Description
The present invention relates to a microfluidic chip. More particularly, the present invention relates to a heating and cooling method involving the transfer of a trace fluid within a microstructure formed in a microfluidic chip.
Recently, microchannels and ports that form channels of a predetermined shape in a substrate, as is known by names such as Micro Total Analysis Systems (μTAS) or Lab-on-Chip (Lab-on-Chip) It has been proposed to perform various operations such as chemical reaction, synthesis, purification, extraction, generation and / or analysis of substances within the microstructure, and some of them have been put into practical use. A structure manufactured for such a purpose and having a microstructure such as a microchannel and a port in a substrate is generally called a “microfluidic chip” or a “microfluidic device”.
Microfluidic chips can be used in a wide range of applications such as genetic analysis, clinical diagnosis, drug screening and environmental monitoring. Compared with the same type of equipment of the common size, the microfluidic chip (1) uses significantly less sample and reagent, (2) analysis time is short, (3) high sensitivity, (4) portable to the field, It has the advantages of being able to analyze on the spot and (5) disposable.
In the conventional
In chemical operations performed within the microstructure such as microchannels and ports of microfluidic chips, the temperature of the material of interest may be an important factor, and heating and cooling means are required as necessary. Will be taken. For example, in order to promote a chemical reaction, heating to a temperature higher than room temperature, or cooling to a temperature lower than room temperature is frequently performed in order to stably store a sample solution or a reagent. Also in the field of microfluidic chips, there is a need to heat and cool a microfluid introduced into a fine structure, and various heating and cooling methods are being studied.
Simultaneously with the chemical operation, it is necessary to transfer the microfluid in the microfluidic chip. That is, a certain amount of fluid dispensed to the input port is transferred from the input port to the reaction chamber, transferred from one reaction chamber to another reaction chamber, or transferred from the reaction chamber to the detection unit, etc. There is a need. When microfluidic chips are integrated with high density and high density, and more complex and advanced operations are required, a variety of processes are required, and the need to properly transport microfluids is increasing. Therefore, a method that fully considers the transfer of a trace fluid simultaneously with the heating and cooling operation is desired.
In discussing the problem of heating and cooling operations and the transfer of the fluid, PCR can be cited as one specific example. In the following description, it will be discussed as an operation for PCR.
PCR is an abbreviation for “Polymerase Chain Reaction”, which translates to “enzyme chain reaction” and the like, and is an important technique in molecular biology for amplifying DNA fragments. PCR is a reaction performed by applying a three-stage temperature cycle to an appropriately formulated PCR solution. In some cases, some PCR reagents can be performed in a two-stage temperature cycle, but the following description assumes that a general three-stage temperature cycle is performed.
The PCR solution is a mixed aqueous solution of (a) primer, (b) enzyme, (c) template DNA, etc., but description on the preparation method and the like is omitted here.
The three-stage temperature cycle is an operation in which different temperature states called (a) heat denaturation, (b) annealing, and (c) elongation are repeated, and temperatures around 95 ° C., 55 ° C., and 72 ° C., respectively. Used. The time for maintaining each temperature is several tens of seconds to several minutes. The number of cycles is generally around 30 times. In addition, the heat denaturation at the beginning of the cycle may be performed for a long time (initial heat denaturation), or the final extension of the cycle may be performed for a long time (final extension).
As a method of operating a desired temperature cycle on the PCR solution, there is a method of sequentially transferring the PCR solution to a plurality of temperature compartments adjusted to a desired constant temperature in advance. In this method, since only the necessary PCR solution needs to be heated and cooled, the reaction time can be shortened, but on the other hand, the PCR solution must be appropriately transferred during the reaction. Non-Patent
このような処理方法はフロースルーPCR(Flow Through PCR)法と呼ばれることがある。
In
In the flow-through PCR method, since the temperature cycle is repeated about 30 times, it is necessary to lengthen the flow path. When the liquid was sent from one end of the flow path with a pump, the flow rate was greatly different between the time when the flow started and the time after the flow path was filled with the solution. This is because the flow rate varies depending on the amount of liquid to be fed, so that the flow path and the volume of the solution change or the discharge flow rate varies depending on the liquid feed pressure depending on the performance of the pump. In particular, in a microchannel such as a microfluidic chip, the action of the liquid interfacial force also greatly affects the liquid flow rate. Further, even after all the channels were filled with the solution, it took time to propagate the pressure over the entire length of the narrow channel, and it took time for the entire fluid to reach a uniform and uniform flow.
As described above, since a stable flow rate cannot be obtained, the PCR solution may be wasted until the target temperature cycle is accurately performed. Further, in order to obtain the desired amount of PCR product, an extra PCR solution is required for at least the volume of the channel in order to flow the PCR product out of the channel. That is, even in the case of PCR performed on a microfluidic chip, the flow-through PCR method can be applied when the amount of solution is relatively large. At present, improvement research of the flow-through PCR method is continued, but it does not change that the liquid is fed at a constant flow rate in one direction, and no great progress has been made regarding the liquid feeding method.
As a PCR method other than the flow-through PCR method in the microfluidic chip, Non-Patent Document 2 describes a reciprocating (shuttle) PCR method. In this method, a flow path that passes only once for three temperature compartments provided in the microfluidic chip is arranged, and a minute droplet in the flow path is reciprocated to realize a desired temperature cycle. It is.
FIG. 8A is a partial schematic plan perspective view of the
FIG. 8C is a partial schematic cross-sectional view showing a state where the
This round-trip (shuttle) PCR method is a method in which PCR can be applied to a small amount of PCR solution as compared with the flow-through PCR method, and the waste of the PCR solution is relatively small. Further, the flow path is short, and the planar occupation area in the microfluidic chip can be small. Furthermore, by providing a large number of the same flow paths in parallel, there is an advantage that a plurality of PCRs can be performed simultaneously by parallel processing.
However, the following problems also exist in the round-trip (shuttle) PCR method.
(1) The flow path is special.
If the exact liquid volume cannot be transferred between the temperature zones, the correct temperature cycle will not be performed. Therefore, in Non-Patent Document 2, a kind of valve structure (Capillary Burst Valve) using a
(2) The actuator is expensive and its driving method is complicated and sophisticated.
In relation to the above, the actuator is not simply that the volume of the chamber should be variable by a constant amount by simply controlling the positioning of the constant displacement, and the displacement speed becomes important. That is, the speed is also controlled. For example, a complicated and advanced driving method is required such that the fluid is changed at high speed in the initial stage of transfer (by applying a suddenly high pressure) and the fluid passes through the valve structure. .
(3) Not suitable for fluid transfer between multiple points.
Since the fluid is transferred between the three temperature compartments by one actuator, the actuator needs analog control in another stage. Simple digital on / off control does not transfer fluid. Furthermore, when the temperature compartment increases to three or more and it is necessary to perform fluid transfer between multiple points, the flow path becomes long, and the transfer of fluid due to the volume change of the chamber disposed at one end of the flow path is technically Have difficulty.
(4) Not suitable for multi-step process.
Since a large chamber is disposed at one end of the flow path, it is difficult to perform fluid transfer between processes other than PCR that performs heating and cooling. Originally, the technique shown in Non-Patent Document 2 is based on the premise that only a PCR process is performed by a temperature cycle using a microfluidic chip. The transfer of fluids such as post-processes such as DNA detection by is not considered.
(5) The volume of the PCR solution cannot be changed.
The volume of the solution for PCR is determined by the size of the flow path in the temperature compartment, and once the microfluidic chip is manufactured, the volume can be changed by changing the operation of the actuator. I can't.
(6) The solution in the other temperature compartment is wasted.
While the target solution is heated and cooled in a certain temperature section, the excess solution is heated and cooled in another temperature section, which is wasted. In order to eliminate the waste of the solution, it is conceivable to perform PCR by introducing only a droplet in which the required amount of the solution is separated by gas into the flow path, but a predetermined amount of liquid droplet is formed in the flow path. That is a very difficult operation.
(7) The evaporation of droplets cannot be prevented.
Further, when heating to a high temperature close to 100 ° C. as in PCR, droplets of the aqueous PCR solution frequently evaporate from the interface with the gas, resulting in changes in the amount and concentration of the solution. Furthermore, the pressure in the flow path is changed by the evaporated gas, which hinders the transfer of droplets. From the viewpoint of evaporation, a liquid that is hermetically sealed and does not contain a gas is preferable, rather than a droplet separated by a gas.
(8) When performing parallel processing, a plurality of actuators and their driving operations are required.
When performing parallel processing by arranging a plurality of the same flow paths in parallel, it is necessary to provide one actuator for each flow path. It is uneconomical to use a plurality of expensive actuators, and the above-mentioned high-function and high-accuracy driving operation must be performed for each actuator, and the control circuit is complicated and expensive. Furthermore, unlike the microfluidic chip microstructure, the actuators arranged outside are relatively large in size, making it difficult to arrange a large number of them at high density.

As a means for solving the above-mentioned problems, the invention according to claim 6 is the microfluidic chip according to any one of
By using the microfluidic chip having the heating and cooling structure as described above, the following effects can be obtained.
(1) The flow path design is easy.
To the extent that the reference volume of the first space (temperature chamber) or the second space (pressure chamber) is obtained, a difficult design method is not required.
(2) The driving source for transferring the fluid is simple and inexpensive, and the driving method is simple.
This can be done by turning on and off the accumulated pneumatic container and solenoid valve.
(3) Suitable for fluid transfer between multiple points.
There is no limit to the number of points due to the transfer principle.
(4) Suitable for multi-step process.
A pressure chamber for fluid transfer is arranged in the upper part of the temperature chamber for heating and cooling, and there is no need to provide a structure that obstructs the transfer in the middle of the microchannel between the preceding and following processes.
(5) The fluid volume can be made variable.
By appropriately selecting the operation pressure, the volume of the fluid to be transferred can be made variable. The micro structure of the conventional microphone mouth fluid chip is formed by an expensive mold, and microchannels having different dimensions require different molds. If the necessary amount of PCR products can be obtained with the same microphone-mouth fluid chip, it is extremely economical.
(6) There is no waste of fluid.
During the forward pressure operation, the volume of the temperature chamber can be made almost zero, so that no solution is accumulated in the unnecessary temperature chamber.
(7) The evaporation of the liquid can be prevented.
The temperature chamber has a sandwich structure sandwiched between the heat transfer member and the pressure chamber. By using a gas barrier material for the heat transfer member and filling the pressure chamber with liquid, evaporation of the liquid in the temperature chamber can be prevented.
(8) Suitable for sealing and preload operation.
Open / close valves are freely provided before and after the temperature chamber, and even when the open / close valve is closed, the fluid can be transferred between the temperature chambers without any problem, so that the fluid can be heated and cooled in a sealed state. In particular, heating and cooling can be performed in a state in which a preload is applied to the fluid, and liquid evaporation and gas expansion can be prevented.
(9) Suitable for parallel processing.
The drive source for liquid transfer can be made common, and even if the number of systems for parallel processing increases, the cost of manufacturing the microphone mouth fluid chip and peripheral devices is not greatly affected, and the drive is only for one system. All systems can be processed by driving.
In addition, the planar occupation area of the pressure chamber, which is a drive unit for liquid transfer in the microphone mouth fluid chip, is small, and multiple systems can be arranged with high density.
(10) Suitable for observation and analysis using light.
Observation and analysis using light from the pressure chamber side by using a gas or transparent fluid as the pressure medium without the need to place a shading object such as a piezoelectric element or an electric motor near the temperature chamber as an actuator for fluid transfer Can be done.
Furthermore, the observation and analysis by transmitted light is possible by devising the arrangement and shape of the transparent heat transfer member and the cooling heat source.
Hereinafter, the microfluidic chip of the present invention will be described in detail with reference to the drawings. FIG. 1 is a partial plan perspective view of an example of a heating / cooling structure used in the microfluidic chip of the present invention, and FIG. 2 is a partial schematic sectional view taken along line II-II in FIG. The

その材料としてPDMS(ポリジメチルシロキサン)が適している。

The forward pressure and the reverse pressure operation of the
As the pressure of the forward pressure operation increases, the amount of decrease in the volume of the
The volume of the fluid to be transferred is equal to the difference between the volume of the
In FIG. 1, the
The dimensions of the
The generation method and control method of the operating pressure will be further described. Here, an air pressure source generally used is taken as an example. In order to operate the predetermined pressure, the compressor may be started / stopped each time, but it takes time to reach the predetermined pressure, and it is necessary to control the compressor so as to maintain the predetermined pressure. . On the other hand, the present inventors recommend the following method. A small number of several 10 cc to several 100 cc pneumatic pressure vessels are prepared, and a predetermined operating pressure is stored in advance in each vessel. The operation of accumulating is appropriately performed by starting / stopping the compressor or adjusting the pressure. The pressure for accumulating pressure may be higher than atmospheric pressure or lower than atmospheric pressure (negative pressure). In the middle of the air pressure container and the

The heating / cooling structure of the present invention is provided with the
One way to prevent evaporation of the liquid maintained at a high temperature is to avoid contact with the gas. For this purpose, it is necessary to keep only the liquid containing no gas sealed. Therefore, the
Furthermore, in order to prevent the liquid from evaporating, it is preferable to pressurize the liquid in advance. By increasing the pressure of the liquid, the boiling point of the liquid increases, and at the same time, the expansion of the gas accompanying evaporation can be suppressed. Even in a sealed liquid, evaporation of the liquid and accompanying gas expansion occur based on a small amount of gas contained in the liquid. It continues until the sealed space reaches a certain pressure. -And, as the volume occupied by the gas increases, the rate at which the gas permeates out of the microphone
One of the features of the
In chemical operation, observation and analysis may be performed after heating and cooling of a substance, but it may be necessary to observe and analyze a reaction process during heating and cooling. For example, real-time PCR. As will be described later, the

In Table 1, in the case of the normal pressure operation, the solution in the temperature chamber is discharged, and in the case of the reverse pressure operation, the temperature chamber is filled with the solution.

During the transition from step 2 to step 3, the solution is transferred through the temperature chamber 7-2. Meanwhile, the pressure chamber 7-2 is operated for normal pressure. Its volume is almost the same. However, unlike the on-off valve, the temperature chamber 7-2 does not completely block the
The above is an example of three types of temperatures, but as is apparent from the principle of transfer, the present invention can be applied to two types of temperatures or four or more types of temperatures.
Furthermore, it is possible to provide a plurality of temperature chambers and selectively use the portion. For example, although three temperature chambers are provided, one or two of them can be selected and used as appropriate. The unused temperature chamber is brought to almost the same volume by a normal pressure operation, so that no solution is wasted. On the other hand, in Non-Patent Document 2 mentioned above, since the fluid is transferred by a subtle volume change of the chamber provided at the minus end of the microchannel, it becomes difficult to transfer between multiple points (for example, three or more temperatures). . In addition, waste of solution may occur when there are multiple points.

(2) Specifications of the used cold heat source A cartridge heater and thermocouple were incorporated in the steel block, and the temperature of the steel block was controlled by an electronic temperature controller and SSR (solid state relay). The temperature settings of the three copper blocks were 95 ° C., 72 ° C., and 60 ° C., respectively, corresponding to denaturation, elongation, and annealing.
(3) Specifications of PCR Solution Used PCR Core System II manufactured by Promega Corporation was used as the PCR reagent.
As template DNA and primers, plasmid DNA and primers for PCR control contained in the above-mentioned products were used. When PCR is performed normally, a 323 bp DNA fragment is amplified.
The PCR solution was prepared as shown in Table 2 below per 50 μL.
(4) Pressure operation The PCR solution was first introduced into a temperature chamber corresponding to denaturation at a preload of 70 KPa. The total reference volume of the temperature chamber and the pressure chamber is about 2 μL. Therefore, about 2 μL per system and about 20 μL total PCR solution was introduced in 10 parallel processing systems. Different PCRs can be performed for each strain, but this time all were made the same PCR solution. The forward pressure operation was performed by pressurizing 110 KPa into the pressure chamber. The reverse pressure operation was performed by releasing the pressure in the pressure chamber to the atmosphere because a preload was applied to the solution.
(5) Temperature cycle The temperature cycle was performed at the following time.
Initial denaturation 2 minutes + (denaturation 30 seconds + annealing 30 seconds + extension 30 seconds) × 30 cycles + final extension 2 minutes in total 49 minutes In addition, the cycle time may be further shortened, but the above is long considering allowance Cycle time.
(6) Confirmation of PCR The temperature chamber during the temperature cycle could be observed from the pressure chamber side.
There was no gas expansion in the PCR solution.
After the temperature cycle, about 20 μL of the PCR product was recovered from the microphone mouth fluid chip in a total of 10 parallel processing systems.
Therefore, it is assumed that the evaporation of the solution by PCR was almost negligible.

It was confirmed that there was a clear band in the vicinity of 323 bp, so that it was proved that PCR was possible under the preload at the same time that PCR was normally performed by the heating and cooling structure of the present invention.
