Description
The present invention relates to an optical measurement apparatus and an optical measurement method for identifying a sample such as a microparticle. More specifically, the present invention relates to a technique for detecting fluorescence or scattered light emitted from a sample when irradiated with light of a specific wavelength and identifying the type thereof.
In general, in order to identify biologically relevant microparticles such as cells, microorganisms, and liposomes, an optical measurement method using flow cytometry (flow cytometer) is used (for example, see Non-Patent Document 1). Flow cytometry irradiates laser light of a specific wavelength to minute particles that flow in a line in a flow path, and detects fluorescence or scattered light emitted from each minute particle, thereby detecting a plurality of minute particles. This is a method for identifying one by one.
このとき、レーザ光により励起されて各微小粒子から発せられた蛍光及び/又は散乱光を、PD(Photo diode;フォトダイオード)、CCD(Charge Coupled Device;電荷結合素子)又はPMT(Photo-Multiplier Tube;光電子増倍管)などの光検出器を用いて検出する。 Specifically, in the flow cell, a laminar flow is formed by a sample liquid containing microparticles to be measured and a sheath liquid flowing around the sample liquid, and a plurality of microparticles contained in the sample liquid are arranged in one row. Line up. When the flow cell is irradiated with laser light in this state, fine particles pass one by one so as to cross the laser beam. At this time, fluorescence and / or scattered light emitted from each microparticle when excited by laser light is converted into PD (Photo diode), CCD (Charge Coupled Device) or PMT (Photo-Multiplier Tube). Detection using a photodetector such as a photomultiplier tube).
Then, the light detected by the photodetector is converted into an electrical signal, digitized, and subjected to statistical analysis, and the type of the microparticle is determined from information such as the size and structure of the microparticle and the constituent protein. Conventionally, a flow cytometer that analyzes the internal structure of living body-related microparticles such as cells by detecting nonlinear Raman scattered light instead of fluorescence or scattered light has also been proposed (see Patent Document 1).
On the other hand, in the above-described flow cytometry, in order to analyze a sample quantitatively and stably, it is desirable to always keep the amount of excitation light irradiated to the sample constant. However, generally, the beam spot of the excitation light is as small as several tens of μm, and even within the beam spot, the power density varies in the three-dimensional direction (the optical axis depth direction and the direction perpendicular to the optical axis). .
For this reason, it is necessary to accurately put the sample to be measured at a specific position of the beam spot of the excitation light, and conventionally, a method for aligning the irradiation position of the excitation light and the flow position of the sample has been proposed. (See
Further, in the fluidized particle analyzer described in Patent Document 3, detection light (scattered light) extracted from forward scattered light, side scattered light, or back scattered light through a light splitter is divided into four-division photodiodes and areas. It is detected by a CCD or the like. Then, a position shift between the center of the excitation light and the center of the sheath flow is detected from the detection position, and the position of the flow cell is adjusted so that the position shift falls within a predetermined range.
Furthermore, in the flat flow cell measuring apparatus described in Patent Document 4, first, in the state where the sample is not flowing, the scattered light is measured by irradiating excitation light while moving in the width direction of the flow path, and the flow path side wall. The position of the flow path is obtained by measuring the scattered light from. Then, based on the position information, the stage is moved so that the beam spot of the excitation light comes to the center of the flow path, thereby aligning the flat flow cell.
However, the conventional techniques described above have the following problems. When analyzing a sample to be measured quantitatively and stably in an analysis method using light such as flow cytometry, the allowable range of the beam intensity even within the beam spot is as small as several μm. Become. For this reason, it is necessary to accurately flow the sample to be measured to a specific position of the beam spot, but it is difficult to always keep the flow position of the sample constant.
In particular, when measurement is performed using an analysis chip on which a microchannel is formed, the flow position of the sample liquid is likely to fluctuate due to variations in the shape of the channel and changes in the liquid feeding speed. On the other hand, in order to eliminate the variation in the shape of the analysis chip, it is conceivable to increase the processing accuracy at the time of manufacture. In this case, however, an increase in cost and a decrease in yield become problems.
In addition, as in the flow cytometer described in
In addition, the scattering intensity of the detection signal depends on the particle diameter of the measurement object, and the maximum detection value differs depending on the microparticles to be measured. Therefore, the method described in
On the other hand, the method of positioning the flow cell based on the detection position of the scattered light as in the device described in Patent Document 3 can detect the particle detection position in the direction orthogonal to the optical axis. There is a problem that cannot be detected. That is, in this method, it is possible to obtain a detection signal having a polarity only in the direction orthogonal to the optical axis, and only the signal intensity of the scattered light emitted from the microparticles is detected in the optical axis direction. There is no polarity in the adjustment signal, and it is necessary to determine whether or not the adjustment signal has the maximum value while moving the actuator. For this reason, it is difficult to instantaneously adjust to the optimum value, and the servo cannot be applied in real time.
Further, the scattered light generated from the fine particles is very weak, and the signal depends on the size of the particle to be measured. For this reason, as the particle to be measured becomes smaller, it becomes more difficult to obtain a sufficient signal intensity, and some measures for obtaining a sufficient detection accuracy are required. As described above, the scattering intensity of the detection signal depends on the particle diameter of the measurement object, and the maximum detection value varies depending on the particle to be measured. Therefore, even in the method described in Patent Document 3, the detection signal has a uniform size. If the particles are not used for adjustment, there is a problem that adjustment cannot be performed with high accuracy.
Further, as in the apparatus described in Patent Document 4, the method of positioning the flow cell based on the scattered light from the side wall of the flow path can find the center of the flow path, but the state of the flow cell and the sheath liquid There is no guarantee that the microparticles to be measured pass through the center due to changes in the flow rate of the sample liquid and the like. For this reason, there is a problem that it is difficult to guarantee sufficient accuracy in an apparatus intended for high-precision analysis.
Therefore, the present invention can obtain a good position signal adjustment signal even if the measurement object is a fine particle, accurately detect the position of the sample (microparticle) to be measured, and emit fluorescence or scattering emitted from the sample. The main object is to provide an optical measuring apparatus and an optical measuring method capable of efficiently measuring light and the like.
An optical measurement apparatus according to the present invention includes a light irradiation unit that irradiates light to a sample, and a scattered light detection unit that detects scattered light emitted from the sample irradiated with light, and the scattered light detection unit Includes at least a polarization beam splitter that splits scattered light emitted from the sample into an S-polarized component and a P-polarized component, a first detector that receives the P-polarized component and detects the intensity of the scattered light, and A second detector that receives the S-polarized light component and detects the sample position from the change in the light-receiving position.
In the present invention, since only the S-polarized light component generated by the scattering phenomenon is extracted and the sample position is detected from the change, the position can be accurately detected even when the sample is small and the scattered light is weak. Is possible. If the sample position can be detected with high accuracy in this way, the sample position can be adjusted based on the detected position, so that fluorescence, scattered light, and the like emitted from the sample can be efficiently measured.
This measuring apparatus may have an analysis chip provided with a microchannel, and in that case, the light irradiation unit irradiates the sample flowing through the microchannel with light.
Further, a cylindrical lens can be disposed between the polarization beam splitter and the second detector, and the sample position in the optical axis direction can be detected by the astigmatism method.
Furthermore, a light shielding mask for removing light transmitted without being irradiated on the sample may be disposed between the sample and the polarizing beam splitter.
Furthermore, a fluorescence detection unit that detects fluorescence emitted from the sample irradiated with light can be provided. In that case, fluorescence and scattered light are placed between the sample, the fluorescence detection unit, and the scattered light detection unit. It is desirable to provide a separating mirror.
Furthermore, a sample position adjusting unit that adjusts the sample position based on the detection result of the second detector may be provided.
The optical measurement method according to the present invention includes a step of irradiating a sample to be measured with light, a step of dispersing scattered light emitted from the sample into an S-polarized component and a P-polarized component, and measuring a P-polarized component. And detecting the intensity of the scattered light and measuring the S-polarized component and detecting the sample position by the astigmatism method.
In the present invention, since the S-polarized light component generated by scattering at the sample is taken out and the sample position is detected by the astigmatism method, the detection accuracy is high. For this reason, the measurement with high sensitivity can be stably performed by adjusting the sample position based on the detection result.
According to the present invention, since the P-polarized component and the S-polarized component are separated and the sample position is detected from the change of the S-polarized component, the position of the sample to be measured can be detected with high accuracy and weak. Since the scattered light can be separated from the excitation light and stray light, a good position detection signal can be obtained even if the measurement target is a minute particle, so that the fluorescence and scattered light emitted from the sample can be efficiently used. It becomes possible to measure well.

FIG. 2 is a diagram schematically showing the configuration of the
Here, the larger the

[Operation of Optical Measuring Apparatus 1]
Next, the operation of the optical measuring device 1 of the present embodiment will be described. When optically measuring the
At that time, the
On the other hand, the
In the optical measurement apparatus 1 of the present embodiment, an adjustment unit (not shown) that adjusts the position of the
As described above, the optical measurement apparatus 1 according to the present embodiment uses the fact that only the scattered
In addition, the optical measuring device 1 of this embodiment can be used not only for analysis devices, such as a microparticle and a cell, but also as a manufacturing device, a test device, a medical device, etc., for example.

FIG. 3 is a perspective view schematically showing a configuration of an optical measuring apparatus according to the second embodiment of the present invention. In FIG. 3, the same components as those of the optical measuring device according to the first embodiment described above are denoted by the same reference numerals, and detailed description thereof is omitted. As shown in FIG. 3, the optical measurement apparatus 11 of the present embodiment performs measurement using an

The detection unit includes a
[Analysis chip 21]
The
Further, the
FIG. 4A is a view showing the flow position of the sample liquid in the
On the other hand, as shown in FIG. 4 (b), the liquid flowing through the
However, it is difficult to always keep the flow rate of the sample liquid constant, and the flow position of the
[Operation of Optical Measuring Device 11]
Next, a method for individually measuring and sorting a plurality of
The
On the other hand, the
Thus, since the optical measuring device 11 of the present embodiment detects the change in the polarization angle of the
In addition, by stabilizing the flow position of the sample liquid based on the detection signal of the change in the polarization angle of the scattered light, it becomes possible to respond to changes in the flow velocity, so the analysis speed range of the device (flow velocity designation range) Can be enlarged. Furthermore, light (
The configuration, operation, and effects other than those described above in the optical measurement device 11 of the present embodiment are the same as those of the first embodiment described above.

<3. Third Embodiment>
[Overall configuration of optical measuring device]
FIG. 5 is a diagram schematically showing a configuration of an optical measuring apparatus according to the third embodiment of the present invention. In FIG. 5, the same components as those of the optical measurement devices of the first and second embodiments described above are denoted by the same reference numerals, and detailed description thereof is omitted. As shown in FIG. 5, in the
[Cylindrical lens 50]
The
[Operation of Optical Measuring Device 12]
Next, a method for detecting the position of the
The elliptical imaging pattern thus obtained is an image obtained by imaging the S-polarized
By using such a detection method using an astigmatism optical system for detection of the light emission point of the
The image formation pattern of the scattered light (S-polarized
For the optical axis direction, the displacement amount FE can be detected as an error signal (Focus Error) by calculating (A + C)-(B + D). However, at this time, in order to determine that the imaging pattern of the
As shown in the excitation light intensity distribution in FIG. 6, when the excitation beam has a flat shape and the range in which the scattered light and the fluorescence signal are output in the flow direction is small, the central portion of the split detector is always provided. It is not necessary to detect for a specific time because it is detected only by the detection. As a servo signal calculation method, the calculation may be performed in real time, or after the values of the respective channels are integrated (AREA), the above-described division may be performed. Furthermore, the calculation can be performed using the maximum value (HIGH) of each channel.
On the other hand, in this embodiment, the flow direction in the microchannel is A → B, and the
In general, the polarization beam splitter has a slight leakage of the P-polarized component as its polarization separation characteristic. When the
As described above, the
Further, in the
