Description
[材料及び方法]
(マウス)
変異マウス(MyD88−、TRIF−、TLR2−、TLR4−、TLR7−、TLR9欠損マウス)を、129/Ola×C57/BL6又はC57/BL6をバックグランドとし、文献(Immunity 9: 143-150, 1999; Immunity 11: 443-451, 1999; Nature 408: 740-745, 2000; Nat. Immunol. 3: 196-200, 2002; Science 301: 640-643, 2003)記載の通りに産出した。同年齢の野生型マウスと変異マウスを実験に使用した。インビボの研究には、P. falciparum培養物から精製したHZ1500μgと合成HZ(βヘマチン)とを、野生型マウス、MyD88−/−マウス又はTLR9−/−マウスの腹腔内に注入した(J. Immunol. 172: 3101-3110, 2004)。サイトカインELISA用に、1、2、4及び6時間後に尾から血清を収集した。
合成CpGオリゴデオキシヌクレオチド(ODN)D35は、北海道システムサイエンス株式会社より購入、合成した。Salmonella minnesota Re-595由来リポポリサッカリド(LPS)、塩化ヘミン(hemin chloride)及びクロロキン(CQ)はSigma-Aldrichより購入した。DNase (Dnase- I) はInvitrogenから購入した。
HZ(不溶性クリスタロイド構造)は、P. falciparum (3D7株)で感染した赤血球から精製した(Infect. Immun. 70: 3939-3943, 2002; Proc. Natl. Acad. Sci. U.S.A 93: 11865-11870, 1996; Proc. Natl. Acad. Sci. U.S.A 88: 325-329, 1991)。要約すると、赤血球をサポニンで溶解した後、原虫を超音波処理し、2%のドデシル硫酸ナトリウム(SDS)で7〜8回洗浄した。次に、ペレットをプロテイナーゼK(2mg/ml)と共に、37℃で一晩インキュベートした。ペレットはその後、2%のSDS内で3回洗浄し、6Mの尿素内で室温で3時間、シェーカー上でインキュベートした。HZペレットを2%のSDSで洗浄した後、蒸留水で3〜5回洗浄し、蒸留水に再懸濁し、使用前に再び超音波処理した。いくつかの実験において、文献(J. Immunol. 167: 2602-2607, 2001)記載の通りに、HZは、95℃で15分間の熱失活を行うか又は、100U/mlのDNase-Iで1時間処理を行った。DNase-I処理は、P.falciparumの粗抽出物から完全にゲノムDNAを除去することによって行った(図5a)。
ンドトキシンの量は、使用したHZ1nmole当たり0.001EU未満であった。
HZ又は合成HZの濃度は、ヘムポリマーを、20mM水酸化ナトリウム/2%SDSの溶液内で、室温で、2時間脱重合して定量し、O.D.は400nmであった(Proc. Natl. Acad. Sci. U.S.A 93: 11865-11870, 1996)。ヘムのモラー吸光係数は400nmで1×105であり、25μgのP. falciparum HZは、ヘム含有量29nmoleと同等である。
脾臓細胞の単一細胞懸濁液(single cell suspension)(5×105細胞/ウェル)を、10%のFCSを添加した完全RPMI 1640培地内で48時間培養した。Flt3リガンド誘導骨髄由来DC(FL−DC)(1×105細胞/ウェル)は、骨髄細胞とFlt3リガンド(100ng/ml;Pepro Tech 社製)とを、10%のFCSを含むDMEM培地内で8〜9日間培養して産出した。細胞を、指定の刺激によって刺激し、サイトカインELISA用に、上澄みを回収した。
マウスのTNF−α、IL−12p40、MCP−1、IL−6(R & D Systems)及びIFNα(PBL Bio. Lab 社製)を、上澄み又は血清のいずれかで、製造者の指示に従ってELISAで測定した。
刺激した細胞の細胞表面の分子発現を、文献(J. Exp. Med. 196: 269-274, 2002)記載の通り測定した。要約すると、刺激した細胞を、冷却したPBS内で洗浄し、固定し、坑CD16抗体の存在下で、FITC標識抗体、PE標識抗体、Cy−Chrome標識抗体及びAPC標識抗体で、室温で30分染色した。染色した細胞を洗浄し、PBS/0.1%BSA/0.1%NaN3内に再懸濁し、FACSCalliberで分析した後、CellQuest software(BD社製)で分析した。全ての抗体は、BDより入手した。
統計的な有意差は、Student’s t-testを用いて分析した。P<0.05は有意とみなした。
(P. falciparum由来の精製HZの、MyD88依存性経路を通してのマウスの脾臓細胞及び樹状細胞の活性化)
P. falciparum由来の精製HZが、マウス免疫システムを活性化するかどうかを調べるために、脾臓細胞及びDC(マウス骨髄樹状細胞)を、インビトロで精製HZで刺激し、培養上澄み中の炎症誘導性サイトカインの産出をELISAによって測定した。FL−DC(Fit3リガンド誘導骨髄由来DC)は、HZに応答して、TNFα、IL−12p40、単球走化性因子−1(MCP−1)及びIL−6を、用量依存的に大量に産出し、その量はCpG ODNとほぼ同様の値だった(図1a)。HZ誘導性の自然免疫活性化における、TLRの役割を調べるために、多数のTLRが介するサイトカイン誘導に必須の分子アダプターであるMyD88が欠損しているマウスを用いた(Nat. Rev. Immunol. 4: 499-511, 2004)。MyD88−/−マウス由来のFL−DCでは、HZ刺激により、TNFα、IL12p40、MCP−1及びIL−6の産出が著しく障害された(図1b)。
HZ誘導性の自然免疫の活性化が、MyD88にのみ依存的であることを確認するために、MyD88非依存性経路に必須のアダプター分子であるTRIF(Toll/IL−1レセプター(TIR)領域含有アダプター)が欠損しているマウスを用いた(Nat. Rev. Immunol. 4: 499-511, 2004)。MyD88−/−マウスとは対照的に、TRIF−/−マウスのFL−DCは、HZに応答し、TNFα及びIL−12p40(p<0.05、TRIF−/−vs培地)を産出し、野生型マウス(p>0.05)の値とほぼ同じであった(図2)。LPS誘導性のTNFα及びIL−12p40は、TRIF−/−マウスのFL−DCで障害され、P. falciparum培養物から精製した多量のHZに、LPSが混入していないことを示唆した。これらのデータは、HZがMyD88を通してマウスにおける炎症誘発性応答を活性化することを示し、MyD88依存性TLRの1つがHZの認識に関与していることを示した。
TLR2−/−マウス、TLR4−/−マウス、TLR7−/−マウス及びTLR9−/−マウスから得た脾臓細胞及びDCを用いて、HZ誘導性の自然免疫活性化が障害されているか、又は変更されているかを調べるために、さらに実験を行った。HZは、野生型マウス、TLR2−/−マウス、TLR4−/−マウス、TLR7−/−マウスにおいて、FL−DCを刺激し、CD11c+、B220+plasmacytoidDCサブセット及びCD11c+、B220−骨髄DCサブセットの両方で、CD40及びCD86をアップレギュレートした(図3a)。対照的に、TLR9−/−マウス由来のFL−DCの両サブセットでは、HZに応答してCD40及びCD86をアップレギュレートしなかった(図3a)。
HZ活性化が、インビボで、TLR9に介され、MyD88に依存的であることを確認するために、P. falciparum精製HZを、野生型マウス及びMyD88−/−マウス又はTLR9−/−マウスの腹腔内に注入し、血清サイトカインの産出をモニターした。HZの注入により、野生型マウスのMCP−1及びIL−6の血清量が著しく上昇し、その値は1〜4時間に最大となり、6時間以内に減少した(図4a)。対照的に、MyD88−/−マウス及びTLR−/−マウスでは、この上昇は、完全に阻害されていた。6時間後、サイトカインの量は、野生型マウスで減少した。これらのデータは、インビボ及びインビトロの両方で、HZ誘導性の炎症誘導性応答が、TLR9及びMyD88によって介されていることを明確に示している。
坑マラリア製剤クロロキン(CQ)は、エンドソーム/リソソームの成熟、マラリアヘモゾインのクリスタル形成(crystal formation)を阻害するほか、マラリア感染中の炎症誘導性応答を阻害することが報告されているが、CQの坑マラリア効果の正確なメカニズムは、未だに論争中である(Int. J. Parasitol. 32: 1645-1653, 2002; Life Sci. 74: 1957-1972, 2004)。最近の論拠は、CQは、TLR9仲介性自然免疫活性化も阻害することを示唆している(J. Immunol. 160: 1122-1131, 1998)。HZ誘導性の炎症誘導性応答に対するCQの効果を調べるために、CQの存在下で、FL―DCをHZで刺激した。HZ誘導性のTNFα及びIL−12p40の産出は、CQにより減少した(図5b)。
