提供される陰電荷の量との間の比により製剤B6を用いた(12カ月前に製剤したプレミックス)、製剤は、複合体形成前に周囲温度で7カ月貯蔵した(定量的な複合体形成);
−8:1の<プレミックス>製剤中のカチオン性界面活性剤による陽電荷の量とsiRNAによって提供される陰電荷の量との間の比により製剤B10を用いた(定量的な複合体形成);
−リポフェクタミン(比較例)(不完全な複合体形成)。
トランスフェクション試験を、Green Fluorescent Protein(GFP)を過剰発現する異種細胞株に対して、このタンパク質のメッセンジャーRNAの配列を特異的に阻害するsiRNAを提供することによっておこなった(GFP−22 siRNAローダミン(カタログn°1022176)(Qiagen))。
−リポフェクタミンRNAimax;
−siRNA/製剤B1複合体。
−siRNA/製剤B6複合体、複合体形成前に周囲温度で7カ月保存した製剤;
−siRNA/製剤B6複合体、複合体形成前に周囲温度で12カ月保存した製剤;
−siRNA/製剤B10複合体;
−siRNA/DOTAP(58wt%)、DOPE(18wt%)、コレステロール(2wt%)及びDSPE−PEG3000(22wt%)を含んで成るカチオン性リポソームの製剤の複合体(比較)。
以下に示した一般手法に従った:
複合体形成は、バッファー中での先に調製したA3プレミックス製剤と、マイクロRNA(miRIDIAN Mimic Human has-miR612(ThermoScientific REF#C-300937-01 Batch n°130611))の溶液との単純混合を伴った。バッファーの選択は、構想している適用に依存した:インビトロ研究向けに、トランスフェクションステップのための最適化した培養、OptiMEMRを使用した。複合体形成に関する試験のために、5mMのHepesバッファーを使用した。
Claims (17)
- 連続水相及び少なくとも1つの分散相を含んで成るナノエマルション形態の製剤であって、
−少なくとも5モル%の両親媒性脂質、
−15〜70モル%の少なくとも1つのカチオン性界面活性剤であって、
−以下から選択される、少なくとも1つの親油性基:
−R又はR−(C=O)-基(式中、Rが、11〜23個の炭素原子を有する直鎖炭化水素鎖である)、
−12〜24個の炭素原子、及びフォスファチジルエタノールアミンを有する脂肪酸のエステル又はアミド、及び
−ポリ(酸化プロピレン)、及び
−以下から選択される、少なくとも1つのカチオン性基を含んで成る、少なくとも1つの親水基:
−1〜12個の炭素原子を有し、且つ少なくとも1つのカチオン性基によって分断及び/又は置換された直鎖又は分岐アルキル基、及び
−少なくとも1つのカチオン性基を含んで成る親水性重合性基、
を含んで成る、カチオン性界面活性剤、
−少なくとも25のエチレンオキシド単位を含んで成る、少なくとも1つのポリ(エチレンオキシド)鎖を含んで成る10〜55モル%の補助界面活性剤、
−可溶性脂質、
−任意にヘルパー脂質(helper lipid)、
−任意の油;
−任意のイメージング剤;及び
−任意の親油性治療剤;
を含んで成り、
両親媒性脂質、カチオン性界面活性剤及び補助界面活性剤のモルパーセンテージが、両親媒性脂質/カチオン性界面活性剤/補助界面活性剤/任意のヘルパー脂質の全量に対してであり、
そして前記分散相に対する可溶性脂質、任意の油、任意のイメージング剤、任意の親油性治療剤の全量の重量比率が10〜60%である、製剤。 - 前記カチオン性界面活性剤が、
−N[1−(2,3-ジオレイルオキシ)プロピル]−N,N,N−トリメチルアンモニウム、
−1,2−ジオレイル−3−トリメチルアンモニウム−プロパン、
−N−(2−ヒドロキシエチル)−N,N−ジメチル−2,3−ビス(テトラデシルオキシ−1−プロパンアニウム(propananium))、
−1−[2−オレオイルオキシ(oleoyloxy)エチル]−2−オレイル-3−(2−ヒドロキシエチル)イミダゾリニウム、及び
−ジオクタデシルアミドグリシルスペルミン、
から選択される、請求項1に記載の製剤。 - ヘルパー脂質を含んで成る、請求項1又は2に記載の製剤。
- 前記両親媒性脂質がリン脂質である、請求項1〜3のいずれか1項に記載の製剤。
- イメージング剤を含んで成る、請求項1〜4のいずれか1項に記載の製剤。
- 治療剤を含んで成る、請求項1〜5のいずれか1項に記載の製剤。
- 所望の分子が結合された補助界面活性剤を含んで成る、請求項1〜6のいずれか1項に記載の製剤。
- 200未満の塩基を含んで成る一本鎖又は200未満の塩基対を含んで成る二重鎖核酸配列であって、RNA干渉の内在性メカニズムを調節することができる一本鎖又は二重鎖核酸配列を含んで成る、請求項1〜7のいずれか1項に記載の製剤。
- 前記核酸配列が、
−低分子干渉RNA、
−ロックド核酸、及び
−合成マイクロRNA、
から選択される、請求項8に記載の製剤。 - 前記核酸配列が低分子干渉RNAである、請求項9に記載の製剤。
- 請求項8〜10のいずれか1項に記載の製剤の調製方法であって、以下の、
(i)可溶性脂質、両親媒性脂質、カチオン性界面活性剤を含んで成る油相を調製する工程、
(ii)補助界面活性剤を含んで成る水相を調製する工程、
(iii)十分な剪断作用下で、上記油相を水相に分散し、ナノエマルション形態の、例えば、請求項1〜7のいずれか1項に記載の製剤を得る工程、続いて
(iv)200未満の塩基を含んで成る一本鎖又は200未満の塩基対を含んで成る二重鎖核酸配列であって、RNA干渉の内在性メカニズムを調節することができる一本鎖又は二重鎖核酸配列を、エマルション形態の請求項1〜7のいずれか1項に記載の製剤に添加する工程、続いて
(v)形成された製剤を回収する工程、
を含んで成る、方法。 - 200未満の塩基を含んで成る一本鎖又は200未満の塩基対を含んで成る二重鎖核酸配列であって、RNA干渉の内在性メカニズムを調節することができる一本鎖又は二重鎖核酸配列の、真核生物細胞へのインビトロ(in vitro)での挿入方法であって、
上記真核生物細胞を請求項8〜10のいずれか1項に記載の製剤と接触させる工程を含んで成る、方法。 - 疾患の予防及び/又は治療のための、請求項8〜10のいずれか1項に記載の製剤を含んで成る、医薬組成物。
- ナノエマルション形態の請求項1〜7のいずれか1項に記載の製剤、及び
少なくとも1つの200未満の塩基を含んで成る一本鎖又は200未満の塩基対を含んで成る二重鎖核酸配列であって、RNA干渉の内在性メカニズムを調節することができる一本鎖又は二重鎖核酸配列、
