Description
The present invention relates to a system for producing an analysis sample of carbon alone used for measuring an isotope ratio of carbon 14 ( 14 C, C-14) contained in an organic sample.
In 14 C dating based on the fact that the abundance ratio (Natural Abundance) of 14 C, which is a radioisotope, is maintained almost constant in the Earth's biosphere, the measurement sensitivity and accuracy of 14 C are subject to analysis. This greatly affects the accuracy and error in determining the age of organic samples. On the other hand, in recent years, in the method using an accelerator mass spectrometer (AMS) that directly counts the number of 14 C atoms, measurement sensitivity (required sample amount), measurement time, measurement resolution, etc. have been greatly improved. It greatly contributes to the high accuracy, high efficiency, and low cost of 14C dating. Not only that, AMS itself has also made significant progress in miniaturization and automation.
Under such circumstances, 14 to further improve the throughput in C dating (generally used in carbon elemental form such as graphite) organic sample (analyte) from 14 C measurement sample pretreatment for preparing the Further efficiency improvement is desired. For example, in Non-Patent
Here, the gas generated by the combustion of the organic sample includes nitrogen oxide (NO x ), sulfur dioxide (SO 2 ), and water (H 2 O) as main components in addition to CO 2 . In order to select CO 2 from these gas components, from the point of excellent selectivity and simplicity, the gas produced by combustion is inserted into a gas holder such as an adsorption column to temporarily hold (carry) the target gas component. And a method using a difference in retention time peculiar to the gas component when the adsorption column is heated can be adopted. In this case, from the viewpoint of gas behavior, NO x is reduced to nitrogen (N 2 ) in advance, and H 2 O is removed with a water-absorbing agent, and then CO 2 and SO 2 are held in the adsorption column (that is, N 2 2 is preferably passed through the adsorption column). In this way, since the selection between CO 2 and SO 2 is relatively easy and the discrimination between the two is high, the selectivity and extractability of CO 2 can be improved.
That is, when the adsorption column holding CO 2 and SO 2 is adjusted to be heated to an appropriate temperature with an appropriate temperature gradient, CO 2 is desorbed before SO 2 . And as described in Non-Patent Document 1 (see Fig. 1 etc.) mentioned above, the adsorption column (Zeorite Co 2 trap) and the graphite production line (Reaction procedure) are connected, and the communication between the two is closed. Then, the inside of the line is evacuated in advance, and the vacuum of the line is released at an appropriate timing before CO 2 is desorbed from the adsorption column. As a result, the volume of CO 2 that is swept from the adsorption column is measured by a pressure sensor, and it is cooled by a cooling medium (Peltier cooler - Water trap) and collected in a dry ice state. it can. The CO 2 thus selected and collected is reduced by an appropriate reducing means and converted to graphite (carbon simple substance).
However, the present inventor has examined in detail the pretreatment of such an organic sample and a method of selectively extracting CO 2 to produce (purify) graphite using the organic sample. It turns out that there is room for further improvement.
That is, normally, NO x contained in the product gas generated by the combustion of the organic sample is reduced in advance as described above. However, when, for example, copper (Cu) or the like is used as the reducing agent, the consumption of the reducing agent is reduced. not completely implemented the reduction of the NO x by, NO x remaining in the product gas also is a fear that held by the adsorption column. Then, since CO 2 and NO x are inferior in discriminability at the time of desorption from the adsorption column, NO x is mixed into CO 2 sent to the graphite production line, and they can be cooled and collected together. As a result, the capacity of the collected gas component is increased as compared with the case of only CO 2, so that the required amount of reducing agent (eg, hydrogen (H 2 )) for generating graphite supplied based on the capacity is required. It will be more wasted.
In
Here, in consideration of the fact that N 2 produced by reducing NO x in the product gas due to the combustion of the organic sample is not collected in the graphite production line, the timing at which the vacuum of the graphite production line is released is the adsorption timing. Any time before desorbing CO 2 from the column should be sufficient. However, when using the LN 2 cooling collection of CO 2, for evaporation of LN 2 is fast at room temperature atmospheric pressure, and the timing of the introduction of CO 2 into the graphite product line, filling of LN 2 to LN 2 container and the timing of the deployment of the LN 2 container lines deviate disadvantageously, before the CO 2 is sent, the liquid surface of the LN 2 in LN 2 in the container will be excessively decreased by the transpiration Is also recalled.
If this happens, the cooling capacity for CO 2 may fluctuate, the CO 2 recovery rate may deteriorate, and the reproducibility of the CO 2 may deteriorate, or LN 2 to prevent such inconvenience. There is also a concern that it is necessary to use more than the necessary amount. In addition, as described above, when NO x is mixed into CO 2 , the refrigerant is also used to cool the NO x , so that more LN 2 is consumed than is necessary and sufficient. Then, in order to sufficiently cool CO 2 , a situation may arise in which extra LN 2 must be additionally used. In addition, the above (waste of refrigerant) is the same in that the electric energy for cooling it is wasted even if it is a cooling method using, for example, the Peltier element other than LN 2. apply.
Accordingly, the present invention has been made in view of such circumstances, and when a carbon simple substance such as graphite is produced from CO 2 contained in a gas generated by combustion of an organic sample, a CO 2 reducing agent or CO 2 is captured. To provide a sample preparation system for 14 C measurement capable of preventing waste of coolant or energy for collecting, and preventing deterioration of CO 2 recovery rate and deterioration of reproducibility thereof With the goal.
In order to solve the above problems, a sample preparation system for 14 C measurement according to the present invention is a system for preparing an analytical sample of simple carbon used for measuring an isotope ratio of 14 C contained in an organic sample, It comprises an element analyzer, a mass spectrometer connected to the element analyzer, and a carbon simple substance generator.
Among them, the elemental analyzer is included in the sample combustion section that generates a gas containing CO 2 , NO x , SO 2 , and H 2 O by burning an organic sample, and the generated gas generated in the sample combustion section A reducing part that reduces NO x to N 2 , a water absorbing part that removes H 2 O contained in the generated gas generated in the sample combustion part, and a conditioning gas containing CO 2 , N 2 , and SO 2 circulate. And a gas holding part that temporarily holds CO 2 and SO 2 contained in the adjustment gas, a heating part that heats the gas holding part, a carbon element (C) provided in the rear stage of the gas holding part and contained in the adjustment gas, An element analysis unit for analyzing nitrogen element (N) and sulfur element (S) is included.
Further, the mass spectrometer is supplied with at least a part of the adjustment gas from the element analyzer, and analyzes the stable isotope ratios of N, C, and S contained in the supplied adjustment gas. is there.
Furthermore, the carbon simple substance generator is supplied with at least a part of the remainder of the adjustment gas from the element analyzer, selectively collects CO 2 contained in the supplied adjustment gas, and from the CO 2 For obtaining carbon simple substance (including graphite and other carbon allotropes), a vent line connected to communicate with the gas holding part of the element analyzer, an open / close valve for opening / closing the vent line, and vent A trap that is provided in the line and collects CO 2 deaerated from the gas holding unit, a cooling unit that cools the trap to liquid nitrogen temperature using, for example, LN 2, and CO trapped in the trap 2 and a reducing agent supply unit that supplies a reducing agent (hydrogen (H 2 ) or the like) for reducing the CO 2 to carbon alone to the port.
The 14 C measurement sample preparation system according to the present invention is connected to at least the heating unit of the element analyzer, and the on-off valve and the cooling unit of the carbon simple substance generator, and holds the gas in which CO 2 is held. A heating command signal for starting heating of the part is sent to the heating part, and a valve opening / closing command signal for opening the opening / closing valve is sent to the opening / closing valve based on the heating command signal.
In the 14 C measurement sample preparation system configured as described above, first, an organic sample that has been subjected to appropriate preanalysis and pretreatment as necessary is subjected to, for example, sample combustion of an element analyzer by an automatic sample supply device. Supplied to the department. Next, when the organic sample is burned by heating with a heater, an electric furnace or the like while supplying oxygen (O 2 ) to the sample burning portion, CO 2 (may contain other CO x ; The same), NO x , SO 2 (may contain other SO x ; the same shall apply hereinafter), and a gas containing H 2 O. In the present invention, the gas generated by the combustion is referred to as “generated gas” for convenience as described above.
The generated gas generated in the sample combustion part is discharged from the sample combustion part to the reduction part by a sweep gas or a carrier gas made of an inert gas such as helium (He). The reducing unit is connected to the sample combustion unit in the form of a reducing tube filled with, for example, copper (Cu) or silver (Ag) grains, and is included in the generated gas when the generated gas circulates in the inside thereof. Of the gas components, NO x is reduced to N 2 . The gas containing CO 2 , N 2 , SO 2 , and H 2 O flows down in the element analyzer together with a sweep gas such as He or a carrier gas, and is discharged to the water absorption section. As this water absorption part, one connected to a reduction pipe in the form of a water absorption pipe filled with a water absorbing agent such as diphosphorus pentoxide (P 2 O 5 , P 4 O 10 ) can be used. When passing through the inside, only the H 2 O component contained in the generated gas is removed. Thus, the gas containing CO 2 , N 2 , and SO 2 (in the present invention, this gas is referred to as “adjustment gas” for convenience as described above) is sent to the gas holding unit.
As the gas holding unit, for example, a temperature programmed desorption (TPD) column having an adsorption tube filled with an appropriate gas adsorbent can be suitably used. Of the gas components contained in the adjustment gas, , CO 2 and SO 2 are adsorbed on the temperature-programmed desorption column, and N 2 is not adsorbed (passes through) there. Thereby, CO 2 and SO 2 derived from the organic sample contained in the adjustment gas are temporarily held in the gas holding unit. Further, an element analysis unit having, for example, a thermal conductivity detector (TCD) is provided downstream of the gas holding unit in the gas flow path, and passes through the temperature-programmed desorption column of the gas holding unit. N 2 is detected by the thermal conductivity detector and qualitative determination of N is performed. At this time, the elemental analyzer, the mass spectrometer, and the carbon simple substance generator are isolated from each other by a valve (such as the on-off valve provided in the above-described ventilation line) so as not to communicate with each other.
In addition, the gas holding unit is configured such that the temperature-programmed desorption column is heated at a predetermined temperature gradient so as to reach a predetermined temperature by a heating unit having heating means such as an electric heater. When the gas holding unit is heated by the heating unit, CO 2 and SO 2 adsorbed and held by the gas holding unit are desorbed in this order with a time difference corresponding to the retention time at a predetermined temperature. Then, by opening the connection between the element analyzer and the mass spectrometer and / or the carbon simple substance generator from the isolated state, N 2 that has reached the element analysis section, and sequentially from the gas holding section by heating with a time difference The desorbed CO 2 and SO 2 are further exhausted together with the sweep gas or carrier gas in that order toward the mass spectrometer and / or the carbon simple substance generator. At this time, CO 2 and SO 2 are detected by the thermal conductivity detector when passing through the elemental analysis unit, and C and S are qualitatively quantified.
Then, at least a part of the adjustment gas sent to the mass spectrometer is obtained by, for example, isotopic ratio mass spectrometry (IRMS), each of the stable isotope ratios of N, C, and S ( δ 15 N, δ 13 C, and δ 34 S) are measured.
On the other hand, before the connection between the ventilation line and the element analyzer is opened, the carbon atmosphere is exhausted to a high vacuum by, for example, a turbo pump, and the on-off valve for opening and closing the ventilation line is opened. Then, when communicating with the gas holding unit, N 2 , CO 2 , and SO 2 which are at least a part of the remainder of the adjustment gas sent from the gas holding unit via the elemental analysis unit are in this order. It is sent to the system at high speed. Further, the ventilation line is provided with a trap for collecting CO 2 , and the trap is cooled to the liquid nitrogen temperature by the cooling unit, for example, with low-pressure He introduced as necessary. The Thus, CO 2 is collected when each gas component of the adjustment gas passes through the cooled trap.
The CO 2 collected in the trap is moved to the port using vacuum distillation using LN 2 and stored therein. Thereafter, a reducing agent such as H 2 is supplied to the port from the reducing agent supply unit, whereby CO 2 is reduced to simple carbon (C) such as graphite, and water generated by the reduction reaction with the simple carbon in the port ( H 2 O) is encapsulated.
It should be noted that a plurality of ports for storing and collecting CO 2 are preferably installed as described above, and when a plurality of organic samples are continuously supplied to the element analyzer and processed, they are different for each organic sample. By assigning ports, a plurality of large quantities of simple carbon samples can be obtained. These carbon simple substance samples are subjected to an appropriate accelerator mass spectrometer (AMS) for 14 C dating after H 2 O is removed by cooling or the like.
And in the sample preparation system for 14 C measurement according to the present invention, the gas holding in which CO 2 is held from the heating part of the elemental analyzer and the control part connected to the on-off valve and the cooling part of the carbon simple substance generator. A heating command signal for starting the heating of the part is sent to the heating part, and a valve opening / closing command signal for opening the opening / closing valve of the ventilation line is sent to the opening / closing valve based on the heating command signal.
At this timing, when the on-off valve of the vent line is opened, the N 2 from the element analyzer is removed from the vent line before the gas holding part of the element analyzer is heated sufficiently and CO 2 is desorbed and released. Exhausted through. At this time, if so CO 2 can not be sufficiently performed cooling trap before being desorbed released, for example, the CO 2 LN 2 or necessary to cool collecting, to run the cooling element The electric energy is not wasted. Then, when the gas holder is sufficiently heated reaches a desorption temperature of CO 2, CO 2 is rapidly desorbed released, drawn into the vent line of the carbon single generator. At this point, if the trap is sufficiently cooled to a temperature suitable for CO 2 capture (for example, liquid nitrogen temperature), CO 2 that has reached the trap can be collected almost certainly.
Further, as described above, it can be recalled that NO x contained in the generated gas generated in the sample combustion part of the elemental analyzer is not completely reduced to N 2 in the reduction part. However, the 14 C measurement sample preparation according to the present invention is possible. Even in such a case, in the system, NO x is effectively prevented from being collected in the trap of the carbon simple substance generator.
That, NO x unreduced, although held by suction, for example, temperature-programmed desorption column of the gas holder with CO 2 and SO 2, the gas holder is heated, desorption before the CO 2 Is done. Then, the NO x previously desorbed from the gas holding unit is exhausted through the ventilation line before the gas holding unit is heated and CO 2 is desorbed and released, as in the case of N 2 described above. Here, even if NO x flows into the trap, as described above, if the trap is not sufficiently cooled at this point, NO x is collected in the trap. Can be prevented. In this way, it is possible to prevent NO x from being mixed into the CO 2 sent to the port, so that it is possible to prevent an increase in the amount of H 2 supplied to reduce the CO 2 thereafter. It becomes.
Further, if attention is paid to the timing of cooling the trap in this way, the cooling that starts cooling the trap from the control unit triggered by the heating command signal sent to the heating unit or the valve opening / closing command signal sent to the on-off valve. It is also preferable that the command signal is sent to the cooling unit. In this case, the cooling command signal may be sent not only simultaneously with or after the heating command signal or valve opening / closing command signal, but also before the heating command signal or valve opening / closing command signal. Good.
That is, “as an opportunity” means that a cooling command signal is sent at the same time as, and before and after, the sending of a heating command signal or a valve opening / closing command signal as a reference time. For example, in a specific system, the time from when the heating command signal or the valve opening / closing command signal is sent until the CO 2 reaches the trap, and the time until the trap reaches a temperature suitable for CO 2 collection from the start of cooling. Can be determined in advance, and the timing for sending the cooling start signal can be appropriately determined in consideration of these times and the reference time.
Further, 14 C measurement sample preparation method according to the invention is, for example, a suitably viable manner using 14 sample preparation system for C measured according to the above present invention, the organic matter sample by burning CO 2, NO A sample combustion step for generating a gas containing x , SO 2 , and H 2 O, a reduction step for reducing NO x contained in the generated gas generated by the combustion to N 2 , and removing H 2 O contained in the generated gas Water removal step, gas holding step for temporarily holding CO 2 and SO 2 contained in the adjustment gas containing CO 2 , N 2 , and SO 2 by the gas holding unit, heating step for heating the gas holding unit by the heating unit, adjustment An elemental analysis process having an elemental analysis step for analyzing C, N, and S contained in the gas, and each of N, C, and S contained in at least a part of the adjustment gas that has passed through the elemental analysis process And selectively collecting CO 2 contained in at least a part of the remainder of the adjustment gas via the mass analysis step for analyzing the stable isotope ratio and the elemental analysis step, and removing carbon from the CO 2 A simple carbon production step.
In addition, in the carbon simple substance production step, an opening / closing step for opening and closing an opening / closing valve provided in the ventilation line connected to communicate with the gas holding unit, and a trap provided in the ventilation line are cooled by the cooling unit and from the gas holding unit. A cooling and collecting step for collecting the deaerated CO 2 , a storage step for storing the CO 2 collected in the trap in the port, and a reducing agent for reducing the CO 2 to simple carbon are supplied to the port. Performing a reducing agent supply step.
Further, in the 14 C measurement sample preparation method according to the present invention, a heating command signal for starting heating of the gas holding unit in which CO 2 is held is sent to the heating unit, and the ventilation line is based on the heating command signal. A control step of sending a valve opening / closing command signal for opening the opening / closing valve to the opening / closing valve, and sending a cooling command signal for starting cooling of the trap to the cooling unit, which is executed.
According to the present invention, based on the heating command signal for starting the heating of the gas holding unit in which CO 2 contained in the generated gas generated by the combustion of the organic sample is held by the control unit, the ventilation line of the carbon simple substance generating device since opening the on-off valve provided in, when generating carbon alone such as graphite from CO 2 of organic origin sample, the waste of coolant or energy for collecting the reducing agent and CO 2 in the CO 2 Can be deterred. This also makes it possible to prevent a reduction in the CO 2 recovery rate and a deterioration in its reproducibility.

An example of the thus constructed 14 C graphite manufacturing procedure as the measurement
First, a plurality of
The generated gas generated in the
When the adjustment gas flows through the temperature-programmed
As described above, at this time, the
Next, when a heating command signal is sent from the
Further, the valve V1b is closed with respect to the on-off valves V1 and V2 provided in the
As a result, the N 2 that has reached the
In the present embodiment, the adjustment gas (N 2 , CO 2 , and SO 2 ) that has reached the
The conditioned gas (N 2 , CO 2 , and SO 2 ) sequentially delivered to the
On the other hand, as described above, when the valve opening / closing command signal from the
Here, the
Thereafter, when the temperature-programmed
In other words, the
Then, after the degassing of CO 2 from the temperature-programmed
Next, the CO 2 collected in the
The
After confirming that the inside of the carbon simple substance production |

Further, as described above with respect to the prior art, it is recalled that NO x contained in the generated gas obtained in the
From the above, when generating graphite from CO 2 contained in the product gas generated by the combustion of the organic sample contained in the
In addition, as above-mentioned, this invention is not limited to said embodiment, A various deformation | transformation is possible in the limit which does not change the summary. For example, as the cooling means for the
As described above, the sample preparation system for 14 C measurement of the present invention can suppress the waste of CO 2 reducing agent (H 2 ), coolant (LN 2 ) for collecting CO 2 or energy. As a result, it is possible to improve the economic efficiency, prevent a reduction in the CO 2 recovery rate and the deterioration of the reproducibility thereof, and the throughput for 14 C dating by the recent AMS. Therefore, it is possible to greatly contribute to the promotion of 14 C measurement with high accuracy with respect to environmental samples and historical samples, and application of 14 C measurement to the medical field, for example, microdose clinical drug It can be widely used for the application of AMS to dynamic research.
