Description
(a)廃液流および窒素源を含む第1段階発酵反応器中にて複数のスラウストキトリッド株を培養すること。
(b)段階(a)の複数のスラウストキトリッド株を、バイオマス採取システムを介して、廃液流のみを有する第2段階発酵反応器へと移送すること。
(c)必要に応じて窒素が無い状態において、第2段階発酵反応器中にて段階(b)のバイオマス培養物を培養すること。
(d)バイオマスと液体培養液の分離、および複数のガス発酵工場への澄んだ培養液のリサイクル。
(e)付加価値のある複数のオメガ3脂肪酸および脂質を得ること。
本発明の複数の目的のために、以下の複数の用語は、そこで特定される意味を有するであろう。
(a)廃液流および窒素源を含む第1段階発酵反応器中にて複数のスラウストキトリッド株を培養すること。
(b)段階(a)の複数のスラウストキトリッド株を、バイオマス採取システムを介して、廃液流のみを有する第2段階発酵反応器へと移送すること。
(c)必要に応じて窒素が無い状態において、第2段階発酵反応器中にて段階(b)のバイオマス培養物を培養すること。
(d)バイオマスと液体培養液の分離、および複数のガス発酵工場への澄んだ培養液のリサイクル。
(e)付加価値のある複数のオメガ3脂肪酸および脂質を得ること。
例1:インドの海洋生物多様性からのスラウストキトリッドの単離
多数のサンプル(土壌、水、分解された葉、等)が、2013年の3月に、Mandovi-Zuariマングローブにわたるリバンダーのマングローブエリアにおいて、30−35kmに及ぶインドの海洋の複数の場所から集められた(収集座標:S15°29'57.39'',E73°52'6.13''、パンジム、ゴア州)。収集場所の地理的位置、pH、温度および湿度のような複数の物理的パラメータが、GPS、pHメータ、温度計および検湿器によってそれぞれ記録された。それぞれのファルコンに100μlの選択的抗生物質混合物が加えられた。複数のサンプルはドライアイスパック中で保管され、処理のために、天然の海水とともに24時間以内に実験室へと移された。単離のために、直接平板法(direct plating method)または釣り餌法(baiting method)の2つのプロトコルに従った。複数の寒天プレート上に播種する前に、滅菌海水によって、複数の土壌サンプルが1000倍に希釈された。または、複数の葉のサンプルが洗浄された。複数の水サンプルが複数の寒天プレート上に直接播種された。これら複数の寒天プレートに選択的抗生物質混合物が補給された。全てのサンプルが、滅菌したマツ花粉またはその他の選択された材料(25−30mg)によって餌を与えられ、25℃において8−10日間、複数の花粉の外縁にコロニー形成が現れるまで、インキュベートされた。コロニー形成は、5日後以降、毎日、光学顕微鏡下で確認された。ひとたび複数の花粉がコロニーを形成すると、100μlの水がファルコンの表面から注意深く採取され、抗生物質を含む複数の寒天プレート上に播種された。これら複数のプレート用の媒体は、グルコース(10g/L)、酵母エキス(1g/L)、ペプトン(1g/L)、寒天培地(10g/L)、濾過された天然の海水(100%v/v)、および抗生物質混合物を含んでいた。これら複数のプレートは、25℃において7−10日間インキュベートされた。複数のプレート上に現れた複数のコロニーが、形態学的調査のため、顕微鏡(40倍)下で観察された。スラウストキトリッド様の複数のコロニーが採取され、さらに、70%の天然の海水とともに上記の媒体組成を有する複数の寒天プレート上に筋状にされた。スラウストキトリッド様の複数の株が、選択された抗生物質混合物を有する複数の寒天プレート上にて3−4回筋状にされた後に精製された。スラウストキトリッドの迅速な同定のためには、複数のオメガ3脂肪酸、特にDHAの存在が重要な複数のパラメータのうちの1つである(図1)。従って、既知の複数の方法を採用することにより、全ての株のバイオマスからの脂質が抽出され、複数の脂肪酸メチルエステルに変換された。これら複数のFAMEが、GC−FIDにより、特にDHAの存在に関して分析された。
複数の株の遺伝子同定のために、5日経過した培養物1mlが採取され、DNeasy blood and tissue kit(Qiagen、米国)に記載される複数のガイドラインに従ってゲノムDNAが抽出された。複数のプライマーT18S1F 5'−CAACCTGGTTGATCCTGCCAGTA−3'およびT18S5R 5'−TCACTACGGAAACCTTGTTACGAC−3'(Honda et al. 1999)を用いた18S rRNA遺伝子のPCR増幅のために、ゲノムDNAが使用された。12.5μLのPCR master mix(Applied Biosystem、米国)、それぞれ0.5μLのプライマー(T18S1F、T18S5R)、1μLのゲノムDNA、10.5μLのmilliQ waterを有する、25μLのPCR反応が設定された。PCRプログラムは、30サイクルにわたる、94℃における3分間の初期変性(initial denaturation)、94℃における45秒間の最終変性(final denaturation)、64℃における30秒間のアニーリング(annealing)、72℃における2分間の伸張(extension)、72℃における10分間の(最終伸張(final extension))を含んでいた。QiAquick gel extraction kit(Qiagen、米国)を用いて、1%のアガロース・ゲルからPCR生成物が精製された。PCR生成物および複数のプライマーの混合物は、配列決定のためにMacrogen (韓国)へと送られた。得られた18S rRNA遺伝子配列は、basic local alignment search tool(BLAST)を用いて、NCBI遺伝子バンクデータベース中の既知の複数のスラウストキトリッド18S rRNA遺伝子配列と比較された。MEGA6ソフトウェアを用いて系統発生樹(NJ樹)を構築すべく、複数の株の配列が、スラウストキトリッドのその他複数の既知の配列とともに使用された(図2)。得られた複数の配列は、(受託番号KF668624、KF668627、KF668629、KF668632として)NCBIデータベースに供託された。複数の株、すなわち、MTCC5890(DBTIOC−1)、MTCC5895(DBTIOC−6)、MTCC5893(DBTIOC−14)、MTCC5896(DBTIOC−18)が、インドのChandigarhにある Institute of Microbial Technology (IMTECH)のMicrobial Type Culture Collection and Gene Bank (MTCC)に供託された。
5g/L、30g/L、50g/L、または100g/Lのような異なる濃度のアセテートが、唯一の炭素源として、または、他の複数の炭素源と混合されて、栄養物の豊富な媒体中に加えられた。酢酸のpHは、水酸化ナトリウムによって7まで上げられ、続いて10g/Lの酵母エキス、1g/Lのペプトンが添加された。海水の50%の強度を模倣すべく、18g/Lの人工的な海塩が媒体中に加えられた。残留アセテートがHPLCによって測定された。振とうフラスコ培養では、最大酢酸利用は15g/Lを越えないことが見出された。しかしながら、より高い濃度のアセテートに対する株の耐性が、50g/Lまたは100g/Lまで試験された。株DBTIOC−18(MTCC5896)は、他の複数の株と比較して、媒体中のアセテートの量に関係なく、炭素源としてのアセテートに対して、より良好に機能した(図3)。
この発明において使用される複数の廃液は、複数の温室効果ガスのガス発酵から排出されたもの、またはリグノセルロース系バイオマスの脱アセチル化、またはリグノセルロース系バイオマスの熱分解、または原油の精製、等から排出されたものであった。これら複数の廃液の化学組成は、エタノールのような他の複数の酸およびアルコール(0.5g/L、または2g/L、5g/L、またはそれ以上)とともに、流れ中では複数の有機酸(特に酢酸、プロピオン酸、酪酸)が支配的に存在することを明らかにした。この流れは、複数の窒素源を含めてあらゆる付加的な栄養源無しに、そのままで、121℃にて20分間、加圧滅菌処理された。5日後、250mlフラスコ中に50mlの培養物が採取され、バイオマスが一晩乾燥された。複数の脂質が抽出され、所与の方法によって複数のFAMEが分析された。複数の対照菌株においては、低速の栄養物隔離により、バイオマスは0.17g/Lから0.61g/Lまでにわたっていた(図4)。複数の廃液流からの栄養物隔離速度を増大させるべく、複数の廃液流への複数の株の適応、複数の窒素源の添加およびより大きな接種物サイズのような、異なる複数の方策が適用された。
スラウストキトリッドの複数の選択された株の接種物が、30g/Lのグルコース、10g/Lの酵母エキス、1g/Lのペプトン、および18g/Lの海塩を有するYPD媒体中で調製され、25℃、150rpmにおいてインキュベートされた。窒素源を有する複数の廃液によって半分が満たされている2Lまたは7Lまたは14Lの複数の反応器中に、48時間経過した接種物の10%が加えられた。培養物は、通常の穴の空けられた複数のパイプスパージャーまたは複数のマイクロスパージャーまたはその他の種類の複数のマイクロスパージャーによって通気された。他方、培養物は、ラシュトンまたはピッチブレードまたは海洋インペラ、あるいはその他複数のインペラによって、100rpm、または200rpm、または300rpm、またはそれ以上で撹拌された。溶存酸素は、空気および酸素供給の組み合わせによって、10%、または20%、または30%、または50%、またはそれ以上に維持された。栄養物隔離、バイオマス生成、脂質含有量およびDHA生成を決定すべく、8から12時間の間隔で定期的にサンプルが取られた。複数の廃液からの栄養物隔離測定用のマーカとして、アセテート消費測定が使用された。反応器のpHは、酸と塩基によって7に維持された。フェドバッチ培養においては、複数の有機酸およびアルコールによって増補された複数の廃液が、培養物に対して直接供給された。
画定された量の乾燥されたバイオマスが脂質抽出用に取られ、修正されたBligh−Dyer法によって脂質が抽出された。(図3および4)。FAME分析用に、乾燥された脂質抽出物に、500μlのトルエン、続いて100μlの内部標準(トリコサン酸メチル C19:0)および100μlのブチル化されたヒドロキシトルエン(BHT)が加えられた。酸性メタノール(400μl)がチューブに加えられ、10−12時間にわたって50℃に保たれた。1mlの5%NaClが加えられ、続いて1mlのヘキサンが添加され、上層が新たなチューブに移された。1mlの2%重炭酸カリウムによって複数のFAMEが洗浄され、これを無水硫酸ナトリウムに通すことによって水分が除去された。複数のFAMEは、窒素下で濃縮され、高速GCキャピラリカラム(Omegawax100、15m×0.1mm、厚さ0.1μm)が備えられたGC−FIDシステム(Perkin Elmer clarus680、米国)によって分析された。1μlのFAMEが、注入器温度250℃で注入された。オーブンの温度は、初期の140℃から、40℃/秒の上昇速度で最終的に280℃まで上げられ始め、2分間保持された。検出器は260℃に設定された。速度50cm・sec−1を有するキャリアガスとして水素が使用された。Totalchromeクロマトグラフィソフトウェア(Perkin Elmer、米国)により、複数の脂肪酸ピークが同定され、定量化された。
Claims (32)
- バイオディーゼル用の高価値のオメガ3脂肪酸および脂質を生成するために、スラウストキトリッドを用いて、ガス発酵工場の複数の廃液から複数の栄養物を隔離するための2段階の連続発酵を含むプロセスであって、
(a)廃液流および窒素源の連続的な供給を含む第1段階発酵反応器中にて複数のスラウストキトリッド株を連続的に培養する段階と、
(b)段階(a)の前記複数のスラウストキトリッド株の培養物を、バイオマス採取/分離システムを介して、廃液流のみを有する第2段階発酵反応器へと移送する段階と、
(c)必要に応じて窒素が無い状態において、前記第2段階発酵反応器中にて、バイオマスを生成する段階(b)のバイオマス培養物を連続的に培養する段階と、
(d)バイオマス採取/分離システムを介して、前記バイオマスを液体培養液から連続的に分離し、澄んだ培養液をガス発酵工場へとリサイクルする段階と、
(e)オメガ3脂肪酸および脂質を連続的に得る段階と、
を含む、プロセス。 - 複数の廃液は、有機酸およびアルコールの混合物を含み、糖または炭水化物を含まない、請求項1に記載のプロセス。

- 前記窒素が無い状態において、DHA含有量を、全脂肪酸の15%から全脂肪酸の50%までの範囲内で前記第1段階発酵反応器から増大させる、請求項1に記載のプロセス。
- 前記窒素が無い状態において、DHA含有量を、全脂肪酸の15%から全脂肪酸の35%までの範囲内で前記第1段階発酵反応器から増大させる、請求項1または請求項12に記載のプロセス。
- 段階(b)において、前記第1段階発酵反応器からの培養物がバイオマス分離反応器に連続的に通され、バイオマスと培養液とを分離する、請求項1に記載のプロセス。
- 前記段階(c)において、分離システムから得られた濃縮されたバイオマスが、前記第2段階発酵反応器へと連続的に移送される、請求項1に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、乾燥バイオマスを8グラム/リットル/日から200グラム/リットル/日までの範囲内に増やす、請求項1に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、乾燥バイオマスを8グラム/リットル/日から150グラム/リットル/日までの範囲内に増やす、請求項1または請求項16に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、前記脂質の含有量を、乾燥質量の20%から乾燥質量の100%までの範囲内に増やす、請求項1に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、前記脂質の含有量を、乾燥質量の20%から乾燥質量の80%までの範囲内に増やす、請求項1または請求項18に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、DHA含有量を、全脂肪酸の15%から全脂肪酸の50%までの範囲内に増やす、請求項1に記載のプロセス。
- 前記第1段階発酵反応器から前記第2段階発酵反応器へのバイオマスの直接の前記移送は、DHA含有量を、全脂肪酸の15%から全脂肪酸の45%までの範囲内に増やす、請求項1または請求項20に記載のプロセス。
- 段階(b)および段階(c)において、細胞中の脂質蓄積を刺激すべく、前記第2段階発酵反応器が連続的な窒素ストレス条件を生成するように使用される、請求項1に記載のプロセス。
- 段階(d)において、前記第2段階発酵反応器から出てくる前記培養物は、バイオマスと培養液水とを分離すべくバイオマス分離システムに連続的に通され、溶存酸素がゼロである前記水は、連続的にリサイクルされてガス発酵処理用に戻される、請求項1に記載のプロセス。
- 段階(a)において、微量の硫化物を除去すべく、前記廃液が空気によって連続的にパージされる、請求項1に記載のプロセス。
- 前記硫化物の除去は、前記複数の栄養物の廃液流からの隔離を、55グラム/リットル/日から90グラム/リットル/日までの範囲内に増大させる、請求項24に記載のプロセス。
- 前記硫化物の除去は、前記バイオマスの生成を、40グラム/リットル/日から55グラム/リットル/日までの範囲内に増大させる、請求項25に記載のプロセス。
- 受託番号MTCC5890(DBTIOC−1)、MTCC5895(DBTIOC−6)、MTCC5893(DBTIOC−14)、およびMTCC5896(DBTIOC−18)を有するスラウストキトリッドの複数の新株。
- バイオディーゼルでの使用のためのオメガ3脂肪酸および脂質を生成することの可能な、請求項27に記載の複数の新株。
- 前記複数の新株は、8グラム/リットル/日から150グラム/リットル/日までの範囲内の乾燥バイオマスを生成する、請求項27に記載の複数の新株。
- 前記複数の新株は、乾燥質量の20%から乾燥質量の80%までの範囲内の脂質を生成する、請求項27に記載の複数の新株。
- 前記複数の新株は、全脂肪酸の15%から全脂肪酸の45%までの範囲内のDHA含有量を生成する、請求項27に記載の複数の新株。
