Description
The present invention relates to a centrifuge rotor and a centrifuge that use a liquid or the like as a sample and perform centrifugal processing such as precipitation separation or concentration by centrifugal force.
A centrifuge, that is, a centrifuge, is used in order to perform a centrifugal process such as precipitation separation, purification, and concentration by applying a centrifugal force to a liquid or a mixture of liquid and solid as a sample. When this centrifuge is used, particles can be separated from a sample containing particles that do not settle or hardly settle in a normal gravitational field, and viruses, fungus bodies, and the like can be targeted for separation. Viruses and fungal cells are indispensable raw materials for producing drugs, vaccines, and the like. In these production processes, continuous centrifuges are widely used as equipment for separating and purifying raw materials.
For example, as described in
The rotor is supported by upper and lower shafts, the upper shaft, that is, the upper shaft is connected to an electric motor, and the rotor is driven to rotate by the upper shaft. The lower shaft, that is, the lower shaft, is rotatably supported by a slide bearing and a damper in order to reduce centering and rotational vibration. The lower shaft is supported by the lower bearing portion, and the upper shaft is supported by the upper bearing portion. A lower connection block to which the lower pipe is connected is incorporated in the lower seal part, and an upper connection block to which the upper pipe is connected is incorporated in the upper seal part. Mechanical seals are respectively provided between the lower connection block and the end of the lower shaft, and between the upper connection block and the end of the upper shaft. As a result, leakage of the sample is prevented by the mechanical seal while the rotor is rotating at high speed, the sample is continuously supplied from the lower shaft, and the processed supernatant is discharged from the upper shaft to the outside. Can do.
The rotor has a rotor outer shell composed of a cylindrical rotor body and a rotor cover attached to both ends, and a core is detachably incorporated in the rotor outer shell. The core has a body portion whose outer peripheral surface is circular, and a plurality of fins that protrude outward in the radial direction and extend in the axial direction, and the tip end surfaces of the fins are in contact with the inner peripheral surface of the rotor body. A plurality of fan-shaped separation spaces partitioned by fins are formed between the inner peripheral surface of the rotor body and the outer peripheral surface of the core. The form of the core includes a solid type in which the body part is solid and a hollow type in which the body part has a hollow structure, and both ends of the hollow type core are closed by end walls. The solid core is manufactured by cutting lightweight plastic, and the hollow core is manufactured by a metal such as a titanium alloy. Further, in addition to the normal shape, the core has a large capacity form in which the outer diameter of the body portion is made smaller than usual to increase the protruding length in the radial direction of the fin and the volume of the separation space is increased.
By the way, the upper rotor cover provided at the upper end portion of the core is provided with a shaft connecting portion connected to the upper shaft, and the lower rotor cover is provided with a shaft connecting portion connected to the lower shaft. To fix the core to each rotor cover, a fitting hole is formed coaxially with each other at both ends of the core, and a protrusion that fits into the fitting hole is connected to the shaft on each rotor cover. It is provided coaxially with the part. Each rotor cover is attachable to and detachable from the core, but it is necessary to process the gap between the protrusion and the fitting hole as small as possible so that the rotation center axis of the rotor body and the core is concentric. There is. If this gap becomes large, the core will be eccentric with respect to the rotation center axis of the rotor body, and if the gap is too small, the assembly workability between the rotor cover and the core will be deteriorated. It is necessary to make adjustments.
However, when the rotor is rotated at a rotational speed exceeding 30,000 rpm, the core is deformed so as to increase in diameter due to centrifugal force, and the gap between the inner peripheral surface of the fitting hole and the outer peripheral surface of the protrusion increases. It will be. When the gap increases, the rotation center axis of the core moves in the radial direction due to the unbalance amount of the core, and an eccentricity amount is generated between the rotation center axis of the protrusion and the rotation center axis of the core. Since the unbalance that affects the entire rotor is determined by the product of the amount of eccentricity and the mass of the core, even a minute deformation results in a large unbalance, and vibration increases when the rotor rotates.
As described above, when a resin-made solid core is used, the specific gravity of the resin, which is the material, is the same as or slightly lower than that of the sample filled in the rotor, so the gap becomes large. However, due to the hydraulic pressure applied to the core by centrifugal force, the core does not move much in the radial direction (see FIG. 10B), and the unbalance amount in the entire rotor does not change greatly. However, the metal hollow core with a large capacity has a small cavity inside the body and the specific gravity of the entire core is heavier than that of the sample. As a result, the core moves in the radial direction (see FIG. 10C), which results in a large unbalance for the rotor, and vibration may increase when the rotor rotates. If vibration increases during rotation, there is a possibility of physical damage to the rotating system of the rotor. Furthermore, there is a possibility of occurrence of liquid leakage at the mechanical seals provided at the ends of the upper shaft and the lower shaft. In order to prevent the vibration from increasing, there is a method of operating with a limitation on the rotational speed of the rotor. However, if the rotational speed of the rotor is reduced, the time required for the centrifugal process must be increased, Even in a centrifugal process using a hollow-type large-capacity core made of metal, it is required to enable a stable centrifugal process at the same rotational speed as that of a solid-type core.
An object of the present invention is to prevent the occurrence of vibration of the rotor even when the rotor is rotated at a high speed, and to enable efficient centrifugal processing.

The centrifuge of the present invention includes the above-described centrifuge rotor, a chamber that accommodates the centrifuge rotor, driving means that rotationally drives the centrifuge rotor within the chamber, and rotation of the centrifuge rotor. And a sample supply / discharge section for supplying and discharging the sample to and from the centrifuge rotor.
The rotor for a centrifuge has a first rotor cover and a second rotor cover provided at both ends of the rotor body, and a first protrusion provided on the inner surface of the first rotor cover is fitted. provided in the first end of the first fitting hole torso portion, the first pointed end abutting the conical shape centering holes of conical shape provided in the first fitting hole is first It provided the projections, a second fitting hole in which the second protrusion provided on the inner surface of the second rotor cover is fitted is provided on the second end of the barrel body, a second the second pointed end abutting the conical shape centering holes of conical shape provided in the fitting hole is provided on the second protrusion. Therefore, when the rotational speed of the rotor is low speed, the center axis of rotation of the core is set by fitting the protrusion and the fitting hole, and when the rotor is rotated at high speed, the respective tip and centering holes Thus, the rotation center axis of the core is positioned on the rotation center axis of the rotor body. As a result, when centrifugal processing is performed using a metal hollow-type large-capacity core, even if the rotor is rotated at a high speed, vibration of the rotor is prevented and the sample is efficiently centrifuged. Can do.
Hereinafter, embodiments of the present invention will be described in detail with reference to the drawings. A

The

As shown in FIG. 3, an
As shown in FIG. 4, a

The core 54 shown in FIGS. 6A and 6B is a solid type in which the

As shown in FIGS. 5 and 7, a conical lower centering
The angles of the conical surfaces of the upper and lower centering
As shown in FIG. 2, the
Since the upper and
In particular, as shown in FIG. 6D, in the
In the solid core shown in FIGS. 6A and 6B, the
The
5 is provided integrally with the
FIG. 9 is a partially cutaway cross-sectional view showing a
Since the unbalance that affects the entire rotating system is determined by the product of the eccentricity E and the mass of the core 54, even a slight deformation results in a large unbalance, and the vibration of the
