Description
本願は、米国仮特許出願第61/248,300号(2009年10月2日出願)に基づく優先権を主張する。該出願は、その全体が参照により本明細書に援用される。
本教示は、様々な生物学的、化学的および/または細胞生物学的用途に有用な試料調製のためのデバイスおよび方法に関する。
例えば、本願発明は以下の項目を提供する。
(項目1)
試料処理のためのデバイスであって、
出口を有する少なくとも1つのチャンバであって、処理用の試料を受け取るように構成されたチャンバと、
該少なくとも1つのチャンバ内の少なくともいくつかの試料部分が通過して流動するフィルタと、
該少なくとも1つのチャンバ内の試料を収容するための第1の状態で配置されたバリア部材と
を備え、
十分な条件になると、該バリア部材は、該チャンバ内に収容される少なくともいくつかの試料部分を、該出口に向かう流動方向に該フィルタを通して流動させるための第2の状態に変化可能である、デバイス。
(項目2)
前記フィルタは、閾値サイズ未満の試料部分を通過させ、少なくとも該閾値サイズの試料部分の通過を阻止するように構成されている、項目1に記載のデバイス。
(項目3)
前記フィルタは、前記少なくとも1つのチャンバ内に導入される試料中に含有される標的分子を通過させるように構成されている、項目1に記載のデバイス。
(項目4)
前記フィルタは、溶解培地に不溶である少なくともいくつかの試料部分の通過を阻止するように構成されている、項目1に記載のデバイス。
(項目5)
前記フィルタは、フリットおよび官能化樹脂のうちの少なくとも一つを備えている、項目1に記載のデバイス。
(項目6)
少なくとも前記第1の状態において、前記バリア部材は、前記フィルタに付着している、項目1に記載のデバイス。
(項目7)
前記バリア部材は、十分な力に供されると前記第2の状態に変化可能である、項目1に記載のデバイス。
(項目8)
前記バリア部材は、膜を備えている、項目1に記載のデバイス。
(項目9)
前記フィルタおよび前記バリア部材は、前記少なくとも1つのチャンバ内において、該少なくとも1つのチャンバの入口と前記出口との間に配置されている、項目1に記載のデバイス。
(項目10)
前記少なくとも1つのチャンバは、変形可能構造によって少なくとも部分的に規定される、項目1に記載のデバイス。
(項目11)
前記少なくとも1つのチャンバは、管によって規定される、項目1に記載のデバイス。
(項目12)
前記少なくとも1つのチャンバは、アレイ状に配置された複数のチャンバを含む、項目1に記載のデバイス。
(項目13)
前記少なくとも1つのチャンバと流体的に直列接続された少なくとも1つの追加的チャンバをさらに備え、該少なくとも1つの追加的チャンバは、前記第1の状態の前記バリア部材を介して該少なくとも1つのチャンバとの流通から分離されている、項目1に記載のデバイス。
(項目14)
前記少なくとも1つの追加的チャンバ内に配置された追加的フィルタおよび追加的バリア部材のうちの少なくとも一つをさらに備えている、項目13に記載のデバイス。
(項目15)
前記少なくとも1つの追加的チャンバは、採取チャンバを含む、項目13に記載のデバイス。
(項目16)
前記少なくとも1つのチャンバおよび前記少なくとも1つの追加的チャンバは、互いに入れ子状に係合するように構成されている、項目13に記載のデバイス。
(項目17)
試料を調製するための方法であって、
流体的に直列接続された複数のチャンバのうちの第1のチャンバ内に試料を配置することであって、連続したチャンバが、第1の状態のそれぞれのバリア部材により互いに分離されている、ことと、
該試料を、該第1のチャンバ内での処理アッセイに供することと、
所定期間後、該第1のチャンバと第2の連続したチャンバとを分離しているそれぞれのバリア部材を第2の状態に変化させることにより、該第1のチャンバから該第2の連続したチャンバに、少なくともいくつかの試料部分を流動させることと
を含み、
該第1の状態のバリア部材は、該バリア部材の場所を越える流動を防止し、該第2の状態のバリア部材は、該第1のチャンバから該第2の連続したチャンバへの流動を可能にする、方法。
(項目18)
前記流動させることは、少なくとも閾値サイズの材料を前記第1のチャンバ内に保持し、該閾値サイズより小さい材料を前記第2の連続したチャンバに通過させるフィルタを通して試料部分を流動させることを含む、項目17に記載の方法。
(項目19)
前記フィルタを通して試料部分を流動させることは、前記フィルタを通した標的分子の通過を可能にする、項目18に記載の方法。
(項目20)
前記バリア部材を変化させることは、前記バリア部材に力を印加することを含む、項目17に記載の方法。
(項目21)
前記試料を処理アッセイに供することは、該試料から標的分子を抽出するために、該試料を破砕に供することを含む、項目17に記載の方法。
(項目22)
前記試料を破砕に供することは、前記試料を溶解に供することを含む、項目21に記載の方法。
(項目23)
前記標的分子は、核酸、ペプチド、タンパク質、およびバイオポリマーから選択される、項目21に記載の方法。
(項目24)
前記試料を、前記第2のチャンバ内で第2の処理アッセイに供することと、
所定期間後、前記第2のチャンバと第3の連続したチャンバとを分離しているそれぞれのバリア部材を、前記第1の状態から前記第2の状態に変化させることにより、該第2のチャンバから該第3のチャンバに試料部分を流動させることと
をさらに含む、項目17に記載の方法。
(項目25)
試料処理のためのデバイスであって、
出口を有する少なくとも1つのチャンバであって、処理用の試料を受け取るように構成された少なくとも1つのチャンバと、
該チャンバに対して配置された第1の状態のバリア膜であって、該第1の状態において該チャンバ内に該試料を収容するバリア膜と
を備え、
該少なくとも1つのチャンバ内で十分な条件になると、該バリア膜は、該出口に向かう流動方向に該チャンバ内の試料部分を流動させる第2の状態に変化可能である、デバイス。
Claims (5)
- 試料を調製するための方法であって、前記方法は、
流体的に直列接続された複数のチャンバのうちの第1のチャンバ内に試料を配置することであって、連続したチャンバが、第1の状態のそれぞれのバリア部材により互いに分離されている、ことと、
前記試料を、前記第1のチャンバ内での処理アッセイに供することと、
所定期間後、前記第1のチャンバと第2の連続したチャンバとを分離している前記それぞれのバリア部材を第2の状態に変化させることにより、前記第1のチャンバから前記第2の連続したチャンバに、少なくともいくつかの試料部分を流動させることと
を含み、
前記第1の状態の前記バリア部材は、前記バリア部材の場所を越える流動を防止し、前記第2の状態の前記バリア部材は、前記第1のチャンバから前記第2の連続したチャンバへの流動を可能にする、方法。 - フィルタを通して前記試料部分を流動させることが、前記フィルタを通した標的分子の通過を可能にする、請求項1に記載の方法。
- 前記バリア部材を変化させることは、前記バリア部材に力を印加することを含む、請求項1に記載の方法。
- 前記試料を処理アッセイに供することは、前記試料から標的分子を抽出するために、前記試料を破砕に供することを含む、請求項1に記載の方法。
- 前記標的分子は、核酸、ペプチド、タンパク質、およびバイオポリマーから選択される、請求項4に記載の方法。
