SILV中の変異をもつ動物は、メルルコートの色パターン表現型とともに存在する。SILV遺伝子における変異を宿す動物の識別のための方法が以下により詳細に記載される。この遺伝子は、複数の生物で記載されているが、メルルパターン化に関与しているとしては決して影響を与えるものではない。実施例1は、SILV中に変異をもつイヌがこのメルルコートの色パターン表現型とともに存在することを示す。1つのこのような変異は、SILVのイントロン10中への短い散在されたヌクレオチド要素(「SINE」)の挿入であり、そしてイヌの複数の血統に存在する。SINEのSILV遺伝子中への挿入は、スプライス部位の変異を引き起こし、異常mRNAを生ずると考えられる。SILV遺伝子における変異の同定は、所有者または飼育者が動物の遺伝子型を確認することを可能にする。なぜなら、それは、メルルに関係するからである。このような情報は、所望コート色パターンおよび動物が、聴覚および眼科学的異常なくして子孫で産出されるように交配を計画するとき、大きな価値をもつ。メルルとともに分離するマイクロサテライトマーカーがまた記載され、これは、連鎖不均衡マッピングを用いて同定された。SILV遺伝子における変異は、ゲノムDNAを含む細胞または組織のような任意の生物学的サンプルから同定され得る。代表的なサンプルは、毛根または頬綿棒で集めた標本(swab)を含む。1つのこのような変異は、SINEのSILVのイントロン10中への挿入であり、そして現在まで分析されたイヌのすべての血統中に存在する。
(定義)

イヌにおいて、完全に色がついた背景上の希釈された色素の大きな領域はメルル(merle)として知られる。このメルルのコートパターンは、多くのイヌ血統で見い出され、そして複数の異常をともなう。
LDデータおよび色素形成におけるSILVの役割に基き、この遺伝子が、メルル遺伝子座の候補して選択された。Minnickら(1992)Gene 110、235〜238によって記載されるイヌSINEのクラスに構造的に類似のSINEが、分析されたすべてのメルルイヌのついてSILV中で同定された。このSINEは、イヌD2ドーパミンレセプター遺伝子中で先に同定されたイヌSINE(Jeoung、D.ら(2000)Anim.Genet.31、334〜335)、ジストロピン遺伝子(Fletcher、S.ら、(2000)Am.J.Vet.Res.12、1964〜1968)、および中心核筋障害において連坐するPTPLA遺伝子(Pele、M.ら、(2005)Hum.Mol.Genet.14、1417〜1427)と高い配列類似性(95%〜97%)を示す。これらのSINEはtRNA由来であり、そしてイヌでは高度に豊富であり、ゲノムの7%になる(Kirkness、E.F.ら(2003)Science 301、1898〜1903)。
SILV遺伝子とともに分離する連鎖不均衡マッピングを用いて同定されたマイクロサテライトマーカーがまた記載される。以下に詳細に記載されるように、連鎖不均衡は、シェトランドシープドッグにおけるメルル表現型およびHSA12q13とのシンテニー(遺伝子相同性)の保存を示すCFA10の領域中に位置するマイクロサテライトマーカーを用いて同定された。
連鎖は、2つ以上の非対立遺伝子の同時遺伝である。なぜなら、これらの遺伝子座は、減数***の後に、それらが非連鎖遺伝子について期待される50%より頻度が多く随伴したままであるように同じ染色体上で緊密に近接しているからである。減数***の間に、遺伝子材料の物理的交差があり、胚細胞の生成の間に、個々のクロマチッド(染色分体)間で母性および親の遺伝子寄与物の物理的交換があることは明瞭である。この交換は、各々の親では連続している染色体領域中の別個の遺伝子を必然的に分離し、そしてそれらを保持された直線状の順序で混合し、「組換え体」を生じる。減数***組換えによる組換え体を形成するプロセスは、遺伝子形質の再取り合わせにおける必須の特徴であり、そして遺伝子の伝達の理解の中枢である。
SILV遺伝子は、上記に記載のような当業者に周知の多くの方法によって任意のタイプの変異についてスクリーニングされ得る。好ましい実施形態では、動物の遺伝子型を決定するための方法は、それがメルル遺伝子に適用されるとき、PCRを基礎にする試験、その後の電気泳動である。例えば、DNAは、イヌからとられた頬の綿棒で集めた標本から抽出される。このDNAは、SILV遺伝子にハイブリダイズするプライマーを用いてPCRによって増幅される。得られる増幅されたDNAフラグメントは、異なるサイズのフラグメントを示す電気泳動によって解像され、より大きなフラグメントは、SILV遺伝子中のSINE変異を宿す。この方法は、2日未満で実施され得る。
好ましい実施形態では、上記生物学的サンプルは、ゲノムDNAを含む任意の組織である。最も好ましくは、この生物学的サンプルは、血液、毛、粘膜かきとり物、***、組織生検物、または唾液である。最も好ましい実施形態では、この生物学的サンプルは血液である。
試薬は、代表的には、以下のいずれかを同定するオリゴヌクレオチドからなる:
(1)SILV遺伝子中に挿入されたSINE;または
(2)SILV遺伝子中に挿入されたSINEに随伴するマイクロサテライトマーカー;そして必要に応じて、
(3)正常表現型を生じるSILV遺伝子中に挿入されたSINEに随伴する多形。
メラノサイトは、表皮、毛包、内耳、および眼の脈絡膜を含む多くの組織中に存在する色素産生細胞である(Steingrimsson、E.ら、(2004)Annu.Rev.Genet.38、365〜411)。メラノサイト細胞集団は、胚形成の間の神経堤から放出される非色素メラニン芽細胞から分化する(Steingrimsson、E.ら(2004)Annu.Rev.Genet.38、365〜411)。メラニン芽細胞がメラノサイトに移動かつ分化する複雑なプロセスは十分に理解されていない;しかし、色素異常の研究は、正常発生のために重要な遺伝子を支援して解明した(Mccallion、A.S.、&Chakravarti、A.(2001)Pigment Cell Res.14、161〜169)。
材料および方法
(サンプル収集)DNAサンプルは、Texas A&M UniversityにおけるCanine Genetics Laboratoryで行われた先の研究から、そして参加した所有者および飼育者を通じて得た。全血または頬の細胞はすべてのイヌから回収され、そしてゲノムDNAは、Puregene DNA Isolation Kit(Gentra Systems、Minneapolis、MN、USA)を用いて単離された。
(メルル表現型をもつLD)279のMSS−2マーカーについての遺伝子型データが、9匹のメルルおよび32匹の非メルルシェトランドシープドッグについて生成された。唯一のマーカーが、メルル集団で見かけ上より共通である対立遺伝子を有していた。このマーカーFH2537について、統計学的に有意なp値(7.2×10−6)が得られた。この結果を有効にするために、さらなる遺伝子型データが、7匹のメルル、2匹の二重メルル、および11匹の非メルルシェトランドシープドッグを用いて前述のマーカーについて生成された。これらのデータは、p値を再計算するために用いられ、これは有意に増加した(9.0×10−8)。
シェトランドシープドッグ集団においてメルルパターン化を引き起こすSILV変異が血統に特定的であったか否かを決定するために、6つのその他の血統を代表するメルルおよび非メルルイヌ(コリー、オーストラリアシェパード、ダックスフンド、カーディガンウェルシュコーギー、ボーダーコリー、およびグレートデン)が、この挿入について分析された。すべての他の血統からのメルルイヌは、この挿入についてヘテロ接合型であった(図4)。
この連鎖研究で用いられた60匹のシェトランドシープドッグが、完全にメルル表現型とともに分離した挿入について分析された。この挿入はまた、コリー、オーストラリアシェパード、ダックスフンド、カーディガンウェルシュコーギー、ボーダーコリー、およびグレートデン血統を代表するイヌの間でメルルとともに分離し、そして配列分析は、それらは、同じ変異を宿すことを確認した。この知見は、上記SINE挿入が、創設者事象として生じた可能性があり、しかも、この研究で分析されたこれら7つの血統が共通の先祖を共有することを示唆する。上記メルル表現型が最近出現したその他の血統は、制限されないで、コッカースパニエル、ポメラニアン、およびチワワを含む。多くの血統におけるメルルの出現、および1800年代の働くシェトランドシープドッグ集団から分かれた最初の血統(コリー、オールドイングリッシュシープドッグ、およびシェトランドシープドッグ)がメルルパターン化を有するという事実は、基礎となる変異が血統の分岐(Neff、M.W.ら(2004)Proc.Natl.Acad.Sci.USA 101、11725〜11730)に先立ち得ることを示唆する。メルル遺伝子座の歴史およびSINE挿入内に引き続く変異を用いる系統発生分析は、血統の進化歴史(Shedlock、A.M.ら(2004)Trends in Ecology and Evolution 19、545〜553)を支援して解明し得る。
Claims (13)
- SILV遺伝子中の変異を有するイヌを識別する方法であって:
該イヌから得られた生物学的サンプルを、該SILV遺伝子内のイントロン10とエキソン11の境界における短い散在されたヌクレオチド要素(SINE)である変異について分析する工程を包含し、該SILV遺伝子内のイントロン10とエキソン11の境界における短い散在されたヌクレオチド要素(SINE)がメルル表現型とともに分離する、方法。 - 前記イヌが、ヘテロ接合型メルルである、請求項1に記載の方法。
- 前記SINEが、配列番号26、27、28、29、30、31、32もしくは33の配列、またはストリンジェントな条件下でそれにハイブリダイズする配列を含む核酸分子である、請求項2に記載の方法。
- 前記イヌが、潜在的メルルである、請求項1に記載の方法。
- 前記SINEが、配列番号34、35もしくは36の配列、またはストリンジェントな条件下でそれにハイブリダイズする配列を含む核酸分子である、請求項4に記載の方法。
- 前記生物学的サンプルが、イヌから得られたゲノムDNAを含む細胞または組織である、請求項1に記載の方法。
- 前記生物学的サンプルが、ゲノムDNAテンプレートを用いるPCRおよびポリアクリルアミドゲル電気泳動を実施することにより分析される、請求項1に記載の方法。
- ヘテロ接合型メルルのイヌについてスクリーニングするためのキットであって、請求項1〜7のいずれか1項に記載の方法において使用するための配列番号26、27、28、29、30、31、32もしくは33の配列、またはストリンジェントな条件下でそれにハイブリダイズする配列を含む核酸、および該イヌから得られたサンプルにおける前記SINEの存在を決定するための試薬を含む、キット。
- 潜在的メルルのイヌについてスクリーニングするためのキットであって、請求項1〜7のいずれか1項に記載の方法において使用するための配列番号34、35もしくは36のいずれかの配列、またはストリンジェントな条件下でそれにハイブリダイズする配列を含む核酸、および該イヌから得られたサンプルにおける前記SINEの存在を決定するための試薬を含む、キット。
- 15bpの二重ヌクレオチド配列が前記SINEの両側に位置する、請求項1に記載の方法。
- 前記15bpの二重ヌクレオチド配列が両側に位置する前記SINEが、配列番号24の43位と44位に対応するヌクレオチド位置間に挿入される、請求項10に記載の方法。
