また、本発明は、当該非ヒト動物を用いた神経変性疾患の治療薬のスクリーニング方法、及び神経変性疾患の治療薬の副作用の検定方法に関する。
ヒトα-synは、140アミノ酸からなる、神経組織に豊富に発現するリン酸化タンパク質であり、主として神経細胞の前シナプスに局在する。α-synは、それぞれ、異なる経緯で同定されアミノ酸配列上相同性の高いβ-シヌクレイン(β-syn)及びγ-シヌクレイン(γ-syn)とともに、シヌクレインペプチドファミリーを構成する(図1)。これらタンパク質は、シナプスの可塑性、ドーパミン放出の制御などに関与することが示唆されているが、詳細な生理的機能は明らかではない(非特許文献1)。
α-synの凝集による神経毒性に関する機序は不明であるが、この機序を明らかにすることは、神経変性疾患の病態を理解し、さらにそれらの治療方法を開発する上で不可欠である。この目的のために、多くの研究室においてα-syn過剰発現型Tgマウスの開発が精力的に試みられた。最初に成功したα-syn過剰発現型Tgマウスは(非特許文献9)、脳切片が、免疫組織染色により、抗α-syn抗体、抗ユビキチン抗体で染まる封入体様構造物を呈するものであった。そして、生化学的には、α-synが不溶分画に集積し、チロシンキナーゼの活性の低下が観察されたこと、行動学的には、ロタロッドで平衡運動感覚能力が有意に低下していることなどから、パーキンソン病の最初のモデル動物と認められた(非特許文献9)。しかしながら、封入体中にあるα-synはアミロイド繊維を形成していないこと、また神経細胞死などの所見が見られないことなど、必ずしもPDやDLBの完全な病態モデルと言えるものではなかった。α-syn Tgマウスの作製は、その後も多くのグループによってなされたが、レビー小体様封入対の形成が認められるものは見出されなかった。
α-synの凝集は神経変性の病態に重要な役割を果たしているため、この凝集を抑制することが神経変性疾患の治療法を開発するための中心課題となる。この見地より、α-synの凝集を抑えるような薬剤開発が重点的に行われてきた。本発明者は、これに関連して、β-synが、α-synとは対照的に、神経保護的に働くことにより、α-synの神経変性促進作用に対する負の調節因子としての役割を担うものであることを提案した(非特許文献10)。β-synは、α-synの中央部にある凝集に本質的な疎水性ドメインが欠損しているため、本来的に、構造上凝集しにくいタンパク質である(図1)。本発明者は、PDのモデル動物であるα-syn Tgマウスとβ-synを過剰発現させたTgマウスとを交配させたバイジェニックマウスにおいて、α-syn Tgマウスに比べてレビー小体様封入体の減少や平衡運動感覚能力の回復などの神経変性病理が改善した所見を得た(非特許文献11)。
β-synの神経保護作用が示される一方で、β-synの変異が関与すると考えられるDLBの症例が報告された(非特許文献16)。この報告において、野生型β-synのアミノ酸配列における第70番目のバリンがメチオニンに置換した変異(V70M)と、第123番目のプロリンがヒスチジンに置換した変異(P123H)とが同定され、それぞれ孤発性及び家族性DLBに連鎖していた(図1)。しかしながら、α-synのミスセンス変異の場合と異なり、P123Hの浸透率が高くないこと、またP123H患者の脳組織におけるレビー小体が、抗β-syn抗体により染色されなかったことなどから、これらの変異が家族性DLBの確かな原因となるかは不明である。それにも関わらず、V70M及びP123Hのいずれのアミノ酸変異も健常人には認められていない。したがって、V70M及びP123Hのアミノ酸変異はSNP多型によるものではないと考えられる。また、変異により置換されたアミノ酸は生物種を越えて保存されている部位であることなどから、V70M及びP123Hのアミノ酸変異は、β-synの機能に重要な変化を及ぼす変異の可能性がある。

本発明の非ヒト動物としては、例えば、変異型βシヌクレイン遺伝子が脳神経細胞及び/又は中枢神経系グリア細胞において発現するものが挙げられる。ここで、変異型βシヌクレインとしては、例えば、自己凝集促進活性及び/又はαシヌクレイン凝集促進活性を有するものが挙げられる。具体的には、変異型βシヌクレインとしては、野生型βシヌクレインのアミノ酸配列において第70番目及び第123番目のアミノ酸のうち少なくとも1つのアミノ酸が他のアミノ酸に置換されたアミノ酸配列、又は当該置換されたアミノ酸配列のうち第70番目及び第123番目のアミノ酸を除く1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなるタンパク質を例示することができ、好ましくは、第70番目のアミノ酸がメチオニンに置換されたもの、及び/又は、第123番目のアミノ酸がヒスチジンに置換されたものが挙げられる。
本発明の非ヒト動物としては、齧歯類動物を例示することができ、具体的にはマウスが挙げられる。
当該方法において、前記非ヒト動物としては、例えば胎生期の非ヒト動物が挙げられ、中枢神経系細胞としては、例えば初代培養細胞が挙げられる。
また当該方法としては、例えば、中枢神経系細胞が脳神経細胞であり、細胞活性が神経突起伸張能及び/又は生存能である方法、あるいは、中枢神経系細胞がグリア細胞であり、細胞活性が増殖能及び/又は生存能である方法が例示できる。
当該方法において、前記非ヒト動物としては、例えば胎生期の非ヒト動物が挙げられ、中枢神経系細胞としては、例えば初代培養細胞が挙げられる。
また当該方法としては、例えば、中枢神経系細胞が脳神経細胞であり、細胞活性が神経突起伸張能及び/又は生存能である方法、あるいは、中枢神経系細胞がグリア細胞であり、細胞活性が増殖能及び/又は生存能である方法が例示できる。
βシヌクレイン(β-syn)の変異による機能変化の有無を解明することは、神経変性の機序の解明において本質的なものである。本発明者は、組換えタンパク質を用いた生化学的実験により、変異型β-syn(P123H及びV70M)がそれ自身で凝集し、またα-synと共存させた場合にはα-synの凝集を促進することを確認した。さらに、変異型β-syn(P123H及びV70M)を過剰発現させた神経芽細胞においては、α-synと共発現させることにより、オートファジー/リソソームによるタンパク質分解が顕著に亢進することを明らかにした。これらの結果から、本発明者は、野生型β-synが神経保護作用を有するものであることとは対照的に、変異型β-synは、神経変性促進的な作用を有するものではないかと考えた(図2)。
(i) 変異型β-synの発現量に応じて、マウスの寿命が顕著に短縮すること(図3D)。
(ii) 複数のライン(F0及びF1マウス)の脳組織標本において、大脳基底核等の特異的な領域にレビー小体様の封入体が形成されること(図3E)。
本発明のトランスジェニック非ヒト動物は、中枢神経系細胞において機能するプロモーター及び変異型βシヌクレイン(β-syn)遺伝子が導入された動物である(以下、「β-synトランスジェニック非ヒト動物」と言うことがある。)。
具体的には、まず、ヒトのcDNA遺伝子ライブラリーからPCR等の方法により野生型β-syn遺伝子断片を得、この遺伝子断片を用いて野生型β-syn遺伝子をスクリーニングする。野生型β-syn遺伝子には、必要により、エピトープタグ等をコードするDNAを連結しておいてもよい。スクリーニングした野生型β-syn遺伝子は、組換えDNA技術を用いて、適当なプラスミドベクターに挿入しておいてもよい。あるいは、上記スクリーニングをする代わりに、予め野生型β-syn遺伝子が挿入された市販のプラスミドベクターを使用してもよい。
中枢神経系細胞において機能するプロモーター(例えば、Thy-1プロモーター)制御下で導入遺伝子が発現制御されるように設計されたベクターに、組換えDNA技術を用いて、変異型β-syn遺伝子を挿入する。上記挿入後のベクターを制限酵素処理等して、目的のDNA構築物、すなわちThy-1プロモーター及びその制御下にある変異型β-syn遺伝子を含むDNA断片を得る。
(iv) 核が融合する前の時期の受精卵に、顕微鏡下で目的のDNA構築物を雄性全核中にマイクロインジェクションにより注入する。
(v) マイクロインジェクション後の受精卵を約20個程度、仮親となる偽妊娠雌マウスの子宮又は卵管内に移植する。
(vi) 移植後の雌マウスを通常の飼育条件下で飼育し、仔マウスを出産させる。
(vii) 仔マウスの一部(尾の先端や耳介片など)からゲノムDNAを抽出し、PCR法又はサザンブロット法等により、Thy-1プロモーターの制御下にある変異型β-syn遺伝子の導入の成否を確認する。
(1) 非ヒト動物を用いる方法
前述の通り、変異型β-synトランスジェニック非ヒト動物は、ヒトの神経変性疾患の病態とよく一致するため、これら疾患の治療薬の開発において極めて有用なものである。そこで本発明は、変異型β-synトランスジェニック非ヒト動物を用いた神経変性疾患の治療薬のスクリーニング方法、並びに当該方法により得られる神経変性疾患の治療薬を提供する。ここで、神経変性疾患としては、例えば、パーキンソン病(PD)、痴呆性レビー小体病(DLB)、多系統萎縮症及びレビー小体亜系型アルツハイマー病等の各種疾患を含む。
(a) 変異型β-synトランスジェニック非ヒト動物に、候補物質を投与する工程(投与工程)
(b) 上記トランスジェニック非ヒト動物について、神経変性疾患の病態を評価する工程
「神経変性疾患の病態を評価する」とは、候補物質を投与した後におけるトランスジェニック非ヒト動物の神経変性疾患に関する病態の表現型を解析することを意味し、候補物質の投与前後のトランスジェニック非ヒト動物の神経変性疾患に関する病態を比較検討すること、あるいは、候補物質を投与した被験動物と投与しない対照動物との比較検討を行うことのいずれをも意味するものである。
「対照動物」は、被験動物との比較対照に使用されるに適している限り限定されるものではなく、変異型β-synトランスジェニック非ヒト動物〔(+/+)又は(+/-)〕であっても、同腹の非トランスジェニック(-/-)の非ヒト動物であっても、トランスジェニック動物でない野生型非ヒト動物であってもよい。
なお、本発明のスクリーニング方法は、必要に応じ、他の工程を含んでいてもよい。
以下に、上記各工程について説明する。
被験動物及び対照動物としては、特に限定されるものではないが、通常、同種の非ヒト動物を用いる。また被験動物及び対照動物は、同腹の動物を用いることが好ましく、同性及び同齢の動物を用いることがより好ましい。さらに被験動物及び対照動物は、飼料の摂取量以外の飼育条件は同様であることが好ましい。
神経変性疾患の治療薬をスクリーニングする場合は、候補物質が投与された被験動物、及び候補物質が投与されていない対照動物について、各種評価項目、例えば、レビー小体様封入体の有無、大小又は個数、寿命、平衡運動感覚能力(ロタロッド)及び記憶能力(水迷路)等を比較評価することが好ましい。これら評価項目の評価(測定方法等)は、公知の手段及び手順により行うことができる。
また本発明は、変異型β-synトランスジェニック非ヒト動物由来の中枢神経系細胞を用いた変性疾患の治療薬のスクリーニング方法、並びに当該方法により得られる変性疾患の治療薬を提供することができる。前記3.(1)の非ヒト動物を用いる方法が、in vivo法であるのに対し、中枢神経系細胞を用いる本方法は、in vitro法と言うことができる。
(1) 非ヒト動物を用いる方法
上述したスクリーニング方法により得られる神経変性疾患の治療薬が優れた効能を示すものであっても、投与した動物に悪影響を及ぼす副作用を持つ場合は有用なものとは言い難いため、上記薬物の副作用の検定は重要である。同様に、公知の又は別途開発された神経変性疾患の治療薬に関しても、その副作用の検定は重要である。そこで本発明は、変異型β-synトランスジェニック非ヒト動物を用いた神経変性疾患の治療薬の副作用を検定する方法を提供する。ここで、神経変性疾患としては、前記3.の項に記載した疾患と同様のものが挙げられる。
(a) 被験動物に、神経変性疾患の治療薬を投与する工程(投与工程)
(b) 被験動物と対照動物とを比較評価する工程、又は被験動物における副作用の有無を検出する工程(評価工程)
なお、本発明の検定方法は、必要に応じ、他の工程を含んでいてもよい。
以下に、上記各工程について説明する。
使用する被験動物及び対照動物については、前記スクリーニング方法と同様である。
各薬物の投与は、その種類に応じ、経口的又は非経口的に行うことができ、投与方法及び投与条件等についても、被験動物の種類や状態を考慮して適宜設定することができる。また各薬物は、薬学的に許容し得る塩又は水和物の状態で投与してもよいし、さらに、薬学的に許容し得る公知の担体とともに投与してもよく、限定はされない。
副作用の有無に関しては、被験動物及び対照動物について、あるいは薬物の投与前後の被験動物について、例えば、体重変化、造血機能、及び生殖機能からなる群より選ばれる少なくとも1つが挙げられる。対照非ヒト動物と、神経変性疾患の治療薬を投与した被験動物とについて、これら評価項目を比較することにより、当該治療薬による副作用の有無、及び副作用の種類や症状の程度等について容易に検定できる。
各評価の方法(測定方法等)は、公知の手段及び手順により行うことができるが、以下に例示して説明する。
本評価方法では、被験動物が、神経変性疾患の治療薬を投与した後、非投与群である対照動物と同様の飼育条件において、対照動物と比べて体重変化に違いが見られるかを検定する。
本評価方法では、被験動物が、神経変性疾患の治療薬を投与した後も末梢血中の各種血液細胞(赤血球、リンパ球及び好中球等)の存在比率と絶対数が正常の範囲にあるかどうかを検定する。
本評価方法では、10週齢を越えた雄と雌の生殖能を検定する。対照動物は、少なくとも遺伝的背景がC57BL/6である限りは、雄も雌も次世代の子供をつくることができるため、神経変性疾患の治療薬を投与した被験動物と有効に比較できる。
また本発明は、変異型β-synトランスジェニック非ヒト動物由来の中枢神経系細胞を用いた神経変性疾患の治療薬の副作用を検定する方法を提供することができる。前記4.(1)の非ヒト動物を用いる方法が、in vivo法であるのに対し、中枢神経系細胞を用いる本方法は、in vitro法と言うことができる。
ここで、中枢神経系細胞の細胞活性については、前記3.(2)で述べた通りである。
野生型β-synの第123番目のアミノ酸であるプロリンをヒスチジンに置換し得る合成プライマーを用いて、野生型β-syn cDNA(pCEP-β-synプラスミド, Takenouchi et al., Mol. Cell Neurosci., 2001)を鋳型にしたPCRを行った。PCRに用いたプライマー、反応液組成及び反応条件は、以下の通りである。

94℃で5分間加熱後、「熱変性・解離:94℃(30sec)→アニーリング:55℃(30sec)→合成・伸長:72℃(30sec)」を1サイクルとして計35サイクル行い、72℃で10分間保温後に4℃で冷却した。
また、常法により、B103神経芽細胞に構築した発現ベクターを導入して形質転換体を得、一過性発現させることにより、β-syn P123HのmRNA及びタンパク質レベルにおける発現を確認した。
生後6、12、18ヶ月のマウスに対し、所定のガイドラインに沿って、安楽死させ、大脳半球を摘出し、左右に分けた。そのうち一方は、病理組織学実験に用い、もう一方は、後日生化学実験に用いるために凍結保存した。

Claims (18)
- 中枢神経系細胞において機能するプロモーター及び変異型βシヌクレイン遺伝子が導入されたトランスジェニック非ヒト動物であって、
該変異型βシヌクレインは、(a)野生型βシヌクレインのアミノ酸配列において第123番目のアミノ酸がヒスチジンに置換されたアミノ酸配列からなるものであるか、又は(b)該置換されたアミノ酸配列のうち第123番目のアミノ酸を除く1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなるものであり、
該非ヒト動物の中枢神経系細胞において発現する変異型βシヌクレインが、自己凝集促進活性を有するものである、
前記非ヒト動物。 - 変異型βシヌクレインが中枢神経系細胞において自己凝集により蓄積したものである、請求項1記載の非ヒト動物。
- 変異型βシヌクレイン遺伝子が脳神経細胞及び/又は中枢神経系グリア細胞において発現するものである、請求項1又は2記載の非ヒト動物。
- 変異型βシヌクレインは、(a)野生型βシヌクレインのアミノ酸配列において第123番目のアミノ酸がヒスチジンに置換され且つ第70番目のアミノ酸がメチオニンに置換されたアミノ酸配列からなるものあるか、又は(b)該置換されたアミノ酸配列のうち第123番目及び第70番目のアミノ酸を除く1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなるものである、請求項1〜3のいずれか1項に記載の非ヒト動物。
- 非ヒト動物が齧歯類動物である、請求項1〜4のいずれか1項に記載の非ヒト動物。
- 齧歯類動物がマウスである、請求項5記載の非ヒト動物。
- 請求項1〜6のいずれか1項に記載の非ヒト動物に候補物質を投与する工程、及び当該候補物質投与後の非ヒト動物の神経変性疾患に関する病態を評価する工程を含む、神経変性疾患の治療薬をスクリーニングする方法。
- 請求項1〜6のいずれか1項に記載の非ヒト動物由来の中枢神経系細胞に候補物質を接触させて当該細胞の細胞活性を測定し、得られる測定結果を指標として神経変性疾患の治療薬をスクリーニングする方法。
- 前記非ヒト動物が胎生期の非ヒト動物である、請求項8記載の方法。
- 中枢神経系細胞が初代培養細胞である、請求項8又は9記載の方法。
- 中枢神経系細胞が脳神経細胞であり、細胞活性が神経突起伸張能及び/又は生存能である、請求項8〜10のいずれか1項に記載の方法。
- 中枢神経系細胞がグリア細胞であり、細胞活性が増殖能及び/又は生存能である、請求項8〜10のいずれか1項に記載の方法。
- 請求項1〜6のいずれか1項に記載の非ヒト動物に神経変性疾患の治療薬を投与する工程、及び当該薬物投与後の非ヒト動物における副作用の有無を検出する工程を含む、神経変性疾患の治療薬の副作用を検定する方法。
- 請求項1〜6のいずれか1項に記載の非ヒト動物由来の中枢神経系細胞に神経変性疾患の治療薬を接触させて当該細胞の細胞活性を測定し、得られる測定結果を指標として神経変性疾患の治療薬の副作用を検定する方法。
- 前記非ヒト動物が胎生期の非ヒト動物である、請求項14記載の方法。
- 中枢神経系細胞が初代培養細胞である、請求項14又は15記載の方法。
- 中枢神経系細胞が脳神経細胞であり、細胞活性が神経突起伸張能及び/又は生存能である、請求項14〜16のいずれか1項に記載の方法。
- 中枢神経系細胞がグリア細胞であり、細胞活性が増殖能及び/又は生存能である、請求項14〜16のいずれか1項に記載の方法。
