(1)神経変性疾患/障害
ある特定の態様では、本明細書において記載される化合物またはその塩および/もしくはプロドラッグは、神経変性疾患、および中枢神経系(CNS)、脊髄または末梢神経系(PNS)に対する外傷性損傷または機械的損傷などのP2Y6受容体関連症状を患っている患者を処置するのに使用することができる。これらおよび本明細書において記載される他の症状の多くは、一定レベルの認知障害および/または認知機能のいくらかの減少もしくは喪失を特徴とする。認知機能および認知障害は、当技術分野において理解されているように使用される。例えば、認知機能は、一般に、それによって考えを認識または理解する心理過程を指す。認知機能は、知覚、思考、学習、推理、記憶、認識および判断能力のすべての面に関与する。認知障害は、一般に、思考過程の問題を伴う症状または症候を指す。これは、より高度な推理力の障害または低下、物忘れ、記憶障害、学習障害、集中力の欠如、知能の低下、および他の精神機能低下などの認知機能の低下を示す1つ以上の症候で現れ得る。
本開示の化合物またはその塩および/もしくはプロドラッグはまた、ダウン症候群(DS)の症候を予防、処置および緩和するのに有用であり得る。ダウン症候群(DS)は、第21染色体のトリソミーを特徴とする遺伝的症状である。DSは、1866年にDSの特徴を最初に説明したイギリス人医師のDr.John Langdon Downにちなんで名付けられたものである。1959年になって初めて、Jerome LeieuneおよびPatricia Jacobsが独立して、原因は第21染色体のトリソミーであることを最初に決定した。
ある特定の態様では、本明細書において記載される化合物またはその塩および/もしくはプロドラッグは、疼痛を有する患者を処置するのに使用することができる。疼痛は、多数の感覚および神経機構が関与する複合的な生理学的過程である。本開示にしたがって使用されるべき化合物またはその塩および/もしくはプロドラッグは、慢性疼痛および/または急性疼痛、特に背痛、線維筋痛症、筋膜痛などの非炎症性筋骨格痛を処置(予防および/または緩和を含む)するために、より具体的には、関連する筋肉痛覚過敏または筋肉アロディニアを軽減するために被験体に投与するのに適切である。本開示の化合物またはその塩および/もしくはプロドラッグ、組成物ならびに方法によって処置することができる疼痛の種類の非限定的な例には、慢性症状、例えば線維筋痛症、筋膜痛、背痛を含む筋骨格痛、月経期間の際の痛み、変形性関節症の際の痛み、関節リウマチの際の痛み、胃腸管の炎症の際の痛み、心筋の炎症の際の痛み、多発性硬化症の際の痛み、神経炎の際の痛み、AIDSの際の痛み、化学療法の際の痛み、腫瘍疼痛、頭痛、CPS(慢性疼痛症候群)、中枢性疼痛、神経因性疼痛、例えば三叉神経痛、帯状疱疹、断端痛(stamp pain)、幻肢痛、顎関節症、神経損傷、偏頭痛、帯状疱疹後神経痛、神経系の傷害、切断術による感染症、代謝障害もしくは変性疾患の結果として引き起こされる神経因性疼痛、糖尿病、偽感覚、甲状腺機能不全症、***、ビタミン欠乏もしくはアルコール症に関連する神経因性疼痛;および急性疼痛、例えば傷害後の疼痛、術後疼痛、急性痛風の際の痛みまたは顎部手術などの手術の際の痛みが含まれる。
本開示の組成物および方法において使用される化合物および薬剤またはその塩および/もしくはプロドラッグは、好ましくは、末梢に投与された場合に、血液脳関門を容易に通過するべきであると認識されよう。しかしながら、血液脳関門を通過することができない化合物は、例えば、脳室内経路によって中枢神経系に直接有効に投与することができる。
化合物6のトリエチルアミン塩およびナトリウム塩の調製
以下のスキーム2は、化合物6のトリエチルアミン(TEA)塩およびナトリウム塩を調製するための一般的な合成経路を提供する。
化合物42(3.0g、8.11mmol)のDMF(90mL)溶液に、Y01(3.0g、16.22mmol)およびK2CO3(4.47g、16.22mmol)を追加し、得られた混合物を70℃で1時間撹拌した。冷却後、この混合物を250mLの水で希釈し、酢酸エチル(EA)(250mL×3)で抽出し、有機層を無水Na2SO4で脱水し、濃縮して、粗生成物を得た。粗生成物をカラム(PE/EA=3:1で溶離)で精製して、無色の油として3.61gの47を得た。収率:94%。1H NMR(300MHz,CDCl3)δ7.36(d,J=8.1Hz,1H),7.32−7.27(m,4H),7.25−7.18(m,1H),5.98(d,J=4.0Hz,1H),5.81(d,J=8.1Hz,1H),5.34(d,J=2.4Hz,2H),4.35(s,3H),4.13(m,2H),3.01−2.84(m,2H),2.14(dd,J=12.1,4.2Hz,9H),1.26(t,J=7.1Hz,1H)。

化合物42(1.061g、1.87mmol)、Y03(930mg、5.73mmol)およびPPh3(1.501g、5.73mmol)の25mL THF溶液に、DIAD(1.159g、5.73mmol)の5mL THF溶液を30分間かけて滴下し、得られた混合物を50℃で3時間撹拌した。反応が終了した後、THFを除去して、粗生成物を得た。粗生成物をカラム(EAで溶離)で精製して、油として1.37gの化合物51を得た。収率:88.8%。

in vitroおよびin vivo研究用の材料および方法
P2Y6受容体の活性化
蛍光Ca2+標識fluo−4を用いて受容体誘導性のCa2+変化を測定することによって、合成リガンドをP2Y6受容体の活性化について試験した。P2Y2、P2Y4またはP2Y6受容体のいずれかを発現する1321N1ヒト星状細胞腫細胞株を24ウェルプレートに蒔いた。蒔いた2日後に蛍光分析測定を行い、リガンドの連続希釈物に対する細胞の反応を決定した。リガンド投与後の3時点の蛍光変化の累積からACSF対照の値を引いて正規化することによって、P2Y6媒介性のCa2+蛍光変化を決定した。GraphPadにおいて、蛍光強度の変化をリガンド濃度に対応させてプロットした。非線形カーブフィッティングおよびS字状用量反応分析を使用して、各リガンドの用量反応曲線およびEC50を推測した。化合物5のナトリウム塩は、12nMのEC50を示した。P2Y2、P2Y4またはP2Y6受容体を発現する3種の1321N1ヒト星状細胞腫細胞株において、そのCa2+動員効果を比較することによって、化合物5のナトリウム塩は、P2Y6受容体を選択的に活性化することが実証された。化合物5のナトリウム塩は、P2Y6受容体を発現する細胞に適用した場合のみ、Ca2+レベルを高めるのに有効であり、P2Y2、またはP2Y4受容体を発現する細胞では、有効ではなかった。化合物5のナトリウム塩が、P2Y6を発現する細胞においてCa2+シグナルを高める能力は、P2Y6アンタゴニストMRS2578を追加することによって弱まった。
ヘテロ変異体(K670N/M671L)APP(50%C57B6、50%SJL)トランスジェニックマウスをヘテロ変異体(A246E)PS−1(50%C57B6、50%SJL)トランスジェニックマウスを交配して、ヘテロPSAPPトランスジェニックマウス(PS−1/APPまたはPSAPP+/+マウスとも称される)を作製した(これは、PS−1 A246E導入遺伝子およびAPP K670N/M671L導入遺伝子についてヘテロな動物を指す)。非トランスジェニック対照動物は、PSAPPトランスジェニック動物の繁殖で作製した同腹子(PSAPP_−/−マウスとも称される)であった。ポリメラーゼ連鎖反応(PCR)によって、マウスの遺伝子型を決定した。6〜7月齢の雄性マウスおよび雌性マウスの両方を以下の実験に使用した。Tufts Animal Care and Use Committeeならびに国家規制および国家政策にしたがって、すべての動物実験を実施した。
本研究では、イソフルランを使用してPSAPPマウスを麻酔し、表面の損傷を最小限に抑えるためにシン−スカル(thin−skull)調製を使用した。メトキシX04標識でアミロイド斑を可視化し、同じイメージング領域の再局在化を容易にするためにローダミンデキストランで血漿を標識した。励起850nmで二光子システム(Prairie Technologies)を使用して、スタック画像を得た。外部光電子増倍管(525/70;DLCP 575;607/45nm)によって、発光を検出した。
動物を麻酔し、定位フレームで固定した。各注射では、以下の座標:AP0.2mm、ML1mm、およびDV2.2mmを使用して、1μlの10mM UDPまたは他の適切な化合物の人工脳脊髄液(ACSF)(ビヒクルとして)を脳室内注射した。
マウスを4%パラホルムアルデヒドで経心臓的に灌流し、40μmの冠状切片を採取した。0.3%H2O2で10分間、ブロッキング溶液で2時間、一次抗体(ウサギ抗β1−42;ウサギ抗β1−40、Chemicon International製およびウサギ抗CD45)を含有するブロッキング溶液で48時間(4℃)、そして、ビオチン化抗体または蛍光標識抗体を含有するブロッキング溶液で2時間(室温)、切片を連続してインキュベーションした。明視野顕微鏡または共焦点顕微鏡で切片を可視化し、MetaMorphソフトウェアを使用して光学密度を取得した。
1日目に、合図および軽度のフットショックからなる2ペアリングパラダイム(30秒間の音による条件付け刺激、80dB;2秒間のショック刺激、0.5mA)を用いて、恐怖条件付け装置で合計7分間にわたって、動物を訓練した。文脈恐怖学習を評価するために、訓練24時間後に動物を訓練文脈に戻し、凍結挙動を5分間にわたってスコア化した。MotorMonitor(Hamilton Kinder)によって凍結挙動をモニタリングし、5秒毎にスコア化した。
6月齢のPSAPPマウスから、海馬スライス(厚さ350mm)を調製した。ベースライン反応を10秒毎に取得し、入出力(I/O)曲線、ペアパルス変化およびLTPの測定に成功した。刺激強度は、得られる最大の30%の値を与えるレベルに設定した。高周波刺激(HFS、100Hzで100パルス、4回)によって、またはシータバースト刺激(TBS、5Hzで10バースト、15秒間隔で10回繰り返す)によって、LTPを誘導した。
P2Y6受容体の用量依存的活性化

プラーク断面積に対するUDPの効果を評価するために、二光子顕微鏡法を使用して、生きているPSAPPマウスのバレル皮質におけるアミロイド斑を評価した。メトキシX04を全身投与することによって、アミロイド斑を染色した。イメージングの1日前に、PSAPPマウスにメトキシX04を注射して、アミロイド斑を標識した。イメージングの日に、同じイメージング領域の再配置を容易にするために、ローダミンデキストランで血漿を標識した。同じ画像ボリュームを確保するために、同じ開始点および終了点から画像を取得した。
P2Y6受容体の活性化は、PSAPPマウスにおいてプラーク断面積を減少させた
3−フェナシル−UDP(PSB0474とも称される)は、強力かつ選択的なP2Y6受容体アゴニストである(EC50=70nM、>500倍選択的)。本研究では、3−フェナシル−UDP(PSB0474)を使用して、P2Y6受容体をin vivoで活性化した。プラーク断面積に対するこの活性化の効果も評価した。
急性UDP投与は、PSAPPマウスにおいて認知機能および海馬LTPを改善した
アミロイドβペプチドは、シナプス伝達に有毒であると報告されており、アミロイドタンパク質の蓄積は、ADの動物モデルおよびAD患者の両方で認知障害に関連している。加えて、アミロイドタンパク質の蓄積は、認知障害に関連する他の症状、例えばダウン症候群で観察されている。したがって、本発明者らは、観察されたプラーク断面積の減少が、AD患者で典型的に観察される認知記憶障害、例えば認知障害、記憶障害および長期増強(LTP)の障害の回復にもつながるかをPSAPPマウスで調査した。
PSB0474の慢性注射によるP2Y6受容体の活性化は、PSAPPマウスの認知機能を改善した
急性UDP処置と同様に、P2Y6アゴニスト3−フェナシル−UDP(PSB0474)の慢性注射は、PSAPPマウスの文脈試験において総凍結率を増加させた(図9(A)〜(C))。本研究では、PSB0474を2つの異なる用量で投与したところ、これらの両方が、PSAPPマウスにおいて認知機能を改善する有益な効果を示した。
化合物5によるP2Y6受容体の活性化は、PSAPPマウスの認知機能を改善し、PSAPPマウスのプラーク断面積を減少させた
本研究では、化合物5を、6〜7月齢のPSAPPおよびWTマウスに、2つの異なる用量(すなわち、1ug/kgまたは1mg/kgの化合物5(1%DMSO/PBS中))で、7日間連続して毎日腹腔内注射した。急性UDPまたはPSB0474処置後に観察された結果と一致して、化合物5による処置は、PSAPPマウスの文脈試験において総凍結率を増加させた(図11)。図11は、ビヒクル対照または化合物5による処置後の、恐怖条件付け研究におけるPASPPマウスの凍結挙動(凍結%)を示す。図11は、ビヒクル対照で処置したPSAPPマウスの文脈恐怖条件付け試験を使用した実験結果を示す(グラフ中央の黒色のバー)。これらのマウスは、年齢適合野生型動物(白色のバー)と比較して有意に低下した凍結率を示した;これは、PSAPPマウスにおける記憶障害および認知障害を示す。試験前に化合物5を投与すると、凍結挙動(グラフ右側の斜線のバー)が、対照処置と比較して有意に改善された。実際、認知機能および記憶を示すこの挙動は、野生型動物で観察されたものと同等のレベルに回復した。この結果は、化合物5が、記憶および/または学習を改善することなどによって、これらのマウスにおいて認知機能を改善した(認知障害を軽減した)という結論と一致している。

- (i)Yが、場合により1個以上のR4で置換されていてもよいC2−脂肪族基であるか、あるいは(ii)Yが−CH2−CH2−であるか、あるいは(iii)Yが−CH2−C(R4)2−であって、出現するR4がそれぞれ、ハロゲンから独立して選択されるか、あるいは(iv)Yが−CH2−C(R4)2−であって、出現するR4がそれぞれ、独立して、(C1−C3)−脂肪族基である、請求項1〜8のいずれか一項に記載の化合物。

- 神経変性障害、外傷性脳損傷、疼痛、またはダウン症候群(DS)の処置を必要とする被験体における、神経変性障害、外傷性脳損傷、疼痛、またはダウン症候群(DS)の処置において使用するための組成物であって、請求項1〜16のいずれか一項に記載の化合物を含む、組成物。
- 前記組成物が、アルツハイマー病、パーキンソン病、軽度認知障害(MCI)、ハンチントン病、多発性硬化症および脳血管障害より選択される神経変性障害の処置において使用するためのものである、請求項19に記載の組成物。
- 前記組成物が、筋骨格痛、線維筋痛症、筋膜痛、月経期間の際の痛み、変形性関節症の際の痛み、関節リウマチの際の痛み、胃腸管の炎症の際の痛み、心筋の炎症の際の痛み、多発性硬化症の際の痛み、神経炎の際の痛み、AIDSの際の痛み、化学療法の際の痛み、腫瘍疼痛、頭痛、CPS、中枢性疼痛、神経因性疼痛、三叉神経痛、帯状疱疹、断端痛(stamp pain)、幻肢痛、顎関節症、神経損傷、偏頭痛、帯状疱疹後神経痛、神経系の傷害、切断術による感染症、代謝障害もしくは変性疾患の結果として引き起こされる神経因性疼痛、糖尿病、偽感覚、甲状腺機能不全症、***、ビタミン欠乏もしくはアルコール症に関連する神経因性疼痛、傷害後の急性疼痛、術後疼痛、急性痛風の際の痛みまたは手術による痛みより選択される疼痛の処置において使用するためのものである、請求項19に記載の組成物。
- 前記組成物が、局所剤形、肺剤形、局所内部(internal topical)剤形、皮内剤形、静脈内剤形、皮下剤形、鼻腔内剤形、表皮剤形、眼剤形、口腔剤形、脳室内剤形および髄腔内剤形からなる群より選択される剤形の一部として使用するためのものである、請求項19〜21のいずれか一項に記載の組成物。
- 認知機能の改善、認知障害の軽減または遅延、海馬長期増強の改善、あるいはβアミロイドクリアランス速度の増強を必要とする被験体における、認知機能を改善する、認知障害を軽減または遅延する、海馬長期増強を改善する、あるいはβアミロイドクリアランス速度を増強するのに使用するための組成物であって、請求項1〜16のいずれか一項に記載の化合物を含み、前記被験体がアルツハイマー病を有する、組成物。
- βアミロイドクリアランス速度の増強を必要とする被験体におけるβアミロイドクリアランス速度を増強するのに使用するための組成物であって、請求項1〜16のいずれか一項に記載の化合物を含み、前記被験体が外傷性脳損傷またはダウン症候群を有する、組成物。
