Description
上記したように、リグニンに、リグニン分解用触媒として、ポルフィリン(ピロール化合物)を添加することによって、アルコール類および有機酸類を製造でき、リグニンの光分解を行って分解生成物を得ることができると共に、水素イオンを遊離することができる。また、ポルフィリン構造を有するテトラピロール化合物や合成ポルフィリンが、ダイオキシンなどの芳香族炭化水素に対する分解用触媒として有効である。この場合に、ポルフィリンとして、例えば、大腸菌を用いて生物学的に製造したものを用いることができる。本製造例では大腸菌を用いるピロール化合物の製造例について説明する。

・その他: NOESYの混合時間は400msecに設定した。
図3に1H NMRスペクトルを示した(溶媒:CD3OD)。その結果、10.0〜10.5ppm付近(d)、4.3ppm付近(f)、3.6ppm付近(g)、および3.2ppm付近(e)に、目的成分に由来すると推測されるシグナルが観測された。それぞれのシグナル強度比は、およそ1:2:3:2であった。このd〜gはその他の図面においても同じである。
図5にHSQCスペクトルを示した。HSQCスペクトルは、J1 CHを検出する測定法である。解析結果を図中に記した。プロトンシグナルについて、直接結合する炭素の番号の大文字で番号付けを行った。
図10に1H NMRスペクトルを、図11に13C NMRスペクトルを示した(溶媒:CD3CN:D2O:CD3COOD=90:10:0.1)。図10中で四角の枠で囲ったシグナルは不純物である。上記した試料1のスペクトルと比較した結果、主成分の構造が同じであると推測された。このことは、COSY、NOESY、HSQC、およびHMBCの各スペクトルの解析結果からも支持された。
・d2は−CH2−CH2−XのみとNOE相関があった。
・d3はメチル基のみとNOE相関があった。
このことと、メチル基同士や−CH2−CH2−X同士にNOE相関が明確に観測されていないことから、図1の側鎖の配置が得られた。
上記で得られた産生物に対して、以下の分析を行った。
TFAなどの除去のため、試料を加熱炉内で、280℃×10min加熱した後、600℃で熱分解させ、分解物を、以下に記載するガスクロマトグラフ/質量分析装置(以下、「GC/MS」と称す。)で分離分析した。

測定試料:試料2mgにAcCNを1mL添加した溶液を試料カップに0.5mL注入し、窒素パージでAcCNを除去した。
溶液中成分の分子量を調査するために、以下に記載するエレクトロスプレー−質量分析装置(以下、「ESI−MS」と称す。)による分析を行った。

導入方法:30μLのループを用いて測定溶液を直接導入した。
測定モード:Positive mode
測定溶液:試料2mgにAcCNを1mL加えた溶液をバイアル瓶に少量採取後、導入溶媒で約100ppmに希釈した。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:2.5mg/mL、0.05M KOH、および上記製造例1に従って得られた、分子中にカルボキシル基を4個持つポルフィリン:50μg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。その後、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析したところ、170μg/mLのメタノールが測定された。この際、ギ酸が140μg/mL、リンゴ酸が25μg/mL、酢酸が19μg/mL、コハク酸が5.4μg/mL、ピルビン酸が6.2μg/mL得られた。この場合、乾燥リグニン重量基準で、6.8重量%に当たるメタノール、5.6重量%に当たるギ酸を得ることができた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:2.5mg/mL、0.05M KOH、および合成ポルフィリンとして、分子内にカルボキシル基を2個含むプロトポルフィリンIX(ALDRICH社製)、分子内にカルボキシル基を8個含むウロポルフィリンI(SIGMA社製)、分子内にカルボキシル基を4個含むコプロポルフィリンI(ALDRICH社製)のそれぞれ50μg/mLを含有する各溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。かくして得られた反応液を80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析した。上記操作を複数回繰り返したところ、プロトポルフィリンIX、ウロポルフィリンI、およびコプロポルフィリンIのそれぞれの場合において、乾燥リグニン重量基準で、参考例1の場合とほぼ同等な量のメタノールおよびギ酸が得られた。すなわち、乾燥リグニン重量基準で、約6〜9重量%がメタノールに、約2〜4重量%がギ酸に変換されていた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:1.8mg/mL、0.05M KOH、およびシグマ社製のプロトポルフィリンIV(カタログNo.258385−1G):50μg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。その後、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析したところ、54μg/mLのメタノールが測定された。すなわち、乾燥リグニン重量基準で3.0重量%のメタノールを得ることができた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:7.5mg/mLおよび0.05M KOHを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析したところ、93μg/mLのメタノールが測定された。すなわち、乾燥リグニン重量基準で1.24重量%のメタノールを得ることができた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:7.5mg/mLおよび0.05M KOHを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を24時間照射した。その後、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析したところ、150μg/mLのメタノールが測定された。すなわち、乾燥リグニン重量基準で2重量%のメタノールを得ることができた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:2.5mg/mL、0.05M KOH、および上記製造例1に従って得られたポルフィリン:25μg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。その後、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAX、を用いて分析したところ、160μg/mLのメタノールが測定された。すなわち、乾燥リグニン重量基準で6.4重量%のメタノールを得ることができた。リグニン−アルカリ化合物溶液に、さらに光触媒としてポルフィリンを添加することによって、リグニンからのメタノールの製造量が飛躍的に促進することが分かる。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:2.5mg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を24時間照射した。その後、80℃で60分間加熱し、揮発したガスを島津GC/MS QP−2010、カラムDB−WAXを用いて分析したところ、21μg/mLのメタノールが測定された。すなわち、乾燥リグニン重量基準で0.84重量%のメタノールを得ることができた。
本参考例では、リグニン分解用触媒を用いて、リグニンから水素イオンを遊離させる方法において、リグニンとして、還元糖セルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン:2.5mg/mL、2.5mM KOH、および上記製造例1に従って得られたポルフィリン:50μg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。このサンプルをHPLC(高速液体クロマトグラフィー)にて、ゲル濾過カラムを用いて分析した。対照として紫外線照射前の溶液についても分析した。分析の結果、照射前の場合は、波長310nmの吸光において、7−9minで溶出されるピークとして検出されるが、リグニンにポルフィリンを添加し、紫外光を照射した場合は、310nmの吸光において、7−9minで溶出されるリグニンのピークの面積が78%まで縮小した。すなわち、リグニンにポルフィリンを添加し、紫外光を照射することによって、22%のリグニンが分解されたことが分かる。また、リグニン反応液のpHが10.7から6.2に降下した。かくして、リグニンから水素イオンが遊離したことが分かる。
リグニン分解用触媒を用いて、リグニンから水素イオンを遊離させる方法において、リグニンとして、還元糖を含みやや純度が低い製品(シグマ社製、カタログNo.471038、分子量52,000)を用いた。

リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を使用した。このリグニン:2.5mg/mL、0.05M KOH、および上記参考例1に従って得られたポルフィリン:50μg/mLを含有する溶液1mLを、ポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を12時間照射した。得られたサンプルをHPLCにて、ゲル濾過カラムを用いて分析した。対照として紫外線照射前の溶液についても分析した。分析の結果、照射後のサンプルは、照射前のサンプルに比べ、310nmの吸光において、7minで溶出されるリグニンのピークの面積から、72%のリグニンが分解されていることが分かった。この分解生成物は、ゲル濾過にて、より低分子量のフラクションに溶出された。このリグニンの分解生成物の吸光度は低いため、310nmの吸光度では描出できなかったが、揮発性ではない分解生成物として、溶出サンプルを凍結乾燥したところ、同部に一致して分解生成物の沈殿が得られた。
リグニンとして、還元糖やセルロースなどの不純物を含まない、純度の高い製品(シグマ社製、カタログNo.471003、分子量60,000)を用いた。このリグニン205mg/mLに対して、0.05M KOH、および合成ポルフィリンとして、分子内にカルボキシル基を4個含むコプロポルフィリンI(ALDRICH社製)50μg/mLを含有する溶液0.5mLを、空気のスペースを残すようにして2mL容量のポリプロピレン製エッペンドルフチューブ程度の光透過性を持つ円筒形チューブに入れ、スペクトロニクス社製のENF型紫外光ランプを用いて紫外光を48時間照射した。かくして得られた反応液を上記参考例の場合と同様に処理した。その結果、リグニンの80%以上が分解されていることが確認できた。
Claims (11)
- ポルフィリン環の中心に遷移金属が配位していないポルフィリンであって、光照射下で触媒機能を発現するポルフィリンを含んでなることを特徴とするベンゼン環を形成する炭素原子に酸素原子が結合した芳香族炭化水素分解用触媒。
- ポルフィリン環の中心に遷移金属が配位していないポルフィリンであって、光照射下で触媒機能を発現するポルフィリンを含んでなる、ベンゼン環を形成する炭素原子に酸素原子が結合した芳香族炭化水素分解用触媒を製造する方法であって、遺伝子ypjD(b2611)が変異により発現できなくなった大腸菌を培地で培養し、得られたポルフィリン環構造を有する化合物であって、ポルフィリン環の中心に遷移金属が配位していないポルフィリンを含んでなる芳香族炭化水素分解用触媒、またはこのポルフィリンを含有する培地からなる芳香族炭化水素分解用触媒を製造することを特徴とする触媒の製造方法。
- 請求項2に記載のベンゼン環を形成する炭素原子に酸素原子が結合した芳香族炭化水素分解用触媒を製造する方法において、前記大腸菌が、遺伝子ypjD(b2611)のトランスポゾン挿入変異株であることを特徴とする触媒の製造方法。
- 請求項2または3に記載のベンゼン環を形成する炭素原子に酸素原子が結合した芳香族炭化水素分解用触媒を製造する方法において、前記ポルフィリンが、ポルフィリン環にメチル基およびエチルエステルまたは酢酸基またはプロピオン酸基を含む化合物であることを特徴とする触媒の製造方法。
- 請求項2または3に記載のベンゼン環を形成する炭素原子に酸素原子が結合した芳香族炭化水素分解用触媒を製造する方法において、前記ポルフィリンが、ポルフィリン環に4つのメチル基および4つのエチルエステルまたは酢酸基またはプロピオン酸基を含む化合物であることを特徴とする触媒の製造方法。
