Description
[1] 5’末端から順に、
(a)レトロウイルス由来の5’LTR(Long Terminal Repeat)配列、
(b)スプライスドナー(SD)配列、
(c)レトロウイルス由来のパッケージングシグナル配列(ψ)、
(d)少なくとも1つのステム−ループ構造を形成し、哺乳動物細胞内でRNA干渉を誘導するRNAが転写されるsiRNA生成配列、
(e)スプライスアクセプター(SA)配列、
(f)所望の遺伝子の配列、
(g)レトロウイルス由来の3’LTR配列、
の各配列の核酸を含むレトロウイルスベクター、
[2] 所望の遺伝子がT細胞受容体(TCR)をコードする遺伝子である[1]記載のレトロウイルスベクター、
[3] 所望の遺伝子がポリシストロニックに連結したTCRα鎖及びTCRβ鎖をコードする遺伝子である[2]記載のレトロウイルスベクター、

(1)本発明のレトロウイルスベクター
本発明のレトロウイルスベクターは、5’末端から順に、
(a)レトロウイルス由来の5’LTR配列、
(b)SD配列、
(c)レトロウイルス由来のパッケージングシグナル配列(ψ)、
(d)少なくとも1つのステム−ループ構造を形成し、哺乳動物細胞内でRNA干渉を誘導するRNAが転写されるsiRNA生成配列、
(e)SA配列、
(f)所望の遺伝子の配列、
(g)レトロウイルス由来の3’LTR配列、
の各配列の核酸を含む核酸構築物である。本発明は、1つのベクターを細胞に導入することにより、当該細胞における特定の遺伝子の発現抑制、及び導入された所望の遺伝子の発現を可能とする。本発明のベクターは、siRNAの生成効率及び/又は所望の遺伝子の発現効率が高い。従って、変異型遺伝子の発現を抑制し野生型遺伝子を発現させるなど、細胞内で発現する遺伝子の機能を新たに導入する別の遺伝子により代替する目的で使用することができる。
本発明の遺伝子細胞の製造方法は、前記(1)に記載する本発明のレトロウイルスベクターを細胞に導入する工程を包含することを特徴とする。当該工程は体外で実施される。
本発明の遺伝子導入細胞は、前記(2)の製造方法により、前記(1)のレトロウイルスベクターが導入された細胞である。本発明の遺伝子導入細胞は、外来性の所望の遺伝子を発現し、特定の内在性遺伝子の発現が抑制されている。
また、本明細書に記載の操作のうち、基本的な操作については2001年、コールド スプリング ハーバー ラボラトリー発行、T.マニアティス(T.Maniatis)ら編集、モレキュラー クローニング:ア ラボラトリー マニュアル第3版(Molecular Cloning:A Laboratory Manual 3rd ed.)に記載の方法によった。
国際公開第2008/153029号パンフレット(出典明示により、本明細書の一部とする)に記載の方法に従い、コドン変換型のヒト抗MAGE−A4 TCRα鎖遺伝子を、野生型の遺伝子の一部を変換して作製した。この遺伝子を含む核酸断片を、pPCR−Script(Stratagene社)の制限酵素KpnI−XhoIサイトにクローニングした。
同様に、コドン変換型のヒト抗MAGE−A4 TCRβ鎖遺伝子を、野生型の遺伝子の一部を変換して作製した。この遺伝子を含む核酸断片を、pPCR−Scriptの制限酵素KpnI−XhoIサイトにクローニングした。
まず、pMSCVneo(Clontech社製)を鋳型に配列番号1記載の3MSCV5プライマー及び配列番号2記載の3MSCV3プライマーでPCRを行い、MSCV3’LTR部位を増幅し、制限酵素XhoIとEcoRIで切断し、pMTベクター[ジーン セラピー(Gene Ther)、第11巻、第94−99頁(2004)に記載されているpMTベクター]のXhoI−EcoRIサイトにクローニングし、pMS−MCを作製した。更に、pMEI−5ベクター(タカラバイオ社製)を制限酵素MluIとXhoIにて切断し、pMS−MCのMluI−XhoIサイトへ挿入し、pMS3−MCを作製した。pMS3−MCは、5’末端から順にMMLV由来の5’LTR、MMLV由来のSD配列、MMLV由来のψ、変異を導入したヒトEF1α遺伝子由来のSA配列、U3領域はMSCV由来で残りの領域はMMLV由来の3’LTRを含む。
図1に示すように、pMS3−MCに挿入するDNA断片を調製した。コドン変換型のヒト抗MAGE−A4 TCRα鎖をコードするDNA(図1中、TCRαと表記する)を配列番号3記載のMAGE−AFプライマー及び配列番号4記載のMAGE−ARプライマーを用いてPCRにて増幅した。同様にコドン変換型のヒト抗MAGE−A4 TCRβ鎖をコードするDNA(図1中、TCRβと表記する)を配列番号5記載のMAGE−BFプライマー及び配列番号6記載のMAGE−BRプライマーを用いてPCRにて増幅した。前記の2種のDNAの増幅には実施例1で作製された組換えプラスミドを鋳型に用いた。更に、配列番号7に示すT2Aペプチド配列を含む人工合成遺伝子(図1中、T2Aと表記する)を鋳型として配列番号8記載のT2A−Fプライマー及び配列番号9記載のT2A−Rプライマーを用いてPCRにて増幅した。これらの増幅産物を、NotI−XhoIにて消化したpMS3−MCベクターにIn−Fusion Advantage PCR Cloning Kit(クロンテック社製)を使用してクローニングし、pMS3−MAGE−A4−b2Aaベクターを作製した。
配列番号10に示す人工遺伝子(図2中、siRNA生成配列と表記する)を合成した。この人工遺伝子は、4種類のステム−ループ構造を形成する一本鎖RNAを転写し、野生型TCRα及び野生型TCRβに対するsiRNA各2種類合計4種類を生成する。この人工合成遺伝子を鋳型とし、配列番号11に示すloop−Mlu−Fプライマーと配列番号12に示すloop−Mlu−Rプライマーを用いたPCRを行って増幅DNA断片を得た。図2に示すように、このDNA断片をpMS3−MAGE−A4−b2AaベクターのMluI消化物へ、In−Fusion Advantage PCR Cloning Kit(クロンテック社製)を使用してクローニングし、pMu1−MAGE−A4−b2Aaベクターを作製した。
プラスミドベクターpMS3−MAGE−A4−b2Aa及びpMu1−MAGE−A4−b2Aaにより大腸菌JM109をそれぞれ形質転換し、形質転換体を得た。これら形質転換体の保持するプラスミドDNAをQIAGEN Plasmid Midi Kit(キアゲン社製)を用いてそれぞれ精製し、トランスフェクション用DNAとして以下の操作に供した。
調製したpMS3−MAGE−A4−b2Aa及びpMu1−MAGE−A4−b2Aaプラスミドを293T細胞にそれぞれトランスフェクトした。この操作にはRetorovirus Packaging Kit Eco(タカラバイオ社製)をその製品プロトコールに従って使用した。得られた形質導入細胞より各種エコトロピックウイルスを含有する上清液を獲得し、0.45μmフィルター(Milex HV、ミリポア社製)にてろ過した。この上清を用いて、ポリブレンを使用する方法によりPG13細胞(ATCC CRL−10686)細胞にエコトロピックウイルスを感染させた。得られた細胞の培養上清を回収し、0.45μmフィルターによりろ過し、コドン変換型TCR発現レトロウイルス溶液(MS3−MAGE−A4−b2Aa)及びコドン変換型TCR−siRNA共発現レトロウイルス溶液(Mu1−MAGE−A4−b2Aa)とした。
インフォームドコンセントの得られたヒト末梢血より分離した末梢血単核球(PBMC)に、実施例4で作製したコドン変換型TCR発現レトロウイルス溶液(MS3−MAGE−A4−b2Aa)及びコドン変換型TCR−siRNA共発現レトロウイルス溶液(Mu1−MAGE−A4−b2Aa)を、レトロネクチン(登録商標、タカラバイオ社製)を用いた標準的な方法で2回感染を行い、コドン変換型TCR導入PBMC及びコドン変換型TCR導入−siRNA共発現PBMCをそれぞれ作製した。2回目のウイルス感染3日後に細胞を回収し、QIAGEN RNeasy Mini Kit(キアゲン社製)にて全RNAの抽出及びDNaseI処理を行った。得られた全RNAを鋳型とし、PrimeScript RT reagent Kit(Perfect Real Time)(タカラバイオ社製)を用いたcDNA合成を行った。更に、このcDNAを鋳型とし、SYBR Premix Ex Taq II(タカラバイオ社製)及び配列番号13、14の野生型TCRα増幅用プライマー、配列番号15、16の野生型TCRβ増幅用プライマーを用いたリアルタイムPCRを行い、野生型TCRα、野生型TCRβ遺伝子発現量を測定してその相対値を算出した。全RNA量の補正は配列番号17、18のGAPDH遺伝子増幅用プライマーを用いた同様のリアルタイムPCRで測定したGAPDH遺伝子発現量に基づいて行った。
実施例5と同様に、コドン変換型TCR導入PBMC及びコドン変換型TCR導入−siRNA共発現PBMCを作製した。2回目のウイルス感染3日後に細胞を回収し、HLA−A2402 MAGE−A4 テトラマー−PE(MBL社製)及びFITC標識抗Human CD8 抗体(べクトンディッキンソン社製)により染色した。フローサイトメーターを使用し、染色後の細胞について、CD8陽性であって、かつテトラマー陽性である細胞の割合及びPEの平均蛍光強度を測定した。図4にMAGE−A4テトラマー陽性細胞率を示し、図5にMAGE−A4テトラマー陽性細胞に由来するフィコエリスリン(PE)の平均蛍光強度を示す。横軸は、導入したレトロウイルス及び希釈率を示し、縦軸はMAGE−A4テトラマー陽性細胞率(図4)及びMAGE−A4テトラマー陽性細胞由来のPEの平均蛍光強度(図5)を示す。なお、図5の平均蛍光強度はMAGE−A4テトラマー陽性細胞における、抗MAGE−A4 TCRの発現量を反映している。図4及び5に示されるように、コドン変換型TCR導入−siRNA共発現PBMC(Mu1−MAGE−A4−b2Aa)は、野生型TCRα及びβの発現が抑制されたことにより、導入されたコドン変換型TCRのα/β複合タンパクの発現率及び発現強度の増強が見られた。
An Jら、インターナショナル ジャーナル オブ ヘマトロジー(Int.J.Hematol.)、第93巻、第176−185頁(2011)(出典明示により、本明細書の一部とする)の記載に従い、腫瘍抗原WT1の235−243のペプチドを認識するTCRα鎖遺伝子又はTCRβ鎖遺伝子を含む核酸断片をそれぞれ調製した。TCRα鎖及びTCRβ鎖をコードする遺伝子のC領域に対し、野生型のTCRの塩基配列を、コドン変換型TCRとする表1に示すそれぞれ2個所の変異を、変異導入プライマーとPCRにより導入した。野生型とコドン変換型の塩基配列がコードするアミノ酸配列は共通であるが、コドン変換型TCRの塩基配列には変異が導入されているので、実際例3記載のDNAより転写される二本鎖siRNAによりコドン変換型TCRの発現は抑制されない。
実施例7で作製したコドン変換型のヒト抗WT1 TCRα鎖遺伝子及びTCRβ鎖遺伝子を含む核酸断片を鋳型としてPCRにより増幅した増幅産物、実施例2に記載するT2Aペプチド配列をコードする人工遺伝子の増幅産物を、ApaI−XhoIにて消化した実施例2のpMS3−MCベクターにIn−Fusion Advantage PCR Cloning Kit(クロンテック社製)を使用してクローニングし、pMS3−WT1−a2Abベクター(ベクターA)を作製した。
実施例3で合成した4種類のsiRNAを生成する人工遺伝子の増幅産物を、図8に示すように、pMS3−WT1−a2Abベクター(ベクターA)の各部位に挿入した。すなわち、TCR遺伝子と3’LTRの間に挿入したpMS3−WT1−a2Ab−loop−siTCR(ベクターB)、SA配列とTCR遺伝子の間に挿入したpMS3−loop−WT1−a2Ab−siTCR(ベクターC)、SD配列とSA配列の間に挿入したpMu1−WT1−a2Ab−siTCR(ベクターD、以下「pMu1−WT1−a2Ab」と称する場合もある)を作製した。
実施例8及び実施例9で調製した各ベクターから、実施例4と同様の方法でレトロウイルス溶液を作製した。実施例6と同様の方法でヒトPBMCに各ベクターを導入し、2回目のウイルス感染3日後に細胞を回収して、WT1テトラマー陽性細胞率、WT1テトラマー陽性細胞の平均蛍光強度、及びゲノムに組み込まれたウイルスコピー数の測定を行った。図9及び図10に結果を示す。図中、横軸はウイルスコピー数を示し、縦軸は、WT1テトラマー陽性細胞率(図9)及びWT1テトラマー陽性細胞の平均蛍光強度(図10)を示す。図9及び図10に示すように、ベクターDが導入された細胞はベクターA〜Cが導入された細胞と比べて、より低いウイルスコピー数でより高いWT1テトラマー陽性細胞率と平均蛍光強度が得られ、導入したコドン変換型抗TCRのα/β複合タンパク質の発現率及び発現強度の増強が見られた。
図11に示すように、実施例2にて作製したpMS3−MCに挿入するDNA断片を調製した。コドン変換型のヒト抗MAGE−A4 TCRα鎖をコードするDNA(図11中、TCRαと表記する)を配列番号31記載のMAGE−AF2プライマー及び配列番号32記載のMAGE−AR2プライマーを用いてPCRにて増幅した。同様にコドン変換型のヒト抗MAGE−A4 TCRβ鎖をコードするDNA(図11中、TCRβと表記する)を配列番号33記載のMAGE−BF2プライマー及び配列番号34記載のMAGE−BR2プライマーを用いてPCRにて増幅した。前記の2種のDNAの増幅には実施例1で作製された組換えプラスミドを鋳型に用いた。更に、配列番号7に示すT2Aペプチド配列を含む人工合成遺伝子(図11中、T2Aと表記する)を鋳型として配列番号8記載のT2A−Fプライマー及び配列番号9記載のT2A−Rプライマーを用いてPCRにて増幅した。これらの増幅産物を、NotI−XhoIにて消化したpMS3−MCベクターにIn−Fusion Advantage PCR Cloning Kitを使用してクローニングし、pMS3−MAGE−A4−a2Abベクターを作製した。
実施例3と同様にして、siRNA生成配列を含むDNA断片を、MluIにて消化した実施例11のpMS3−MAGE−A4−a2Abベクターへクローニングし、pMu1−MAGE−A4−a2Abベクターを作製した(図12)。
実施例7で作製したコドン変換型のヒト抗WT1 TCRα鎖遺伝子及びTCRβ鎖遺伝子を含む核酸断片を鋳型としてPCRにより増幅した増幅産物、実施例2に記載するT2Aペプチド配列をコードする人工遺伝子の増幅産物を、ApaI−XhoIにて消化した実施例2のpMS3−MCベクターにクローニングし、pMS3−WT1−b2Aaベクター(ベクターE)を作製した(図13)。
実施例3で合成した4種類のsiRNAを生成する人工遺伝子の増幅産物を、実施例13のpMS3−WT1−b2Aaベクター(ベクターE)のSD配列とSA配列の間に挿入し、pMu1−WT1−b2Aa(ベクターF)を作製した(図13)。
実施例2、3、11、12で調製した各ベクター(pMS3−MAGE−A4−b2Aa、pMu1−MAGE−A4−b2Aa、pMS3−MAGE−A4−a2Ab、pMu1−MAGE−A4−a2Ab)を用いて実施例4と同様の方法でレトロウイルス溶液を作製した。実施例6と同様の方法でヒトPBMCに各ベクターを導入し、2回目のウイルス感染3日後に細胞を回収して、MAGE−A4テトラマー陽性細胞率、MAGE−A4テトラマー陽性細胞の平均蛍光強度、及びゲノムに組み込まれたウイルスコピー数の測定を行った。図14にpMS3−MAGE−A4−b2AaとpMS3−MAGE−A4−a2Abの結果を示す。図15にpMu1−MAGE−A4−b2AaとpMu1−MAGE−A4−a2Abの結果を示す。図中、横軸はウイルスコピー数を示し、縦軸は、MAGE−A4テトラマー陽性細胞率(図14左、図15左)及びMAGE−A4テトラマー陽性細胞の平均蛍光強度(図14右、15右)を示す。図14及び図15に示すように、MS3−MAGE−A4−b2Aaベクターが導入された細胞はMS3−MAGE−A4−a2Abベクターが導入された細胞と比べて、より低いウイルスコピー数でより高いMAGE−A4テトラマー陽性細胞率と平均蛍光強度が得られ、また、Mu1−MAGE−A4−b2Aaベクターが導入された細胞はMu1−MAGE−A4−a2Abベクターが導入された細胞と比べて、より低いウイルスコピー数でより高いMAGE−A4テトラマー陽性細胞率と平均蛍光強度が得られ、導入したコドン変換型抗TCRのα/β複合タンパク質の発現率及び発現強度の増強が見られた。

- 5’末端から順に、
(a)レトロウイルス由来の5’LTR(Long Terminal Repeat)配列、
(b)スプライスドナー(SD)配列、
(c)レトロウイルス由来のパッケージングシグナル配列(ψ)、
(d)少なくとも1つのステム−ループ構造を形成し、哺乳動物細胞内でRNA干渉を誘導するRNAが転写されるsiRNA生成配列、
(e)スプライスアクセプター(SA)配列、
(f)所望の遺伝子の配列、
(g)レトロウイルス由来の3’LTR配列、
の各配列の核酸を含むレトロウイルスベクターであって、所望の遺伝子がポリシストロニックに連結したTCRα鎖及びTCRβ鎖をコードする遺伝子である前記ベクター。 - siRNA生成配列が野生型TCRの定常領域をコードするmRNAに作用して発現を抑制するsiRNAを生成する配列であり、所望の遺伝子が定常領域の塩基配列に変異を導入したTCRをコードする遺伝子であり、前記変異を導入したTCRをコードする遺伝子は前記siRNAにより発現の抑制を受けない、請求項1記載のレトロウイルスベクター。
- レトロウイルスがオンコレトロウイルス又はレンチウイルスである請求項1記載のレトロウイルスベクター。
