本発明は、病原微生物によって分泌される細胞障害性因子及び細胞障害性因子の阻害剤及び壊死及びアポトーシスの両方によって細胞死を調節する際のそれらのその使用に関する。本発明は、又、アポトーシスを調節する際に有用な細胞障害性因子を生成、単離及び同定する方法、及び細胞死を調節する際に有用な実質的に純粋な細胞障害性因子を組み込む組成物に関する。本発明は、又、アポトーシスに関連した状態を治療する方法に関する。より詳細には、本発明は、癌細胞のアポトーシスを誘発する方法における実質的に純粋な細胞障害性因子の使用及び感染又はその他の病原体誘発性状態を治療する方法における細胞障害性因子の阻害剤の使用に関する。
感染症は多くの環境因子の産物であると思われる。任意の感染症の原因は原因となる病原体である。病原体は、典型的には病原微生物、例えば病原菌である。防衛機構を克服し、疾病を生じる病原微生物の程度及び能力は、その病原性に関連する。病原微生物は細胞障害性因子を発現することが知られ、病原体に、宿主免疫システムから自らを守らせ、身体から病原体を取り除くことから食細胞(例えば、マクロファージ及び肥満細胞)を防がせる。病原微生物が生き残る場合、微生物は宿主組織に侵入及び増殖でき、重篤な病徴を生じる。病原菌は、種々の衰弱又は死病、例えば結核、コレラ、百日咳、ペスト等の根本的原因として同定されている。そのような重篤の感染を治療するために、薬剤、例えば抗生物質を投与して、病原菌がもはや宿主免疫システムに対して自らを守ることができないように病原菌を殺し、細胞障害性因子を無力にする。しかしながら、病原菌は、一般に抗生物質耐性になり、そのような微生物による感染の蔓延を防止するために、改良剤が必要である。
本発明は、壊死又はアポトーシスによって細胞死を刺激する細胞障害性因子に関する。一の局面において、実質的に純粋な細胞障害性因子は特徴付けられ、単離される。実質的に純粋な細胞障害性因子は、病原微生物にさらされた増殖培地のカラムクロマトグラフィーのフラクションによって得られる。好ましくは、前記細胞障害性因子の生成及び分泌は、哺乳類のタンパク質の存在下で病原体の増殖中に刺激される。
(定義)
本明細書に記載した目的で、用語“細胞障害性因子”は病原微生物によって分泌される因子を意味し、それは壊死又はアポトーシスによって細胞死を刺激する。用語“ATP-依存性”は、用語“細胞障害性因子”を修飾するために使用される場合、アデノシン5'-三リン酸塩(ATP)の存在下で細胞死を誘発するために作用する細胞障害性因子を意味する。用語“ATP-非依存性は、用語“細胞障害性因子”を修飾するために使用される場合、ATPの非存在下で細胞死を誘発するために作用する細胞障害性因子を意味する。
本発明は、病原微生物によって分泌され、壊死又はアポトーシスによって細胞死を刺激する細胞障害性(病原性)因子を与える。病原微生物がヒト又は動物組織に侵入する場合、食細胞は宿主免疫システムにおける防御の第一線である。典型的には、食細胞は身体に侵入する外来性病原体を見つけ出し、破壊する。しかしながら、細菌病原体によって分泌される細胞障害性因子は食細胞の細胞死を刺激することができる。従って、食細胞はその防御免疫機能を遂行できないように妨げられる。
本発明の一の局面において、本発明の細胞障害性因子は、細菌及び原生動物を含む多くの異なる病原微生物によって分泌される。細胞障害生因子を与えるのに適した病原菌の例としては、これに限定されないが、緑膿菌、セパシア菌(Burkholderia cepacia)、コレラ菌、及びウシ結核菌が挙げられる。さらに、細胞障害性因子はレーシュマニア(Leishmania amazonensis)及びマレー糸状虫(Brugia malayi)のような病原体によって分泌される。
本発明の別の局面において、細胞障害性因子の生成及び分泌は、哺乳類のタンパク質の存在下で病原微生物の増殖中に刺激される。例えば、緑膿菌、ウシ結核菌及びセパシア菌のような病原微生物による細胞障害性因子の分泌は、κ-カゼイン、ウシ血清アルブミン、オボアルブミン又はα2-マクログロブリンのような哺乳類のタンパク質の存在によって刺激される。病原微生物が哺乳類の宿主環境の指標として特定の哺乳類のタンパク質の存在を感知し、それにより宿主の防御に対抗し、それを打ち破るために細胞障害剤の分泌機構が開始することが示唆されるが、本明細書においてはそれに頼らない。
本発明の別の局面において、実質的に純粋なATP-非依存性細胞障害性因子は、分泌する微生物の増殖培地のカラムクロマトグラフィー分別により得られる。好ましくは、細菌細胞は分別前に増殖培地から取り除かれる。これは増殖培地の初期遠心分離及び続く濾過により達成することができる。適したフィルターは、典型的には約0.5μm以下の細孔サイズであり、好ましくは約0.2μm以下である。しかしながら、病原体除去の他の方法は当業者によく知られている。
本発明のさらなる局面において、分別した増殖培地はATP-非依存性細胞障害性因子の存在を決定するために試験される。細胞死の程度は、Zaborinaら、Infection and Immunity, 67, 5231-5242 (1999)及びZaborinaら、Microbiology, 146, 2521-2530 (2000)に記載されるように細胞内酵素乳酸デヒドロゲナーゼ(LDH)の放出によって測定することができる。
別の局面において、本発明は、ATP-非依存性アポトーシス誘発細胞障害性を示すことを特徴とする細胞障害性因子を提供する。本発明者らは、緑膿菌及びセパシア菌のQSETフラクションにより2つのタンパク質、アズリン及びシトクロムc551が富化されることを見出した。これらの2つのタンパク質の同定は、SDS-PAGEにおけるその分離及びそれらのN-末端アミノ酸配列の同定に基づく。その一方、ウシ結核菌のQSFTフラクションのSDS-PAGE分析は、ウシ結核菌を増殖するために使用される7H9培地の成分である濃い65kDaバンドのウシ血清アルブミン(BSA)を示し、同様に45kDaよりも大きい分子質量のいくつかのバンドを示すが、シトクロムc551又はアズリンの特徴的なバンドではない(実施例9を参照のこと)。
本発明は、癌細胞のアポトーシス細胞死を誘発するためのATP-非依存性細胞障害性因子の使用方法を提供する。アズリン及びシトクロムC551のようなATP-非依存性細胞障害性因子は、細胞死の異常不全に関連する状態を治療するために使用できる。癌細胞がアポトーシスを受けないことはよく知られている。本発明の一の局面に従って、癌細胞のアポトーシスを誘発するのに十分な量の細胞障害性因子の分泌を刺激する細胞障害性因子又は活性剤を投与することは、インビボで腫瘍サイズを小さくし、腫瘍の増殖を抑える際に役立つであろう。例えば、アズリン及びシトクロムC551を公知の抗メラノーマ制癌剤[5-(3,3'-N,N'-ジメチルトリアゼン-1-イル)-イミダゾール-4-カルボキシアミド](DTIC)と比較した検査は、アズリン及びシトクロムC551の混合物がヌードマウスにおけるインビボの腫瘍退行を促進する強力な無毒性組成物を提供することを示す。
本発明の別の局面において、細胞障害性因子の特徴は、例えば感染症の細胞死を阻害する新しい物質を同定するのに有用である。例えば、ATP-利用細胞障害性因子の分泌又は活性の阻害、又はATPの産生は、病原体によって細胞障害性活性を低下させ、又は除去できる。
細胞障害性因子を含む医薬組成物は、任意の従来の方法、例えば従来の混合、溶解、粒状化、糖衣錠形成、乳化、カプセル化、取りこみ(entrapping)、又は凍結乾燥プロセスで製造できる。実質的に純粋な細胞障害性因子又はその他の薬剤は、当該技術においてよく知られる医薬的に許容される担体と容易に組み合わせることができる。このような担体は、製剤が錠剤、ピル、糖衣錠、カプセル、液体、ゲル、シロップ、スラリー、懸濁液等として処方されることを可能にする。また、適切な賦形剤も、例えばフィラー及びセルロース製剤を含むことができる。その他の賦形剤として、例えば香料添加剤、着色剤、粘着除去剤(detackifiers)、増粘剤、及びその他の許容される添加剤、アジュバント、又は結合剤を挙げることができる。
また、細胞障害性因子の同定及び特徴付けは、細胞障害性因子の分泌の刺激方法の開発をもたらし得る。病原体は、哺乳類のタンパク質の存在下で多量の細胞障害性因子を分泌することを示した。この原理は人体において修正されて、所望の細胞障害性因子の産生を刺激し、望まれていない細胞障害性因子の産生を阻害する新しい方法を提供する。このような方法は細胞のアポトーシスを阻害又は刺激するのに有用である。細胞障害性因子の理解、及びその特徴付け及び開発は、また細胞障害性因子の活性又は分泌を調節するのに適した活性剤又は化合物の薬剤開発及びスクリーニングを可能にする。細胞における細胞障害性因子の分泌に関連する分泌機構の理解は、細胞障害性因子の有用な阻害剤又は刺激剤の設計を開発及び同定する新しい道をさらに提供する。哺乳類のタンパク質の受容体の存在の有無及び同定は、また病原微生物と非病原微生物とを区別する手段として使用でき、疾病の治療に特異性を与えることができる。分泌機構の成分、及び細胞障害性因子自体は、ワクチンとして使用できる。
細胞障害性因子は、また化学的に修飾され、又は遺伝学的に変更されて、ATP-利用酵素又はレドックス活性を欠いているが毒性を維持する変異体を産生することができる。遺伝子の変異及び/又は切断は、組成物を変化させ、また機能活性を示す細胞障害性剤を産生することができる。特に、高い効力及び低い抗原性を有する切断された誘導体は、元の細胞障害性因子から産生できる。このような修正又は変更された細胞障害性因子、及びこのような細胞障害性剤は、また本発明の範囲に包含される。
(実施例1.哺乳類のタンパク質による細胞障害性因子の分泌の刺激)
α2-マクログロブリン(1mg/ml)を加えて、及び加えないでプロテオースペプトン-酵母抽出物(PPY)ブロス中で、セパシア菌の臨床及び環境単離物(それぞれ5個)を増殖した。34℃で10時間、振盪機で増殖した後、各培養物由来の増殖培地の一部を遠心分離し、その上澄みを0.22μmのミリポアフィルターにより濾過し、全細胞及び細片を除去した。次いで濾過した上澄みを、Melnikov A.ら、Mol. Microbiol.m 36: 1481-1493 (2000)に記載されるアデニル酸キナーゼ活性について試験した。アデニル酸キナーゼは、末端ホスフェートを[γ-32P]ATPからADPを生み出すAMPに移動させる。次いで、この反応生成物を薄層クロマトグラフィーにより検出した。アデニル酸キナーゼの分泌は、セパシア菌細胞がPPYブロスで増殖される場合に最小となった。しかしながら、臨床単離物からの分泌は(環境単離物についてはないが)、α2-マクログロブリンの存在下で刺激された。
セパシア菌の臨床株(菌株38-収集番号95828、D.G. Allison、マンチェスター科学技術大学、マンチェスター、イギリス)を34℃でTBブロス(水1リットル当たり10gのバクト(Bacto)トリプトン、3gのバクト(Bacto)牛肉エキス)中振盪機で1.3のOD550nmに増殖した。次いで、増殖培地遠心分離し、この上澄みを0.22μmのミリポアフィルターにより濾過して全細胞及び細片を除去した。マクロファージ細胞をJ774細胞株から単離し、Zaborina O.ら、Infect. Immun. 67: 5231-5241 (1999)に記載されるようにRPMI培地1640(GIBRO-BRL、グランドアイランド、ニューヨーク)中で増殖した。濾過した増殖培地をヒドロキシアパタイトカラム、ATP-アガロースカラム、及びQ-セファロースカラムに順に加えた。ヒドロキシアパタイトカラム(HAFT)を貫流したフラクションをATP-アガロースカラム(AAFT)で分別した。次いで、AAFTフラクションをQ-セファロースカラム(QSFT)で分別した。
セパシア菌増殖培地の上澄み(SUP)及びカラムクロマトグラフィーフラクション(HAFT、AAFT及びQSFT)は実施例2と同様にした。マクロファージ単離物は実施例2と同様にした。マクロファージの細胞死の程度は、実施例2と同様にLDHの放出によって決定し、それを図2に示す。QSFTフラクションのみがマクロファージに関して高いATP-非依存性細胞障害性を示す。
緑膿菌をLブロス中12時間37℃で1.2のOD550nmに増殖した。次いで、増殖培地を遠心分離し、この上澄みを0.22μmのフィルターにより濾過した。上澄み(SUP)及びカラムクロマトグラフィーフラクション(HAFT、AAFT及びQSFT)を実施例2と同様に収集した。マクロファージ単離物を実施例2と同様にした。上澄み又は貫流フラクションの1つからの2μgのタンパク質を200μlのRPMI培地中1×105のマクロファージに加え、この混合物を一晩インキュベートした。SUP又はHAFT、AAFT又はQSFTフラクションと共に一晩インキュベーションすることにより処置された又は処置されないマクロファージにおけるアポトーシスの誘導を、Zaborina O.ら、Microbiology 146: 2521-2530 (2000)に記載されるようにApoAlertミトコンドリア膜センサーキット(クローンテックラボラトリーズ社、パロアルト、カリフォルニア、米国)を用いて共焦点顕微鏡法により測定した。
肥満細胞を、Melnikov A.ら、Mol. Microviol. 36: 1481-1493 (2000)に記載される方法により単離した。セパシア菌のフラクション増殖培地を実施例2と同様に調製した。セパシア菌細胞障害性因子による肥満細胞におけるアポトーシスの誘導を、実施例4に記載されるように、共焦点顕微鏡法を用いて決定した。
マクロファージ単離物を実施例2と同様にした。セパシア菌及びウシ結核菌の細胞障害性因子によるマクロファージにおけるアポトーシスの誘導を、実施例4の方法を用いて決定した。マクロファージのアポトーシスの誘導は、それらがセパシア菌及びウシ結核菌のQSFTフラクションにより処置された場合に観測された。
マクロファージの単離を実施例2と同様にした。マクロファージを、実施例2に記載される方法を用いてセパシア菌のQSFTフラクションにより一晩処置する。マクロファージサイトゾル抽出物の調製及びカスパーゼアッセイは、Zaborina O.ら、Microbiology 146: 2521-2530 (2000)に記載される通りであった。
分別したセパシア菌増殖培地を、実施例2に記載される方法を用いて得た。ウシ結核菌BCGを、2%グリセロール、0.02%TWEEN(登録商標)80及びADC(アルブミン/デキストロース/シトレート)(ディフコラボラトリーズ、メリーランド、米国から入手可能)により補充されたMiddlebrook 7H9ブロス(ディフコラボラトリーズ、メリーランド、米国)中で増殖した。細菌を、収集前に32℃で数日間、振盪機で増殖した。分別したウシ結核菌増殖培地を、実施例2に記載される方法を用いて得た。マクロファージの単離は実施例2と同様にした。未処置の、又はSUP若しくはHAFT、AAFT又はQSFTフラクションの一晩のインキュベーションにより処置されたマクロファージにおけるアポトーシスの誘導を、ApoAlert DNA断片化キット(クローンテックラボラトリーズ社、パロアルト、カリフォルニア、米国)でアポトーシス誘導核DNA断片化を検出することによる共焦点顕微鏡法を用いて測定した。このアッセイを末端デオキシヌクレオチジルトランスフェラーゼ(Tdt)-媒介dUTPニック-末端ラベリング(TUNEL)に基づき、ここでTdtは、アポトーシスを起こす細胞において断片化DNAの遊離3'-ヒドロキシル末端でのフルオレセイン-dUTPの組み込みを触媒する。断片化核DNAにおけるフルオレセイン-dUTPの組み込みは、共焦点顕微鏡法により検出される緑色の蛍光を生じる。
SDS-PAGE分離は、緑膿菌、セパシア菌及びウシ結核菌の上澄み及びAAFT、HAFT及びQSFTフラクションに存在するタンパク質を示した。ムコイド緑膿菌の菌株8821由来のQSFT培地フラクションは、2つのバンド、N-末端分析によりアズリンに対応する18kDaとシトクロムc551に対応する9kDaの存在を示した。セパシア菌のQSFTフラクションは、75kDa、20kDa及び8kDaの3つの主なバンドの存在を示した。エドマン分解により決定される20kDaバンドの10個のアミノ酸のN-末端アミノ酸配列(AHHSVDIQGN)は、緑膿菌のアズリンのN-末端の10個のアミノ酸配列と80%の配列相同性を示し、8kDaバンドの10個のアミノ酸のN-末端アミノ酸配列(EDPEVLFKNK)は、緑膿菌のシトクロムc551のそれと100%の一致を示した。従って、緑膿菌及びセパシア菌の両方の高い細胞障害活性を有するQSFTフラクションは、レドックスタンパク質のアズリン及びシトクロムc551タイプを豊富に含むことを示す。その一方、ウシ結核菌のQSFTフラクションはウシ血清アルブミン(BSA)の薄い65kDaバンドを示し、それはウシ結核菌を増殖するために使用される7H9培地の成分であり、又同様に45kDaよりも大きい分子量のいくつかのバンドも示すが、8kDa又は22kDaのシトクロムc551又はアズリンタイプのタンパク質ではない。
精製したアズリン及びシトクロムc551(シグマケミカルズ、セントルイス、米国)を、実施例2と同様に調製したマクロファージに加え、この混合物を2時間インキュベートした。アズリン及びシトクロムc551の濃度は、実施例4と同様にした。数値はμgのタンパク質を表す。マクロファージの細胞死を、実施例2の方法を用いて細胞内酵素乳酸デヒドロゲナーゼ(LDH)の放出により測定した。アズリン及びシトクロムc551の両方はマクロファージ細胞死を引き起こす。アズリン及びシトクロムc551の組み合わせは、より大規模なマクロファージの細胞死を引き起こす。緩衝剤コントロール(緩衝剤)は右側に示す(図4)。
マクロファージの単離は実施例2と同様にした。このマクロファージをアズリン/シトクロムc551(50/25μg)で4及び6時間処置し、次いで実施例4と同様にApoAlertミトコンドリア膜センサーキットを用いて共焦点顕微鏡法により試験し、アポトーシスの程度を決定した。マクロファージは、アズリン/シトクロムc551混合物の存在下でインキュベーションの期間を長くすると、アポトーシスのレベルは増加した。処置しない(リン酸緩衝生理食塩水で6時間処置した)コントロールマクロファージはアポトーシスを示さなかった。
マクロファージの単離は実施例2と同様にした。マクロファージを、精製したアズリン/シトクロムc551混合物(50/25μg)、又はラビットにおいて調製した抗-アズリン及び抗-シトクロムc551抗体の混合物の存在下及び非存在下でセパシア菌又はウシ結核菌のQSFTフラクションで処置した。抗体を1:1の比で混合し、この混合した抗体(1、2、3、又は4mg)をマクロファージの処置で使用した。
H460肺癌、PA-1卵巣癌、NCF乳癌、HT-29大腸癌及びHT-1080白血病細胞株をthe American Type Culture Collection(マナッサス、ヴァージニア、米国)から得た。MDD7及びMN1乳癌細胞株をAndrei Gudkov, Ph.D., Cleveland Clinic Foundation(クリーヴランド、オハイオ、米国)から得た。UISO-BCA-9乳癌及びUSIO-MEL-1、MEL-2、MEL-6及びMEL-29メラノーマ細胞株をRauth, SらのIn vitro Cellular and Developmental Biology, 30a(2): 79-84 (1994)及びRauth, SらのAnticancer Research, 14(6): 2457-2463 (1994)に記載されるように発生させ、維持した。約1×105個の各細胞タイプを、セパシア菌のQSFTフラクション(5μgのタンパク質)又はアズリン/シトクロムc551混合物(50/25μg)の存在下、0.15mm厚のdTC3皿(Bioptech、バトラー、ペンシルヴェニア、米国)で一晩培養した。続いて、細胞を、実施例4と同様に共焦点顕微鏡により試験して、アポトーシスの程度を決定した。セパシア菌のQSFTフラクションとアズリン/シトクロムC551混合物の両方は、一晩のインキュベーション後にH460肺癌、HT-29大腸癌、HT-1080白血病、PA-1卵巣癌、MDD7、NCF及びNM1乳癌、及びUSIO-MEL-1、MEL-2、MEL-6及びMEL-29メラノーマ細胞における広範なアポトーシスを誘導した。いずれの場合にも、細胞障害性因子(リン酸緩衝生理食塩水を加えた)で処置されない細胞は広範なアポトーシスを示さなかった。
USIO-Mel-6メラノーマ細胞をRauth, SらのAnticancer Research, 14(6): 2457-2463 (1994)に記載されるように調製した。ウシ結核菌のQSFTフラクションを実施例8と同様に調製した。ウシ結核菌のQSFTフラクション(5μgのタンパク質)で処置したメラノーマ細胞及び未処置のコントロール細胞を12時間インキュベートした。アポトーシスの誘導は、実施例8と同様にTUNELアッセイを用いて測定し、アポトーシス誘導核DNA断片化を検出した。ウシ結核菌のQSFTフラクションで処置したメラノーマ細胞は赤色の細胞質バックグラウンドに黄緑色の核を示し、核DNA断片化を示す。断片化は未処置のメラノーマ細胞では少ないか、又は観測されなかった。
約106個のUSIO-Mel-2細胞をヌードマウス(フレデリック癌研究開発センター、フレデリック、メリーランド、米国から入手可能)に皮下に注入した。約3週間後小さな腫瘍が発生した。その後、このマウスは、公知の抗-メラノーマ薬、DTIC[5-(3,3'-N,N-ジメチルトリアゼン-1-イル)-イミダゾール-4-カルボキシアミド](7.5μg)(AhlmaisらのCancer 63: 224-7 (1989))の腹腔内注入を週1回又は高(150μgのアズリン/75μgのシトクロムc551)、低(10μgのアズリン/5μgのシトクロムc551)用量のアズリン/シトクロムc551混合物の腹腔内注入を週3回又はコントロール(シトレート緩衝剤)を週4回受けた。腫瘍の体積は、コントロール、DTIC処置、及び高並びに低用量アズリン/シトクロムc551処置マウスで、時間をあけて決定した。
約106個のUSIO-Mel-6細胞を3匹のヌードマウス(フレデリック癌研究開発センター、フレデリック、メリーランド、米国)の皮下に注入した。小さな腫瘍は約3週間後発生した。その後、1匹のマウスに、リン酸緩衝生理食塩水を腹腔内に注入し(コントロール)、別の1匹のマウスに、ウシ結核菌のQSFTフラクション(5μgのタンパク質)を注入し、さらに別の1匹のマウスに、ウシ結核菌のQSFTフラクション(5μgのタンパク質)とアズリン(50μg)の混合物を注入した。ウシ結核菌のQSFTフラクションは実施例8と同様に調製した。コントロール、ウシ結核菌のQSFTフラクション処置及びウシ結核菌のQSFTフラクション/アズリン処置マウスの腫瘍のサイズ(腫瘍体積)は、30日間に渡って決定した。これらのデータを図8に示した。両処置マウスでは、コントロールマウスと比較して腫瘍増殖が減少することを示した。
Claims (10)
- 癌細胞の細胞死増加用医薬組成物を調製するための、アズリンの使用であって、前記アズリンが緑膿菌又はセパシア菌から得られたものである、使用。
- 癌細胞がメラノーマ細胞、白血病細胞、乳癌細胞、卵巣癌細胞、肺癌細胞、間葉癌細胞、大腸癌細胞、及びアエロ消化管癌細胞からなる群より選ばれる、請求項1に記載の使用。
- 癌細胞がメラノーマ細胞である、請求項2に記載の使用。
- 前記化合物が細胞アポトーシスを増加させる、請求項1に記載の使用。
- 前記医薬組成物がアズリン及びシトクロムC551を含む、請求項1に記載の使用。
- 癌細胞増殖阻害用医薬組成物を調製するための、アズリンの使用であって、前記アズリンが緑膿菌又はセパシア菌から得られたものである、使用。
- 前記癌細胞がメラノーマ細胞、白血病細胞、乳癌細胞、卵巣癌細胞、肺癌細胞、間葉癌細胞、大腸癌細胞、及びアエロ消化管癌細胞からなる群より選ばれる、請求項6に記載の使用。
- 前記細胞がメラノーマ細胞である、請求項7に記載の使用。
- さらにシトクロムC551を含む請求項6に記載の使用。
- アズリン、及び薬剤的に許容される賦形剤を含む医薬組成物であって、癌細胞の細胞死調節のために使用され、前記アズリンが緑膿菌又はセパシア菌から得られたものである、医薬組成物。
