Description

本発明は一般に分子生物学およびメタボロミクスの分野に関する。より具体的には、本発明はリガンド結合を分子内蛍光共鳴エネルギー移動(FRET)を用いて測定および検出するためのバイオセンサーおよび方法に関する。

本発明はそれゆえ、分析対象濃度の変化の検出および測定のための改変された分子内バイオセンサーおよびナノセンサーを提供し、特にトランスポーターバイオセンサーおよび細菌周辺質結合タンパク質(PBP)を用いて構築されたバイオセンサーを提供する。具体的には、本発明は、少なくとも1つの内部的に融合したフルオロフォア部分を含む分子内バイオセンサー、ならびに、硬直性が向上したフルオロフォアをコードするFRET 融合コンストラクトを提供する。

以下の記載は本発明の理解に有用であり得る情報を含む。本明細書により提供される情報はいずれも従来技術であるとも本願請求項の発明と関連するものとも認めるものではなく、明示的または黙示的に引用するいずれの刊行物も従来技術であると認めるものではない。
上記のように、本発明者らは驚くべきことに、リガンド結合タンパク質の内部部分への蛍光ドメインの融合が、PBP センサーの同じ葉の中にある場合でも、その末端に融合した対応物と同様のリガンド親和性および対応物に比較して有意に大きいデルタ比を示す有効なバイオセンサーの設計を可能にするということを見いだした。これはドナーおよびアクセプター分子が、リガンド結合の際にドナーおよびアクセプター発色団の配向および/または距離の変化を最大にするために、タンパク質の別々の葉にある末端に典型的に融合される分子内 FRET センサーの一般的モデルの視点に反するものである。
上記のように、本発明はまた、末端および内部的に融合したバイオセンサーを含む分子内バイオセンサーの感受性を向上させる方法も提供する。例えば、かかる方法は以下の工程を含みうる:(a)リガンド結合タンパク質部分、および該リガンド結合タンパク質部分の2つの末端にそれぞれ融合したドナーおよびアクセプター蛍光タンパク質部分を含む分子内 FRET バイオセンサーを提供する工程、ここでドナー部分とアクセプター部分との間の蛍光共鳴エネルギー移動 (FRET)が、ドナー部分が励起され、該リガンドがリガンド結合タンパク質部分に結合すると、変化する;および(b)フルオロフォアとリガンド結合部分との間の融合ドメインを改変または変更する工程、ここで該改変により、該改変を施さない該バイオセンサーと比較して分子内 FRET バイオセンサーの感受性が向上する。改変は、フルオロフォア結合の強剛性を上昇させるリンカー、フルオロフォアまたはリガンド結合ドメインからの1以上のアミノ酸の欠失、挿入または突然変異であり得る。
本発明による好ましいリガンド結合タンパク質部分はとりわけ、トランスポータータンパク質およびそのリガンド結合配列であり、例えばチャンネル、単輸送体、共輸送体(coporter)および逆輸送体からなる群から選択されるトランスポーターが挙げられる。また好ましいものは、周辺質結合タンパク質(PBP)、例えば、 以下の表1に含まれるあらゆる細菌 PBPである。上記のように、細菌 PBPは2つの球状ドメインまたは葉を含み、FRET センサーの設計に便利な足場である(Fehr et al.、2003)。結合部位はドメインの間の間隙に位置し、結合すると、2つのドメインが基質を飲み込み、ヒンジ-捻れ運動が起こる(Quiocho and Ledvina、1996、Mol. Microbiol. 20: 1725)。 I型PBP、例えば、GGBP (D-ガラクトースD-グルコース 結合タンパク質)においては、末端はリガンド結合の際に動いて離れる2つの葉の近位の末端に位置している (Fehr et al.、2004)。II型 PBP、例えば、 マルトース 結合タンパク質 (MBP)においては、末端はヒンジ領域と比較して葉の遠位の末端に位置しており、リガンド結合の際に互いに近づく。したがって、PBPの型および/または内部的に融合したドナーまたはアクセプター部分の位置に応じて、FRETはリガンド結合の際に上昇または低下し得、その両方の場合が本発明に含まれる。
本発明の単離核酸は、組み合わせた場合にFRETにおけるドナーおよびアクセプター部分として作用することができるいずれの好適なドナーおよびアクセプター蛍光タンパク質部分を組み込んでいてもよい。好ましいドナーおよびアクセプター部分は、GFP (緑色蛍光タンパク質)、CFP (シアン蛍光タンパク質)、BFP (青色蛍光タンパク質)、YFP (黄色蛍光タンパク質)、およびそれらの増強された変異体からなる群から選択され、特に好ましい態様はドナー/アクセプター対CFP/YFP-Venusにより提供され、FP-VenusはpH 耐性および熟成時間が改良されたYFPの変異体である (Nagai、T.、Ibata、K.、Park、E.S.、Kubota、M.、Mikoshiba、K.、and Miyawaki、A. (2002) A variant of yellow fluprescent protein with fast and efficient maturation for cell-biological applications. Nat. Biotechnol. 20、87-90)。あるいは好ましくはより高いpH 安定性およびより大きいスペクトル分離を示すMiCy/mKO 対である(Karasawa S、Araki T、Nagai T、Mizuno H、Miyawaki A. Cyan-emitting and/or ange-emitting fluprescent proteins as a donor/acceptor pair for fluorescence resonance energy transfer. Biochem J. 2004 381:307-12)。ドナーまたはアクセプターのいずれかとして好適なものとしてはまた、 Discosoma 種からのネイティブな DsRed、別の属からのDsRedのオルソログまたはネイティブな DsRedの性質を最適化した変異体(例えば、K83M 変異体またはDsRed2 (Clontechから入手可能))が挙げられる。ドナーおよびアクセプター蛍光部分を選択する際の考慮すべき基準は当該技術分野において知られており、例えばUS 6,197,928に開示されており、その内容全体を引用により本明細書に含める。
シグナルペプチドを有さない成熟タンパク質をコードする切型グルタミン酸-アスパラギン酸結合タンパク質配列 (配列番号4)を、大腸菌K12ゲノム DNAをテンプレートとして用いてPCRにより増幅した。用いたプライマーは、5'- ggtaccggaggcgccgcaggcagcacgctggacaaaatc -3' (配列番号5) および5' accggtaccggcgccgttcagtgccttgtcattcggttc 3' (配列番号6)であった。 PCR 断片をpRSET ベクター (Invitrogen)中の消化したFLIPmal-25μ (Fehr et al. 2002)のKpnI 部位にクローニングし、マルトース結合タンパク質配列をYbeJ 配列に交換した。その結果得られたプラスミドをpRSET-FLIP-E-600n (配列番号9)と命名した。
成熟 YBEJ タンパク質をコードするDNA 断片をN- およびC-末端にてそれぞれECFPおよびVenus 配列に融合させた(図 1)。発光スペクトルおよび基質滴定曲線をモノクロメータ マイクロプレートリーダー Safire (Tecan、Austria)を用いて得た。 励起フィルターは433±12nmであり、CFP および YFP 発光についての発光フィルターはそれぞれ485±12、528nm±12nmであった。すべての分析は20mM リン酸ナトリウムバッファー、pH 7.0中で行った。
FLIP-E ナノセンサーのインビボ特徴決定のために、FLIPE-600n およびFLIPE-10μを哺乳類発現ベクター pDisplay (Invitrogen、USA)にクローニングした。pDisplay ベクター はタンパク質を分泌経路に向けるリーダー配列、およびタンパク質を原形質膜にアンカーする膜貫通ドメインを含み、タンパク質を細胞外表面に提示する。ラット海馬細胞およびPC12 細胞にpDisplay FLIPE-600n(配列番号11)および -10μ(配列番号12)コンストラクトをトランスフェクトした。FRETをトランスフェクションの24 48時間後に冷却 CoolSnap HQ デジタルカメラ (Photometrics)を備えた蛍光顕微鏡 (DM IRE2、Leica)で画像化した。二重(dual)発光強度比を436nmでの励起の後に同時に記録し、OI-5-EM フィルターセット (Optical Insights) およびMetafluor 6.1r1 software (Universal Imaging)を備えたDualViewによりCFPとVenus 発光とを分割した。
YbeJについての結晶構造は現在得られていない。本発明者らは、関連するアミノ酸結合タンパク質(HisおよびGln)の既存の構造に基づいて可能性のある構造をホモロジーモデリングした。本発明者らは次いで、許容的であり得る位置、即ち、挿入によりタンパク質の全体の構造が影響を受けない部位を予測した。次に本発明者らはかかる位置に部位特異的突然変異誘発により制限部位を導入した(以下の表3参照)。次いでeCFPのコード領域をかかる部位に挿入した。本発明者らは次いで蛍光を示す細菌コロニーを探索した。(配列番号28に示すYbeJ 配列に基づいて)eCFP が挿入されたN58V-Q59Nのみが蛍光性であった。本発明者らは次いでC-末端にVenusを結合させた(FLIP-E 分子内融合(intermol)) (図 6参照)。親和性を試験し、本発明者らはより大きいデルタ比変化を見いだし、親和性はおよそ 1μMであり、これはフルオロフォアを末端に担持するYbeJ の600n バージョンよりもわずかに高いだけである(図 7参照)。
内部的に融合した FRET バイオセンサーをその他のタンパク質を用いて構築できることを示すために、本発明者らは、大腸菌グルコース/ガラクトース結合タンパク質 (GGBP)を結合ドメインとして含み、かつ、オワンクラゲ緑色蛍光タンパク質変異体CFPおよびYFPをレポータードメインとして含むナノセンサーを構築した。YFPは結合タンパク質のC-またはN-末端のいずれかに融合させたが、CFPは結合タンパク質の様々な位置に挿入し、内部的に融合したセンサーのセットを作成した。これらセンサーのそれぞれは、発色団の異なる相対的空間的配向によって特徴づけられる。
発色団挿入を許容するGGBPの内部の許容的部位をスキャンするために全部で13の異なる部位を選択した。これら部位はループ上またはコアタンパク質から突出しており結晶構造において高いB-因子を示す二次構造要素の末端に優先的に位置させた。GGBPの両方の葉上の部位を選択した。CFP 挿入を可能にするために、Kunkel's法を用いてGGBP コード配列におけるそれぞれの位置に部位特異的突然変異誘発によりNru I 制限認識配列を導入した。表4はGGBPにおける選択した部位およびNru I 認識配列により導入された突然変異を示す。
CFP コード配列を、分子クローニングによりGGBPにおけるNru I 部位に挿入した。コンストラクトはすべての開発段階で未完成のセンサーの発現を可能とするように設計した。同一の挿入部位を担持する2セットのコンストラクトを操作して作成した。一方のセットはYFP とのN-末端融合を可能にするように設計し、他方のセットはYFP のC-末端融合を可能にするように設計した。
YFPのコード配列を分子クローニングにより工程 2のCFP 挿入を含む発現カセットに挿入した。2セットの CFP 挿入を用いて、同じCFP挿入を担持するがN- またはC-末端のいずれかにYFPが結合している2セットの蛍光ナノセンサーを得た。ライゲーション反応を発現株 BL21(DE3)goldに移した。選抜条件下での培養の後、結果として得られたコロニーを用いてマイクロタイタープレート中の200 μl培養を開始し、両方の発色団を発現するクローンをスクリーニングした。培養を2日間室温で増殖させ、2日間4 ℃で静止状態にして発色団の成熟を促した。次いで培養をCFP 励起波長 (433 nm)にて励起し、発光強度をCFPとYFPの発光ピークを包含する460 nm 〜560 nmで記録した。両方の発色団の存在を示した各ナノセンサー発現カセットのうち2、3のクローンをさらなる分析のために選択した。スモールスケール培養を開始してタンパク質をNi-NTA アフィニティークロマトグラフィーにより回収した。新しいナノセンサーの比変化を分析するために、精製タンパク質のスペクトルを10 mM グルコースの非存在下および存在下にて記録し、YFP/CFP 発光強度比の相違を計算した。表6は測定された比変化を示す。
比変化が0.2より大きいナノセンサー(表6において太字で示す)をさらなる分析のために選択した。タンパク質をラージスケール培養からNi-NTA アフィニティークロマトグラフィーを用いて精製した。その結果得られたタンパク質抽出物を、マイクロプレートに基づくFRETアッセイにおいて様々な濃度のグルコースにより滴定した(titrated)。ナノセンサーの親和性を滴定曲線の非線形回帰により測定した。さらに、スペクトルをグルコース非存在、半飽和グルコース濃度、および飽和グルコース濃度にて記録した。対照として、元のナノセンサーであってGGBPがCFPとYFPとの間にあるFLIPmglBF16Aを含める。比変化 (デルタ比)を規準化するために、比変化をグルコースの非存在下における比で割った (表7) (図8および9参照)。
22の挿入のうち、6つの機能的グルコースセンサーが同定された。4つのセンサーはグルコースの添加により正の比変化を示した。2つのみが元のセンサー FLIPmglBF16Aと同様に負の比変化を示した。4つのセンサーはFLIPmglBF16A と比較して大きな相対比変化を示した。2つのセンサーはFLIPmglBF16A と同様の相対比変化を示した。したがって発色団の結合タンパク質への挿入は、ナノセンサーの設計および改良に有効な戦略であることが判明した。さらに、発色団は、機能的センサーを得るために結合タンパク質の異なる葉の上に位置している必要はない。
フルオロフォアの内部挿入における回転性の平均化の低下が、高い比変化を有するセンサーの作成のための一般的戦略であることを知ったため、本発明者らは分析対象結合ドメインとフルオロフォアとの間の結合の回転の自由を低減することによっても同様の結果が得られるのではないかと仮定した。したがって本発明者らは、結合ドメインのコアタンパク質構造とフルオロフォアを連結している配列を系統的に除去した。即ち、それはリンカー配列の除去および分析対象結合部分とフルオロフォアの末端から両方のアミノ酸を欠失させることにより行った。本発明者らは緊密な結合(close coupling)によってもより高い比変化が導かれることを見いだした。この概念をFLIPgluを用いて例証する。
材料および方法:
FLIPglu 内部的に融合したセンサーについてのリンカー欠失
2つの内部的に融合したグルコースセンサー、FLII12Pglu-600μおよびFLIIP275Pglu-4.6mをそのΔ比と親和性に基づいて選択した。FLII12Pglu-600μについては、リンカーおよびmglBおよびCitrineの末端 のあまり構造化されていないドメイン (共に17アミノ酸の「複合リンカー」を含む)をKunkel 突然変異誘発 (Kunkel et al.)を用いてmglBから出発して系統的に欠失させた。FLII12Pglu-600μからアミノ酸残基の欠失数を増やすよう設計した17のプライマーを用いて、FLII12Pglu-1aaからFLII12Pglu-17aaを作成した。 さらに、16アミノ酸の欠失である、FLII12PgluΔ16も、mglBの残基 305にXhoI 部位を付加し、短くしたCitrine (アミノ酸7-238)を XhoIおよびHindIIIを用いてクローニングすることにより作成した。 FLII12Pglu-16aaおよび FLII12PgluΔ16はしたがってmglB の位置 305 における1つのアミノ酸残基において異なる(FLII12Pglu-16aaではAla であり、FLII12Pglu δ16ではLeuである)。2つのさらなるプライマーを用いて、mglBおよびCitrineをインタクトに維持しmglB とCitrineの間のプラスミド由来 リンカーの4および6 アミノ酸残基、 Gly-Gly-Thr-Gly-Gly-Ala (配列番号32) (GGTGGTACCGGAGGCGCC (配列番号33))を欠失させた(FLII12Pglu δ4 およびFLII12Pglu δ6)。Citrine がN-末端にあるFLIIP275Pglu-4.6mの場合、一つのプライマーを用いて15アミノ酸残基を欠失させた(Citrine のC-末端の不要な部分から9アミノ酸およびCitrine とmglBとを連結するプラスミド由来リンカーから6アミノ酸) (図14)。
コンストラクトをエレクトロポレーションを用いて大腸菌 BL21(DE3)Gold (Stratagene、USA)に移し、以前に記載されているようにして抽出および精製した (Fehr et al.、2002、Proc. Natl. Acad. Sci. USA 99: 9846-9851)。発光 スペクトルおよびリガンド滴定曲線をモノクロメータマイクロプレートリーダー (Safire、Tecan、Austria)を用いて得た。励起フィルターは433/12nmとし; ECFP および EYFP発光についての発光フィルター (CitrineおよびVenusも同様)はそれぞれ485/12および 528/12 nmとした。FLIPE コンストラクトおよび直線的に融合した FLIPglu コンストラクトについてのすべての分析は、20mM リン酸ナトリウムバッファー、pH 7.0中で行った; FLIIXPgluの分析は20 mM MOPS バッファー、pH 7.0中で行った。FLII12Pglu-600μとFLIIP275Pglu-4.6m 欠失をよりよく比較するために、それぞれについてのCitrine 発光値は約 20000と一定に維持し、発光増加は80と一定に維持した。MOPS pH 7.0におけるアッセイに用いたのと同じ量のタンパク質を用いて、センサーを、ハンクスバッファー (pH 7.2)、合成哺乳類細胞質ゾル (pH 7.2)、合成植物細胞質ゾル (pH 7.2)およびMOPS pH 5.0中でも分析した。各センサーのKdは、単一部位結合等温式: S = (r - rapo)/(rsat - rapo) = [L]/(Kd + [L])に適合させることによって決定した。ここでSは飽和; [L]はリガンド濃度; rは比; rapoはリガンド の非存在下における比; およびrsatはリガンド飽和での比である。測定は少なくとも3つの独立のタンパク質抽出物を用いて行った。ECFP 発光は485 と502 nmとの2つのピークによって特徴づけられる; ここで規定する比は、528 nmでの非集光蛍光強度を485 nmでの強度で割ったものとした。
環境条件のセンサーへの影響を調べるために、センサーを以下の様々な条件下で分析した:哺乳類細胞培養液中(ハンクスバッファー: 137mM NaCl、5.4mM KCl、0.3mM Na2HPO4、0.4mM KH2PO4、4.2mM NaHCO3、0.6 MgSO4、10mM 乳酸、1mMピルビン酸、pH 7.4)、合成哺乳類細胞質ゾル中(135mM K(グルコン酸)、4mM KCl、12mM NaHCO3、0.8mM MgCl2、0.2μM CaCl2 pH 7.4)、合成植物細胞質ゾル中(10mM NaCl、150mM K(グルコン酸)、1mM MgCl2、100mg/mL BSA、10mM HEPES pH 7.5 、BTP含有)およびMOPS バッファー pH 5.0中。タンパク質量をMOPS バッファー pH 7.0中での分析に関しては一定に維持した。スペクトルをグルコース無し、10mM グルコースおよび100mM グルコースで3連で測定し、分析は各センサーについて2 つの独立のタンパク質調製物を用いて行った。
FLIPglu リンカー変異(variation)
シグナル対ノイズ比をさらに改善し、環境に安定なセンサーを開発するために、分子内 FRET センサー FLII12Pglu-600μ (Deuschle et al.、2005、Protein Science 14:2304-2314)中のリンカーの系統的欠失分析を行った。グルコースナノセンサーであるFLII12Pglu-600μは、その位置 12にCFPが挿入された大腸菌由来成熟グルコース/ガラクトース-結合タンパク質 mglBと、C-末端に6- アミノ酸リンカーを介して直線的に融合した EYFPからなる (Deuschle 2005)。リンカーおよびmglBおよびEYFP変異体の末端のあまりよく構造化されていないドメイン(「複合リンカー」を一緒に含む)は、結合タンパク質に対するフルオロフォアの、および互いの(自由ではないとしても) 可動性の回転を可能にすると考えられる。この複合リンカーを回転性の平均化を低下させ、アロステリックカップリングを増強させるために系統的に切断した。FPは、フォールディングおよび蛍光に絶対的には必要ではない 末端領域を有する(N-末端ヘリックスおよびC-末端 コイル) (Li et al.、1997、J. Biol. Chem. 272: 28545-28549)。さらに、5アミノ酸を結合に影響を与えることなくmglB 結合タンパク質のC-末端領域から欠失させうる。これらを合わせると17アミノ酸となり、これらを除去しても経験的に結合および蛍光が保存されると予測される(図14)。複合リンカー領域を段階的にFLII12Pglu-600μから欠失させた。
ほとんどのFLII12Pglu-600μ 欠失は、全長センサーと比べてFRETの低下を示した。欠失コンストラクトのΔ比は0.52 (FLII12Pglu-17aa、78% 低下) から2.26 (FLII12Pglu-12aa、5%低下)の間で変動した。20の欠失コンストラクトのうち、14は1を超えるΔ比を有しており、そのなかで5つのコンストラクトは1.3 以上のΔ比を有していた(FLII12Pglu-6aa 1.32、FLII12Pglu-7aa 1.31、FLII12Pglu-10aa 1.3、FLII12Pglu-12aa 2.26、FLII12Pglu-16aa -1.40 )。 FLII12Pglu δ4およびFLII12Pglu δ6はFLII12Pglu-600μと比べてΔ比がわずかに上昇していた(4% 上昇)。興味深いことに、FLII12Pglu-16aa およびFLII12Pglu δ16はリガンド結合の際に比の低下を示したが、FLII12Pglu-600μおよび他の欠失のすべてはリガンド結合の際に比の上昇を示した。それぞれのセンサーの親和性をグルコースで滴定することにより測定した (表8)。グルコースに対する親和性は、2アミノ酸の欠失、FLII12Pglu-2aa以降に低下し、FLII12Pglu-13aaまでは1.5-2.0 mMの範囲の結合定数を有し、13を超えるアミノ酸の欠失ではさらに親和性が低下した(FLII12Pglu-14aa 3.4mM、FLII12Pglu-15aa 2.6mM)。FLII12Pglu-17aa では親和性は劇的に低下し、6.8mMであった。FLII12Pglu δ4およびFLII12Pglu δ6はしかし、FLII12Pglu-600μに匹敵するKd を有する(図15、表8)。
バッファーが比変化に影響を与えうることが以前に指摘されている。さらに、インビボでは比変化は常に様々な因子により弱められ、かかる因子としては、例えば、pH、イオン、糖等の存在が挙げられる。それゆえ、インビボへの適用にもっとも好適なセンサーを同定するために、細胞培地 (ハンクス)を模倣する様々なバッファー、哺乳類細胞質ゾル、植物細胞質ゾルおよび小胞、液胞または細胞壁の内側に類似した低 pHについて試験した(表8)。FLII12Pglu-600μは、MOPS pH 5.0および植物細胞質ゾルにおいて57%〜74%の比変化の低下を示す。ほとんどの欠失コンストラクトは様々なバッファー中で20-70%低下したΔ比を有する。Δ比が1.3以上である5つのコンストラクトのうち、FLII12Pglu-6aaおよびFLII12Pglu-7aa は試験したすべてのバッファーにより非常に影響を受け、20-61%の低下したΔ比を示した。FLII12Pglu-10aaはハンクスバッファーにより影響を受けず、哺乳類細胞質ゾル中では非常にわずかに影響を受け (10% 低下)、植物細胞質ゾル中では28% のΔ比の低下を示し、低 pHでは 66%の低下を示した。FLII12Pglu-12aaはすべてのバッファー中で52-59% の低下を示したが、依然としてΔ比は1.0であった。 FLII12Pglu-16aaはハンクスバッファーおよび哺乳類細胞質ゾル中で約 30%の低下を示し、植物細胞質ゾルおよびMOPS pH 5.0中では影響を受けなかったが、それは異なるイオンに応答して完全に配向が変化した。それはハンクスバッファーおよび哺乳類細胞質ゾル中では比の上昇を示し、植物細胞質ゾルおよびMOPS pH 5.0中では比の低下を示した(FLII12Pglu δ16と同じ)。FLII12Pglu-15aaはしかしすべてのバッファー中でほとんど影響を受けず、ハンクスバッファーおよび哺乳類細胞質ゾル中でΔ比の向上を示した(図16)。
ほとんどのFLII12Pglu-600μ 欠失コンストラクトは元のセンサーよりも低いΔ比を示したが、それらは試験した環境条件に対してより耐性であった。したがってもっともイオンに感受性の残基が露出されなくなるようにセンサーを再編成すると考えられる残基の欠失により、センサーはより環境条件に耐性になる。
本発明者らは、高-シグナル変化ループ-挿入型グルコースセンサー FLII12Pglu-600μにおける結合タンパク質 (BP)と蛍光タンパク質 (FP)との間の境界からの一連の欠失の効果を1残基毎に追跡する大きなデータセットを蓄積した。欠失はナノセンサーファミリーのシグナル変化とグルコース-結合親和性との両方に対し同時の効果を有し、これは粗分子モデリングからの予測と一致する。すべてのセンサー修飾のうち、mglB C-末端とYFP N-末端とを連結する合成リンカーの中央からの1または2アミノ酸の欠失のみが、より高いシグナル 変化またはより高いリガンド結合親和性を示すセンサーをもたらした (この場合、その両方)。その他のすべての欠失ではグルコースに対する親和性およびグルコース-依存的シグナル変化が低下した。しかしセンサーのなかには、異なるバッファー条件においては元の センサーよりも高いシグナル変化をもたらすものもあり、それはインビボでのセンサーとして有用であろう。おそらくもっとも重要なことに、リンカー-欠失センサーのファミリーは、さらなるリンカー変異体の合理化および設計のための有用なデータセットを提供し、これは非機能的なセンサーから高応答性センサーを作るのに役立つであろう。
生細胞におけるグルコース検出のための改良されたセンサーの検証のため、および、センサーがその他の細胞タイプにも利用できるか否かを検証するために、3つの分子内センサー(FLII12Pglu-600μ; FLII12Pglu δ4aa-593μ; FLII275Pglu-4600μ;図17)をpcDNA3.1 (-)にクローニングした(図18)。図 19は、改良グルコースセンサーで形質転換されたNIH3T3 細胞において観察されたFRET変化を示す。
Claims (39)
- 以下をコードするポリヌクレオチド配列を含む単離核酸:
目的のリガンドに特異的に結合する周辺質結合タンパク質 (PBP)部分;および
該PBPに融合したドナーフルオロフォアタンパク質部分または該PBPに融合したアクセプターフルオロフォアタンパク質部分の少なくとも1つ;
ここで、該ドナーフルオロフォアタンパク質部分のコード領域または該アクセプターフルオロフォアタンパク質部分のコード領域が、該PBPのコード領域の内部部位にて挿入されている。 - 該ドナーフルオロフォアタンパク質部分のコード領域が該PBPのコード領域の内部部位にて挿入されている、請求項1の単離核酸。
- 該アクセプターフルオロフォアタンパク質部分のコード領域が該PBPのコード領域の内部部位にて挿入されている、請求項1の単離核酸。
- 該ドナー蛍光タンパク質部分のコード領域および該アクセプター蛍光タンパク質部分のコード領域が該PBPの同じ葉に融合している、請求項1の単離核酸。
- 該PBPがアミノ酸に特異的に結合する、請求項1の単離核酸。
- 該アミノ酸が、グルタミン酸、アスパラギン酸、γ-アミノ酪酸 (GABA)、アミノ酢酸 (グリシン)およびタウリンからなる群から選択される、請求項5の単離核酸。
- 該PBPが糖に特異的に結合する、請求項1の単離核酸。
- 該糖が、グルコース、ガラクトース、マルトース、スクロース、トレハロース、アラビノース、フルクトース、キシロース、セロビオースおよびリボースからなる群から選択される、請求項7の単離核酸。
- 該アミノ酸がグルタミン酸である、請求項6の単離核酸。
- 該グルタミン酸に結合するPBPが配列番号28の配列を含む、請求項9の単離核酸。
