Description
本発明は、RPEを使用するリーシュマニア症などの寄生虫性疾患の診断に関する。
イヌの内臓リーシュマニア症(CVL)は、地中海沿岸地方周辺の国々、中近東およびメキシコにおける重要な新生の動物原性感染症である(20)。この重症疾患は、地中海地方、中東およびアジア諸国においては、リーシュマニア・インファンツム(Leishmania infantum)、中南米およびメキシコにおいてはL.チャガシ(L. chagasi)に起因する(20、21)。それらの遺伝子型の関係のために、異なる大陸において両種が引き起こすCVLは同じものであると考えられる(26)。
この研究において、我々は、RPE、特に、リーシュマニアRPE(LRPE)がリーシュマニア症などの寄生虫性疾患の診断のために有利に使用できることと;この新規の診断方法が公知の診断方法、例えば、例において示すようなSLAに基づく方法などに比べて、より特異的であることとを示す。この新規の方法は、リーシュマニア症の前症候性診断を可能にし、これは、イヌからヒトへのリーシュマニアの感染を防止または減少するために極めて重要である。
第1の態様において、対象における寄生虫性疾患を診断するためのリボソームタンパク質抽出物の使用が提供される。
a.寄生細胞と溶解緩衝液とを混合物すること、
b.得られた混合物を遠心分離し、細胞質抽出物を得ること、
c.得られた細胞質抽出物からリボソームタンパク質抽出物を調製すること。
典型的には、エッペンドルフピペットを使用してこの溶解緩衝液と穏やかに混合される。
第2の態様において、対象においてRPEを使用して寄生虫性疾患を診断するための方法を提供し、当該方法は、対象から得られた試料においてRPEを認識する抗体が存在するか否かを決定することを含む。本発明の好ましい方法は、本発明の好ましい使用に関する場合と同様に、好ましくはインビトロまたはエキソビボにおいて行われる。定義は、より早期にここにおいて示されている。
第3の態様において、対象における寄生虫性疾患を診断するためのアッセイデバイスまたはアッセイが提供され、デバイスまたはアッセイは、RPEを含む。RPEを明確に認識する抗体の存在は、当業者に公知の何れかの標準方法により検出されてよい(例えば、Harlow and Lane, Antibodies: A Laboratory Manual, Cold Spring Harbor Laboratory, 1988を参照されたい。この文献は、引用によりここに組み込まれる)。適切な方法は、アフィニティクロマトグラフィコエレクトロフォレシス(ACE)アッセイおよび(酵素結合性免疫吸着アッセイ)ELISAを含む。好ましくはアッセイはELISAを含む。幾つかのアッセイが、より広範に以下で記載される。
材料および方法
寄生生物. リーシュマニア・チャガシ(MOM/BR/1970/BH46)およびL.・インファンツム(MCAN/ES/1996/BCN/150、MON−1)を20%加熱非働化胎仔ウシ血清(Sigma, St. Louis, MO, USA)、20mMのL−グルタミン、200U/mLのペニシリン、100μg/mLのストレプトマイシンおよび50μg/mLのゲンタマイシンで補足されたpH7.4のシュナイダー培地(Sigma, St. Louis, MO, USA)中、24℃で培養した。
イヌ感染中のL.インファンツムLRPの抗原性. イヌ感染中のLRPの抗原性を分析するために、L.インファンツムに自然感染した10例のイヌからの血清を、この寄生生物からのLRP抽出物を含むニトロセルロースメンブランとインキュベートした。全ての感染動物の血清が、この寄生生物精製タンパク質断片を認識した(図1A、レーン4〜13)。健康なイヌからの血清は、陰性であるか、粗リボソーム調製物中の幾つかのポリペプチドを微かに染色した(図1A、レーン1〜3)。大部分のCVL血清は、多数のタンパク質バンドを認識したが、認識パターンの複雑度および強度は、個々のイヌ血清間で異なっていた。観察された変動性にも拘らず、2つの免疫優性領域がウェスタンブロットにおいて観察された:それぞれ45〜36kDaおよび25〜22kDaポリペプチド。
多くの細胞内細胞質性または核性のリーシュマニアタンパク質様ヒストン、ヒストンプロテイナーゼまたはキネシンが、ヒトまたはイヌ内臓リーシュマニア症(VL)における抗原性であると同定されている(2、13、14、30、32、35)。本研究において、我々は、寄生生物リボソームタンパク質もまたCVL疾患中の抗原性であることを示す。幾つかの個々の変動性が個々のイヌ間で観察されたが、症候性イヌからの全ての血清は、寄生生物リボソーム抗生物質の幾つかに対して反応性を示した。寄生生物のリボソームタンパク質の抗原性がまた、2つの異なる皮膚リーシュマニア症のマウスモデルにおいて示されたことから(22)、我々の結果は、寄生生物リボソームが、リーシュマニア自然感染および実験感染中の脊椎動物ホストの免疫系と相互作用することを示す。

対象における寄生虫性疾患を診断するためのリボソームタンパク質抽出物(RPE)の使用。

(1)に記載の使用であって、当該RPEが、少なくとも2つのリボソームタンパク質、および/またはリボソームタンパク質の少なくとも2つの抗原、および/またはリボソームタンパク質の少なくとも2つのタンパク質断片を含む使用。

(2)に記載の使用であって、当該タンパク質断片が、リボソームタンパク質に対応する少なくとも2、3、5、7、10、15、20、25、30またはそれ以上の連続したアミノ酸を含む断片である使用。

(1)または(3)の何れか1項に記載の使用であって、当該RPEが、配列番号1で示されるEP824699に開示されるリーシュマニア・チャガシからの酸性リボソーム抗原LcPoを起源とするエピトープを含まない、または当該エピトープからなるものではなく、より好ましくは、当該RPEがLcPoのC末端に位置する17アミノ酸、即ち、配列番号1で示されるLcPoのアミノ酸306−322を含まない、または当該アミノ酸からなるものではない使用。

(1)〜(4)の何れか1項に記載の使用であって、当該リボソームタンパク質抽出物が、対象において存在する寄生虫性疾患を引き起こす寄生細胞を使用して以下の工程を行うことにより得られる使用:
a.寄生細胞と溶解緩衝液とを混合すること、
b.得られた混合物を遠心分離して細胞質抽出物を得ること、
c.当該得られた細胞質抽出物からリボソームタンパク質抽出物を調製すること。

(1)〜(5)の何れか1項に記載の使用であって、当該リボソームタンパク質抽出物がリーシュマニア種、好ましくはリーシュマニア・メジャーから得られる使用。

- 対象における寄生虫性疾患を診断するためのリボソームタンパク質抽出物(RPE)の使用であって、
当該RPEが、少なくとも2つのリボソームタンパク質、および/またはリボソームタンパク質の少なくとも2つの抗原、および/またはリボソームタンパク質の少なくとも2つのタンパク質断片を含み、
当該リボソームタンパク質抽出物が、対象において存在するときに寄生虫性疾患を引き起こすリーシュマニア種由来の寄生細胞を使用して以下の工程を行うことにより得られる、使用:

c.当該得られた細胞質抽出物からリボソームタンパク質抽出物を調製すること。 - 請求項1に記載の使用であって、当該タンパク質断片が、リボソームタンパク質に対応する少なくとも2、3、5、7、10、15、20、25、30またはそれ以上の連続したアミノ酸を含む断片である使用。
- 請求項1または2に記載の使用であって、当該リーシュマニア種がリーシュマニア・メジャーである使用。
- 請求項1〜3の何れか1項に記載の使用であって、当該寄生虫性疾患がリーシュマニア症またはマラリアである使用。
- 請求項1〜4の何れか1項に記載の使用であって、当該寄生虫性疾患がリーシュマニアまたはプラスモディウム種により引き起こされる使用。
- 請求項1〜5の何れか1項に記載の使用であって、当該寄生虫性疾患が、当該リボソームタンパク質抽出物を誘導する当該種とは異なる種により引き起こされる使用。
- 請求項1〜6の何れか1項に記載のRPEを使用して、対象における寄生虫性疾患を診断する方法であって、対象から得られた試料において、前記RPEから得られた抗原に対する免疫応答が存在するか否かを決定することを含む方法。
- 請求項1〜6の何れか1項に記載のRPEを含む、対象における寄生虫性疾患を診断するためのアッセイであって、前記アッセイは、対象から得られた試料を前記RPEと接触させ、続いて前記RPEから得られた抗原に対する免疫応答を測定する工程を含む、アッセイ。
- 請求項8に記載のアッセイであって、当該アッセイがELISAであるアッセイ。
