Description
筋肉疲労を改善する方法としては、マッサージ、ビタミンやタンパク質等の栄養補給が知られている。
チドであり、生体内では速やかにアラニンとグルタミンに分解される(非特許文献2参照)。グルタミンに由来する生理機能として、骨格筋タンパク代謝の調節、小腸粘膜の修復、免疫機能の向上など多くの作用が知られており、アラニンに由来する生理機能としては糖尿病患者の血糖値抑制作用が報告されている(非特許文献3参照)。
小麦グルテン由来のグルタミンペプチドが、運動負荷後の疲労感と筋肉痛を軽減する作用を有することが知られている(特許文献1参照)。
(1)アラニルグルタミンまたはその塩を有効成分として含有する筋肉疲労改善剤。
(2)アラニルグルタミンまたはその塩の有効量を、必要とする対象に投与することを特徴とする、筋肉疲労改善方法。
(3)筋肉疲労改善剤の製造のための、アラニルグルタミンまたはその塩の使用。
アラニルグルタミンの塩としては、酸付加塩、金属塩、アンモニウム塩、有機アミン付加塩、アミノ酸付加塩等があげられる。
酸付加塩としては、塩酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩、酢酸塩、マレイン酸塩、フマル酸塩、クエン酸塩、リンゴ酸塩、乳酸塩、α−ケトグルタル酸塩、グルコン酸塩、カプリル酸塩等の有機酸塩があげられる。
アンモニウム塩としては、アンモニウム、テトラメチルアンモニウム等の塩があげられる。
有機アミン付加塩としては、モルホリン、ピペリジン等の塩があげられる。
アラニルグルタミンは、合成法、酵素法または発酵法など、いずれの製造方法でつくられたものでもよい。
アラニルグルタミンの製造方法としては、たとえばブレティン・ザ・ケミカル・ソシエティー・オブ・ジャパン(Bull.Chem.Soc.Jpn.), 34,739(1961)、35,1966(1962) 、37,200(1964)、欧州特許第311057号、ドイツ特許第3206784号、特開平6−234715号、WO2004/058960に記載された方法があげられる。
筋肉疲労を有する者に対して、本発明の筋肉疲労改善剤を投与することにより、筋肉疲労を改善させることができる。
本発明の筋肉疲労改善剤としては、アラニルグルタミンまたはその塩をそのまま投与することも可能であるが、各種の製剤として提供するのが望ましい。
製剤の投与形態は、治療に際し最も効果的なものを使用するのが望ましく、経口投与または、例えば静脈内、腹膜内もしくは皮下投与等の非経口投与をあげることができるが、経口投与が好ましい。
経口剤を製剤化する際には、賦形剤、結合剤、崩壊剤、潤沢剤、分散剤、懸濁剤、乳化剤、希釈剤、緩衝剤、抗酸化剤、細菌抑制剤等の添加剤を用いることができる。
また、経口投与に適当な製剤には、一般に飲食品に用いられる添加剤、例えば甘味料、着色料、保存料、増粘安定剤、酸化防止剤、発色剤、漂白剤、防かび剤、ガムベース、苦味料、酵素、光沢剤、酸味料、調味料、乳化剤、強化剤、製造用剤、香料、香辛料抽出物等が添加されてもよい。
経口投与に適当な製剤は、そのまま、または、例えば粉末食品、シート状食品、瓶詰め食品、缶詰食品、レトルト食品、カプセル食品、タブレット状食品、流動食品、ドリンク剤等の形態としては、筋肉疲労改善用の健康食品、機能性食品、栄養補助食品、特定保健用食品等の飲食品として用いてもよい。
また、これら非経口剤においても、経口剤で例示した希釈剤、防腐剤、フレーバー類、賦形剤、崩壊剤、潤沢剤、結合剤、界面活性剤、可塑剤などから選択される1種またはそれ以上の補助成分を添加することができる。
本発明の筋肉疲労改善剤の投与量および投与回数は、投与形態、患者の年齢、体重、治療すべき症状の性質もしくは重篤度により異なるが、通常、成人一日当り、アラニルグルタミンまたはその塩として、通常は5mg〜10000mg、好ましくは50mg〜5000mg、より好ましくは500mg〜3000mgとなるように、一日一回ないし数回投与する。投与期間は、特に限定されないが、通常は1日間〜1年間、好ましくは2週間〜3ヶ月間である。
試験例1
21-60歳の健常男性26名に、アラニルグルタミン(協和発酵工業社製)を1ヶ月間継続して摂取させ、被験者の筋肉疲労症状を評価した。
以下に、本発明の実施例を示す。
アラニルグルタミン136.2kg、微結晶セルロース36.0kg、ショ糖脂肪酸エステル6.6kg、リン酸カルシウム1.2kgおよびβ−シクロデキストリン20.0kgを、コニカルブレンダー(CB−1200ブレンダー、日本乾燥機株式会社製)を用いて混合する。得られる混合物をロータリー圧縮成形機(VIRGO524SS1AY、菊水制作所社製)を用いて、圧縮成形圧10kNで圧縮成形して、直径8mm、250mgの筋肉疲労改善用錠剤を製造する。
実施例1で調製した混合物20kgと0.2kgの二酸化ケイ素とを混合攪拌して得られる混合物をカプセル充填機に投入し、ゼラチン製2号ハードカプセル20,000錠に充填し、ハードカプセルを得る。得られるハードカプセルの表面を、ハイコーターHCT−48型(フロイント産業社製)により、ツェイン溶液を用いてコーティングし、筋肉疲労改善用腸溶カプセル20,000錠を製造する。
実施例1で調製した錠剤の表面を、ハイコーターHCT−48型(フロイント産業社製)により、シェラック溶液を用いてコーティングし、筋肉疲労改善用腸溶錠剤を製造する。
アラニルグルタミン1.28kg、エリスリトール3kg(日研化学社製)、クエン酸0.05kg、人工甘味料3g、香料0.06kgを液温70℃で水50Lに攪拌溶解し、クエン酸でpHを3.3に調整後、プレート殺菌を用いて滅菌して瓶に充填後、パストライザー殺菌し、筋肉疲労改善用飲料を製造する。
Claims (1)
- 1日あたりのアラニルグルタミンの摂取量が500mg〜3000mgとなるように製剤化されたアラニルグルタミンまたはその塩を有効成分として含有する筋肉疲労改善剤。
