Description
本出願は、2007年3月5日出願の米国仮出願60/905227及び2007年11月30日出願の米国仮出願60/991654の優先権を主張し、これらの出願の内容は全て引用をもって本書に繰り込むものとする。
本発明は、膜組み込みペプチド、特に生体内でそう(膜組み込み)でなければ非選択的に細胞に有毒な膜組み込みペプチドの輸送に関する。特に本発明は、疎水性コアが脂質/界面活性剤層でコーティングされたナノ粒子の自己配列エマルジョンを用いた上記ペプチドの輸送に関する。
また、本発明に係るナノ粒子を含む組成物において、該ナノ粒子は脂質/界面活性剤層によってコーティングされた液状の疎水性コアを含み、該脂質/界面活性剤層は1以上の膜組み込みペプチドを含むことが好ましい。(形態1)
また、前記疎水性コアは1以上の含ハロゲン炭素化合物を含むことが好ましい。(形態2)

リポソームの調製
パーフルオロカーボンナノ粒子ビヒクルへのペプチドキャリヤとしての従来型リポソームの挙動を比較するため、リポソーム(98モル%卵レシチン、2モル%DPPE)をSaito, M.ら(Nat. Cell. Biol.(2000)2:553-555)による方法で合成した。Avanti Polar Lipid, Inc.からクロロホルム溶解脂質を得て、高真空中で3時間乾燥させることにより溶媒の痕跡を飛ばし、脂質膜を得た。脂質をジエチルエーテルに溶解し、等容量の緩衝液(PBS)に懸濁させ、全脂質濃度を10mMとした。混合液は超音波槽(Laboratory Supplies Co, Hicksville, NY)にフラスコを沈めて20秒間超音波処理し、安定なエマルジョンを得た。減圧下で有機溶媒を除去した後、得られたリポソームを200nmのポリカーボネートのメンブレンフィルタで押出し、4℃で保存した。
パーフルオロカーボンナノ粒子の調製
パーフルオロカーボンナノ粒子をWinter, P.M.らの方法で合成した(Arterioscler. Thromb. Vasc. Biol.(2006)26:2103-2109)。簡単にいうと、卵レシチン(98モル%)の脂質界面活性体混合体とジパルミトイル−ホスファチジルエタノールアミン(DPPE)2モル%(Avanti Polar Lipids, Piscataway, NJ)とをクロロホルムに溶解し、減圧蒸留し、50℃の真空オーブンで乾燥させ、超音波処理して水に分散させた。懸濁液はパーフルオロオクチルブロマイド(PFOB)又はパーフルオロ−15−クラウンエーテル(CE)(Gateway Specialty Chemicals, St. Peters, MO)と結合させ、脱イオン水で蒸留し、S110マイクロフルイディクス懸濁装置(Microfluidics, Newton, MA)により20000lbf/in2で4分間連続処理した。αVβ3−インテグリンをターゲットとしたナノ粒子は、等モルのレシチンに置換するポリエチレングリコール(PEG)2000−ホスファチジルエタノールアミン(Avanti Polar Lipids, Inc.)と結合した0.1モル%ペプチド類似体のビトロネクチンアンタゴニストを導入することにより調製した。
ナノ粒子へのメリチンの導入
メリチンを担持したナノ粒子は、既知の量のメリチンをパーフルオロカーボンナノ粒子に混合することにより形成した。固体状ペプチド合成法により製造される、純粋なメリチンペプチド物質は、ワシントン大学医学校細胞生物学及び生理学部のRobert Mecham博士から得た。メリチンを100mMKCl(pH7,10mMHEPES)に0.1mMで溶解し、50μlのナノ粒子懸濁液に混合しつつ2−20mL加えた。室温で10分間保温処理(インキュベート)した後、ナノ粒子を2回の遠心分離(100g、10分間)で洗浄し、未結合のメリチンを除去した。上澄み液中のメリチンを、トリプトファン蛍光法(下記)で定量した。加えたメリチンの量により、メリチンを担持したナノ粒子が、脂質/メリチンモル比で1000から40の範囲で得られた。
メリチンナノエマルジョンの性質
A.粒度分布とゼータ電位
メリチンを担持したナノ粒子の粒度分布は、Malvern Zetasizer 3000HS(Malvern Instruments, Malvern, UK)を用いて光子相関法(PCS)により測定された。ナノ粒子のゼータ電位(ζ)は、Brookhaven Instruments PALS Zeta Potential Analizer(Brookhaven Instruments)を用いて測定された。データは、25℃の溶液平衡ののち、相解析光分散法(PALS)により得られた。測定されたナノ粒子の電気泳動移動度(μ)からζを計算するにあたってスモルコフスキー近似を用いた。

ここでεとηはそれぞれ溶媒の誘電率と絶対粘度である。
二重層リポソームと単層パーフルオロカーボンナノ粒子に与えるメリチンの効果を、透過型電子顕微鏡により確認した。メリチンを担持したパーフルオロカーボンナノ粒子は実施例2のように調製した。粒子は、0.1Mカコジル酸ナトリウム緩衝液に2.5%グルタルアルデヒドを含む液に30分間氷上において固定した。遠心分離後のペレットをすすぎ、粒子を順に1.25%四酸化オスミウム、2%タンニン酸、及び酢酸ウラニルで染色し、ペレットを脱水し、Polybed812(Polysciences, Inc, Warrington, PA)に包埋した。次にペレットをReichert-Jung Ultracutで薄く切片化し、さらに酢酸ウラニルとクエン酸鉛で染色した。
パーフルオロカーボンナノ粒子の脂質単層へのメリチン挿入動力学を、表面プラズモン共鳴(SPR)を用いて調査した。SPRは表面の屈折率の変化を測定するものである。ビアコア−Xバイオセンサとカルボキシメチル化デキストランチップL1をビアコア(Biacore, Inc)(Piscataway, NJ)より得た。溶液はすべて脱ガスし、推奨どおり0.22μmメンブレンでろ過した。35μlのPFOBナノ粒子(3μl/分)で注入することにより、L1チップの上に均一な脂質単層を生成した。2分間流量を1500μl/分に増加させ、次いで水酸化ナトリウム(50μl、10mM)を注入することにより、緩く沈着しているナノ粒子を除去し、安定なベースラインを得た。陰性対照試料であるウシの血清アルブミン(25μl、0.1mg/μlPBS)を注入することにより、完全被覆が確認された。メリチンをいろいろな濃度で流量30μl/分(30μlPBS;15nMから1000nM)で注入し、60分間反応を記録した。各試験終了後、チップは3−[(3−コールアミドプロピル)ジメチルアンモニオ]−1−プロパンスルフォネート(CHAPS)(50μl、100μl/分)を2回続けて注入して再生した。選択したメリチン濃度の一連のセンサーグラムが得られた。データは、BIAevaluation(登録商標)ソフトウェア(Biacore, Inc, Piscataway)を用いて、種々の濃度のペプチドのセンサーグラムを同時にフィッティングすることにより、包括的に解析された。脂質単層に対するメリチン相互作用の明らかな結合親和性(Kd)が、注入したメリチンの濃度(μM)関数としての平衡メリチン結合応答(応答単位, RU)のプロッティングと、ラングミュア等温式モデルへのフィッティングにより、推定された。
ナノ粒子の蛍光スペクトル分析が脂質/メリチン比80において行われた。脂質二重層に挿入されたペプチドの立体配列が、内因性トリプトファン蛍光測定により調査された。メリチンは19位置にトリプトファン残基を持つ。臭素を含む分子はトリプトファン放出の波長に強い吸収を示すため、トリプトファンの蛍光を、トリプトファンと臭素との接触が必要な重元素衝突クエンチ、又はフォルスター(Forster)エネルギー転移のいずれかによってクエンチすることが知られている。パーフルオロカーボンナノ粒子の脂質単層へのメリチンの挿入は、ミニサンプル攪拌器を備える蛍光スペクトル分析計(Varian, Inc, Palo Alto, CA)中で280nmで励起した後、トリプトファン蛍光放射(350nm)の動力学によって調査された。強度は、メリチンを含まないナノ粒子を含むキューベットからの信号を差し引くことにより、光散乱の補正を行った。パーフルオロ−15−クラウンエーテルコアにより合成されたナノ粒子を陰性対照(臭素を含まない)として用いた。メリチン濃度は10μMであった。
円偏光二色性(CD)分光分析は、ペプチド及びたんぱく質の二次構造に関する情報を与える。この方法は、一般に脂質膜に取り込まれたペプチドに研究に用いられる。アミド発色団はペプチド主構造の構造的変化に対して敏感である。ジャスコ(Jasco) J-810分光偏光計(Jasco, Inc., Eastern, MD)を用いて、遊離メリチンとナノ粒子脂質単層内に入り込んだメリチンのCD測定を行った。1cm光路長の石英キューベットを200−260nmの範囲の遠UVで50nm/分のスキャン速度でスキャンし、すべてのスペクトルはアルゴン下で採取した。ナノビーズと、脂質/メリチン比40でメリチンを含むナノビーズとを遠心分離で洗浄し、5mMのリン酸塩緩衝液でpH7に緩衝した150mMの塩化ナトリウム液に分散させ、20℃、レスポンス4秒、バンド幅1nmで測定した。すべてのデータは15回のスキャンを平均したものであり、またブランクを差し引くことにより、バックグラウンド信号を補正している。データはモル楕円率[θ]で表している。

ここでθobsは測定された楕円率(mdeg)、Lは光路長(cm)、Cはメリチン濃度(モル/l)である。
メリチンを担持したナノ粒子と細胞との相互作用
メリチンを担持したナノ粒子の赤血球とC−32メラノーマ細胞に与える影響が、溶血分析と細胞増殖(MTT)分析によって調査された。メリチンは、赤血球を溶解し、細胞死をもたらすことが知られている。
同意を得た後、ヒトの臍帯血が健康なドナーから採取された。赤血球が200gで10分間の遠心分離により分離され、通常の生理食塩水に再分散させた。さまざまな濃度のメリチン又はメリチンを担持したナノ粒子を決まった数の赤血球(5×107細胞)に加え、37℃で3時間保温処理(インキュベート)した。遠心分離後、Microplate Reader(Model 550, BioRad)で上澄み液の540nmの吸収を測定することにより、ヘモグロビンの放出量を測定した。同じ条件下で水中で保温処理した赤血球の上澄み液の吸収を100%とした。
αVβ3をターゲットとしたメリチンナノ粒子の、C−32がん細胞増殖に与える効果を、3−[4,5−ジメチルチアゾール−2−イル]2,5−ジフェニルテトラゾリウムブロマイド(MTT)分析によって調査した。この分析法は、テトラゾリウム塩であるMTTを、570nmに吸収を持つホルマザン結晶に変換するミトコンドリアエンザイム活性を測定するものである。
メリチンによる細胞死がアポトーシスによるのかネクローシスによるのかを決定するため、アネキシン−V FITCのホスファチジルセリンへの結合がフローサイトメトリにより測定された。アポトーシスの初期の事象のひとつとして、ホスファチジルセリンが細胞膜の外面部(outer cell membrane)に移動し、アネキシン結合部位が露出する。膜の健全性はアポトーシスでは最後の段階まで維持されるが、ネクローシスでは早い段階で弱められる。アネキシン−V FITCの結合と7−AADの染色のフローサイトメトリによる解析により、メリチンを含むパーフルオロカーボンナノ粒子による細胞死の支配的な機構は、アポトーシスであることを示している。

細胞内に輸送されるメリチンを追跡し、位置を同定するため、蛍光標識メリチンを作成した。FITCをメリチンのN末端に結合させるため、Fluoro Tag(登録商標)FITC結合キット(Sigma, St. Louis, MO)を用いた。簡単に言うと、フルオレセイン−イソチオシアネートとメリチンを、0.1Mの炭酸塩−重炭酸塩緩衝液(pH9.0)中にモル比で10:1で混合した。定常的に攪拌しつつ、室温で一晩保温処理し、混合反応液をG−25セファデックス(sephadex)カラムに通し、フラクションを集めて保存した。結合していないFITCを除去するため、カラムをPBSで洗浄して再生した。A280>0.4であるフラクションを集め、結合率を以下の式で評価した。

をそれぞれ示す。
細胞膜に対するメリチン活性の調整に関するコレステロールの役割を、細胞増殖分析及びフローサイトメトリ(上述)の前にコレステロール細胞を欠乏させることにより評価した。C−32メラノーマ細胞を0.25mM又は0.5mMメチルベータシクロデキストリン(Sigma, St. Louis, MO)で37℃で15分間処理し、コレステロールを減少させた。メチルベータシクロデキストリンにより減少したコレステロール量の定量は、標準的操作によりメラノーマ細胞から全脂質を抽出することで行った。つまり、6孔組織培養プレートで培養した106個のC−32メラノーマ細胞をゴム製ポリスマンで掻き取り、遠心分離後のペレットを200μlのクロロホルム−メタノール(2:1)溶液で処理した。最高速度でマイクロ遠心分離した後、有機相を集めて真空乾燥した。乾燥した脂質を、10%のトリトンX100(triton-X 100)を含む、20μlの2−プロパノールに溶解して分析試料とした。細胞内の全コレステロール含有量は、Amplex Red Cholesterol Assay kit (Sigma, St. Louis, MO)を用いて測定した。製造社の推奨に基づき、標準コレステロールを用いて校正した。
チトクロームCは水に可溶の15kDaのタンパク質であり、ミトコンドリア膜間スペース内に存在し、アポトーシス早期に放出される。メリチン担持ナノ小滴がチトクロームC放出の引き金を引くかどうかを確認するため、C−32メラノーマ細胞をさまざまな濃度のメリチンナノ粒子で37℃、1時間処理し、それをMitochondrial Isolation Kit (Pierce Biotechnology, Rockford, IL)を用いてミトコンドリアを分離した。こうして得られたミトコンドリアフラクション及び細胞質ゾルフラクションに存在するチトクロームCを、チトクロームC ELISA kit (Invitrogen, Carlsbad, CA)を用いて分析した。
乳酸デヒドロゲナーゼ(LDH)は、安定なエンザイムであり、原形質膜の損傷によって細胞からすみやかに放出される。Bio Vision (Mountain View, CA)より得た乳酸デヒドロゲナーゼ放出分析キットを用いて、操作マニュアルに従って分析した。簡単に言うと、C−32メラノーマ細胞をさまざまな濃度の遊離メリチン又はメリチンナノ粒子で37℃、1時間処理し、放出された乳酸デヒドロゲナーゼ量を定量した。0.1%トリトンX100で処理した細胞を100%放出とした。
腫瘍モデルに与えるメリチンナノ粒子の効果:B16メラノーマ
このモデルでは、100万個のB16F10メラノーマ細胞をC57BL/6マウスの右わき腹に移植した(第0日)。マウスを3つのグループに分けた。対照グループは生理食塩水を与えた。第2のグループは、αVβ3ターゲットリガンドが脂質/界面活性剤層に含まれていない点を除き、実施例1に従って調製したターゲット化していないメリチンナノ粒子を与えた。第3のグループは、実施例1に従って調製したαVβ3ターゲット化メリチンナノ粒子を与えた。
対照グループ、生理食塩水:2170(±995);
非ターゲット化メリチンナノ粒子:285(±207);
αVβ3ターゲット化メリチンナノ粒子:337(±198)
であった。同様に、最終的腫瘍重量(g)は図9Bに示すように、
生理食塩水:1.90(±0.88);
非ターゲット化メリチンナノ粒子:0.28(±0.18);
αVβ3ターゲット化メリチンナノ粒子:0.33(±0.16)
このように、非ターゲット化した、またターゲット化したメリチンを含むナノ粒子によって、腫瘍が顕著に縮小した。

アスパラギン酸アミノトランスフェラーゼ(AST)は、肝臓への転移のため生理食塩水処置対照マウスで増加した。
MDA435異種移植片に対する生体内効果
50μlのマトリゲル(matrigel)、100ng/mlのVEGF、100ng/mlのbfGFからなる50μlに含んだ200万個のMDA435細胞を、舌脂肪体(linguinal fat pad)に挿入して、異種移植片を持つマウスを用意した(第0日)。先と同様、3つのグループに分け、生理食塩水、ターゲット化及び非ターゲット化メリチン担持ナノ粒子を第7,10,13,16,19,22日に投与した。投与量は、グループ2,3で5mg/kgであった。マウスを犠牲にし、腫瘍の体積を測定した。グループ3では、対照グループに比較して腫瘍の成長速度が24.68%減少した。ターゲット化ナノ粒子では11.18%低下した。しかし図10に示すようにグループ間でばらつきがあった。
Claims (10)
- ナノ粒子を含む組成物であって、該ナノ粒子は脂質/界面活性剤層によってコーティングされた液状の疎水性含ハロゲン炭素コアを含み、該脂質/界面活性剤層は、膜溶解性又は細孔形成ペプチドである、1以上の膜組み込みペプチドを含むことを特徴とする、組成物。
- 前記含ハロゲン炭素(化合物)は1以上のパーフルオロカーボンを含み、前記脂質/界面活性剤層は1以上のリン脂質を含むことを特徴とする、請求項1に記載の組成物。
- 前記脂質/界面活性剤層は、ターゲット組織又は細胞に特異的なターゲットリガンドを含むことを特徴とする、請求項1又は2に記載の組成物。
- 前記膜組み込みペプチドは、細胞毒性を持つことを特徴とする、請求項1〜3のいずれか一に記載の組成物。
- 前記膜組み込みペプチドは、3−6個のアミノ酸からなるカチオン性アミノ酸配列に隣接する、10−30個のアミノ酸からなる疎水性アミノ酸配列を含むことを特徴とする、請求項4に記載の組成物。
- 前記膜組み込みペプチドはメリチンであることを特徴とする、請求項5に記載の組成物。
- 前記脂質/界面活性剤層は、さらに治療用又は診断用薬剤を含むことを特徴とする、請求項1〜6のいずれか一に記載の組成物。
- 前記膜組み込みペプチドは、治療用又は診断用薬剤と共有結合していることを特徴とする、請求項1〜6のいずれか一に記載の組成物。
- ターゲット細胞又は組織の破壊又は成長の抑制により恩恵を受ける対象の症状の治療に使用するための、請求項1〜8のいずれか一に記載の組成物。
- ターゲット細胞又は組織への治療薬剤の輸送により恩恵を受ける対象の症状の治療に使用するための組成物であって、前記脂質/界面活性剤層はさらに該治療薬剤を含むことを特徴とする、請求項7又は8に記載の組成物。
