忠実度:正常的な序列に対する正確性が通常的なPCRを制限する。例えば、DNAの増幅のために通常的に利用される耐熱性重合酵素であるTaqは、PCRの間に約1×10−4個のエラー/塩基対のエラー率を表す。これは、400個の塩基対から構成されたDNA序列のPCR増幅は、20サイクルの間にPCR生成物の全ての分子のうち、約40,000個のエラーを任意的に導入するということを意味する。

好熱性(すなわち、耐熱性)DNA重合酵素を利用した標準温度循環式のPCRを行うために、通常的に下記のステップを実行する:1)チューブにPCR用の緩衝溶液、dNTP混合物、プライマー対、DNA重合酵素、及び二重脱イオン水を含むカクテルを準備するステップと、2)前記チューブに増幅されるDNAを添加するステップと、3)前記チューブを温度循環器(例えば、Perkin−ElmerTM9600または9700 PCR Thermal Cycler)の温度ブロックに配置するステップと、4)約25回ないし30回のサイクルの間に反復される特定の反応条件(例えば、約1ないし2分間、約90℃以上まで加熱して二重ストランドDNAの熱変性を行う期間、2分間徐々に約50ないし65℃まで冷却してアニーリングする期間、及び数分間約70ないし75℃まで加熱してプライマー延長と呼ばれる重合を行う期間)で温度循環器をプログラミングするステップ。前記方法を実行すれば、開始物質の約2n倍の増幅を生成し、前記で、nは、行われる増幅サイクルの総数である。
Nano−PCRTM方法及び装置は、通常的な方法によって賦課される色々な限界点を克服することによって、PCRの検出及び増幅能力を飛躍的に拡張させうる。表1は、現在のPCRの一般的な性能パラメータをNano−PCRTMと比較する。
核酸に対する制御された張力の適用は、二重ストランドDNA(dsDNA)の熱変性に対する代案だけでなく、PCR過程の各ステップを正確に制御できる独特の能力を提供する。Nano−PCRTMは、DNA/RNA分子及び/または重合酵素に対する機械的応力、水力学的応力、または電磁気的応力のような正確に制御される力の効果を利用してDNAまたはRNAを増幅する新たな接近方法を導入する。
プライマー延長ステップの間に鋳型DNAに張力が適用される場合、重合酵素は、“逆方向”へ誘導され、重合酵素のエキソヌクレアーゼ活性が優勢でありうる。原子水準及び単一重合酵素/エキソヌクレアーゼステップの順序に対して、経時的にその過程は確率的であると理解されるであろう。しかし、色々なステップの時間規模に対する平均から考慮すれば、重合酵素は、適用される張力が所定の温度及び溶液条件で理論的に予測されうるしきい値より大きい時、持続的なエキソヌクレアーゼ活性を表すと見なされる。
増幅効率性が式N1=N2(l+Y)nによって定義され、N1は、生成物コピーの数であり、N2は、鋳型オリゴヌクレオチドコピーの数であり、nは、サイクルの数、Yは、効率性の場合、80%より高い効率性が達成される。特定条件下で、85%、90%、95%、96%、97%、98%、ひいては99%より高い増幅効率が達成されうる。そのような増幅効率性はまた、オリゴヌクレオチドのGC含量が55%、60%、65%、70%、ひいては75%より高い場合でも収得されうる。オリゴヌクレオチド断片のGC含量が50%、55%、60%、65%または70%より高い場合で、50%より高い増幅効率性が達成されうる。60%、70%、80%、90%、95%、97%、ひいては99%より高い効率性が収得されうる。通常的なPCRで、このようなGC−高含量領域は、水酸化ナトリウム、塩化TMA、シュウ酸TMA、硝酸TMA、TMA硫酸水素、塩化アンモニウム、ベンジルジメチルヘキサデシル塩化アンモニウム、臭化HTA、シュウ酸HTA、ベタイン一水化物、DMSO、及びホルムアミドを含むが、これに限定されない多様な変性剤の添加と共に増幅され、その序列が決定される。Nano−PCRTMの特定の具体例では、そのようなPCR添加剤の不在下に本明細書に記載された方法を利用した効率的な増幅が観察される。
通常的なPCR方法は、一般的に温度の正確な制御及び循環に依存する。また、通常的なPCR方法は、多様な変性剤及び退屈な最適化のような追加的な因子を必要とするであろう。しかし、本明細書に記載されたように、増幅を推進するための張力循環の利用は、PCR過程に対して温度循環単独によって許容されるよりはるかに高い正確度及び制御を可能にする。また、本明細書に記載された方法は、選択された重合酵素が作用できる温度範囲及び所定の張力でプライマー/鋳型結合の融点温度のような因子によってのみ限定され、広範囲な温度条件下で作用できる。
Nano−PCRTM方法は、DNA変性及び/またはDNA重合酵素活性の正確な制御を提供できる非温度駆動過程であって、DNAストランドに機械的な張力を直接適用する多様な方法を利用して行われうる。二重ストランドオリゴヌクレオチドに張力を適用する色々な相異な方法がある。例えば、DNAストランドは、移動可能な要素、個別的に制御可能な要素、または操作されうる粒子にアレイとして固定されうきる。
一例であって、図2Aないし図2Cに示したように、対向するコーティングされた表面に固定された核酸を利用する方法が行われうる:コーティングされた表面は、各表面に対して同一かまたは相異なる第1複合体化分子(例えば、ストレプトアビジン)201,207を二つの基板表面203,209に付着させることによって製造される。コーティングされた基板表面は、適した距離ほど離隔されて相互間に逆に配列される。一つの両末端が第1複合体化分子を認識して結合できる第1複合体化分子(例えば、ビオチン)に相補的な複合体化分子を含むものである二重ストランド核酸205がコーティングされた表面に固定される。コーティングされた表面間の距離を増大させるか、または表面のうち一つまたは両者を側面移動させることによって固定された核酸の末端に力が適用されうる。例えば、基板のうち一つまたは両者は、圧電素子のような移動可能な要素であるか、または移動可能な要素を含みうる。dsDNAを融解させるのに十分な張力(例えば、室温で65pNを超える)が核酸に適用され、固定されたストランド及び遊離したストランドを生成しうる。固定されたストランド及び遊離したストランドは、何れも適したプライマー及び重合酵素を利用して複製されうる。望ましくは、固定されたストランドのみが複製されるように遊離したストランドは、フラッシュングされて除去され、選択的に回収されうる。対向する表面の位置は、固定された鋳型ストランドに適用される張力の量を調整するために複製の間に制御されうる。サイクルは、所望するほど反復されうる。
DNAに張力を直接的に適用するさらに他の方法は、DNAが固定された粒子を操作するために光学的なトウィーザーまたは磁気的トウィーザーまたはその他のトラップを利用しうる。光学的なトウィーザーは、光学強度の勾配によって生成される力で粒子を捉える。光の電場によって両極化された誘電体粒子は、勾配を通じて最も明るい地点に引かれる。これと対照的に、反射しかつ吸収する低誘電体粒子は、放射圧によって最も暗い地点に押される。三次元トラップを形成するほど十分に強い選択的に生成された力が、適した形状の波面を有するレーザビームを高い開口数値のレンズによる厳格なフォーカスでもたらすことによって収得されうる。図3Aは、レーザビーム303のフォーカスにトラップされたビード301の間に延長されたDNAストランドを示す。図3Bに示したように、光学的トウィーザーのアレイ307を利用して複数の粒子を操作することが可能である。購買可能な光学的トウィーザーアレイは、Arryx,Inc.によって生産されるアレイを含む。光学的トウィーザーのアレイのさらに他の具現は、E.R.Dufresne and D.G.Grier,Rev.Sci.Instr.69:1974(1998)、及び米国特許第6,055,106号(2000)を参照する。光学的トウィーザーアレイは、約103、104、105、106、またはそれ以上の対の光学的トウィーザーまたは磁気的トウィーザーを含みうる。
Nano−PCRTM方法は、制御された流体流れで水力学的な応力によって張力をDNAに適用することを利用して行われうる。この方式を利用する方法は、ベンチトップ装置であるか、または代案的に携帯用装置に含まれうるような移動可能なサイズに縮少しうる微細流体装置で行われうる。制御された流体流れで水力学的な応力によってDNAに張力を適用することを利用するNano−PCRTM方法は、制御された流体の流速を提供する任意の配列を利用して行われうる。
Nano−PCRTM方法で電場及び磁場の利用を通じてDNAストランドに力を適用することが可能である。これが達成されうる多様な方式がある。例えば、電場は、微細流体装置で流体流れを駆動させることによってDNAに間接的に力を適用するために利用されうる。前述したように、流体流れは、DNA、例えば、表面及び/または粒子に固定されたDNA、または表面に固定されたDNA重合酵素に結合されたDNAに水力学的な応力を適用するために利用されうる。電気泳動的な力が直接DNAストランドに力を適用しうる。
Nano−PCRTM方法は、病原菌及び生物学的な武器検出用キット及びシステムに採用されうる。そのような病原菌の例は、次のものを含むが、これに限定されていない:アデノ−関連ウイルス(Adeno−AssociatedVirus:AAV)、アデノウイルス、サイトメガローウイルス(Cytomegalovirus:CMV)、エプスタイン−バールウイルス、エンテロウイルス、A型肝炎ウイルス(HepatitisA Virus:HAV)、B型肝炎ウイルス(Hepatitis B Virus:HBV)、C型肝炎ウイルス(Hepatitis C Virus:HCV)、人間ヘルペスウイルスタイプ6(HumanHerpes Virus Type 6:HHV−6)、人間免疫欠乏症ウイルスタイプ1(Human Immunodeficiency Virus Type1:HIV−1)、人間免疫欠乏症ウイルスタイプ2(HumanImmunodeficiency Virus Type2:HIV−2)、ヘルペスシンプレックスウイルスタイプ1及びタイプ2(Herpes Simplex VirusType 1andType2:HSV−1及びHSV−2)、人間T−細胞リンパ営養性ウイルスタイプI及びタイプII(Human T−CeIl LymphotropicVirus TypeI and TypeII:HTLV−I及びHTLV−II)、マイコバクテリア結核、マイコプラズマ、パーボウイルスB−19、呼吸器細胞融合ウイルス(RespiratorySynctitial Virus:RSV)及び豚内因性レトロウイルス(Porcine Endogenous Retrovirus:PERV)。Nano−PCRTM方法は、この方法が提供できる敏感度、正確度及び堅固性の強化のために、全ての環境で全ての病原菌の検出のために利用されうる。
本明細書に記載されたような非温度−駆動PCRを行うために利用される複数の相異なる装置の種類及び構成がある。そのような装置のうち一つは、装置上にある微細流体チャンネル内の流速が制御可能で可変的な微細流体装置である。望ましい具体例で、装置は、反応チャンバを有し、前記反応チャンバは、チャンネル、チャンネルの配列、または密閉された空間でありうる。反応チャンバは、一般的に核酸を維持する手段及びその内部に維持された前記核酸に応力または張力を適用する手段を含む。したがって、本明細書に記載の方法のうち一つを行うように設計された配列は、その内部に、核酸に結合できる粒子またはそれらの相補的な複合体化分子を獲得できる複合体化分子、複合体化分子を有する表面、移動可能な要素、流体流れを配向し、流速勾配を形成するチャンネル、ポンプ、弁及び膜を有するチャンネル及び密閉された空間の組合わせを含むと予想しうる。チャンバは、例えば、光学的微細操作器が利用される場合、光学的に透明な窓を含みうる。装置は、携帯用ユニットに含まれうる微細流体装置として製造されうる。所望の場合、Nano−PCRTMは、1μl未満、例えば、約50−1000nL、望ましくは、約100−500nLの溶液容量で行われうる。
標準方法によって一対のストレプトアビジン−コーティングされた表面を製造した(Sabanayagam,Smith,andCantor.“Oligonucleotide immobilization on micropatterned streptavidin surfaces.”NucleicAcids Res.2000,Vol.28,No.8 ppi−iv)。ビオチン化されたdsDNA(一ストランドの両末端でビオチン化された)を前記表面に添加し、表面の間に前記dsDNAを固定化させた(JeffreyM.Rothenberg and Meir Wilchek.p−Diazobenzoyl−biocytin:a new biotinylating reagentfor DNA Nucleic Acids Research Volume 16 Number 14 1988)。表面に適用される鋳型の濃度を調整して、DNA分子の表面密度は調節されうる。室温で、コーティングされた表面の間の距離を増大させて約65pNより大きい張力をdsDNAに適用した。これは、dsDNAを変性させ、増幅のための標的ssDNAのみを残した。結合されたDNAを束縛されたビオチン基を含むプライマーと接触させ、表面の間の距離を縮小させて30pNないし60pNの力が固定されたssDNAに適用させた。プライマーを標的DNAにアニーリングさせ、DNA重合酵素及びヌクレオチドを結果物である複合体に添加した。プライマー延長は、適用される張力を30pN未満までさらに減少させることによって開始された。プライマー延長が完了すれば、表面の間の距離を増大させて結果物である二重ストランドに65pNより大きい力を適用させた。この力の適用は、複製ヌクレオチドストランドをその鋳型から分離(変性)させた。束縛されたビオチンモイエティを含む複製ストランドを光活性化させ、ストレプトアビジン−コーティングされた対向する表面に結合させた。標的ヌクレオチドに対する所望の程度の増幅を収得するまで前記ステップを反復した。

二重ストランドDNA複合体を周囲温度で適した媒質に込められたポリスチレンビードの間に固定化させた(“OverstretchingB−DNA:the Elastic Response of Individual Double Stranded and Single Stranded DNAMolecules”by Steven B.Smith,Yujia Cui,and Carlos Bustamante Science(1996)vol.271,pp795−799)。約65pNの伸張力を光学的トウィーザーの利用を通じてDNAに適用した(Rouzina,L,and V.A.Bloomfield.2001b.Force−inducedmeltingof the DNA double helix 2.Effect of solution conditions.Biophys.J.80:894−900)。その力は、DNA変性を招いた。プライマーを媒質に添加し、伸張力を60pN未満に減少させた。これは、プライマーを変性された単一ストランドDNAにアニーリングさせた。DNA重合酵素及びヌクレオチドを媒質に添加して伸張力を0ないし30pNに減少させて高度に正確な複製を開始させた。複製が完了すれば、約65pNの伸張力をそれぞれの二重ストランドDNA分子に適用して、単一ストランドDNA分子を放出させた。これは、操作器のアレイを利用することによってその規模が拡張されうる。例えば、Arryx,Inc.によって製造された光学的トラップアレイのようなアレイが利用されうる。
Claims (54)
- 核酸を増幅する方法であって、
(a)単一ストランド核酸の一つ又はそれ以上の鋳型ストランドを、前記一つ又はそれ以上の鋳型ストランドの一部に相補的な一つ又はそれ以上のオリゴヌクレオチドプライマーと接触させるステップと、
(b)前記一つ又はそれ以上のプライマーのうちの少なくとも1つのプライマーを前記プライマーが相補的である前記一つ又はそれ以上の鋳型ストランドの一部にアニーリングさせるステップと、
(c)前記一つ又はそれ以上の鋳型ストランドを核酸重合酵素及び少なくとも4種類の異なるヌクレオシド三リン酸と接触させるステップと、
(d)前記核酸重合酵素によって前記少なくとも一つのアニーリングされたプライマーを延長し、これにより、前記一つ又はそれ以上の鋳型ストランドに結合された一つ又はそれ以上の延長生成物を形成するステップと、
(e)前記一つ又はそれ以上の延長生成物を前記一つ又はそれ以上の鋳型ストランドから分離させるステップと、
(f)前記核酸を増幅するために前記ステップ(a)、(b)、(c)、(d)、及び(e)を反復するステップと、を有し、
前記ステップ(a)〜(e)の各サイクルにおける前記(e)の前記一つ又はそれ以上の延長生成物の少なくとも一つは、ステップ(a)〜(e)の次のサイクルにおける鋳型ストランドとして使用され、
前記ステップ(b)または(d)のうちの少なくとも一つは前記一つ又はそれ以上の鋳型ストランドに張力を適用するステップを含むことを特徴とする方法。 - 更に、前記ステップ(e)は前記一つ又はそれ以上の鋳型ストランドに張力を適用するステップを含むことを特徴とする請求項1に記載の方法。

- 前記張力を適用する、前記張力を減少させる、および前記張力を制御するステップのうちの少なくとも1つは、前記張力が適用される前記核酸分子を一つ又はそれ以上の粒子に結合させるステップ及び光学的な操作または磁気的な操作によって前記一つ又はそれ以上の粒子の運動を制御するステップを含むことを特徴とする請求項28に記載の方法。
- 前記張力を適用する、前記張力を減少させる、および前記張力を制御するステップのうちの少なくとも一つは、前記張力が適用される前記核酸分子に対する流体の流れを調整するステップを含むことを特徴とする請求項28に記載の方法。
- 前記核酸分子のうちの少なくとも一つは、前記基板に結合された重合酵素によって結合され、流体が前記基板上を流れていることを特徴とする請求項28に記載の方法。
- 前記核酸分子のうちの少なくとも一つは、基板に結合されることを特徴とする請求項28に記載の方法。
- 前記核酸分子のうちの少なくとも一つは、流体の流れの速度勾配で、又は逆伝搬性の流体の流れによって伸びることを特徴とする請求項28に記載の方法。
- 前記ステップ(d)は、複製することが難しい前記一つ又はそれ以上の鋳型ストランドの序列が複製される時期に対応する時点で前記張力を増大させるステップを含むことを特徴とする請求項28に記載の方法。
- 前記の全体プロセスは、周囲の温度で行われることを特徴とする請求項28に記載の方法。
- 前記の全体プロセスは、20℃より高く、60℃より低い温度で行われることを特徴とする請求項28に記載の方法。
- 前記の全体プロセスは、室温で行われ、前記ステップ(b)で、前記張力は、65pNより大きく、前記ステップ(c)で、前記張力は、0pN〜60pNの範囲に減少し、前記ステップ(d)で、前記張力は、0pN〜45pNの範囲に調整されることを特徴とする請求項28に記載の方法。
- 重合酵素連鎖反応によって核酸を増幅する方法であって、
(a)二重ストランド核酸を変性させて単一ストランド鋳型ストランドを生成するステップと、
(b)前記鋳型ストランドにプライマーをアニーリングさせるステップと、
(c)鋳型ストランドで駆動された重合酵素によって前記プライマーを延長して二重ストランド核酸分子を生成するステップと、
(d)前記ステップ(a)〜(c)を反復して前記二重ストランド核酸分子を増幅するステップと、を有し、
前記ステップ(a)〜(c)の各サイクルにおける前記(c)で生成した前記二重ストランド核酸分子の少なくとも一つは、ステップ(a)〜(c)の次のサイクルのステップ(a)で変性され、
前記ステップ(b)と(c)のうちの少なくとも一つは、前記核酸に張力を適用するステップを含むことを特徴とする方法。 - 前記張力を適用するステップは、機械的張力、水力学的応力、または電場力またはそれらの組み合わせを適用するステップを含むことを特徴とする請求項39に記載の方法。
- 前記ステップ(a)〜(d)は、80℃以下、30℃以下および周囲温度から選択された温度で行われることを特徴とする請求項39に記載の方法。
- 前記ステップ(a)で適用される張力は、65pNまたはそれより大きい、または、
前記ステップ(b)で適用される張力は、(i)50pNまたはそれより小さい、(ii)30pN〜50pN、(iii)35pN〜45pN、または、
前記ステップ(c)で適用される張力は、45pNまたはそれより小さいことを特徴とする請求項39に記載の方法。 - 核酸増幅を行う方法であって、
(a)二重ストランド核酸を変性させて単一ストランドの鋳型ストランドを生成するステップと、
(b)前記鋳型ストランドにプライマーをアニーリングさせるステップと、
(c)鋳型ストランドで駆動された重合酵素によって前記プライマーを延長して二重ストランド核酸分子を生成するステップと、
(d)前記ステップ(a)〜(c)を反復して前記二重ストランド核酸分子を増幅するステップと、を有し、
前記ステップ(c)は等温であり、前記核酸に機械的張力または水力学的張力を適用するステップを含み、
前記ステップ(a)〜(c)の各サイクルにおけるステップ(c)で生成された前記二重ストランド核酸分子の少なくとも1つは、前記ステップ(a)〜(c)の次のサイクルのステップ(a)で変性されることを特徴とする方法。 - 核酸増幅を行う方法であって、
(a)二重ストランド核酸を変性させて単一ストランドの鋳型ストランドを生成するステップと、
(b)前記鋳型ストランドにプライマーをアニーリングさせるステップと、
(c)鋳型ストランドで駆動された重合酵素によって前記プライマーを延長して二重ストランド核酸分子を生成するステップと、
(d)前記ステップ(a)〜(c)を反復して前記二重ストランド核酸分子を増幅するステップと、を有し、
前記ステップ(c)は前記核酸に変更可能な張力を適用するステップを有し、
前記変更可能な張力は、前記核酸の重合酵素のプルーフリーディング・エキソヌクレアーゼ活性を調製することを特徴とする方法。 - 核酸増幅を行う方法であって、
(a)二重ストランド核酸を変性させて単一ストランドの鋳型ストランドを生成するステップと、
(b)前記鋳型ストランドにプライマーをアニーリングさせるステップと、
(c)鋳型ストランドで駆動された重合酵素によって前記プライマーを延長して二重ストランド核酸分子を生成するステップと、
(d)前記ステップ(a)〜(c)を反復して前記二重ストランド核酸分子を増幅するステップと、を有し、
前記ステップ(a)〜(c)の各サイクルにおけるステップ(c)で生成された前記二重ストランド核酸分子の少なくとも1つは、前記ステップ(a)〜(c)の次のサイクルのステップ(a)で変性され、
前記ステップ(b)と(c)のうちの少なくとも1つは、前記核酸に張力を適用するステップを含み、
前記鋳型ストランドの複製の数は、ステップ(a)〜(c)がn回繰り返された後で、2n倍に増加し、ここで、nは1より大きい整数であることを特徴とする方法。 - 前記ステップ(a)は前記核酸に張力を適用するステップを含むことを特徴とする請求項39に記載の方法。
- 核酸ストランドへのプライマーのアニーリングの緊縮調節を制御する方法であって、
複数の核酸増幅サイクルにおいて、核酸ストランドにプライマーをアニーリングするステップであって、前記核酸ストランドへの前記プライマーのアニーリングの緊縮調節を制御するために張力が前記核酸ストランドまたは前記プライマーに調整可能に適用されるステップを含み、
核酸重合酵素によって前記プライマーを延長させることによって形成された延長生成物が、核酸増幅の次のサイクルにおいて核酸鋳型ストランドとして使用されることを特徴とする方法。 - 核酸重合酵素によってプライマーを延長するときの誤り率を低下する方法であって、
複数の核酸増幅サイクルにおいて、核酸重合酵素によって核酸鋳型にアニーリングされたプライマーを延長して二重ストランドの核酸分子を生成するステップであって、前記核酸重合酵素によって前記プライマーを延長するときの誤り率を低下させるために張力が前記核酸鋳型または前記プライマーに調整可能に適応されるステップを含み、
核酸重合酵素によって前記プライマーを延長させることによって形成された延長生成物が、核酸増幅の次のサイクルにおいて核酸鋳型ストランドとして使用されることを特徴とする方法。 - 核酸重合酵素によってGCに富む鋳型でプライマーを延長するときの誤り率を低下する方法であって、
複数の核酸増幅サイクルにおいて、核酸重合酵素によってGCに富む核酸鋳型にアニーリングされたプライマーを延長して二重ストランドの核酸分子を生成するステップであって、前記GCに富む鋳型で前記プライマーを延長するときの誤り率を低下させるために張力が前記GCに富む核酸鋳型または前記プライマーに調整可能に適応されるステップを含み、
核酸重合酵素によって前記プライマーを延長させることによって形成された延長生成物が、核酸増幅の次のサイクルにおいて核酸鋳型ストランドとして使用されることを特徴とする方法。 - 核酸増幅の忠実度を改善する方法であって、
(a)二重ストランド核酸を変性させて単一ストランドの鋳型ストランドを生成するステップと、
(b)前記鋳型ストランドにプライマーをアニーリングさせるステップと、
(c)鋳型ストランドで駆動された重合酵素によって前記プライマーを延長して二重ストランド核酸分子を生成するステップと、
(d)前記ステップ(a)〜(c)を反復して前記二重ストランド核酸分子を増幅するステップと、を有し、
前記ステップ(a)〜(c)の各サイクルにおけるステップ(c)で生成された前記二重ストランド核酸分子の少なくとも1つは、前記ステップ(a)〜(c)の次のサイクルのステップ(a)で変性され、
前記ステップ(a)〜(c)の少なくとも1つは、前記核酸に張力を調整可能に適応するステップを含むことを特徴とする方法。 - 核酸序列に張力を適用するための装置であって、
1つまたはそれ以上の流体チャンネルと、
前記1つまたはそれ以上の流体チャンネル内で核酸分子を維持する手段と、
前記維持された核酸分子に可変的に制御された量の張力を適用する手段と、
核酸プライマーと、ヌクレオシド三リン酸と、核酸増幅酵素とを含む試薬を、反応させ、格納し、又は導入する1以上のチャンバーと、
を有することを特徴とする装置。 - 核酸分子に張力を適用するための微細流体装置であって、
a)基板と、
b)前記基板内に配置された流れチャンネルと、
c)核酸試料を前記流れチャンネル中に導入することができる前記流れチャンネルと流体連結する入口と、
d)前記核酸分子に張力を適用する手段と、
e)前記微細流体デバイス内で前記核酸分子を維持する手段と、
を有し、
核酸プライマーと、ヌクレオシド三リン酸と、核酸増幅酵素とを含む試薬を、反応させ、格納し、又は導入する1以上のチャンバーを有するか、又は前記微細流体デバイスは等温で操作されるように構成されていることを特徴とする微細流体装置。 - 核酸分子の序列を判定する方法であって、
(a)標的核酸序列と、核酸重合酵素と、前記標的核酸序列と相補的なプライマーと、1以上のヌクレオチドと、を含む試料を提供するステップと、
(b)前記プライマーを該プライマーと相補的な標的核酸序列にアニーリングさせるステップと、
(c)前記プライマーを延長して延長生成物を形成するステップと、
(d)さらなる延長生成物を形成するために前記ステップ(b)〜(c)を繰り返すステップと、
(e)前記標的核酸序列の序列を判定するステップと、
を有し、
前記ステップ(b)〜(c)のサイクルのうちの少なくとも1回は、前記標的核酸序列と前記プライマー延長生成物とのうちの少なくとも一方に張力を適用する工程を含み、
核酸重合酵素によって前記プライマーを延長させることによって形成された延長生成物が、核酸増幅の次のサイクルにおいて核酸鋳型ストランドとして使用されることを特徴とする方法。 - 核酸分子の序列を判定する方法であって、
(a)4種類の試料を提供するステップであって、当該試料のそれぞれは、一重鎖又は二重鎖の標的核酸序列と、核酸重合酵素と、前記標的序列と相補的なプライマーと、少なくとも4つの異なるヌクレオシド三リン酸と、それぞれについてddATP、ddCTP、ddGTP、及びddTTPの中から選択されたジデオキシヌクレオシド三リン酸と、を含むステップと、
(b)前記二重鎖の標的核酸序列を、一重鎖の標的核酸序列へと変性させるステップと、
(c)前記プライマーを該プライマーと相補的な一重鎖の標的核酸序列にハイブリダイズさせるステップと、
(d)前記プライマーを延長して延長生成物を形成するステップと、
(e)さらなる延長生成物を形成するために前記ステップ(b)〜(d)を繰り返すステップと、
(f)前記標的核酸序列の序列を判定するステップと、
を有し、
前記ステップ(b)〜(d)のうちの少なくとも1回は、標的核酸序列の1以上のストランドに張力を適用する工程を含み、
核酸重合酵素によって前記プライマーを延長させることによって形成された延長生成物が、核酸増幅の次のサイクルにおいて核酸鋳型ストランドとして使用されることを特徴とする方法。
