Description
The present invention relates to a microscope.
Conventionally, there has been known a microscope in which fluorescence emitted from a sample is divided into spectral components according to wavelength and detected by a multichannel photodetector (see, for example, Patent Document 1).
In this microscope, the larger the number of detectors, the higher the wavelength resolution. For example, when a 32-channel photodetector is used, it is responsible for 10 nm per channel in order to cover the visible region.
On the other hand, since the emission band of the dye extends over a wavelength region of several hundreds of nanometers, the channels are summed according to the fluorescence band of the dye used, and the summed image is displayed on the GUI (Graphical User Interface). Is done. According to such a microscope, the wavelength range to be observed can be switched at a higher speed than in the case of performing spectral detection using a dichroic filter or the like.
By the way, even if the output of a certain channel is saturated among the summed channels, the summed result may not be saturated. In this case, since the microscope disclosed in Patent Document 1 displays only the summed result, the user cannot know that the output of the channel is saturated. Therefore, the sensitivity of the photodetector is increased too much, and as a result, there is a problem that an image of inaccurate luminance data is displayed.
The present invention has been made in view of the above circumstances, and a microscope capable of observing fluorescence emitted from a specimen with an accurate intensity in a microscope that displays luminance data detected by a multi-channel photodetector. The purpose is to provide.
In order to achieve the above object, the present invention employs the following means.
The present invention provides an objective lens that collects fluorescence generated in a specimen, a spectroscopic element that separates the fluorescence collected by the objective lens into spectral components, and a plurality of spectral components that are detected by the spectroscopic elements. A detector having a plurality of channels, an input unit that receives selection of a fluorescent dye to be observed, a dye selection processing unit that determines a summed channel corresponding to the selected fluorescent dye among the plurality of channels, and the fluorescence intensity calculating unit for calculating the fluorescence intensity by summing the detected spectral components by the sum channel of the plurality of channels of the photodetector, an image based on the fluorescence intensity calculated by the fluorescence intensity calculating unit and generating an image generating unit, and a display unit for displaying an image generated by the image generating unit, it is detected by each channel of the photodetector A determination unit that determines whether or not the luminance value of the spectral component exceeds a predetermined threshold value for each of the channels to be added in the fluorescence intensity calculation unit, and at least one of the channels to be added by the determination unit When it is determined that the luminance value of one channel exceeds a predetermined threshold value, a microscope is provided that includes notification means for notifying the determination result.
According to the present invention, the fluorescence generated in the specimen and collected by the objective lens is split into spectral components by the spectroscopic element. The spectral components thus separated are respectively detected by a photodetector having a plurality of channels, and are added together by a fluorescence intensity calculation unit to calculate the fluorescence intensity. In this case, the determination unit determines whether or not the luminance value of the spectral component detected by each channel of the photodetector has exceeded a predetermined threshold, and the luminance value of at least one channel has exceeded the predetermined threshold. When the determination is made, the determination result is notified by the notification means.
Thereby, the user can know that the luminance value of at least one channel is saturated. Therefore, by adjusting the sensitivity of the photodetector, it is possible to accurately detect the spectral components in all the channels without saturating them, and it is possible to perform observation with an accurate intensity of the fluorescence emitted from the specimen.

In this way, the sensitivity of the photodetector can be adjusted by the sensitivity adjustment means so that the spectral components in all channels do not saturate, and observation with an accurate intensity of the fluorescence emitted from the sample is performed. Can do.

By doing in this way, the image produced | generated by the image generation part based on the fluorescence intensity calculated by the fluorescence intensity calculation part is displayed on a display part. At this time, on the screen displayed on the display unit, the notification unit displays a region where the luminance value exceeds a predetermined threshold so as to be visible. As a result, it is possible to know in which area the luminance value exceeds a predetermined threshold on the screen, and it is possible to know whether or not the luminance value is saturated for the area of interest to the user.
In the above invention, the notification means includes an image generation unit that generates an image based on the fluorescence intensity calculated by the fluorescence intensity calculation unit, and a display unit that displays the image generated by the image generation unit. FIG. 1 is a configuration diagram of a microscope 1 according to the present embodiment. As shown in FIG. 1, the microscope 1 according to the present embodiment is a laser scanning microscope that irradiates a specimen A with laser light and detects fluorescence generated in the specimen A. ダイクロイックミラー111は、光源200からのレーザ光を反射する一方、標本Aにおいて発生して対物レンズ114により集光された蛍光を透過するようになっている。

These electric signals are respectively amplified by the
This saturation determination will be described below using the flowchart shown in FIG.
In FIG. 3, in step S <b> 1, the user operates the
The above settings are sent to the
In step S2, the
In step S4, it is determined whether there is any channel that exceeds the threshold among the channels to be added. As a result of the determination, when none of the channels to be added is saturated, the processing of Step S5 and Step S6 described later is executed, and there is even one channel exceeding the threshold value. In step S7 and step S8, which will be described later, are executed.
If none of the channels is saturated, in step S5, summing is performed on the channels to be summed. In step S <b> 6, a value summed as one pixel data of the two-dimensional image is output to the PC
On the other hand, if there is even one channel that exceeds the threshold, data for indicating saturation is generated in step S7. In step S8, the data is output to the PC
The process from step S3 to S6 or step S3 to S8 is repeated for the number of pixels to obtain a two-dimensional image (step S9). As a result, the PC
As described above, according to the microscope 1 according to the present embodiment, the fluorescence generated in the specimen A is dispersed into spectral components by the
Thereby, the user can know that the luminance value of at least one channel is saturated. Therefore, by adjusting the sensitivity of the
Further, according to the microscope 1 according to the present embodiment, the scan image generated by the PC
In other words, if saturation occurs in a region that is not focused, it is only necessary to perform observation with matching sensitivity to a desired location and then cut out only the focused region later. For example, in FIG. 4, although there is a saturated portion in the region ROI2, if attention is paid to the region ROI1, the observation may be continued with the sensitivity adjusted to the region ROI1 as it is.
As a modification of the microscope 1 according to the present embodiment, as shown in FIG. 5, the luminance value of the spectrum component detected by the
By doing in this way, even when the saturated portion is minute in the scan image, the saturated portion can be made conspicuous, and the visibility can be improved.
As mentioned above, although embodiment of this invention was explained in full detail with reference to drawings, the specific structure is not restricted to this embodiment, The design change etc. of the range which does not deviate from the summary of this invention are included.
For example, in the present embodiment, it has been described that the user is notified by displaying on the
In addition, as a method of displaying the saturated portion on the
Moreover, the processing method demonstrated in FIG. 3 is an example, and you may process by a different structure. For example, in step S7, the summation process may be performed in the same manner as in step S5. In that case, in the next step S8, data for indicating saturation is generated, and processing for replacing the summed result is performed.
