Description
The present invention relates to a measurement substrate used in a biochemical measurement method called an immunoassay, and a biochemical bond formation and biochemical bond amount measurement method using the same.
The immunoassay is a biochemical measurement method such as an immunological measurement method that detects and quantifies a trace amount substance using an immune reaction, a measurement method that uses an antigen-antibody reaction, and the like.
In recent years, as one of the biochemical measurement methods, an assay using a light emission phenomenon caused by singlet oxygen generated by exciting fine particles containing a fluorescent dye with light oscillated from a laser diode has been put into practical use. Compared with the enzyme immunoassay (ELISA), which is still commonly used today, this assay can detect these antigens and antibodies only by mixing the antigen and the two antibodies sandwiching it in one step. Therefore, there is an advantage that the measurement time and labor can be greatly reduced. Furthermore, since this assay does not emit light unless the primary antibody-antigen-secondary antibody is in close proximity to each other, the background can be kept low compared to enzyme immunoassays and conventional fluorescent immunoassays, thus increasing the sensitivity. It is also an advantageous technique. In addition, in this assay, singlet oxygen that excites the fluorescent dye reacts with molecules in the living body and superoxide to reduce the concentration, so the fine particles with the fluorescent dye protected by silica have the effect of impurities during measurement. It has also been reported that it can be suppressed (see, for example, Non-Patent Document 1).
Chemical amplification using reactive oxygen species (singlet oxygen) having the above-mentioned advantages can be used for clinical tests (POCT, Point of Care Testing) and on-site biochemical measurements performed in the field of medical care and nursing care. Expected to be suitable. However, conventionally, since a laser diode is used, it is difficult to achieve downsizing and cost reduction of the measuring apparatus, and there is a limit to the installation location of the measuring apparatus. Therefore, instead of laser diode, it is possible to generate singlet oxygen by using a light emitting diode that can be incorporated into a small and inexpensive measuring device. The amount of was not sufficient for biochemical analysis.
The present invention has been made in view of the above circumstances, and is a chemical amplification type using singlet oxygen as a medium by applying a surface plasmon resonance and using a simple measuring device mainly composed of a light emitting diode and a CCD camera.

Then, an evanescent wave is generated on the surface of the gold
