Description
本出願は、2007年3月14日出願の米国仮出願第60/894,786に基づく優先権を主張するものであり、その全てを参照により本明細書に組み込まれるものである。
CD26の構造は、3つの領域−細胞外領域(その細胞外領域は、membrane-proximalグリコシル化ドメイン、システインに富むドメインおよびDPPIV酵素活性を含む260アミノ酸C末ドメインを含む)、22残基の疎水性膜貫通領域および6アミノ酸の細胞質領域から成り立っている。我々の以前の報告では、ヒトCD26の20アミノ酸の柔軟幹領域から始まる細胞membrane-proximalグリコシル化ドメインを認識する、マウス抗CD26mAb14D10がG1/S細胞周期停止を含む直接的な抗腫瘍効果およびそれに付随して癌細胞のECMへの癒着を防ぐことを示した。しかし、システインに富むドメインがみられる、5F8と名付けられた別のマウス抗CD26mAbは、この生物学的活性を欠いている(非特許文献18)。
ヒト悪性中皮種(MM)は明らかな従来治療法に対して高い悪性腫瘍抵抗性があるので、MMにおける新規な標的および新規な治療戦略が早急に開発されることを必要とされている(非特許文献4および19)。
(1)細胞外マトリックスへのCD26の結合を阻害する物質を含有する、悪性中皮腫、肺癌、腎臓癌、肝臓癌又はその他のCD26の発現を伴う悪性腫瘍の治療用の医薬組成物。
(2)悪性中皮腫、肺癌、腎臓癌、肝臓癌又はその他のCD26の発現を伴う悪性腫瘍のための成長阻害剤である、(1)記載の医薬組成物。
(3)前記物質がCD26cDNAを標的とするsiRNAである、(1)記載の医薬組成物。
(4)前記物質が抗CD26抗体である、(1)記載の医薬組成物。
(5)前記抗CD26抗体がモノクローナル抗体である、(4)記載の医薬組成物。
(6)前記抗CD26抗体が14D10である、(4)記載の医薬組成物。
(7)前記抗CD26抗体がヒト化抗体である、(4)記載の医薬組成物。
(8)前記ヒト化抗体が14D10由来の可変領域を含む、(7)記載の医薬組成物。
(9)前記ヒト化抗体がAmerican Type Culture Collection(ATCC)の受託番号PTA−7695を有し、s604069.YST−pABMC148(x411)と命名された株により生産される、(7)記載の医薬組成物。
寄託は、2006年6月30日、特許手続上の微生物の寄託の国際的承認に関するブダペスト条約の規定の下でATCCに寄託され、受託番号PTA−7695と指定された。寄託された試料は、菌株表示がs604069.YST−pABMC148 (x411)を有する、「ヒトCD26cDNAに対するヒト化モノクローナル抗体の重鎖および軽鎖のcDNAが挿入されたプラスミドを有するDH5α大腸菌」とされた。
(10)前記抗体に特異的なエフェクター細胞を更に含む、(4)記載の医薬組成物。
(11)抗CD26抗体および該抗体に特異的なエフェクター細胞を含有する、悪性中皮腫、肺癌、腎臓癌、肝臓癌又はその他のCD26の発現を伴う悪性腫瘍の治療用キット。

(18)抗CD26抗体を含有する、悪性中皮腫、肺癌、腎臓癌、肝臓癌又はその他のCD26の発現を伴う悪性腫瘍の診断剤。

試薬および抗体
抗CD26マウスmAb(IgGl)14D10、5F8、および抗CD45 RAマウスmAb(IgGl)2H4は、以前に記載した(非特許文献20および21)ように、当研究室で開発されたものであり、後者を対照として使用した。正常なヒトIgG(Sigma Aldrich, St. Louis, ミズーリ州)も対照として使用した。ヒト化抗CD26mAbは、14D10コード配列から構築した。簡単に説明すると、前記エピトープに対する高い結合親和力を有する幾つかのFabクローンを選択した。それらは、in vitro増殖アッセイを用いて、生物学的有効性について試験した。それらのうちの一つをヒト化抗CD26mAb(humAb)として構築するために選択した。PKBα/Akt、CDK2、CDK4、CDK6、cyclinE、およびβアクチンに対するマウスmAbをCell Signaling Technology Inc.(Beverly、マサチューセッツ州)から入手し、そしてp27kip1、p21cipl/wafl、cyclinDl、および活性caspase 3に対するマウスmAbをBD PharMingen(レキシントン、ケンタッキー州)から入手した。抗ヒトIgG、Fcγフラグメント特異的ヤギF(ab’)2フラグメントおよび抗マウスIgG、Fcγフラグメント特異的ヤギF(ab’)2フラグメントをJackson ImmunoResearch (West Grove, ペンシルベニア州)から入手した。
JMN細胞は、Brenda Gerwin博士(Laboratory of Human Carcinogenesis, National Institute of Health, Bethesda, メリーランド州)から謹呈された。NCI−H2452細胞および293T細胞は、American Type Culture Collection(Rockville, メリーランド州)から入手した。JMN細胞株およびNCI−H2452細胞株は、悪性中皮腫の患者から採取した。10%の熱非動化牛胎児血清(FBS) 、ペニシリン(100 units/ml)およびストレプトマイシン(100μg/ml)(Life Technologies Inc.、Gaithersburg, メリーランド州)、またはG418(500μg/ml)(Sigma-Aldrich, St. Louis, ミズーリ州)を含有するRPMIメディウム(Life Technologies Inc.、Grand Island, ニューヨーク州)で、すべての細胞を生育した。FuGENE6試薬(Roche Diagnostics, Indianapolis, インディアナ州)を使用してpEB6ベクター(非特許文献22)にサブクローニングした完全長のCD26を293T細胞にトランスフェクトした。pEB6ベクターはY.Miwa博士(筑波大学、茨城、日本)から謹呈された。
総体積100μL(1ウェルあたり5x103個の細胞)の培養液中で、細胞を単独またはhumAb(0.1、1.0もしくは10μg/ml)もしくは2H4(0.1、1.0もしくは10μg/ml)の存在下で培養した。37℃で24時間のインキュベーションの後、2−(2−メトキシ−4−ニトロフェニル)−3−(4−ニトロフェニル)−5−(2,4−ジスルホフェニル)−2H−テトラゾリウム(生化学工業、東京、日本)を各ウェルに加えた。更に2時間のインキュベーションの後、電子伝達体である1−メトキシ−5−メチルフェナジニウムの存在下での生体還元の後に水溶性ホルマザン染料をマイクロプレートリーダー(BioRad、Hercules、カリフォルニア州)を使用し450nmで測定した。全てのサンプルを3つの重複する系で試験した。測定値は、3つの重複するウェルの平均値で表示し、その平均標準誤差(SE)は15以内であった。
免疫組織化学において、12人の患者の外科標本(7名からのMM、3名からの反応性中皮腫細胞、および2名からの類腺腫瘍)を評価した。それぞれに対して、癌とその隣接している非新生物組織の両方を含む10%ホルマリン固定パラフィン包埋標本を作製した。パラフィン包埋組織を、それぞれキシレンおよびエタノールを使用して脱ろうおよび再水和した。スライドを脱パラフィンし、次に、抗原回復のために10分間10mMクエン酸塩緩衝液(pH6.0)中でマイクロ波プロセッサにて加熱した。3%(vol/vol)BSAでブロッキング後、スライドを一次抗体(抗CD26 mAb)と共に4℃で終夜インキュベートし、PBSで洗浄し、二次抗体をビオチンでラベル化し、30分間適用した。ストレプトアビジン−LSA増幅法を30分間行った後にペルオキシダーゼ/ジアミノベンジジン基質/クロマゲンで処理した。スライドをヘマトキシリンで対比染色した。2人の異なる病理学者が、得られた結果の妥当性を点検した。全てのヒト標本は、慶応義塾大学(東京、日本)病理学教室から入手するとともに、全ての患者から治験審査委員会(IRB)の形式に沿ったインフォームドコンセントを得た。
内因性CD26を欠損させるため、CD26 cDNA(アクセッション番号NM_001935)を標的とするsiRNA−オリゴをタカラバイオのデザインサイト(http://www.takara-bio.co.jp/RNAi.htm) に従って作製した(センス鎖: 5'-GAAAGGUGUCAGUACUAUU TT-3’(配列番号30)およびアンチセンス鎖:3'-TT CUUUCCACAGUCAUGAUAA-5'(配列番号31))。そしてヒトCas−Lを標的としたスクランブル対照siRNA−オリゴ(センス鎖: 5'-UAAUUAGGGUCGGGUAAAC TT-3’(配列番号32)とアンチセンス鎖: 3'-TTAUUAAUCCCAGCCCAUUUG-5'(配列番号33))を対象として用いた。TransIT-TKO(登録商標)トランスフェクション試薬(Mirus Bio Corporation、Madison、ウィスコンシン州)をメーカーのプロトコルに従って用いてCD26 siRNAオリゴ(siCD26)をトランスフェクションした。
全細胞溶解液の調製と細胞分画は、他に記載されているように行った(非特許文献23)。タンパク質サンプルをSDS-PAGEで泳動し、ポリビニリデンジフルオリド膜(Immobilon-P; Millipore、Bedford、マサチューセッツ州)に転写した。対応するmAbで特異的抗原をプローブし、次いで、HRP結合の二次Ig(Amersham Pharmacia Biotech, Piscataway, ニュージャージー州)でプローブした。ウエスタンブロット産物を増強化学発光法(ECL法)(NEN, Boston, マサチューセッツ州)により可視化した。
腫瘍細胞のエフェクター細胞依存性溶解に対するmAbをCalcein−AM放出アッセイにおいて評価した。NK Cell Isolation Kit II Miltenyi Biotec(Bergisch Gladbach, ドイツ)によって、健常ドナーのNK細胞をPBMCから単離し、エフェクター細胞として使用した。37℃、1時間、振とう下で、標的細胞(1×106個の細胞)を10μM Calcein−AM (Dojindo、熊本、日本)で標識した。細胞をPBSで3回洗浄し、培地(1×105 細胞/ml)に再懸濁した。標識した細胞を96ウェルU字型底プレート(50ul/ウェル中、5×103)に分注し、0.1pg/mlから0.1mg/ml(最終濃度)の範囲でhumAbまたは14D10を培地で7倍段階希釈した溶液50μlでプレインキュベーション(37℃、30分)した。Calcein−AM自然放出量を測定するためにmAbの代わりに培地を加えるとともに、Calcein−AMの最大放出量を測定するためにTriton X−100(1%の最終濃度)を加えた。その後、ヒトのエフェクター細胞をウェル(5×105 細胞/ウェル)に加え、細胞を37℃で終夜インキュベートした。そして、上清をCalcein−AM放出量の測定のために回収した。特異的溶解の割合を以下の公式を使用して計算した:特異的溶解率(%)=(実験的放出量−自然放出量)/(最大放出量−自然放出量)×100。ここで、最大放出量はTriton X−100を加えることによって測定し、自然放出量は増感性Ab(sensitizing Ab)およびエフェクター細胞の非存在下で測定した。
先の文献に記載されているようにCDCアッセイを実施した(非特許文献24)。標的細胞を96ウェルU字型底プレートに1x105細胞/ウェルで分注し、30分間、4℃で様々な濃度のmAbとインキュベートした。その後、ヒトの血清を加え、2時間、37℃で細胞をインキュベートした。CDC特異的細胞死およびADCC特異的細胞死の評価をAnnexin V-FITC Apoptosis Detection Kit(Bio Vision、Mountain View、CA)および活性化カスパーゼ3の検出によって評価した。
全てのin vivo試験が実験動物委員会(Institute Animal Care and Use Committee)において承認された。6週齢雌のNOD−SCIDマウスは、チャールズ・リバー(神奈川、日本)から購入し、mAb処置の1日前に抗アシアロGMlポリクローナル抗血清25%(v/v、和光、大阪、日本)を腹腔内投与で前処置した。
腫瘍形成性に対するhumAbの効果を評価するために、JMN細胞(1×106)をマウスの左横腹の皮下に接種した。腫瘤(5mmの大きさ)が目に見えるようになった癌細胞接種後14日目に、アイソタイプ一致の対照mAb、5F8、14D10、またはヒト化抗CD26 mAb(10μg/各注射)の腫瘍内注射でマウスを処置した。各mAbを1週間に3回投与した。そして、担癌マウスを腫瘍成長と進行について観察した。腫瘍の大きさは、最も大きい垂直方向の直径(x)と最も小さい垂直方向の直径(y)のカリパス測定によって測定し、式V=π/6×xy2によって計算した。
腫瘍転移に対するhumAbの効果を評価するために、JMN細胞(1×105)を尾静脈内に注入した。次いで、癌細胞注入日から、アイソタイプ一致の対照mAb、5F8、14D10、またはヒト化抗CD26 mAb(10μg/各注射)の静脈内注射によってマウスを処置した。各mAbを1週間に3回投与した。累積生存割合はカプラン-マイヤー法で評価した。
6週齢雌のBalbマウスをチャールズ・リバー(神奈川、日本)から購入し、マウスのエフェクター系の結合を維持するために、抗アシアロGMlポリクローナル抗血清処置は行わなかった。
腫瘍形成性に対するhumAbの効果を評価するために、JMN細胞(1x106)をマウスの左横腹の皮下に接種した。腫瘤(5mmの大きさ)が目に見えるようになった癌細胞接種後14日目(サイズにおける5mm)に、アイソタイプ一致の対照mAb、5F8、14D10、またはヒト化抗CD26 mAb(10μg/各注射)の腫瘍内注射でマウスを処置した。各mAbを1週間に3回投与した。そして、担癌マウスを腫瘍成長と進行についてモニターした。腫瘍の大きさは、最も大きい垂直方向の直径(x)と最も小さい垂直方向の直径(y)のカリパス測定によって測定し、式V=π/6×xy2によって計算した。
最初のmAb処置後35日目に、皮下腫瘍形成モデルにおける切除標本の顕微鏡的特徴評価のため、全てのマウスを安楽死させた。
腫瘍播種に対するhumAbの効果を評価するために、JMN細胞(1×105)を尾静脈内に注入した。次いで、癌細胞注入日から、アイソタイプ一致の対照mAb、またはヒト化抗CD26 mAb(10μg/各注射)を静脈内注射でマウスに処置した。各mAbを1週間に3回投与した。累積生存割合は、カプラン-マイヤー法で評価した。更に、ヒト化抗CD26 mAbの遠隔転移形成効果を評価するため、処置したマウスを安楽死させ、肺と肝臓の複数の転移形成を別の腫瘍転移モデルにおいて計算した。JMN細胞(1×105)を各グループのマウスに静脈内注射した。癌細胞注射日に、アイソタイプが一致する対照のmAb(レーン1、n=4)、5F8(レーン2、n=4)、14D10(レーン3、n=4)、またはヒト化抗CD26 mAb(レーン4、n=4)をマウスに静脈内注射処置した。各mAbを1週間に3回投与した。癌細胞注射後の35日目に、マウスを安楽死させ、肺と肝臓の複数の転移形成を計算した。
NOD/Shi−scid.IL−Rγnull(NOGマウス)を実験動物中央研究所(CIEA)(神奈川、日本)から入手した。ヒトのPBMCは健常ドナーの末梢血からLymphoprep(AXIS-SHIELD, Oslo,ノルウェ−)を使用して単離し、HuECとして使用した。その後、無菌条件下、HuEC(5x106)をPBSに懸濁した0.2mlの容量でNOG−SCIDマウスに腹腔内に注入した。但し、マウスは、HuEC注射の1日前に0.2mLの抗アシアロGMlのポリクローナル抗血清25%(v/v、和光、大阪、日本)の腹腔内投与により前処理した。HuEC注射の1日後にヒトHuECを移植したSCIDマウスへNCI−H2452細胞(5x104)を腹腔内に注入した。1、3、および5日後に、humAbを腹腔内に注入した。腹水症腫瘍成長による死亡をモニターするため、マウスを毎日観察した。累積生存割合はカプラン-マイヤー法によって算定した。
細胞表面CD26はヒト悪性中皮腫に高発現している
最初に、患者から外科的に切除されたヒトMM組織におけるCD26発現レベルを評価した。外科的に切除された原発部位からの12個の連続標本を細胞表面CD26発現について調べた。CD26は全てのMM組織で高発現していた(図1A)。類腺腫瘍、または反応性中皮腫細胞においては、CD26発現は非常に弱かった(図lA−a、b)。それに対して、局在化したMM、分化の進行した乳頭のMM、および拡散MMを含む様々な病理タイプのMMで、CD26が高発現していた(図.lA-cからh)。これらの結果は、CD26が良性の中皮腫組織ではなく、MMで高発現していることを示唆した。
MM細胞株、JMNおよびNCI−H2452は、高い表面CD26発現を示した(図1B)。
CD26が、細胞外マトリックス(ECM)タンパク質への細胞接着において役割を果たすことが報告されているため(非特許文献13および25)、CD26が細胞との相互作用において役割を果たすかどうかを調べた。図1Cで見られるように、siRNAオリゴを用いて内因性のCD26を欠損させたNCI−H2452は、フィブロネクチンおよびコラーゲンIを含む細胞外マトリックスタンパク質に結合するCD26の有意な減少を示した。これらの結果とは対照的に、CD26の欠損は、ラミニン(CD26への結合能のないECMのタンパク質)またはヒアルロナン(CD44に対するリガンド)に対する結合を変化させなかった(図1C)。これらの知見の更なる裏づけとして、pEB6ベクターにサブクローンした完全長CD26 cDNAをトランスフェクションした293T細胞が、対照のpEB6をトランスフェクションした293T細胞と比べて、より高いフィブロネクチン及びコラーゲンIへの結合能を示した(図1C)。そのうえ、CD26の欠損は、p27kip1(図1D)のアップレギュレーションと関連していた。したがって、これらの知見により、CD26が特定のECMタンパク質への結合分子として作用すること、ならびに接触阻害が、CD26の欠損と関連して、観察されたCD26媒介性のp27kip1(非特許文献26および27)のアップレギュレーションに寄与する役割をが果たし得ることが示唆された。
CD26がECM結合タンパク質であると証明されたため、抗CD26 mAbがECMに細胞接着を阻害するかどうかを更に評価した。この目的のために、アイソタイプ一致の対照mAb、5F8、14D10、およびヒト化抗CD26 mAb(humAb)を用いて、ECMへの細胞接着を阻害する可能性について評価した。図2Aに示したように、14D10で処置したJMN細胞およびhumAbで処置したJMN細胞では、フィブロネクチンとコラーゲンIに対する結合が減少し、一方、対照mAbと5F8(生物学的機能のない抗CD26 mAb)ではフィブロネクチンやコラーゲンIに対する結合に影響を及ぼさなかった。更に、14D10およびhumAbは、in vitroの増殖アッセイにおいて用量依存的にJMN細胞に対して直接増殖阻害を与え、とりわけ、humAbは14D10よりも強い増殖抑制効果を示した(図2B)。重要なことに、14D10およびhumAbは、p27kip1のアップレギュレーションおよびCDK2のダウンレギュレーションを引き起こした。これらの結果は、14D10とhumAbの両方が劇的にp27kip1のアップレギュレーションおよびCDK2のダウンレギュレーションを通して接触阻害に関連した増殖阻害を伝達することを示唆するものであった。
14D10とhumAbの両方が癌細胞に同様の直接効果を持っている一方、本研究は、ヒトのエフェクター細胞を用いたADCCアッセイの利用を通じて、14D10との比較におけるhumAbの生物学的効果の差異に重点を置いた。エフェクター/ターゲット(E/T)比を50に一定に保ったとき、humAbで処理したJMN細胞は、抗体用量依存的にADCCを通して特異的溶解を示した(図3A、左パネル)。重要なことに、14D10で処理したJMN細胞は、ADCC特異的溶解を示さず(図3A、左パネル)、これらの結果によって14D10をヒト化してhumAbにすることにより、ヒトのエフェクター系の関与を介して強力なADCC活性が誘導されることが示唆された。そのうえ、図3A右パネルで見られるように、humAbはエフェクター用量依存的にADCC特異的溶解を誘導した。JMN(NCI−H2452)以外のCD26陽性MM株が標的細胞として使用されたときにもこれらの結果が認められた。これらのデータにより、humAbが、14D10で見られた標的細胞に対する直接効果以外の新規な生物学的機能、即ちADCC特異的溶解作用を有することが示唆された。humAb媒介性ADCCをより詳細に特徴付けるために、Pl-annexinV染色および切断されたカスパーゼ3の検出を用いたアポトーシスアッセイを採用した。これらのアッセイでは、抗ヒトIgGのFcγ断片特異的なヤギF(ab’)2断片および抗マウスIgGのFcγ断片特異的なヤギF(ab’)2断片を用いた架橋法により、それぞれをhumAbおよび14D10に対するヒトのエフェクターの擬似体として使用した。図3B(上側3つのパネル)に見られるように、架橋されたhumAbは遅延型アポトーシスを誘導した一方で、架橋された14D10は遅延型および即時型のどちらのアポトーシスをも誘導しなかった。重要なことに、humAbおよび14D10のいずれもが、ヒト補体を使用することによってCDCを誘導しなかったことである(図3B、下段3個のパネル、補助資料の図1)。これらの結合を更に裏付けるのは、架橋されたhumAbだけがJMN細胞においてカスパーゼ3の活性化を誘導し、一方、架橋された14D10、humAb+ヒト補体、および14D10+ヒト補体のいずれもが、カスパーゼ3の活性化を誘導しなかったことである(図3C)。したがって、これらの結果は、humAbがCDC特異的溶解ではなく、ADCC特異的溶解を誘導することを示すものであった。
本発明者らは、近年、14D10が固形腫瘍に対する直接的in vivo抗腫瘍効果を示す(非特許文献24)ことを実証したので、humAbが同様のin vivo抗腫瘍効果があるかどうかをさらに調べた。この目的のため、機能的なB細胞およびT細胞ならびにナチュラルキラー(NK)細胞活性をほとんど欠損しているNOD−SCIDマウスを使用した(非特許文献28)。マウスのエフェクター細胞の効果を最小限にするために、NOD−SCIDマウスは、humAb機能評価を行う前に抗アシアロGMlポリクローナル抗血清で前処置した。図4AおよびBで見られるように、humAbおよび14D10は皮下に接種したJMNの腫瘍形成性を減少させたが、腫瘍形成の抑制においてはhumAbがより強力であった。これらの観察された結果により、humAbには14D10より強い直接的抗腫瘍効果があることが示唆された。さらに、腫瘍拡散におけるhumAbの直接的抗腫瘍活性を調べるために、JMN異種移植モデルにおいて、静脈内投与された抗体の効果を調べた。図4Cで見られるように、humAbと14D10は、両抗体を静脈内投与した場合、マウスの生存を延長させ、余命の延長という観点では、humAbがより効果的であった。以上をまとめると、これらの観察結果により、直接的抗腫瘍活性においてhumAbが、14D10と比較して、より強力であることが示唆された。
humAbおよび14D10の両方が直接的in vivo抗腫瘍効果を示したことから、次に、抗CD26 mAb活性により誘導された抗腫瘍作用におけるマウスのエフェクター系の潜在的関与を調べた。この目的のために、強力なNK細胞活性を有するBalbマウスを我々は利用した。図5Aで見られるように、humAbおよび14D10は、皮下に接種したJMNの腫瘍形成性を減少させた。特筆すべきは、14D10およびhumAbの両方がマウスのエフェクター系の存在下において腫瘍形成を低下させたことである(図5A)。図5Bで見られるように、humAbを処置した腫瘍および14D10を処置した腫瘍の両方が顕微鏡分析で結果的に死組織として観察された。これらの結果は、humAbおよび14D10の両方がマウスのエフェクター系を利用することを示唆するものであり、マウスのエフェクター欠損異種移植モデルにおいてhumAbと14D10との間に差異が観察されたこととは著しく対照的である。。JMN細胞の静脈内投与を利用した更なる実験では、humAbの静脈内注射がマウスのエフェクター系存在下でマウスの生存を効果的に促進したことを示した(図5C)。重要なことに、遠隔JMNの形成はhumAbと14D10の両方で同様に抑制された(図5D)。これらのデータにより、マウスのエフェクター系が抗CD26 mAb媒介性の直接的抗腫瘍効果を増強することが示された。
次に、抗CD26 mAb誘導性の抗腫瘍作用におけるヒトのエフェクター系の潜在的関与について評価した。この目的のために、T細胞、B細胞、およびNK細胞の活性が有意に欠損しているNOD/Shi−scid.IL−Rγnull(NOGマウス)をNCI−H2452異種移植モデル構築に使用した。ヒトのPBMCをヒトのエフェクター細胞(HuEC)としてこのin vivoモデルで使用した。マウスのエフェクター系を完全に欠損させるために、NOGマウスを腹腔内HuEC移植する前日に、抗アシアロGMl抗血清で前処理した。図6で見られるように、humAbの腹腔内投与によってHuECの非存在下でNCI−H2452異種移植マウス余命が著しく延長された。また、14D10でもマウス生存が延長したが、その効果はHuEC非存在下のhumAbよりもはるかに弱かったことに注目すべきである(図6)。これらの結果により、humAbがより強い直接的抗腫瘍効果を有すことが示された。重要なことに、HuECの存在下において、14D10の抗腫瘍効果が著しくは変化しなかったのに対して、humAbの抗腫瘍効果は増強された。以上の観察された結果により、CD26が中皮腫治療のための適切な分子標的であり、そしてhumAbが、少なくとも2つの異なる作用機序、その直接的抗腫瘍活性およびヒトのエフェクター系と連携する能力によって腫瘍増殖を制御することが示唆された。
本研究において、in vitroおよびin vivoモデルにおける抗CD26 mAbの抗腫瘍効果が証明された。重要なことに、本研究結果は、抗CD26 mAbのヒト化により、p27kip1誘導に関連した接触阻害以外の付加的な抗腫瘍効果が与えられることを示唆している。本研究結果は、CD26のヒトMMにおけるECMへの結合タンパク質としての機能的役割を示すものでもある。
免疫組織化学的分析により、ヒトMM細胞が非悪性組織より高レベルの表面CD26を発現することが示され、これによってCD26が癌の成長および進行における役割を担うことが示唆される。NCI−H2452におけるsiRNAオリゴを使用した内因性CD26の欠損により、該細胞のフィブロネクチンおよびコラーゲンIを含むECMへの結合の有意な減少がもたらされた結果は特筆すべきである。更に、完全長のCD26 cDNAをトランスフェクトした293T細胞は、対照のモックトランスフェクトした293T細胞よりもフィブロネクチンおよびコラーゲンIに対して強い結合親和力を示す。そのうえ、CD26の欠損は、p27kip1のアップレギュレーションを引き起こす。したがって、これらの知見は、CD26がECMへの癌細胞接着に関与していること、ならびに接触阻害が観察されたCD26欠損により媒介されるp27kip1のアップレギュレーションに対して寄与的な役割を果たすことを示唆するもである。特筆すべき事実としては、接触阻害の間にp27kip1がアップレギュレーションされることが先に報告されていることである(非特許文献26)。
HumAbと14D10の両方がp27kip1のアップレギュレーションおよびECMへの結合阻害を介したMM増殖の直接阻害を示す。したがって、これらの抗CD26モノクローナル抗体の使用による結果は上記のsiRNA研究から得られた結果と一致しており、humAbと14D10の両方がMMの接着性に対して拮抗作用を有することを示すものである。
抗CD26 mAbを介す抗腫瘍効果と関連するエフェクター機能の更なる試験により、14D10ではなくhumAbがADCC誘導性の細胞溶解を誘発することが示される。HumAbの架橋により、annexin V陽性およびPI陽性のポピュレーションが蓄積し、そして活性化カスパーゼ3が切断された。これらのデータは、抗CD26 mAbのヒト化が直接的抗腫瘍効果に加えてADCCからより大きな寄与を引き起こすことを示すものである。一方で、humAbがCDC活性を誘導しない正確な理由は、現時点で明らかでないが、理由の1つには、ヒト補体タンパク質に対して拮抗作用を有すDAFおよびCD59が表面に高発現していることである(データは示されていない)。あるいは、CDC活性化の誘導には、本in vitroシステムが適切でない可能性も考えられる。
NOD−SCIDマウスを用いた本in vivo研究において、humAbおよび14D10が皮下に接種したJMN細胞の腫瘍形成能を減少させることが示され、これによって、humAbが直接的な抗腫瘍効果も有することが示唆される。また、本結果により、humAbが、CD26に対してより高い結合親和力を有するために、腫瘍形成の抑制において14D10より強力であることも示唆される。
その一方で、Balbマウスを用いたin vivo研究は、humAbおよび14D10が皮下接種したJMN細胞の腫瘍形成能の抑制において同等の効果を有することを示すものである。これらのデータは、マウスのエフェクター系がhumAbより14D10の抗腫瘍効果を増強する可能性を示唆する。実際に、これらのマウスでは、humAb処置腫瘍検体だけではなく、14D10処置腫瘍検体もまた、腫瘤中の生存細胞の減少を示す。humAbと14D10の両方が遠隔転移の形成を抑えることも注目に値し、これらの知見はCD26が特定のECMタンパク質に対して結合タンパク質として機能するという本in vitroの結果から部分的に説明され得る。
機能的なマウスのエフェクターを欠損しているNOG−SCIDマウスによるin vivoの研究において、ヒトのエフェクター細胞と標的細胞との二重異種移植が、humAbを組み合わせた際、標的細胞の単一異種移植の場合よりもマウスをより長く生存させることを示した。これらのデータは、明確に、humAbがヒトのエフェクター系と共に二相性の抗腫瘍作用を誘導することを示唆するin vitroのデータを実証している。
CD26の状態は、癌において変化し、様々な腫瘍の増殖や腫瘍転移能に影響を与え得る。CD26陰性は、幾つかの癌発生に関連している一方で、CD26陽性はその他の新生物の侵襲的な表現型と関連している。例えば、非小細胞肺癌細胞株では、CD26をトランスフェクトした細胞は、形態学的変化を起こし、接触阻害を変化させ、足場非依存的増殖能も低下させる (非特許文献29)。また、CD26再発現は、p21発現上昇と関連しており、アポトーシスの誘導やGl期での細胞周期停止を引き起こす。Wesleyらは、CD26をトランスフェクトしたDU−145転移性前立腺癌細胞においてはCD26/DPPIVの発現によってCDKI p27kip1の発現が、親株およびベクターをトランスフェクトしたDU1−45細胞との比較において、4−6倍増大することを報告した(非特許文献30)。また、卵巣癌のCD26過剰発現によってE−カドヘリンとMMPの組織阻害物質が増加し、結果として侵襲性が減少することも報告されている(非特許文献31)。従って、CD26/DPPIVは上記の場合においては腫瘍のサプレッサーとして機能し、CD26/DPPIVのダウンレギュレーションは増殖制御の欠陥につながり得る。対照的に、CD26発現はT細胞大型顆粒リンパ球白血病(T−LGLL)において、より侵襲的な臨床経過と関連している(非特許文献32)。
非ホジキンリンパ腫(NHL)におけるCD26発現は、主にT−リンパ芽球性リンパ腫(LBL)/急性リンパ性白血病(ALL)やT細胞CD30陽性未分化大型細胞リンパ腫(ALCL)などの侵襲的なサブタイプで見られる。先の報告は、より進行の遅い疾患由来の細胞ではCD40Lが発現しているものの、CD26とCD40Lの発現が互いに排他的であることを示した。特筆すべきは、T細胞LBL/ALLにおけるCD26発現が生存率の悪化と関連していることである(非特許文献33)。腎細胞癌(RCC)では、RCC組織やCaki−2、VMRC−RCWおよびCaki−1などのRCC由来の多くの細胞株の細胞表面上にCD26が高発現している。そのうえ、抗CD26 mAbを介したCaki−2細胞株の増殖害がGl/S細胞周期停止、p27kip1発現量の増大、およびCDK2のダウンレギュレーションと関連している(非特許文献18)。そこで本研究においては、CD26がMM組織において高発現しており、CD26陽性のMM細胞株において抗CD26 mAb処置およびRNAiによるCD26のダウンレギュレーションによって接触阻害およびp27kip1アップレギュレーションが誘導されることを示す。
したがって、T細胞リンパ腫、腎細胞癌および悪性中皮腫などの悪性腫瘍の場合には、CD26は、腫瘍増殖に寄与し、浸潤や転移に関与している可能性がある。これまでに知られている複雑な作用から見て、癌におけるCD26の役割は、各腫瘍のタイプごとに個別に評価する必要がある。
悪性中皮腫は、予後の見通しの悪い侵襲的な新生物であり、化学療法に対する感受性が比較的低い。1つの研究が、系統的に、1965年から2001年6月まで化学療法効果の証拠を再検討し、88トリートメントアームで83試験を見出だした(非特許文献34)。シスプラチンは最もよく作用する単剤で、ドキソルビシンと併用したシスプラチンは、最大応答率(28.5%の応答率、21.3−35.7%の信頼区間)を示した。この報告以来、シスプラチン(cisplatis)/pemetrexed(代謝拮抗物質)の併用あるいはシスプラチン単独を用いたphaseIII無作為試験(切断不能の中皮腫の化学療法ナイーブ患者448人に対する試験)の結果によって、余命の中央値が、シスプラチン単独治療患者では9.3ヶ月であったが、両方の薬剤で治療された患者では12.1ヶ月まで延長することが証明された(非特許文献35)。しかしながら、MMに対する標準的治療法は生存率という観点ではまだ満足できるものではない。したがって、MMの新規な治療方法が緊急に求められている。
したがって、本データによって、本発明の新規なヒト化抗CD26 mAb humAbは、直接的抗腫瘍効果に加え癌細胞をターゲッティングするヒトのエフェクター系も利用可能なため、MMを含む癌治療に対して有効な治療ツールであることが示される。
Claims (17)
- in vivoにおいて悪性中皮腫細胞の増殖を阻害する抗CD26ヒト化抗体又はその断片を含有する、悪性中皮腫の治療用の医薬組成物。
- 前記ヒト化抗体が、CD26に対する14D10モノクローナル抗体由来の可変領域を含む、請求項1記載の医薬組成物。
- 前記ヒト化抗体が、配列番号8〜14からなる群から選択されるアミノ酸配列と80%以上の同一性を有するアミノ酸配列を含む重鎖可変領域、および配列番号1〜7からなる群から選択されるアミノ酸配列と80%以上の同一性を有するアミノ酸配列を含む軽鎖可変領域を含むものである、請求項1記載の医薬組成物。
- 前記ヒト化抗体が、配列番号1〜7からなる群から選択されるアミノ酸配列と80%以上の同一性を有するアミノ酸配列を含む重鎖可変領域、および配列番号8〜14からなる群から選択されるアミノ酸配列と80%以上の同一性を有するアミノ酸配列を含む軽鎖可変領域を含むものである、請求項1記載の医薬組成物。
- 前記ヒト化抗体が、配列番号17で示されるアミノ酸配列と90%以上の配列同一性を有するアミノ酸配列を含む少なくとも1本の重鎖、および配列番号18で示されるアミノ酸配列と90%以上の配列同一性を有するアミノ酸配列を含む少なくとも1本の軽鎖を含む、請求項1記載の医薬組成物。
- 前記ヒト化抗体が、配列番号17で示されるアミノ酸配列のうち第20〜465番目のアミノ酸配列を含む少なくとも1本の重鎖、および配列番号18で示されるアミノ酸配列のうち第21〜234番目のアミノ酸配列を含む少なくとも1本の軽鎖を含む、請求項1記載の医薬組成物。
- 前記ヒト化抗体が、配列番号19〜28からなる群から選択される1つ以上のペプチドに結合する、請求項1〜7のいずれか1項に記載の医薬組成物。
- 前記ヒト化抗体が、American Type Culture Collection (ATCC)の受託番号PTA-7695を有し、s604069.YST-pABMC148(x411)と命名された株により生産される、請求項1記載の医薬組成物。
- 前記断片が、請求項1〜8のいずれか1項に記載の抗体のFab、Fab’、Fab’‐SH、Fv、scFv、およびF(ab’)2からなる群から選択されるものである、請求項1に記載の医薬組成物。
- 請求項1〜9のいずれか1項に記載の抗体又はその断片に特異的なエフェクター細胞を更に含む、請求項1記載の医薬組成物。
- 請求項1〜9のいずれか1項に記載の抗体又はその断片および該抗体又はその断片に特異的なエフェクター細胞を含有する、悪性中皮腫の治療用キット。
- 請求項1〜9のいずれか1項に記載の抗体又はその断片を含む、悪性中皮腫細胞の増殖阻害剤。

- 中皮腫の患者から採取された中皮腫検体と請求項1〜9のいずれか1項に記載の抗体とを反応させてCD26陽性細胞を検出する工程を含み、CD26陽性細胞が検出されることをもって前記中皮腫検体が悪性であるとする、悪性中皮腫の検出方法。
