Description
免疫系の細胞は先天性免疫系の細胞と適応(後天性または特異的)免疫系の細胞とに分けられる。先天性免疫系の細胞には、末梢血中に存在する単球、顆粒球、ナチュラルキラー細胞や、皮膚、気道、消化管、尿生殖路などの末梢組織、各種内臓器、ならびに脾臓、リンパ節(LN)、パイエル板などの二次リンパ組織のような血管外コンパートメントに存在するマスト細胞、マクロファージ及び樹状細胞(DC)などがある。先天性免疫細胞の主な機能は、a)感染性粒子の伝播を中和、制限し、腫瘍細胞を除去することによる直接防御機構の提供、b)健康な組織の免疫監視機能、c)適応免疫応答の開始、である。適応免疫系の細胞としてはT細胞やB細胞などのリンパ球が挙げられる。これらの細胞にはT細胞抗原受容体(TCR)及びB細胞抗原受容体(BCR)と呼ばれるクロノタイプの細胞表面抗原受容体が存在することから先天性免疫細胞と区別される。各リンパ球は特定の抗原を認識する異なるTCRまたはBCRを有している。抗原認識の特異性は、T細胞及びB細胞の分化過程と、エフェクターT細胞及びB細胞が分化する際の抗原親和性の成熟過程(アフィニティーマチュレーション)において、多数のTCRまたはBCR可変遺伝子セグメントの再構成が起こることで決定される。抗原刺激を受けていない末梢血中のナイーブなT細胞は、それぞれのTCRの抗原選択性において互いに異なっており、ナイーブT細胞はそれぞれが特異的に認識する抗原を有する物質によって免疫活性化されることによって増殖する。その結果、適応性免疫応答においては、潜在的な感染性物質に対して特異的なTCRを有するナイーブT細胞群が細胞***によって増殖し、a)潜在的感染性物質に対する防御に直ちに参加するエフェクターT細胞へと分化するとともに、b)この特定の潜在的感染性物質に対する長期的な防御を与えるメモリーT細胞へと分化する。エフェクターT細胞の寿命は短く、免疫応答の消失期には消失するのに対し、メモリーT細胞は長寿命であり、初期にこれらの細胞が存在した組織や選択的再循環経路に応じてメモリーT細胞のサブセットへと分化する(Moser et al.,2004)。T細胞は、へテロ二量体であるTCRの構成要素によってαβT細胞とγδT細胞とに更に分けられる(下記参照)。αβ−TCRはα及びβタンパク鎖からなり、γδ−TCRはγ及びδタンパク鎖からなる。正常で健康なヒトでは、CD3+T細胞の大半(>80%)はαβT細胞である。TCRには無変異のCD3分子が結合しており、これによってT細胞はB細胞や他のすべての免疫細胞と区別される。αβT細胞の多くは、いわゆる主要組織適合遺伝子複合体(MHC)分子拘束性により抗原を認識する。これは、わずかな抗原とMHC非拘束的に結合するB細胞のBCRとは対照的である。「MHC拘束」とは、TCRによる抗原の認識の形態をいい、抗原性ペプチドをMHC分子とともにいわゆるMHC−ペプチド複合体として、DCなどの抗原提示細胞(APC)上に提示することを含む(下記参照)。MHC分子はMHCクラスI(MHC−I)とMHCクラスII(MHC−II)に大別され、これらはαβT細胞の2つの主要なサブクラスであるCD8+αβT細胞とCD4+αβT細胞のTCRを活性化させる。CD4+αβT細胞上のTCRがMHC−II−ペプチド複合体を認識するのに対し、CD8+αβT細胞上のTCRはMHC−I−ペプチド複合体を認識する。これと大きく異なり、ヒトの末梢血中に存在する主要なサブセットであるγδT細胞はCD4、CD8のいずれも発現せず、また、そのTCRは、抗原認識についてMHC拘束を必要としない(下記参照)。
γδT細胞は、Vγ及びVδ遺伝子セグメントによってコードされるTCRを有するCD3+細胞のサブセットである(Morita et al.,2000;Carding and Egan,2002)。それらは、初期にこれらの細胞が存在する血液または組織、細胞のVγVδ−TCRのタンパク鎖の構成や抗原選択性によって更に分類される。ヒトの皮膚の上皮組織や上皮関連/粘膜組織、気道、消化管、尿生殖路及び幾つかの内臓ではVδ1+−TCR鎖を発現しているγδT細胞(Vδ1+T細胞)が大部分を占めるが、末梢血のγδT細胞ではそのごく一部(<20%)がVδ1+T細胞である。Vδ1+T細胞のTCRはMHC関連CD1分子が提示する脂質抗原を認識する。更にVδ1+T細胞は、MHC関連分子であるMICA、MICB、及びヒートショックタンパクなどのストレス関連タンパクに対して応答する。この細胞は、腫瘍やストレスを受けた細胞に対する最初の防御ラインとして機能すると考えられており、更には外傷の治癒、組織修復や自己免疫に関与しているものと考えられている。正常なヒトの末梢血では、γδT細胞は全CD3+細胞の2〜10%程度であり、末梢血のγδT細胞の大半(>80%)はVγ2Vδ2+−TCR鎖を発現しているγδT細胞(Vγ2Vδ2+γδT細胞)である(Morita et al.,2000;Carding and Egan,2002)。この細胞は、ほとんどが微生物に由来する小型の非ペプチド抗原に対して高い選択性を示し、ペプチド選択的αβT細胞では一般的である(上記参照)古典的MHC分子による抗原提示を必要としない。Vγ2Vδ2+γδT細胞の抗原としては、イソペンテニルピロリン酸(IPP)などのプレニルピロリン酸、アルキルアミン、ならびに多くのヒトの微生物病原体や共生細菌で新たに発見されたイソプレノイド生合成経路の代謝産物などがある(Morita et al.,2000;Eberl et al.,2003)。IPPやアルキルアミンなどのVγ2Vδ2+γδT細胞の抗原の一部は壊死性組織細胞からも放出される。微生物由来の小型の非ペプチド抗原に対する選択性を有する相同的VγVδ−TCRを有するヒトVγ2Vδ2+γδT細胞の相同タンパクは、アカゲザルなどの高等霊長類にも存在するが、マウスやウサギなどのげっ歯類には存在しない。Vγ2Vδ2+γδT細胞の存在がなぜ高等霊長類に限定されているかは明らかではないが、こうした細胞は、微生物の特徴的な種特異的選択に対する細胞防御機構に対する特定の必要性を満たすべく進化したものと考えられる。Vγ2Vδ2+γδT細胞は、in vitroでの末梢血Vγ2Vδ2+γδT細胞の組織培養や、in vivoでのアカゲザルなどの高等霊長類におけるワクチン接種実験で、モデル抗原としてのIPPや微生物抽出物に応答して急速に増殖する(Chen and Letvin,2003)。また、ヒトにおける微生物感染では、末梢血中のVγ2Vδ2+γδT細胞がしばしば急激に増殖し、末梢血中の全CD3+細胞の60%以上を占めるレベルに達する。これらの知見は、ヒトVγ2Vδ2+γδT細胞が微生物感染における免疫プロセスにおいて重要な役割を担っているという考え方を支持するものである(Morita et al,2000;Carding and Egan,2002;Chen and Letvin,2003)。病原体や共生細菌などの微生物に一般的に見られる非ペプチド抗原に対するVγ2Vδ2+γδT細胞の特有の選択性は、Vγ2Vδ2+γδT細胞のTCRが微生物由来の多様なリガンドに応答して、樹状細胞(DC)や他の抗原提示細胞(APC)の活性化及び成熟を引き起こすToll様受容体(TLR)と同様の機能を有すること、γδT細胞が先天性免疫系の細胞を動員するケモカインや、抗原提示細胞を刺激して顆粒球、マクロファージ、NK細胞による細菌への攻撃を促す炎症性サイトカインを速やかに分泌することによって病原体の除去に寄与していることを示唆するものである(Morita et al.,2000;Carding and Egan,2002;Chen and Letvin,2003)。Vγ2Vδ2+γδT細胞はまた、感染または腫瘍性組織細胞を殺すナチュラルキラー細胞受容体を発現する。これらの知見は、TNF−αなどの炎症性サイトカインの分泌が局所的な適応免疫応答に寄与することも知られているが、γδT細胞は自然免疫機能を第一に行うものであるという考え方を支持するものである。これに対して、適応免疫応答におけるγδT細胞の直接的な関与を示す証拠は明らかなものではない。例えば、CD1拘束性T細胞はCD1−脂質複合体を提示する樹状細胞(DC)の成熟を誘導することが報告されている。また、更なる説明はなされていないものの、マウスでの実験によって、γδT細胞のB細胞応答における一定の役割が示され、加えて、ヒトγδT細胞はin vitroでの共培養でB細胞の応答を調節することが示された(Brandes et al.,2003)。更に、アカゲザルでの研究によってVγ2Vδ2+γδT細胞はMycobacterium bovis 抗原に対してin vivoで記憶応答を起こすことが示された(Chen and Letvin,2003)。これらの知見は、γδT細胞がB細胞や樹状細胞といった適応免疫系の細胞と相互作用を行うことの根拠を与えるものである。こうした免疫調節機能の多くはγδT細胞によるサイトカイン産生によるものと結論づけられたか、あるいは説明がなされていない。これらの知見はいずれも、抗原提示におけるγδT細胞の役割を支持するものではない点は重要である。
樹状細胞(DC)は白血球の一クラスであり、骨髄の造血前駆細胞から誘導され、上皮/粘膜組織(皮膚、気道、消化管、尿生殖路など)や二次リンパ組織(脾臓、リンパ節(LN)、パイエル板(PP)など)のような血管外部位に主に存在する(Banchereau and Steinman,1998;Steinman et al.,2003;Banchereau et al.,2004)。末梢血では、樹状細胞または樹状細胞の前駆細胞が単核白血球に占める割合は1%未満である。異なる樹状細胞のサブセットは、上皮の境界を形成する軟部組織に主として存在する間質性樹状細胞、真皮に存在するランゲルハンス細胞(LC)、及びリンパ節に対する選択的ホーミング能を有する形質細胞様樹状細胞などのように、組織における局在化の仕方が異なっている。これらの樹状細胞のサブセットは、完全に分化した非増殖性細胞であり、数日から数週間と限られた寿命を有するが、このことはこれらの細胞が安定的状態で骨髄由来の前駆細胞によって常に置き換えられていることを示している。対照的に、ヒトメモリーT細胞は何年にもわたって生き続け、安定的(恒常的)増殖によって維持されている。組織に存在する樹状細胞の主な機能は、局所での抗原の吸収とプロセシング、求心性リンパ管を介した流入領域リンパ節への抗原の移送、及び抗原特異的適応免疫応答の開始である(Banchereau and Steinman,1998;Steinman et al.,2003;Banchereau et al.,2004)。樹状細胞はまた、免疫寛容誘発的な条件下(炎症誘発性T細胞が共刺激されない)でT細胞に抗原が提示されると免疫寛容を誘導する。同様に抗原提示B細胞も免疫寛容を誘導することが示されている。自己抗原に対する免疫寛容の破綻はしばしば自己免疫疾患の原因となると考えられていることから、自己抗原を提示する免疫寛容誘発性樹状細胞は免疫恒常性の不可欠な調節因子である。正常な末梢組織では、樹状細胞は完全に分化しているが「未成熟」な状態で存在している。未成熟な樹状細胞は、局所的な感染、炎症または組織損傷部位へと速やかに動員されるように炎症性サイトカインに対する受容体を発現している。これらの細胞はそれ自体の免疫寛容誘発性は低い(一次適応免疫応答を誘導しない)が、(受容体を介したエンドサイトーシスや液相のピノサイトーシス機構による)抗原の取込み、抗原のプロセシング、細胞内のMHC−I/II分子へのペプチドの結合、及びその細胞表面への提示に特化している。未成熟樹状細胞はリンパ節ホーミング受容体であるCCR7を通常発現しておらず、このタイプの樹状細胞がどのようにして脾臓、リンパ節やパイエル板のT細胞領域に到達するかは現時点では解明されていない。Toll様受容体(TLR)を活性化するウイルスまたは細菌による刺激、ホスト細胞由来の炎症性メディエーター(特にインターフェロン[IFN]−g、腫瘍壊死因子[TNF]−a、インターロイキン[IL]−I、プロスタグランジンE2[PGE2]、組織増殖因子)、及びT細胞共刺激分子(CD40リガンド/CD154)など多くの成熟シグナルによって樹状細胞の「成熟」が誘導される(Banchereau and Steinman,1998;Steinman et al.,2003;Banchereau et al.,2004)。樹状細胞成熟の早期では、更なる樹状細胞や先天性免疫系の細胞(単核球、顆粒球、ナチュラルキラー細胞)の動員によって免疫応答を増強するために樹状細胞は高濃度の炎症性ケモカインを分泌する。この後、こうした炎症性遊走プログラムは、炎症性ケモカインに対する受容体がCCR7に置換されることを特徴とするリンパ節ホーミングプログラムに次第に移行していく。CCR7は、リンパ管及び脾臓、リンパ節やパイエル板のT細胞領域に存在する2種類のCCR7選択的ケモカインであるELC/CCL19及びSLC/CCL21に応答して、感作された樹状細胞が末梢組織から流入領域リンパ節へと有効に移動するうえで不可欠である。したがってCCR7の発現は成熟した、または成熟しつつある樹状細胞をマーキングするものである。リンパ節ホーミング能を有する以外に、成熟樹状細胞はその細胞表面に多数のMHC−I/II−ペプチド複合体と、ナイーブな(抗原刺激を受けていない)αβT細胞の適正な刺激に必要とされる多様な共刺激分子を安定的に発現している(Banchereau and Steinman,1998;Steinman et al.,2003;Banchereau et al.,2004)。樹状細胞は一次免疫応答においてナイーブαβT細胞を刺激する能力を有することから「プロフェッショナル」な抗原提示細胞とも呼ばれる。(抗原刺激を受けた)メモリーT細胞は活性化閾値が低く、弱い刺激レジメンに対しても応答する。a)末梢組織に存在する抗原プロセシングを行う未成熟樹状細胞と、b)組織からの流入領域リンパ節に移動した、抗原提示及び共刺激を行う成熟樹状細胞という2つの分化状態を区別する樹状細胞の機能的二重性は、樹状細胞の生理学における大きな特徴であり、局所の炎症、感染や組織損傷と密接な関連がある。最後に、適応免疫応答の結果(質及び量)は、主として応答開始の「モード」によって決定される。樹状細胞はリンパ節やパイエル板のT細胞領域内で、ナイーブT細胞に病原体の排除に必要な免疫応答のタイプについての「指示」を与えることが知られている(Banchereau and Steinman,1998;Steinman et al.,2003;Banchereau et al.,2004)。したがって組織の炎症環境は樹状細胞の成熟に直接影響し、樹状細胞が移動することから流入領域リンパ節内でのT細胞の分化も決定する。ナイーブT細胞は、特化したヘルパーT細胞のサブセット(特にIFN−g/TNF−aを産生する1型ヘルパーT(Th1)細胞、IL−4/IL−5/IL−13を産生する2型ヘルパーT(Th2)細胞)、調節性T細胞、細胞障害性T細胞(CTL)などのエフェクターへと分化する。エフェクターT細胞は炎症部位にホーミングし、迅速なエフェクター機能(サイトカイン分泌、感染/腫瘍細胞の溶解/障害)を行う細胞であって、その寿命は短い。一方、メモリーT細胞は一次免疫によって生じる長寿命の細胞であり、リコール抗原に対して優れた免疫応答を示す。
樹状細胞は、「天然の免疫賦活剤(アジュバント)」であり、言い換えれば、防御免疫応答の誘導を行う最も特化した細胞系を構成しているため、ヒトの免疫療法における使用が検討されている(Fong and Engleman,2000;Steinman et al.,2003;Schuler et al.,2003;Figdor et al.,2004)。考えられる応用領域としては、癌治療、病原体に対するワクチン接種(例えばヒト免疫不全ウイルス[HIV]−IやC型肝炎ウイルス)、自己免疫疾患の治療などがあげられる。現在用いられている樹状細胞療法のプロトコールでは以下の段階を行う。
1.患者の血液からの前駆細胞の単離精製(骨髄由来CD34+造血前駆細胞や末梢血のCD14+またはCDHc+細胞)。
2.in vitro細胞培養による樹状細胞の生成。
3.成熟樹状細胞にペプチドを提示させるためのin vitroでの抗原のローディング。
4.ペプチド提示樹状細胞の単回または複数回注入による患者の治療。
本発明は、抗原提示を行うヒトγδT細胞の簡便な単離方法及びin vitro調製方法、ならびに免疫療法における有効である抗原提示細胞(APC)としての当該細胞の使用を開示するものである。ヒトγδT細胞は、その能力と効果において樹状細胞と同様であり、抗原をプロセスして、抗原性ペプチドをαβT細胞に提示し、ナイーブαβT細胞に抗原特異的応答(増殖及び分化)を誘導する。γδT細胞は末梢血中に比較的高頻度で見出され(CD3+T細胞の2〜10%)、多くの簡便な方法によって末梢血から容易に精製することが可能であり、簡単な刺激条件化でのin vitro培養で一日以内に「成熟」状態に達し(MHC−II及び基本的な接着分子及び共刺激分子の発現)、in vitro培養で7日間以上にわたって有効である抗原提示能を維持し、ヘルパーT細胞の強力な一次及び二次応答を誘導する。更に、γδT細胞は、保存して後に使用する目的でin vitro培養で容易に増殖させることができる。
「スキーム」に示したフロー図は、ヒト末梢血のγδT細胞を単離及びin vitro処理することによる、有効である抗原提示ヒトγδT細胞(Vγ2Vδ2+γδT細胞、以下「γδT細胞」と称する)を調製するための本発明の方法を要約したものである。γδT細胞の単離及びin vitro増殖のプロトコールはそれ自体知られているものであるが、刺激と抗原の投与またはペプチドローディングとの特定の組合わせについてはこれまでに述べられていない。開始物質はヒト末梢血であり、これを分画遠心法または免疫吸着法を用いて処理することによって、それぞれ高純度の、増殖または新鮮単離γδT細胞を得る。抗原の投与、またはペプチドの負荷とともに短時間(例えば24時間)の刺激を加えることにより、免疫療法で使用するための有効である抗原提示能を有するγδT細胞が得ることができる。

1.細胞の単離及び生成
γδT細胞
ヒト末梢血単核細胞(PBMC)を標準的プロトコールにしたがって、ヘパリン処理したドナー血液のバフィーコート、または新鮮血からFicoll−Paque遠心分離によって単離した(Brandes et al.,2003)。Miltenyi Biotec社の磁気細胞分離装置を用い、ヒトVγVδ−TCRに対する抗体によってPBMCからγδT細胞をポジティブ選択した。この方法によれば通常、50mlの新鮮血から2〜5×106個の細胞を98〜99%のγδT細胞の純度で得ることができる。
抗原刺激を受けていないナイーブなCD4+またはCD8+αβT細胞(純度98〜99%)を、それぞれVγVδ−TCR、CD1c、CD14、CD16、CD19、CD25、CD45RO、CD56、HLA−DR及びCD4またはCD8に対する特異的抗原を用いたネガティブ磁気細胞分離によってPBMCから単離した後、これらのマーカーについて細胞をネガティブに染色する蛍光励起細胞分離を行った。
ヒトCD4に対する抗体を用いて磁気細胞分離によりPBMCからCD4+αβT細胞をポジティブ選択した。CD4+αβT細胞を、TTまたはPPDを提示する放射線照射(30Gy)自己PBMCによって最初のサイクルで1:100、続くサイクルで1:10の比となるように刺激した後、IL−2含有培地で増殖させた。抗原性に基づく選択及び増殖を3サイクル繰り返した後、CFSEに基づく増殖アッセイ(後述)を行って抗原特異性を確認した。
ネガティブ磁気細胞分離によってPBMCからB細胞を単離した。B細胞はγδT細胞の刺激に直接用いるか(後述)、あるいは標準的なプロトコールにしたがったEBV誘発形質転換によりB細胞系を確立してからγδT細胞の刺激に用いた。
ヒトCD14に対する抗体を用いたポジティブ磁気細胞分離によってPBMCから単核球を分離した。磁気的に捕集された細胞を分離カラムに流す前にカラムを厳密に洗浄することでCD14高発現(CD14high)単核球を高濃度で得た。
IL−4(10ng/ml)及びGM−CSF(25ng/ml)の存在下、10%FCSを含有する培地中でCD14高発現細胞を培養することによって単核球由来の樹状細胞を得た。6〜7日間の培養後、HLA■DR、CD1a、CD14、CD80、CD83、CD86及びCCR7に対する細胞表面の染色によって評価した結果、大半の細胞は未成熟であった。100ng/mlのLPS(Salmonella abortus equi由来)の存在下、更に8時間培養することによって樹状細胞の成熟を誘導した(Langenkamp et al.,2000)。細胞表面に存在する樹状細胞の成熟を示すマーカーであるHLA■DR、CD80、CD83、CD86及びCCR7についてフローサイトメトリー染色を行って樹状細胞の成熟を確認した。
γδT細胞
異種または自己のEBV形質転換B細胞系または初期B細胞によって提示されるイソペンテニルピロリン酸(IPP)50μMを1:10の希釈度で用い、前述したように(単離したばかりの)休眠状態γδT細胞を活性化した(Brandes et al.,2003)。γδT細胞を、8%ヒト血清を加えた培地中でIL−2(20または200IU/ml)の存在下培養した。
ポジティブ選択したαβT細胞を、10μg/mlの抗CD3抗体(OCT3)及び250ng/mlの抗CD28抗体(28.2)、または10ng/mlのフォルボール12−ミリスチン酸13−酢酸(PMA)及び1μg/mlのイオノマイシン、または1μg/mlのPHAでコーティングしたプレート上で活性化し、8%ヒト血清を加えた培地中でIL−2(200IU/ml)の存在下培養した。
細胞をPBS―で洗浄し、1%FCSを加えたPBS−中で4分間、室温で2.5μMのCFSE(モレキュラー・プローブズ社、オレゴン州ユージーン)で標識した。5%FCSを加え、氷で冷やしたPBS−で細胞を繰り返し洗浄することにより、標識を停止した。CFSE標識した細胞は直ちに細胞活性化及び増殖アッセイに使用した。
αβT細胞系に対する抗原提示
血液のγδT細胞を、10〜20μg/mlのTT(ベルナ・バイオテック社、ベルン、スイス)または20μg/mlのPPD(スタテンス・シーラム・インスティテュート社、コペンハーゲン、デンマーク)の存在下、IPPを提示する放射線照射(100Gy)異種EBV−B細胞系CP−EBV(上記参照)によって24〜60時間活性化した。単核球由来樹状細胞をTTまたはPPDと同じ時間培養し、最後の8時間に成熟させた(上記参照)。放射線照射(γδT細胞及び樹状細胞に対してそれぞれ26及び40Gy)及び集中的な洗浄の後、これらの抗原提示細胞を用いてTT及びPPD特異的CD4+αβT細胞のクローンを(特に断らない限り)1:5の比で刺激した。培養中の異なる時点で、フローサイトメトリーで応答性細胞を活性化マーカー(HLA−DR、ICOS及びCD25)、TCRの内在化(細胞表面CD3の消失)及び細胞増殖(CFSEシグナルの低下)について調べた。
γδT細胞及びαβT細胞の刺激後、または単核球由来樹状細胞の刺激後、またはPBMCからの単核球の単離後、これらの潜在的抗原提示細胞を異なる濃度の毒素性ショック症候群毒素(TSST−1)(トキシン・テクノロジー社、フロリダ州サラソタ)で1時間、37℃でパルスした。次いで潜在的抗原提示細胞を12Gy(樹状細胞は40Gy)で放射線照射し、よく洗浄してからCFSE標識したナイーブCD4+αβT細胞と混合(通常1:5の比)した。一般的には96穴丸底プレートに外因性サイトカインを添加しない培地中、8×103個の抗原提示細胞と4×104個のCFSE標識した応答性細胞を入れる。4日後に細胞の増殖をフローサイトメトリーにより調べた。次いでTh細胞分化についてアッセイを行った。このアッセイでは、培養5日目に100IU/mlのIL−2を加えた後、10〜16日間、応答性細胞が増殖を停止して休眠状態に戻るまで細胞を増殖させた(Langenkamp et al.,2000)。21日目にTh細胞の分化を細胞内のサイトカイン産生を測定することによって調べた。細胞を10μg/mlのブレフェルジンA(シグマ・アルドリッチ社)の存在下、PMA/イオノマイシンで6時間刺激した後、2%パラホルムアルデヒドで固定し、2%FCSを加えたPBS中で0.5%サポニンによって透過性を与え、IL−2、IFN−γ、IL−4及びVβ2−TCRに対する抗体で染色してからフローサイトメトリーで分析を行った。
混合白血球応答において、放射線照射したIPP刺激γδT細胞、スーパー抗原で活性化したαβT細胞またはLPSで成熟させた樹状細胞を、CFSE標識した異種ナイーブCD8+αβT細胞と共培養した。一般的には96穴丸底プレートに1穴当たり4×104個のCFSE標識した応答性細胞と、4×104個〜4個の抗原提示細胞を入れる。これにより抗原提示細胞:応答性細胞の比は1:1〜1:10,000となる。6日間の培養後、フローサイトメトリーで細胞の増殖を分析した。14日間のナイーブCD8+αβT細胞との混合白血球応答後に、高濃度(1μM)及び低濃度(0.05μM)のCFSEでそれぞれ標識した異種(真の標的)及び自己(ネガティブコントロール)CD4+T細胞の混合物と12時間共培養を行った後、フローサイトメトリーでCFSEシグナルを調べる細胞溶解アッセイを行ってCD8+エフェクター細胞の生成を調べた。真の標的細胞のカウントの減少は、CD8+エフェクター細胞による抗原特異的な細胞障害を示し、ネガティブコントロール細胞のカウントの減少はCD8+エフェクター細胞による非特異的な細胞障害を示す。
実験全体を通じて使用した培地は、2mMのL−グルタミン、1%非必須アミノ酸、1%ピルビン酸ナトリウム、50μg/mlペニシリン/ストレプトマイシン、5×10−5Mの2−メルカプトエタノール、及び10%FCS(ハイクローン・ラボラトリーズ社、ユタ州ローガン、またはギブコ ビー・アール・エル社)または8%ヒト血清(スイス・レッドクロス社、ベルン、スイス)を補ったRPMI1640培地である。ヒト組換えIL−2はミエローマ発現系を用いて産生されたものを使用した。
細胞の調製
細胞を2%FCS及び0.01%ナトリウム−酸を補った氷冷PBS中で2回洗浄した。この細胞を10mg/mlヒト免疫グロブリンで10分間ブロックした後、多様な細胞タンパク質に対して特異性を有する一次抗体またはアイソタイプが一致したコントロール抗体とともに氷上で20分間インキュベートしてから、洗浄し、非標識一次抗体の場合には蛍光標識した二次試薬とともに更にインキュベートした。最後に洗浄した後、細胞の蛍光をFACSCaliburフローサイトメーター(ベクトン・ディキンソン社、カリフォルニア州サンホセ)によって測定し、記録されたデータをCellQuestProソフトウェア(ベクトン・ディキンソン)によって分析した。
抗体は以下から入手した。マウスモノクローナル抗体である抗CD1a(HI149)、CD3(UCHT1)、CD4(RPA−T4)、CD8(HIT8α)、CD11b(D12)、CD14(MФP9)、CD16(3G8)、CD19(HIB19)、CD20(2H7)、CD25(M−A251)、CD40(5C3)、CD45RA(HI100)、CD45RO(UCHL−1)、CD50(TU41)、CD54(HA58)、CD56(B159)、CDw70(Ki−24)、CD80(L307.4)、CD83(HB15e)、CD86(2331;FUN1)、CD134(L106)、CD205(MG38)、HLA−DR(G46−6)、pan−VγVδ−TCR(11F2)、IL−2(MQ1−17H12)、IL−4(8D4−8)、IFNγ (B27)及びIL-10(No20705A)はBDファーミンゲン社(カリフォルニア州サンディエゴ)より;マウスモノクローナル抗体である抗CD1a(Nal/34−HLK)及びCD19(HD37)はダコ・ダイアグノスティクス社(グラストラップ、スウェーデン)より;マウスモノクローナル抗体である抗TCRVβ2(MPB2D5)はイミュノテック社(マルセーユ、フランス)より;マウスモノクローナル抗体である抗CD138(B−B4)はジアクローン社(ベサンコン、フランス)より;モノクローナル抗体である抗CD102(B−T1)はレインコ・テクノロジーズ社(ミズーリ州セントルイス)より;マウスモノクローナル抗体である抗CD11a(TS1−22)及びCD18(TS1−18)はR.パルディ社(ミラノ、イタリア)より;マウスモノクローナル抗体である抗ICOS(F44)は、R.A.クロツェック社(ベルリン、ドイツ)より;ラットモノクローナル抗体である抗CCR7(3D12)はM.リップ社(ベルリン、ドイツ)より入手した。これらの抗体をフローサイトメトリー分析または細胞の単離に使用した。また以下の二次抗体、接合体、及びコントロール抗体を使用した。RPE標識ヤギ抗マウスIgGはシグマ・アルドリッチ社(ミズーリ州セントルイス)より;RPE標識ロバ抗ラットIgGはジャクソン・イミュノリサーチ・ラボラトリーズ社(ペンシルベニア州ウエストグローブ)より;RPEならびにRPE−Cy5標識ストレプトアビジン(SA)はダコ社より;APC標識SAはBDファーミンゲン社より;マウスコントロールIgGI(MOPC21)はシグマ・アルドリッチ社より;他のアイソタイプコントロール抗体はBDファーミンゲン社より入手した。
刺激された腫瘍抗原提示γδT細胞を調製するため、腫瘍患者(または慢性/再発性感染症患者、下記参照)から50〜150mlの末梢血を採取し、γδT細胞の単離と抗原のロードを上述のようにして行った。また、単離したばかりのγδT細胞を20〜1000IU/mlのIL−2の存在下、Vγ2Vδ2+TCR刺激条件下(前述のIPPによるγδT細胞の活性化の方法を参照)でin vitro培養することによって増殖させ、液体窒素中で保存し、後で腫瘍(またはワクチン、下記参照)抗原提示γδT細胞の調製に使用してもよい。定義された腫瘍/ワクチンタンパク質以外にも、腫瘍細胞の粗(未定義の)抽出物や感染細胞からの抽出物(下記参照)、あるいは生きた血液や組織細胞のトランスフェクションやトランスダクションに通常用いられる組換えRNA/DNA技術によるγδT細胞の処置など、γδT細胞に抗原を与える他の多くの方法が可能である点は重要である。また、細胞表面のMHC分子に直接ロードする腫瘍(またはワクチン、下記参照)ペプチドが既知のものである場合、こうしたペプチドを0.1〜10μg/mlの濃度で1〜10×106細胞/mlの細胞密度の刺激されたγδT細胞に添加し、この細胞を20〜37℃で短時間インキュベートしてから、等張リン酸緩衝溶液で2回洗浄し、直ちに治療に用いることも可能である。1回の投与で使用するγδT細胞の数、γδT細胞の投与の経路及び頻度は、使用される個々の腫瘍(またはワクチン、下記参照)抗原による免疫応答誘導の有効性、及び個々の患者における腫瘍の種類や位置に応じて異なる点は重要である。各プロトコールは樹状細胞による免疫療法で現在用いられているものにしたがう(Fong and Engleman,2000;Steinman et al.,2003;Schuler et al.,2003;Figdor et al.,2004)。すなわち、1〜20×106細胞/0.5〜2mlの投与の後、同量もしくはそれ以下の量の細胞を2週間から2ヶ月の間隔で1回〜6回、追加投与する。

Claims (10)
- 樹状細胞と同等に抗原をプロセスして、抗原性ペプチドをαβT細胞に提示し、ナイーブαβT細胞に抗原特異的増殖及び分化を誘導する抗原提示能を有する抗原提示ヒトγδT細胞の調製方法であって、
ヒト末梢血の単核細胞からγδT細胞を選別し、
前記選別された細胞を、イソペンテニルピロリン酸、4−ヒドロキシ−3−メチル−ブト−2−エニルピロリン酸、(E)−4−ヒドロキシ−3−メチル−ブト−2−エニルピロリン酸(HMB−PP)、エチルピロリン酸(EPP)、ファルネシルピロリン酸(FPP)、ジメチルアリルリン酸(DMAP)、ジメチルアリルピロリン酸(DMAPP)、エチル−アデノシン三リン酸(EPPPA)、ゲラニルピロリン酸(GPP)、ゲラニルゲラニルピロリン酸(GGPP)、イソペンテニル−アデノシン三リン酸(IPPPA)、モノエチルリン酸(MEP)、モノエチルピロリン酸(MEPP)、3−ホルミル−1−ブチル−ピロリン酸(TUBAg1)、X−ピロリン酸(TUBAg2)、3−ホルミル−1−ブチル−ウリジン三リン酸(TUBAg3)、3−ホルミル−1−ブチル−デオキシチミジン三リン酸(TUBAg4)、モノエチルアルキルアミン、アリルピロリン酸、クロトイルピロリン酸、ジメチルアリル−γ−ウリジン三リン酸、クロトイル−γ−ウリジン三リン酸、アリル−γ−ウリジン三リン酸、エチルアミン、イソブチルアミン、sec−ブチルアミン、イソ−アミルアミン、及び窒素含有ビスホスホネートからなる群から少なくとも1つ選択される抗原提示能を誘導するための刺激で処理し、
該細胞によって取込み、提示されるための抗原を細胞に投与する調製方法。 - γδT細胞の選別を、標準的プロトコールにより、ヒトVγVδT細胞受容体に対する抗体を用いた磁気細胞分離によって行う請求項1に記載の方法。
- γδT細胞の選別を、Vγ2Vδ2+T細胞受容体鎖を発現しているγδT細胞の選択的増殖を誘導する、イソペンテニルピロリン酸の存在下、新鮮単離末梢血リンパ球を培養することによって行う請求項1に記載の方法。
- 前記投与される抗原が、タンパク質、タンパク質混合物、または腫瘍もしくは感染細胞からの粗もしくは濃縮抽出物である請求項1に記載の方法。
- 前記投与される抗原が、病原体または腫瘍細胞由来のペプチドである請求項1に記載の方法。
- 抗原が病原体由来のタンパク質であり、該タンパク質の内在的発現を可能とする条件下でそれをコードするDNAまたはRNAとして投与される請求項4に記載の方法。
- 前記DNAまたはRNAが、精製されたDNAまたはRNA、またはDNAまたはRNAを含む導入ベクターである請求項6に記載の方法。
- 請求項1の方法によって調製された樹状細胞と同等に抗原をプロセスして、抗原性ペプチドをαβT細胞に提示し、ナイーブαβT細胞に抗原特異的増殖及び分化を誘導する抗原提示能を有する抗原提示ヒトγδT細胞。
- 免疫療法またはワクチン接種に用いる薬剤の製造における、樹状細胞と同等に抗原をプロセスして、抗原性ペプチドをαβT細胞に提示し、ナイーブαβT細胞に抗原特異的増殖及び分化を誘導する抗原提示能を有する、請求項1に記載の方法によって調製された抗原提示ヒトγδT細胞の使用。
- 新たな腫瘍関連または病原体由来の抗原を特定するための方法であって、
ヒト末梢血の単核細胞からγδT細胞を選別し、
選別された細胞を、イソペンテニルピロリン酸、4−ヒドロキシ−3−メチル−ブト−2−エニルピロリン酸、(E)−4−ヒドロキシ−3−メチル−ブト−2−エニルピロリン酸(HMB−PP)、エチルピロリン酸(EPP)、ファルネシルピロリン酸(FPP)、ジメチルアリルリン酸(DMAP)、ジメチルアリルピロリン酸(DMAPP)、エチル−アデノシン三リン酸(EPPPA)、ゲラニルピロリン酸(GPP)、ゲラニルゲラニルピロリン酸(GGPP)、イソペンテニル−アデノシン三リン酸(IPPPA)、モノエチルリン酸(MEP)、モノエチルピロリン酸(MEPP)、3−ホルミル−1−ブチル−ピロリン酸(TUBAg1)、X−ピロリン酸(TUBAg2)、3−ホルミル−1−ブチル−ウリジン三リン酸(TUBAg3)、3−ホルミル−1−ブチル−デオキシチミジン三リン酸(TUBAg4)、モノエチルアルキルアミン、アリルピロリン酸、クロトイルピロリン酸、ジメチルアリル−γ−ウリジン三リン酸、クロトイル−γ−ウリジン三リン酸、アリル−γ−ウリジン三リン酸、エチルアミン、イソブチルアミン、sec−ブチルアミン、イソ−アミルアミン、及び窒素含有ビスホスホネートからなる群から少なくとも1つ選択される抗原提示能を誘導するための刺激で処理し、
タンパク質混合物の画分、あるいは腫瘍及び感染細胞からの粗または濃縮抽出物、あるいは腫瘍及び感染性物質から得られたRNAまたはDNAライブラリーを投与し、
自己ナイーブαβT細胞のin vitroでの活性化について試験し、
高い抗原性を示したタンパク質試料を選別する方法。
