Description
(A)材料
DMEM、RPMI-1640、ペニシリン、ストレプトマイシン、およびゲンタマイシンは、Gibco社(エッゲンシュタイン,ドイツ)から購入した。FCSは、Biochrom社(ベルリン,ドイツ)から入手した。DEAE-デキストランは、Sigma社(タウフキルヘン,ドイツ)から得た。制限酵素のT4-DNAリガーゼおよびポリヌクレオチドキナーゼは、New England Biolabs社(シュワルバッハ,ドイツ)から得た。[3H]-チミジン、ECL-試薬、およびX線フィルムは、Amersham Bioscience社(フライブルク,ドイツ)から入手した。クイックチェンジ部位特異的変異誘発キットは、Stratagene社(アムステルダム, NL)から得た。抗体は、Cell Signalling Technology社(フランクフルト,ドイツ)から購入した。BAF/gp130細胞、組換えIL-6およびハイパー-IL-6(H-1L-6)、IL-6+sIL-6Rを含む融合タンパク質(Rakemannら, J. Biol. Chem. 274 (1999), 1257)は、Stefan Rose-John氏(生化学研究所所長、キール,ドイツ)から入手した。ヒトsgp130Fcは、上記したように通りに作成した(Atreyaら, Nat. Med. 6 (2000), 583)。
BAF/gp130細胞を、水飽和大気(water saturated atmosphere)内において5%CO2、37℃でDMEMの中で増殖させた。細胞培養培地に10%FCS、100 mg/lのストレプトマイシンおよび60mg/lのペニシリンを添加した。10ng/mlのH-IL-6が存在する中でBAF/gp130細胞を培養した。
標準的なプロトコールに従ったPCRによって、gp130のコード配列の1番目から978番目までの塩基(Met1からPro326までのアミノ酸に相当する(図2))を増幅することによって、gp130(D1-D3)のリガンド結合ドメインのクローニングを行った。pSVL-sgp130-Fc(Atreyaら, Nat. Med. 6 (2000), 583)を鋳型として採用した。得られたDNA断片を、1%アガロースゲル上で精製し、Qiagen MiniEluteキットを用いて単離し、発現プラスミドpQE60、pQE70(Qiagen)、pBAD/Myc-His(Invitrogen)、pET-3、pET-11、pCAL-c、およびpCAL-kc(Stratagene)にクローニングした。すべての構築物を制限酵素分解によって同定し、標準的な技術によって挿入配列の配列を確認した。
標準的なクローニング技術に従った部位特異的変異誘発法によって、1個以上のシステイン残基を含むさまざまな長さのポリペプチド(Pn)をsgp130(D1-D3)のC末端に付加した。すべての構築物を制限酵素分解によって同定し、挿入配列の配列を確認した。
単離されたばかりのヒトゲノムDNAを鋳型として使用して、PCRによってvIL-6のcDNAを増幅した(図3のコード配列)。vIL-6-HisをCOS-7細胞で発現させるために、viL-6のcDNAを、ポリヒスチジン(His)タグの前で、哺乳動物発現プラスミドであるpEF11myc-His、pUB6N5-His(Invitrogen)またはpQE-TriSystem(Qiagen)の中に挿入した。vIL-6-Hisを細菌で発現させるために、vIL-6のcDNAを、ポリヒスチジンタグの前で、原核生物発現ベクター(pQE60、pQE70(Qiagen)、pBAD/Myc-His(Invitrogen)、pET-3、pET-11、pCAL-c、およびpCAL-kc(Stratagene)の中に挿入した。すべての構築物を制限酵素分解によって同定し、標準的な技術によって挿入配列の配列を確認した。
組換えviL-6-Hisを、Muellbergら; J. Immunol. 164 (2000), 4672に記載されている通りに精製した。
精製された組換えVIL-6-Hisを以下のようにしてNi-NTAアガロースカラム(Qiagen)に結合させた。カラム材料は、5容積倍量50 mMのリン酸緩衝液(pH 7.5)、500 mM NaCl、および20 mMイミダゾール(平衡用緩衝液、EB)で平衡させた。vIL-6-Hisをカラムにかけて、未結合タンパク質を5容積倍量のEBで洗浄して除去した。sgp130(D1-D3)-Pnを含むタンパク質懸濁液をカラムにかけ、続いて、そのカラムを10容積倍量のEBで洗浄した。最後に、クエン酸緩衝液(pH 1.4)でsgp130(D1-D3)-Pnを溶出し、溶出液を直ちに中和した。溶出物を、モノQファーストタンパク質液体クロマトグラフィー(Pharmacia)によって精製してから、PBSに対して透析した。
sgp130(D1-D3)-PnおよびvIL-6-Hisをコードする原核生物発現プラスミドを、細菌株のTOP10(Invitrogen)、XL1-Blue、BL21(DE3)(Stratagene)、M15[pREP4]またはSG13009[pREP4](Qiagen)に同時形質移入した。形質転換から24時間後、1 mMのイソプロピル-β-D-チオガラクトピラノシド(IPTG)によってタンパク質産生を開始させ、6時間後にタンパク質を抽出した。タンパク質抽出物を、予め5容積倍量のEBで平衡しておいたNi-NTAアガロースカラム上に負荷した。カラムを10容積倍量のEBで洗浄して、未結合のタンパク質を除去した。最後にgp130(D1-D3)-Pnを溶出させて、上記のようにして精製および透析した。
vIL-6-Hisについて述べた技術と同じ技術によって、sgp130(D1-D3)-Hisを精製した。
ここ数年、ポリエチレングリコール(PEG)によって生体高分子の共有的修飾についての興味の高まりが起きている。この種の修飾は、医薬およびバイオテクノロジーに応用する上で非常に重要である。ペグ化(PEGの共有的結合)は、例えば、抗原性または免疫原性のエピトープの遮蔽をもたらす。さらに、細網内皮系によるレセプター介在型取り込みを抑制し、タンパク質分解酵素による認識と分解を防止する(5)。タンパク質のペグ化は、腎臓濾過を低下させることによって、その生物利用能を高めることが分かっている。sp130(D1-D3)は、9個のシステイン(C)残基を含み、そのうちの8個がジスルフィド架橋に関与する(C28-C54、C48-C103、C134-C144、C172-C182)。最後のC301は、遊離のスルフヒドリル基を含むため、部位特異的なペグ化に適している。3倍モル過剰のmPEG-MAL(分子量20,000)(Nektar Therapeutics, San Carlos, CA, USA)をsgp130(D1-D3)-PnにpH 7.2で付加してsgp130(D1-D3)の修飾を行った。この反応を室温で60分間行ってから、標準的な条件による最終的なゲル濾過工程で生成物を単離した。このタンパク質を、3倍モル過剰の分岐型mPEG(MAL)2(分子量20,000)(Nektar)とpH7.2、室温で90分間インキュベートして、ペグ化sgp130(D1-D3)-Hisを生成させた。次に、標準的な技術によって、この生成物をゲル濾過した。
3 mMのL-グルタミン、10 mMのHEPES緩衝液、10μg/mlのゲンタマイシン、100 U/mlのペニシリン、100U/mlのストレプトマイシン、50μMの2-メルカプトエタノールおよび10%の熱不活性化FCSを加えたRPMI-1640からなる完全培地の中でLPMCを培養した。図に示すように、1から10μg/mlの濃度のsgp130Fc、中和IL-6R特異的抗体(ドイツ国キール大学教授であるRose-John博士より提供された)、sgp130PEGまたはsgp130TagPEGの存在下または非存在下で細胞を10μg/mlのC反応性タンパク質(Sigma)、50 ng/mlのホルボール-12-ミリステート-13-アセテート(PMA)、および10μg/mlのフィトヘマグルチニン(PHA)(Sigma)によって刺激した。48時間後、ApoAlertアネキシンV-FITCアポトーシス検出キット(BD Bioscience社、ハイデルベルク,ドイツ)を用いて、アネキシンVとヨウ化プロピジウムで細胞を染色し、FACSによる分析を行った。
増殖因子を除去するためにBAF/gp130細胞をPBSで入念に洗浄してから、サイトカインを含まない培地に再懸濁した。96ウエルプレートの1ウエルにつき5×103個の細胞を、サイトカインを入れて最終用量100μlにして、図に示すようにsgp130Fcまたはsgp130PEGまたはsgp130TagPEGの量を増加させながら68時間培養した後、0.25 mCiの[3H]-チミジンで4時間パルス標識した。細胞をガラスフィルター上に回収して、取り込まれた[3H]-チミジンをシンチレーション計測法によって測定した。
BAF31gp130細胞を、レポーター遺伝子プラスミドのpSTAT3-TA-Luc(BD Bioscience社、ハイデルベルク,ドイツ)およびpCMV-Lucによって同時形質転換して、24時間インキュベートした。そして、形質転換された細胞を、10μg/mlのsgp130Fc、sgp130PEGまたはsgp130TagPEGがそれぞれ存在するかしない中で、5 ng/mlのH-IL-6とともにさらに20時間インキュベートした。Promega社(マンハイム,ドイツ)から購入した二重ルシフェラーゼレポーター遺伝子アッセイ(Dual Luciferase Reporter Gene Assay)を用い、製造業者の使用説明書に従って、MicroLumatPlus LB96Vマイクロプレート発光測定装置(EG&G Berthold, ウェルズリー, MA, USA)で測定してルシフェラーゼ活性の抽出および検出を行った。
標準的なプロトコールに従ったPCRによって、gp130のコード配列の70番目から966番目までの塩基(Leu 24からTyr 322までのアミノ酸に相当する(図2))を増幅することによって、gp130(D1-D3).1のリガンド結合ドメインのクローニングを行った。pSVL-sgp130-Fc(2)を鋳型として採用した。得られたDNA断片を、1%アガロースゲル上で精製し、Qiagen MiniEluteキットを用いて単離し、適当な発現プラスミドにクローニングした。標識されたタンパク質を発現させるために、His(4-6)、FLAG、Step-Tag、GFP、GST、HA CBP、またはその他、抗体を利用できるエピトープなど、適当な標識を含むベクターを用いた。あるいは、所望の標識をsgp130(D1-D3).1のc-DNAのすぐ後ろにクローニングした。すべての構築物を制限酵素分解によって同定し、標準的な技術によって挿入配列の配列を確認した。
製造業者の使用説明書に従って、対応する発現プラスミドを部位特異的変異誘発することによって、1個以上のシステイン残基を含むさまざまな長さ(Pn)のポリペプチドをsgp130のC末端に付加した。すべての構築物を制限酵素分解によって同定し、標準的な技術によって挿入配列の配列を確認した。
単離されたばかりのヒトゲノムDNAを鋳型として使用して、PCRによってvIL-6のcDNAを増幅した(図3のコード配列)。vIL-6-HisをCOS-7細胞で発現させるために、vIL-6のcDNAを、ポリヒスチジン(His)タグの前で、適当な哺乳動物発現プラスミド、例えば、pcDNA3.1/myc-His、pEF1/myc-His、pUB6/V5-His(Invitrogen)、pQE-TriSystem(Qiagen)などの中に挿入した。vIL-6-Hisを細菌で発現させるために、vIL-6のcDNAを、ポリヒスチジンタグの前で、適当な原核生物発現ベクター、例えば、pQE60、pQE70(Qiagen)、pBAD/Myc-His(Invitrogen)、pET-3、pET-11、pCAL-c、pCAL-kc(Stratagene)などの中に挿入した。すべての構築物を制限酵素分解によって同定し、標準的な技術によって挿入配列の配列を確認した。
ウイルスのインターロイキン-6(vIL-6)(4)は、さらにIL-6レセプター(IL-6R)を必要とせずにgp130に特異的結合することが分かっている(3)。vIL-6のこの相互作用を用いて、vIL-6-Hisアフィニティーカラムによってsgp130を精製した。精製された組換えVIL-6-Hisを以下のようにしてNi-NTAアガロースカラム(Qiagen)に結合させた。カラム材料は、5容積倍量50 mMのリン酸緩衝液(pH 7.5)、500 mM NaCl、および20 mMイミダゾール(平衡用緩衝液、EB)で平衡させた。vIL-6-Hisをカラムにかけて、未結合タンパク質を5容積倍量のEBで洗浄して除去した。sgp130-Pnを含むタンパク質懸濁液をカラムにかけ、続いて、そのカラムを10容積倍量のEBで洗浄した。最後に、クエン酸緩衝液(pH 1.4)でsgp130-Pnを溶出した。溶出液を直ちに中和して、モノQファーストタンパク質液体クロマトグラフィー(Pharmacia)によって精製してから、PBSに対して透析した。

vIL-6-Hisについて述べた技術と同じ技術によって、sgp130-Tag(His6の標識を有する)を精製した。His(4-6)、FLAG、Step-Tag、GFP、GST、HA CBP、またはその他、抗体を利用できるエピトープなど、別の標識の場合には、アガロースなどの基質上に固定した適当な抗体によってsgp130Tag分子を分離した。

IL-6Rに対する中和抗体は、CD患者由来の粘膜固有層T細胞におけるアポトーシスを誘導することができる(Atreyaら, 2000)。抗IL-6R mAbの作用は、大腸炎の3つの異なったモデル、すなわち、CD62L+ CD45RBhighCD4+T細胞によって再構築された重症複合型免疫不全症マウス、IL-10欠損マウス、およびトリニトロベンゼンスルホン酸(TNBS)処理されたマウスでも実証された。すべての場合で、大腸炎の活性が、抗TNF mAb処理した後に観察されたレベルと同じレベルにまで下方制御された(Atreyaら, 2000)。この実験の目標は、本発明に係るsgp130PEGおよびsgp130TagPEGが、LPMCにおけるアポトーシスの誘導について、例えばsgp130Fcなどと同じ効率を示すか否かを明らかにすることであった。実施例1に記載した条件下、中和抗IL-6R抗体、sgp130Fc、sgp130PEG、またはsgp130TagPEGの存在下でLPMCを培養した。FACS分析によって、アネキシンV陽性でヨウ化プロピジウム陰性の細胞をアポトーシス細胞と判定した。未処理の試料は、約60%のアポトーシス細胞を含んでいたが、抗IL-6R抗体、sgp130Fc、sgp130PEG、またはsgp130TagPEGで処理すると、11%から16%までアポトーシス細胞が増加した。ペグ化していないsgp130二量体は効果が低く、分子の生物学的活性にとってペグ化が重要であることを示している(図4および8)。
IL-3依存性前駆B細胞株BAF/3はgp130を発現しないため、IL-6にもIL-6/IL-6Rにも応答できない。これに対し、ヒトgp130のcDNAで形質転換されたBAF/3細胞(BAF/gp130細胞)は、IL-3非存在下で、IL-6/IL-6Rまたはハイパー-IL-6(H-IL-6)に応答して増殖することができる。IL-6/IL-6R(図5Aおよび9A)またはH-IL-6(図5Bおよび9B)で刺激されたBAF/gp130をsgp130Fc、sgp130PEG、またはsgp130TagPEGで量を増加させながら処理し、細胞増殖に対する両sgp130分子の効果を測定した。どちらの実験においても、sgp130Fc、sgp130PEG、またはsgp130TagPEGの量を増加させると、[3H]-チミジンの取り込みが有意に低下する結果となった。しかし、7.5から10 ng/mlのsgp130Fcを細胞に投与すると、最大半量取り込みに達した(約7.500 cpmにおける点線)。これに対し、sgp130PEGおよびsgp130TagPEGの濃度が1から5 ng/mlのときに、同様の取り込みの低下が既に見られた。このことは、sgp130PEGおよびsgp130TagPEGによる処理が、sgp130Fcよりも1.5から3倍強く作用することを示している。さらに、パネルBでは、ペグ化されていないsgp130二量体を用いて、タンパク質の生物学的活性に対するペグ化の重要な役割を明らかにしている。
転写の潜在型細胞質転写因子シグナル伝達因子および活性化因子(STAT)3が、IL-6処理によっていくつかの異なった細胞型において活性化されることが知られている。STAT3の機能が、さまざまな細胞型において盛んに研究されている。これらには、肝癌細胞における急性期反応の誘導、Bリンパ球における増殖刺激、最終分化および増殖アレスチン単核細胞(growth arrestin monocytes)の活性化、および胚幹細胞の多分化能の維持などが含まれる(Levy and Lee, J. Clin. Invest. 109 (2002), 1143における概説)。IL-6誘導型STAT3活性化がsgp130PEGまたはsgp130TagPEGによって影響を受けるか否かを判定するために、実施例1に記載されているように、pSTAT3-TA-LucによってBAF/gp130細胞を形質転換した。24時間後、10μg/mlのsgp130Fc、sgp130PEG、またはsgp130TagPEGそれぞれの存在下または非存在下において細胞を5 ng/mlのH-IL-6でさらに20時間処理した。細胞を抽出して、ホタルルシフェラーゼ活性を測定した。H-IL-6だけで処理した細胞における相対的ルシフェラーゼ活性を100%とした(図6Aおよび10A)。これに対して、STAT3活性は、それぞれ、26%(sgp130Fc)、18%(sgp130PEG)、および19%(sgp130TagPEG)に低下した。第2の実験において、細胞をsgp130Fc、sgp130PEG、またはsgp130TagPEGの量をそれぞれ増加させながら処理して、上記した作用の用量依存性を測定した(図6Bおよび10B)。sgp130PEGおよびsgp130TagPEGを2.5μg/mlの濃度で用いて、STAT3の最大半量活性化を測定したが、これと同じ効果は、約7.5μg/mlの濃度のsgp130Fcで見られた。ここでも、前に示したように、sgp130PEGおよびsgp130TagPEGの効率は、sgp130Fcで見られる効率よりも約2〜3倍高かった。
Claims (10)
- 2つの可溶性gp130分子を含むポリペプチド二量体であって、該可溶性gp130分子の少なくとも一方がポリエチレングリコールに共有結合しており、該可溶性gp130分子の各々が、gp130の細胞外ドメインD1〜D3からなる、ポリペプチド二量体。
- 前記可溶性gp130分子の各々がポリエチレングリコールに共有結合している、請求項1記載のポリペプチド二量体。
- 前記2つの可溶性gp130分子の少なくとも一方が、配列番号:2に示したアミノ酸配列を含む、請求項1または2記載のポリペプチド二量体。
- 前記可溶性gp130分子の各々が、配列番号:2に示したアミノ酸配列を含む、請求項3記載のポリペプチド二量体。
- 2つの可溶性gp130分子が、1個以上のジスルフィド架橋を介して互いに結合している、請求項1〜4いずれか記載のポリペプチド二量体。
- 2つの可溶性gp130分子が、分岐型ポリエチレングリコールを介して互いに結合している、請求項1〜4いずれか記載のポリペプチド二量体。
- 2つの可溶性gp130分子が、フレキシブルなペプチドリンカーを介して互いに結合している、請求項1〜4いずれか記載のポリペプチド二量体。
- 請求項1〜7いずれか記載のポリペプチド二量体または該二量体の単量体をコードするポリヌクレオチドを含む発現ベクターを含む宿主細胞を培養すること、該宿主細胞または培養物からポリペプチド単量体または二量体を回収すること、および該単量体または二量体をペグ化することを含む、請求項1〜7いずれか記載のポリペプチド二量体の作製方法。
- 請求項1〜7いずれか記載のポリペプチド二量体を含有してなる医薬組成物。
- 骨吸収、高カルシウム血症、悪液質、腫瘍、自己免疫疾患、炎症性疾患、または細菌もしくはウイルス感染の治療用または予防用医薬組成物の調製のための請求項1〜7いずれか記載のポリペプチド二量体の使用。
