本発明に係る微生物の検出方法は、ファンネル及びフィルターを使用して、微生物を含有する液体サンプル、該微生物を染色する染色液、該染色液を洗浄する洗浄液を順次ろ過し、フィルター上に捕集された微生物を染色して検出する方法であって、微生物を含有する液体サンプルをろ過し、フィルター上に微生物を捕集する工程(1)と、微生物を捕集後に、染色液をろ過し、捕集された微生物を染色する工程(2)と、微生物を染色後に、洗浄液をろ過し、フィルターを洗浄する工程(3)と、を有し、工程(3)において洗浄液のろ過面を、工程(1)における液体サンプルのろ過面及び工程(2)における染色液のろ過面を包含するように、これらろ過面よりも拡大して、洗浄液をろ過することを特徴とするものである。
本発明の検出方法によれば、ファンネルとフィルター、並びにフィルターとベースとの間への染色液の滲出が抑制され、調整具とフィルターとの間に染色液が滲出しても、洗浄液で除去でき、微生物の染色以外でフィルター上に残留している余剰の染色液を除去できるので、微生物検出時のノイズを大幅に低減でき、目的とする微生物を高精度に検出できる。
以下、各工程について順次説明する。

[実施例1]
ADVANTEC社製ファンネル(d1;16mm)と、図3に示すステンレス製調整具(D5;25mm、d5;12mm、R51;6mm、r51;3.5mm)を適用した図1に示すろ過システムと、ポアサイズ0.4μmのポリエステル製フィルターを使用して、ろ過装置を組み立て、以下の試験を行った。なお、ファンネル、調整具、ベース及びフィルターは、クランプで一体に固定した。
まず、大腸菌(Escherichia coli)を分散させたPBS(Phosphate Buffered Saline)100mlをファンネルに注ぎ、減圧ろ過して、フィルター上に大腸菌を捕集した。
次いで、200μg/mlのCFDA染色液(Molecular probes社製)2mlをファンネルに注ぎ、そのまま室温で10分間保持して染色してから、CFDA溶液を減圧ろ過し、さらに20mlのPBSをファンネルに注ぎ、減圧ろ過した。
次いで、リングを取り外し、それ以外は上記と同様にろ過装置を組み立てた。そして、PBS30mlをファンネルに注ぎ、減圧ろ過して、フィルターを洗浄した。
次いで、洗浄済みフィルターを、マイクロファインダー(アサヒビール株式会社製)で解析した。解析画像を強調処理した結果を図8に示す。図8に示すように、フィルター上に大腸菌が検出された。この時、染色ノイズは発生せず、大腸菌を正確に検出できた。
