Description

本発明の一実施形態において、細胞質膜の正常な構成が乱された細胞に、化学化合物を選択的に標的化する方法が提供され、このような細胞は、死プロセス(例えばアポトーシス)を受けた細胞、および、活性化を受けた血小板から選択される。以下、このような細胞を包括的に、PNOM(perturbations of the normal organization of the cell plasma membrane: PNOM)細胞という。本方法は、細胞群中に存在する、または散在する上記細胞に選択的に化学化合物を標的化することに関する。本方法は:
(i)細胞群を、乱された膜に結合する化合物(perturbed membrane binding compound:PMBC)と称される化学化合物、または、PMBCを含む結合体と接触させる工程、を含む。PMBCは、式(I):
bは、整数であり、ここで、Wが酸素であるか、または、存在しない場合、1であり;Wが窒素の場合、2であり;または、Wが炭素原子である場合、3であり;
AおよびA’は、それぞれ独立して、以下の4種の基のいずれか一種から選択されるラジカルであり:
i)水素;
ii)SO3H、および、L−SO3H、ここで、Lは、C1、C2、C3、C4またはC5アルキレンリンカーを意味し;
iii)式IIに記載の構造:

(i)PMBCまたはPMBCを含む結合体、および、イメージングのためのマーカーを、ヒトまたは動物に投与する工程、ここで、前記PMBCは、式(I)に記載の構造によって示され、式中、A、A’、X、Z、R1、R2、R3、R4、R5、R6、L、cおよびdは上記で定義された通りであり;および、(ii)組織中の細胞に結合したPMBCの量を決定する工程、ここで、組織中の細胞に結合した化合物の量がコントロールより有意に高いことは、前記組織が、PNOM細胞を含むことを示す。

本発明の実施形態において、細胞質膜の正常な構成が乱された細胞に選択的に結合する化合物、および、それらの使用が提供され、このような細胞は、死プロセス(例えばアポトーシス)を受けた細胞、および、活性化を受けた血小板から選択される。以下、このような細胞は、包括的に、PNOM細胞という。
本発明のアルキル、アルキレン、アリール、および、アラルキルのいずれかは、1またはそれ以上のアルキル、ニトロ、ハロまたはヒドロキシ基で置換されていてもよい。
i.イメージングのためのマーカー、これは、色、蛍光、X線、CTスキャン、磁気共鳴イメージング(MRI)、および、放射性同位体スキャン、例えば単光子放射型断層撮影法(SPECT)または陽電子放射断層撮影法(PET)の検出器から選択され;
ii.PNOM細胞の出現を示す特定の病気を、予防、回復または治療するための薬物。このような薬物としては、これらに限定されないが、以下のようなあらゆる薬物が挙げられる:(i)アポトーシス阻害剤(例えば、カスパーゼ阻害剤、抗酸化剤、Bcl−2系のモジュレーター);(ii)細胞死の活性化因子、アポトーシス誘発物質(例えば、抗ガン薬);(iii)血液凝固のモジュレーター、これは、凝固防止剤、血小板凝集阻止薬、または、血栓溶解薬から選択される。本発明の実施形態によれば、上記薬物は、抗血小板薬、ヘパリン、低分子量ヘパリン、糖タンパク質IIb/IIIaのアンタゴニスト、組織プラスミノゲン活性化因子(tPA)、または、凝固因子の阻害剤、例えばトロンビン阻害剤、または、Xa因子阻害剤から選択してもよく;(iv)抗炎症薬、または、免疫調整薬;または、(v)また、特定の放射性原子も、十分な用量で運搬される場合、細胞毒性となり得る;
iii.固体支持体。
Xは、原子(C、N、OまたはS)を示し、ここで、これらの原子はそれぞれ、Zの意味に応じて0、1または2個の水素原子を有していてもよく;

過剰なアポトーシスの出現を特徴とする病気、例えば、変性性の障害、神経変性障害(例えば、パーキンソン病、アルツハイマー病、ハンチントン舞踏病)、AIDS、骨髄異形成症候群、虚血性または毒性の発作、移植による拒絶反応の際の移植細胞の損失; 腫瘍、および、特に極めて悪性の/高悪性度の腫瘍もまた、過剰な組織の増殖に加えて、促進されたアポトーシスを特徴とすることが多い。このような実施例によって、医学的な障害は、中枢神経系腫瘍、乳ガン、肝臓ガン、肺ガン、リンパ腫または黒色腫のようなガンが挙げられる。
本発明を理解し、実際的にどうように実行可能であるかを観察するために、いくつかの実施形態を以下で説明するが、ここでは、本発明の化合物の、アポトーシスによるPNOMを受けた細胞への結合の検出を評価した。
a.5−ブチルアミノ−ナフタレン−1−スルホン酸(1):
ジメチルホルムアミド(2.2L)中の、1−アミノ−ナフタレン−1−スルホン酸(111g,0.497モル)、炭酸水素ナトリウム(124g,1.48モル)、および、1−ブロモブタン(79mL,0.734モル)の混合物を撹拌し、125℃で2.5時間加熱した。この混合物を、水(13L)と塩化ナトリウム(2.90kg)の混合物に注入し、pHを濃塩酸で8.9から3.0に低くし、形成された固体をろ過によって単離し、水で洗浄し(250mLで3回)、乾燥させ、53.9gの茶色の粉末を得た。この固体を、炭酸水素ナトリウム(22.4g,0.267モル)を加えながら、水(2.5L)に溶解させた。pHを7.9〜3.0に低くするために、塩酸(3M,78mL)を添加した。沈殿をろ過によって単離し、50℃で乾燥させ、47.9gの茶色の固体を得た。再結晶(イソプロパノール:水=2:1)により、44.5gの1を得た(32%)。
1(42.7g,0.153モル)を、水(850mL)に懸濁し、および、炭酸水素ナトリウム(38.6g,0.459モル)を添加した。得られた溶液を、15℃で撹拌し、硫酸ジメチル(15mL,0.16モル)を添加した。6時間後に、さらに硫酸ジメチル(15mL,0.16モル)を添加した。この溶液を一晩撹拌し、80℃で0.5時間加熱した。室温に冷却した後に、pHを3.0に調節した。形成された固体を一晩ろ過し、水で洗浄し、50℃で乾燥させ、32.8g(73%)の茶色の粉末2を得た。
窒素下で、2(23.9g,0.081モル)を氷で冷却し、五塩化リン(18.7g,0.0896モル)と混合した。この混合物を水浴で60℃で2時間加熱し、氷(164g)と水(341g)の混合物に注入した。酢酸エチル(400mL)と、炭酸水素ナトリウム(50g,0.60モル)を添加した。相分離を行い、酢酸エチルで水相を抽出した。合わせた有機相を洗浄し、乾燥させ(硫酸ナトリウム)、活性炭(1.1g)で処理した。溶媒を蒸発させることにより、20.8gの3を油として得た。この生成物は、クロマトグラフィーで精製できる。
2ミリモルのL−グリシン、および、3ミリモルのNaHCO3を、5mLの水に溶解させた。1ミリモルの3を含むアセトン溶液をゆっくり添加した。さらに溶液が透明になるまでアセトンを添加した。この混合物を一晩室温で撹拌し、光を遮断した。アセトンをフラッシュ蒸発により除去し、残存する水溶液を、2Mクエン酸を加えることによってpH4.5〜5に酸性化した。次に、NaClを飽和するまで添加し、この溶液を、酢酸エチルを3回(それぞれ20mL)用いて抽出した。抽出物を合わせ、MgSO4上で乾燥させ、溶液をろ過した。溶媒を除去し、残留物をシリカゲルを用いた液体クロマトグラフィーで精製した。溶出液として、ジクロロメタン中のメタノール勾配0〜5%を用いた。望ましい生成物を含む分画を合わせ、溶媒を除去した。1:1のエーテル:石油エーテル中で残留物を粉砕した。沈殿物をろ過して除き、真空中で一晩乾燥させることによって、5−ブチルメチルアミノ−ナフタレン−1−スルホニル−グリシン(NST703)を得た。

2ミリモルのアスパラギン酸と、4ミリモルのNaHCO3を、5mLの水に溶解させた。1ミリモルの5−ジブチルアミノ−ナフタレン−1−スルホニル塩化物(塩化バンシル(bansyl chloride),TCIから購入した)を含むアセトン溶液をゆっくり添加した。さらにアセトンを、溶液が透明になるまで添加した。この混合物を一晩室温で撹拌し、光を遮断した。アセトンをフラッシュ蒸発により除去し、残存する水溶液を、2Mクエン酸を加えることによってpH4.5〜5に酸性化した。次に、NaClを飽和するまで添加し、この溶液を、酢酸エチルを3回(それぞれ20mL)用いて抽出した。抽出物を合わせた、MgSO4上で乾燥させ、溶液をろ過した。溶媒を除去し、残留物をシリカゲルを用いた液体クロマトグラフィーで精製した。溶出液として、ジクロロメタン中のメタノール勾配0〜5%を用いた。所望の生成物を含む分画を合わせ、溶媒を除去した。1:1のエーテル:石油エーテル中で残留物を粉砕した。沈殿をろ過して除き、真空中で一晩乾燥させることによって、5−ジブチルアミノ−ナフタレン−1−スルホニル)−L−アスパラギン酸(NST705)を得た。
ESI-MS: Consistent. m/z + H+= 451.4。
培養したJurkat細胞(ヒト成人のT細胞性白血病細胞)を、10%のウシ胎仔血清(FCS)、4mMのL−グルタミン、1mMのピルビン酸ナトリウム、1mMのCaCl2、および、抗生物質(100ユニット/mlのペニシリン;100μg/mlのストレプトマイシン、および、12.5ユニット/mlのナイスタチン)が添加されたRPMI培地(Beit−Haemek,イスラエル)の懸濁液中で成長させた。アポトーシスの誘発の前に、培地を、HBS緩衝液(10mMのHEPES;140mMのNaCl、1mMのCaCl)で置き換えた。次に、アポトーシスを、抗FasAb(0.1μg/ml;3時間)での処理によって発生させた。その結果、かなりの割合の細胞がアポトーシスを起こし、これらは新たに形成された初期のアポトーシス細胞とみなされる。未処理細胞をコントロールとして用いた。次に、コントロール細胞とアポトーシス細胞の両方を、100μMのNST705と共に20分間インキュベートした。その後、細胞を、ヨウ化プロピジウム(PI)で共染色し、細胞死の後期の膜の崩壊のマーカー、および、NST705化合物のコントロールとアポトーシス細胞への選択的な結合を、ベクトン・ディッキンソンのセルソーターとCellQuestソフトウェアを用いたフローサイトメトリー(FACS)解析によって決定した(356nmで励起させ、発光を530nmで測定した)。図1に示される解析は、コントロールと抗Fas処理した培養物中のアポトーシス細胞群へのNST705化合物の取り込みを説明する。

8週齢の雄DBA/2マウス(ハーラン・ラボラトリーズ(Harlan laboratories),イギリス)に、リンパ腫細胞を皮下注射することによってリンパ腫を誘発させた。

腫瘍、特に攻撃性の悪性腫瘍(例えば黒色腫)は、異常な組織の増殖に加えて、腫瘍細胞の著しいアポトーシスによっても特徴付けられる。従って、腫瘍中のこれらのアポトーシス細胞を選択的に標的化するDCの性能を試験した。マウス(c57/黒;8週齢の雄マウス)に、マウス黒色腫から誘発されたB16−F10細胞(ATCC CRL−6475;容積100μlで、105細胞/マウス)を両側のわき腹の皮下に注射した。注射の前に、細胞系を、4mMのL−グルタミン;100ユニット/mlのペニシリン;100μg/mlのストレプトマイシン;12.5ユニット/mlのナイスタチン、および、10%のウシ胎仔血清(FCS)が添加されたダルベッコ改変イーグル培地(DMEM)の培養中で維持した。腫瘍を14日間成長させたところ、腫瘍は直径5〜7mmに達した。
18Fの短い半減期(約110分間)が、この同位体が、本発明のPMBCのPETイメージングのためのマーカーとして付着するための特定の条件を規定している。この条件としては、なかでも、塩基性溶液中での迅速な反応、および、比較的高温(「PETホットボックスケミストリー(PET Hot Box chemistry)」)が挙げられる。これらの条件は、本発明のPMBCの比較的感受性のあるスルホンアミドサブユニットに適合していない場合がある。また、望ましくない副反応も生じる可能性がある。これら起こり得る難点を克服するために、末端ダンシル化標識法を開発した(TDLM)。この方法によれば、PMBCは、2つのサブユニット:アミンサブユニット(これに18Fが連結されると予想される)と、スルホン酸サブユニット(例えばダンシル)を含むとみなされる。この方法によれば、18Fは、まずアミンサブユニットに付着される。精製前の最終工程としてのみ、フッ素化アミンサブユニットにスルホン酸サブユニットを付着させ、それによって、望ましい化合物を形成する。この反応条件を最適化することにより、ダンシル化の最終工程の実行を10分間にすることができ、それによって、「PETホットボックス」計画における18F付着の時間枠に適した合成が可能になる。
NST730の、腫瘍中のアポトーシス細胞へ選択的に標的化する性能を、実施例7で説明されているようにして試験した。マウス(c57/黒;8週齢の雄マウス)に、マウス黒色腫から誘発されたB16−F10細胞(ATCC CRL−6475;容積100μlで、105細胞/マウス)をわき腹の両側の皮下に注射した。腫瘍を14日間成長させたところ、腫瘍は直径5〜7mmに達した。
DBAマウス(5〜6週齢の雄,ジャクソン・ラボラトリーズ(Jackson laboratories))に、50μlの塩類溶液中の106個のlyS細胞を皮下注射し、腫瘍形成に関して毎日試験した。8、9および10日目に、マウスをパースペックス製の箱に置き、6MVのX線装置(Linac)で、それぞれ6グレイを数回にわけて照射した。各マウスに、約1.0センチグレイ/モニターユニットの線量が照射された。13日目(最後の放射線照射から72時間)に、マウスに、NST732(70mg/kg,25%クレモフォア(cremophore)RH410、pH9を含むトリス塩基に溶解させた)を静脈注射した。2時間後に、腫瘍を切り出し、液体窒素中で凍結させ、組織病理学的な評価を行った。
Balb/Cマウス(10〜12週間)で、中大脳動脈を焼灼することによって脳虚血を誘発した。マウスを麻酔し、側頭骨を露出させた。骨を最小限の大きさの穴まで削り取って、焼灼を受ける動脈に接触できるようにした。22時間後、NST732(70mg/kg)を静脈注射した。2時間後(傷害の誘発から24時間で)、マウスを麻酔し、殺して、凍結させた切片を組織病理学的に解析するために、脳を採取して液体窒素中に入れた。蛍光顕微鏡による解析のために、4μmの薄片を作製し、それにより、細胞をNST732ではっきりと染色した。
Balb/Cマウス(10〜12週間)で、中大脳動脈を焼灼することによって脳虚血を誘発した。マウスを麻酔し、側頭骨を露出させた。骨を最小限の大きさの穴まで削り取って、焼灼を受ける動脈に接触できるようにした。22時間後、NST730(70mg/kg)を静脈注射した。実施例11で説明されているように、2時間後(傷害の誘発から24時間で)、マウスを麻酔し、殺して、凍結させた切片を組織病理学的に解析するために、脳を採取して液体窒素中に入れた。

ジメチルホルムアミド(2.2L)中の、5−アミノ−ナフタレン−1−スルホン酸(111g,0.497モル)、炭酸水素ナトリウム(124g,1.48モル)、および、1ブロモブタン(79mL,0.734モル)の混合物を撹拌し、125℃で2.5時間加熱した。この混合物を、水(13L)と塩化ナトリウム(2.90kg)の混合物に注入し、pHを濃塩酸で8.9から3.0に低くし、形成された固体をろ過によって単離し、水で洗浄し(250mLで3回)、乾燥させ、53.9gの茶色の粉末を得た。この固体を、炭酸水素ナトリウム(22.4g,0.267モル)を加えながら、水(2.5L)に溶解させた。pHを7.9〜3.0に低くするために、塩酸(3M,78mL)を添加した。沈殿をろ過によって単離し、50℃で乾燥させ、47.9gの茶色の固体を得た。再結晶(イソプロパノール:水=2:1)により、44.5gの1(32%)を得た。
化合物1(42.7g,0.153モル)を、水(850mL)に懸濁し、炭酸水素ナトリウム(38.6g,0.459モル)を添加した。得られた溶液を、15℃で撹拌し、硫酸ジメチル(15mL,0.16モル)を添加した。6時間後に、さらに硫酸ジメチル(15mL,0.16モル)を添加した。この溶液を一晩撹拌し、80℃で0.5時間加熱した。室温に冷却した後に、pHを3.0に調節した。形成された固体を一晩ろ過し、水で洗浄し、50℃で乾燥させ、32.8g(73%)の茶色の粉末2を得た。
HeLaS3細胞(ATCC CCL−2.2)を、2mMのL−グルタミン;100ユニット/mlのペニシリン;100μg/mlのストレプトマイシン;12.5ユニット/mlのナイスタチン、および、10%のウシ胎仔血清(FCS)が添加されたダルベッコ改変イーグル培地(DMEM)中で成長させた。細胞を、5×106細胞/プレートの密度で、10cm3の培養プレート(容積10ml)上に植え、培養物を、増殖培地を交換しないで96時間インキュベートすることによって、老化させた。その結果、かなりの割合の細胞がアポトーシスを起こした。細胞スクレーパーを用いて細胞を回収し、18G注射針を備えた注射器で植え継ぐことによって単一の細胞に分離し、106細胞/mlの密度でpH7.4のPBS緩衝液中に再懸濁した。NST601化合物のアポトーシス細胞への選択的な結合を、ベクトン・ディッキンソンのセルソーターとCellQuestソフトウェアを用いたフローサイトメトリー(FACS)解析によって決定した(356nmで励起させ、発光を530nmで測定した)。図10に示される解析は、NST601化合物のアポトーシス細胞群への取り込みを説明する。
培養したJurkat細胞(ヒト成人のT細胞性白血病細胞)を、10%のウシ胎仔血清(FCS)、2mMのL−グルタミン、1mMのピルビン酸ナトリウム、1mM 1mMのHEPES、および、抗生物質(100ユニット/mlのペニシリン;100μg/mlのストレプトマイシン、および、12.5ユニット/mlのナイスタチン)が添加されたRPMI培地(Beit−Haemek,イスラエル)の懸濁液中で、成長させた。アポトーシスの誘発の前に、培地を、HBS緩衝液(10mMのHEPES;140mMのNaCl、1mMのCaCl)で置き換えた。次に、アポトーシスを、抗FasAb(0.1μg/ml;3時間)での処理によって発生させた。その結果、かなりの割合の細胞がアポトーシスを起こした。未処理細胞をコントロールとして用いた。次に、コントロール細胞とアポトーシス細胞の両方を、250μMのDAで20分間インキュベートした。その後、細胞を、ヨウ化プロピジウム(PI)で共染色した。DAのコントロールとアポトーシス細胞への結合を、ベクトン・ディッキンソンのセルソーターとCellQuestソフトウェアを用いたフローサイトメトリー(FACS)解析によって決定した(356nmで励起させ、発光を530nmで測定した)。解析は、図11に示される。
