[製造例2]タルトチェリー果実抽出物の製造
タルトチェリー果実抽出物の調製例として、溶媒抽出による調製方法を以下に示す。なおタルトチェリー果実抽出物の効果はその調製方法によらない。
タルトチェリー(Prunus cerasus)の果実10gを破砕し、50vol%1,3−ブチレングリコール溶液100mLを加え、室温にて3日間抽出を行った後、ろ過してタルトチェリー果実抽出物を得た。

実施例1
<美白効果の評価>
マウス由来のB16メラノーマ培養細胞を使用し、製造例1で得られたタルトチェリー果実搾汁液凍結乾燥物のメラニン生成抑制効果を調べた。具体的には、2枚の6穴プレートに10%FBS含有MEM培地を適量とりB16メラノーマ細胞を播種し、37℃、二酸化炭素濃度5%中にて静置した。翌日、製造例1で得られた果実搾汁液凍結乾燥物を、その最終濃度が0μg/mL(対照)、30μg/mL、100μg/mL、300μg/mLとなるように水に溶解して添加し混和した。培養5日目に培地を交換し、再度検体調製液を添加した。翌日、培地を除き、1枚のプレートについて、細胞をリン酸緩衝液にて洗浄した後回収し、B16メラノーマ培養細胞の白色化度を以下の基準にて評価した。対照には検体を添加しないものを用意した。
(判定基準)
++:コウジ酸200μg/mLを添加した試料と同程度の美白効果を示す。
+ :コウジ酸100μg/mLを添加した試料と同程度の美白効果を示す。
± :コウジ酸50μg/mLを添加した試料と同程度の美白効果を示す。
− :対照と同じ黒色を示す。
また残りの1枚のプレートについて、細胞をホルマリン固定後、1%クリスタルバイオレット溶液に添加し染色した。各検体濃度に対する細胞生育率(%)をモノセレーター(オリンパス社製)で測定した。
結果を下記表1に示す。
