期の注目がなされた。しかしながら、治癒組織への骨髄由来細胞の組み込みが実証されたのに対して、骨髄由来のどのタイプの細胞が組織へ組み込まれたのか、及びこれらの細胞が損傷組織の機能的回復にどの程度寄与したのかということを含む多くの疑問が依然として明らかになっていないままである。それにもかかわらず、ヒトの実験を含むこの分野における研究を進行させる治療上の効果における十分な潜在能力が見られた。骨髄由来細胞療法に対する実験は、未分画骨髄としても既知である骨髄全体、あるいはその内部に含まれる内皮前駆体、造血幹細胞(CD34+、AC133+)及び非造血幹細胞(CDstrol+)を利用している。未分画骨髄、骨髄由来前駆体及び幹細胞が実験的に使用され続けている一方で、これらの使用に関する初期の結果は、ほんの限られた改善又はネガティブな結果であり、期待していたものではなく、初期の動物実験と骨髄由来細胞の治療上の価値の関連性に疑問を投げかけた。
本発明は、損傷組織の機能を改善させるための組成物及び方法を提供することにより上述の問題に対処する。本発明は、プレプロB細胞、プロB細胞、プレB細胞、未熟B細胞及びいくつかの成熟B細胞を含むB細胞系列の細胞が、損傷組織へ移植されると、損傷後の回復を促進する能力を有することを初めて実証する。

本発明は、有効な量のB細胞の投与により損傷組織を治療する新たな有効且つ効率的な方法を提供する。組織損傷は、非限定的に虚血、糖尿病及び他の慢性疾患、臓器及び組織移植、外傷、火傷、卒中(虚血性及び出血性)、感染、炎症、手術並びに他の原因などの多数の原因から生じる。投与されるべきB細胞は、好ましくは自己細胞であり、当業者に既知の手法を使用してドナーから回収される。かかるB細胞の供給源は、一般的に既知である骨髄、血液、脾臓、リンパ節及び同種異系供給源が挙げられるが、これらに限定されない。続いて、比較的純粋なB細胞の集団を得るために、B細胞は不均質細胞集団から精製される。かかる精製手法は本出願に記載されている。不均質細胞集団からB細胞を精製するためのキットが提供される。
B細胞の任意の供給源が本発明で有用である。かかるB細胞は当業者に既知であるように、骨髄、脾臓、リンパ節、血液又はB細胞の供給源である他の同種異系組織に由来する。B細胞の好ましい供給源は骨髄及び血液である。かかる方法に関する参照文献としては、Funderud S 他, 「免疫磁気分離によるバフィコートからポジティブ選択されるCD19+Bリンパ球の機能特性」, Eur J Immunol, 1990, 20(1):201-6、Monfalcone他, 「マウスリンパ節を酵素的に解離することにより調製される細胞懸濁液におけるB細胞及びT細胞マイトジェンに対する白血球多様性並びに応答の増加」, J Leukoc Biol 1986 39(6):617-28が挙げられ、Miltenyi Biotecは、組織からB細胞及び形質細胞を単離するためのプロトコルと共にキットを販売している。
不均質細胞集団からB細胞又は前駆B細胞を得る方法は当業者に既知である。これらの
手法の多くが目的のB細胞又はB細胞前駆体の表面上の分子を認識する一次抗体を用い、これらの抗体を使用して細胞をポジティブ選択し、望ましくない細胞と分離させる。この手法はポジティブ選択として既知である。当業者により一般的に用いられる他の手法は、目的のB細胞又はB細胞前駆体と分離されるべき細胞の表面上の分子を認識する一次抗体を使用する。この方法によると、これらの望ましくない細胞上の分子が抗血清に結合して、これらの細胞は不均質細胞集団から除去される。この手法はネガティブ選択として既知である。ポジティブ選択手法及びネガティブ選択手法を組み合わせて、比較的純粋なB細胞又は前駆B細胞の集団を得ることもできる。本出願では、「比較的純粋な」とは少なくとも純度80%、純度85%、純度88%又はより高い純度、例えば少なくとも純度90%、少なくとも純度95%、少なくとも純度97%又は少なくとも純度98%であることを意味する。
間葉細胞へのB細胞接着を増大させるため、及びB細胞活性を促進するために、B細胞は任意に低酸素条件へ暴露させることにより前処理されてもよい。上述するように、いったん骨髄が回収されて、B細胞が単離されると、この前処理を行うことができる。B細胞の送達に先立って、B細胞は、生理学的レベル未満のレベルの酸素を含有する閉鎖系内でインキュベートされる。
投与されるべき細胞数は、治療されるべき損傷組織の面積又は容積、送達の方法、及び場合によっては治療対象の種に関連する。例えば、ラットは、比較的小さい心臓を有するため、心筋は、イヌ、ブタ及びヒトのようなより大きな種よりもはるかに小さく薄い。ラット研究で使用される細胞の数は、実施例1及び図面に記載されている。本明細書中で報告されるラット研究における注入は、それぞれ4回の20μlの注入から構成された。したがって、終了時に、80μl中に含まれる総計106個の細胞が、およそ1.2gmの重量のラット心臓へ注入された。
匹当たりの総注入容量は、種、送達方法及び治療対象の組織の容量に応じて、10μl〜10mlの範囲である。
B細胞は、損傷組織又は損傷組織の周辺へ任意の方法を用いて投与される。B細胞は、シリンジ及び適切な大きさの針を介した注入により投与される。
抗原へ接着し、同様に移植表面又は損傷組織へ接着して、当該部位でB細胞局在化を引き起こす、バイファンクショナル抗体のような物質を全身的に送達することができる。このアプローチは、動物又はヒトから回収される自己B細胞の投与と併用することができる。このアプローチはまた、自己B細胞が動物又はヒトから回収されない状況であるが、損傷組織での内因性B細胞の増加が望ましい場合に使用することができる。例えば、損傷組織を有する動物又はヒトへ投与されたかかる物質は、循環するB細胞のような利用可能なB細胞に、及び同様に損傷組織の部位に結合することができ、それによりその部位でB細胞の数を増加させる。
注入は、好ましくは32ゲージ〜21ゲージの範囲、あるいは30ゲージ〜23ゲージの範囲の小ゲージ針によりなされる。針サイズは、注入される組織のタイプ、深さ及び厚さに応じて多様である。例えば、ラットでは、27ゲージ針を用いる。ヒトでは、様々なゲージの針又はカテーテルが使用される。27ゲージ針を利用するBoston Scientific Stiletto(商標)心筋注入カテーテルが用いられてもよい。一実施形態では、注入は、心膜を通して心筋の梗塞を生じた領域へなされる。
投与されるべきB細胞は、薬学的に許容可能な液体のような、薬学的に許容可能なキャリア中に懸濁される。かかる液体としては、生理食塩水、細胞培養培地及び血漿が挙げられるが、これらに限定されない。さらなる薬学的に許容可能なキャリアとしては、サイトカイン及び成長因子を有する又は有さない、スキャフォールド、マトリックス、グルー、ゲル及び他の組織保持キャリアが挙げられる。
損傷組織へのB細胞の投与の時期は、患者の状態に応じて様々である。概して、B細胞は、損傷後に数時間、数週、数ヶ月又は数年投与される。本発明はまた、慢性状態を治療するのに使用されてもよく、その場合、投与スケジュールは、医師及び獣医により決定される。細胞は、患者の状態の重篤性、及びB細胞の投与後の機能の回復に応じて、損傷後
に1回又はそれ以上、患者へ投与される。
組織損傷の治療のための細胞単独の使用に加えて、本発明は、他の物質と細胞との併用療法を念頭に置いている。かかる物質としては、組織機能の回復、あるいは投与されるべき細胞の機能及び/又は生存(送達の成功)を促進する物質が挙げられるが、これらに限定されない。かかる物質としては、成長因子VEGF、FGF、IGF−1;ニトログリセリンのような酸化窒素ドナー;COX−2阻害剤;抗炎症薬;HMGコレダクターゼ阻害剤;スタチン;アンジオテンシン変換酵素(ACE)阻害剤、トロンビン、並びに非限定的にCXCL12、CXCL13、CCL19、CCL21、CCL25、CXCL9及びCXCL10を含むケモカイン;NSAIDSケモカイン拮抗薬、トロンビン、細胞外分子、並びに非限定的にMADCAM1(粘膜アドレシン細胞接着分子1)及びVCAM1(血管細胞−接着分子1)を含むインテグリンリガンド;インターロイキン−4;血管拡張薬、浸透作用物質、抗凝結剤、酸/塩基改質剤及び界面活性剤といった血管灌流を改変する物質、又はそれらの組合せが挙げられるが、これらに限定されない。これらの物質は、当業者に既知の投与量且つレジメンで投与される。
B細胞は、任意の損傷組織、あるいは機能低下組織へ投与される。好ましくは、B細胞は、任意の損傷後の組織へ投与される。組織損傷は、多様な原因に起因する。本発明の方法で治療される組織の損傷につながる疾患又は状態としては、虚血又は出血、臓器移植、外傷、手術、炎症、感染、心血管疾患、並びに急性梗塞、慢性虚血及び灌流が適切でない心筋を含む心臓病、末梢血管疾患、糖尿病に起因する灌流の低下、並びに慢性腎虚血を含む腎臓病、うっ血性心不全及び虚血性卒中が挙げられるが、これらに限定されない。
本発明は、ヒトにおける損傷組織を治療するためのシステムを包含する。このシステムは、損傷組織を有するヒトからB細胞を含有するサンプルを得ること、比較的純粋なB細胞の集団を得るために当該サンプルを精製すること、組成物を生成するために当該比較的純粋なB細胞の集団を薬学的に許容可能なキャリアと組み合わせること、及び当該ヒトにおける損傷組織を治療するのに有効な量の組成物を、損傷組織を有するヒトへ投与することを含む幾つかの工程を含む。
本発明は、ヒトB細胞の単離のために使用するキットを包含する。これらのキットは、不均質細胞集団からのB細胞を単離するのに有用である。キットを使用するための説明書が提供される。
ラットにおける虚血後の心機能の回復
骨髄細胞はドナーラットから回収され、幹細胞及びB細胞集団へ分離され、梗塞を生じたレシピエントラット心臓へ直接注入された。これらの動物は極度の近交系であり、多くの場合ヒトの条件に代えてこのようにして使用される。ベースライン及び2週にわたる追跡心エコー検査を実施して、左心室収縮末期径及び左心室拡張末期径並びに短縮パーセント(短縮パーセントは、心臓の短軸「拍動径」(拡張末期径と収縮末期径との間の距離)を拡張期における直径で除算したものである)を測定した。この測定は、全体的な左心室機能を正確に反映して、駆出分割率と強く相関する。
とも純度80%、純度85%、純度88%、又はより一層高い純度、例えば少なくとも純度90%、好ましくは少なくとも純度95%、好ましくは少なくとも純度97%、又は好ましくは少なくとも純度98%であることを意味する。いったん集団を分離して、純度を蛍光励起細胞走査分析(図1)により確認されると、細胞は、心筋注入の当日まで37℃で保持されるか、あるいはすぐに動物へ注入された。細胞注入の当日に、HSC、NHSC及びB細胞をDPBSで洗浄して、注入用の滅菌生理食塩水中に再懸濁させた。
これらの実験の別の態様では、動物に、37℃で一晩培養したB細胞を投与した。個々に培養することに加えて、幹細胞及びB細胞の両方を共培養させて、B細胞はトランスウェルフィルタ(孔径0.4μm)の上部に、幹細胞はトランスウェルフィルタの下に配置させた。細胞は、B細胞対幹細胞の1:1の比で播種して、1%ペニシリン及び1%ストレプトマイシンを含有するDMEM中で一晩培養した。B細胞又は幹細胞のいずれかを翌日回収して、注入用に調製した。

t検定を実施して、p<0.05で両側検定に基づいて有意性を評価した。分散分析(ANOVA)もまた、全ての種々の心機能測定に関して実施されて、群間の最小二乗平均における差が検出された場合、Tukey posthoc解析を実施して、p値を付与した。
心筋梗塞が生じた後のヒト患者の治療
55歳男性は、アンギナを訴えて彼の心臓専門医を訪ねた。動脈造影図により、左前下行枝において90%閉塞が明らかとなっている。トレッドミル試験中に、患者は、急性心筋梗塞を患い、生き延びている。続く試験により、左心室心筋の一部が、梗塞により損傷
を受けていることが明らかとなっている。
心筋梗塞が生じた後のヒト患者の治療
右冠動脈の梗塞を伴う62歳女性患者は、虚血性損傷を患っている。B細胞を患者の骨髄から得て、細胞の投与前に一晩培養した(1億2千万個の細胞を含有する1000μl)。当業者により梗塞が生じた後の患者へ提供されるものに類似したプロトコルを使用して、患者は薬物のレジメン下に置かれる。翌日、患者の冠動脈に、大腿動脈を介して鼠径部に導入されるガイドカテーテルをカニューレ挿入する。PTCAバルーンを、このカテーテルを通じて冠動脈へ導入して、十分な圧力で膨らませて、冠血流を塞いだ。PTCAワイヤを除去した後、細胞を、バルーンワイヤ管腔を通して注入して、膨らませたバルーンに対して遠位に送達させた。およそ90秒後、バルーンをしぼませて、手順を終了させた。完了後、患者は、その後数ヶ月モニタリングされた。CD19+細胞の注入を施した虚血領域は、この期間中に血管分布及び収縮機能の改善を示した。
心筋梗塞が生じた後のヒト患者の治療
左心室において梗塞を伴う35肥満男性は、外科的バイパスではなくPTCAが適用された。B細胞を腸骨から回収して、大動脈弁を交差させ、左心筋を嵌め込んで、心筋へ直接細胞を注入することを可能にする針を配置させる大腿経由デバイスを用いて送達させた。注入に先立って、B細胞は、細胞保持、生存性及び有効性を高めるフィブリンシーラント、生体ポリマー又はコラーゲンのようなマトリックス物質と混合された。さらに、梗塞の周辺に配置されるステントは、マトリックス、及びこれらの細胞を優先的に誘引及び保持するB細胞抗原と反応する抗体でコーティングされた。
虚血後のうっ血性心不全を伴うヒト患者の治療
患者は、虚血により引き起こされると考えられるうっ血性心不全に罹患している。この手順は、患者の腸骨稜から骨髄50〜500mlを回収することから始まり、キット(密閉バッグ系)を用いて比較的純粋な配合物中からCD19+細胞を単離した。これらの細胞の送達に先立って、患者の冠動脈に、PTCA業界に共通した診断用カテーテルをカニューレ挿入した。精製した細胞の動脈への送達は、診断用カテーテルへの細胞の注入により達成された。心機能の改善は、細胞の投与の数日後に測定された。
出血性卒中後の中枢神経系に対する損傷を伴うヒト患者の治療
患者は、左中大脳動脈から分岐している小動脈からの出血に起因する出血性卒中に罹患している。中心後回周辺での出血は、運動機能に大きな悪影響を及ぼす。この手順は、患者の腸骨稜から骨髄50〜500mlを回収することから始まり、キット(密閉バッグ系)を用いて比較的純粋な配合物中からおよそ2億個のCD19+B細胞を単離した。これらの細胞の送達に先立って、患者の内頸動脈に、インターベンション外科医に一般的に既
知の診断用カテーテルをカニューレ挿入した。デバイスは、左内頸動脈へ配置されて、中大脳動脈の原点へと装着した。薬学的に許容可能なキャリア中に懸濁させた精製したB細胞は、中大脳動脈へ徐々に注入されて、血流により出血の部位へ運搬された。運動機能の改善は、B細胞の投与の数日後に測定された。
出血性卒中後の中枢神経系に対する損傷を伴うヒト患者の治療
患者は、後大脳動脈から分岐している小動脈からの出血に起因する出血性卒中を患っていた。後頭皮質の18野及び19野付近での出血は、視覚機能に大きな悪影響を及ぼす。この手順は、患者の血液からB細胞を回収すること、及びキット(密閉バッグ系)を用いて比較的純粋な配合物中からCD19+B細胞を単離することから始まる。これらの細胞の送達に先立って、患者の頭蓋を定位固定装置で固定して、後頭骨における開頭手術を、神経外科医に一般的に既知の手法により実施した。硬膜を穿孔して、薬学的に許容可能なキャリア中に1億個を越えるB細胞を含有する注入シリンジに接続された針を、マイクロマニピュレータを使用して血塊を取り囲む18野へ導入した。続いて、B細胞を1〜5μlの容量で徐々に注入した。この手順を、血塊を取り囲む19野への注入においても繰り返した。視覚機能の改善は、B細胞の投与の数日後に測定された。
PTCA手順後の腎臓に対する損傷を伴うヒト患者の治療
患者は、透視診断における可視化のための造影剤を使用するPTCA手法を用いた、心筋の血管を再生させるための一般的な手法を受ける。患者の腎臓は、造影剤に反応して、造影誘導ネフロパシーと呼ばれる腎臓の一部の機能不全を引き起こす。この手順は、患者の後腸骨から骨髄50〜500mlを回収することから始まり、キット(密閉バッグ系)を用いて比較的純粋な配合物中からCD19+細胞を単離した。これらの細胞の送達に先立って、患者の腎動脈に、腎動脈への溶液の送達用に設計された送達カテーテルをカニューレ挿入した。精製した細胞の動脈への送達は、カテーテルへの細胞の注入により達成された。腎機能の改善は、B細胞の投与のその後数週間にわたって観察された。
腎臓移植を受けるヒト患者の治療
末期腎疾患を患う患者は、組織適合ドナーから腎臓の移植を受ける。移植後、骨髄50〜500mlを患者の後腸骨から回収して、キット(密閉バッグ系)を用いて比較的純粋な配合物中からCD19+細胞を単離した。続いて、これら単離されたB細胞は、腎臓を外科的に移植する前に、ドナー腎臓全体にわたって注入される。腎機能及び臓器拒絶反応の改善は、B細胞の投与のその後数週間にわたって観察された。
ヒト骨髄からのB細胞の単離
この実施例では、B細胞の供給源として骨髄が選択され、骨髄がポジティブ免疫磁気分離により選択される。ヒト自己骨髄B細胞は、密閉且つ滅菌環境で流体移行及び分離プロセスの両方が可能である回収バッグが取り付けられた、特別に設計された分離チャンバー中で単離された。このB細胞選択プロセスで使用されるU材料は滅菌されている。回収された骨髄は、分離チャンバーへ入れられて、リン酸緩衝生理食塩水(PBS)で1:2に希釈され、チャンバーの遠心分離セクション内に含有されるフィコール密度勾配上へ層状にされた。骨髄細胞を密度勾配遠心分離に付した。上部フィコール層は、廃棄バッグに誘導された。中間層内に含有される骨髄単核球(MNC)は、光学センサにより検出、回収され、PBSで洗浄された。洗浄したMNCは、B細胞特異的抗原であるCD19に対して誘導され、マイクロ磁気ビーズへ結合した抗体を含有する回収バッグに誘導された。細胞は、抗体−マイクロ磁気ビーズと共に制御温度で20分間インキュベートされた。続い
て、磁石を回収バッグに適用して、その結果、抗体−マイクロ磁気ビーズに結合した細胞が、回収バッグの側面に維持される。抗体へ結合しなかった細胞を除去した。続いて、磁石を回収バッグから分離して、抗体−マイクロ磁気ビーズへ結合した細胞を、細胞を抗体から移動させるCD19により認識されるペプチドを含有する緩衝液中に懸濁させた。磁石を再度回収バッグに適用して、放出されたCD19+B細胞を取り出し、回収した。B細胞は、細胞を未結合ペプチドと分離させる滅菌フィルタ上に洗浄されて、未結合ペプチドは、フィルタを通って廃棄管へと流れる。B細胞を緩衝液中に再懸濁させて、患者へ移植し戻す準備が整う。
この実施例で使用される材料の幾つかとして、遠心分離チャンバー、活栓マニホールド、接続ユニット、接続ライン、投入生成物用ポート、洗浄溶液用ポート、再懸濁用ポート、針注入部位を有する排出生成物用ポート、無針ルアー、回収バッグ又は管、廃棄物バッグ、投入ライン離脱装置、最終生成物用のバイアル、0.2μmフィルタ、リン酸緩衝生理食塩水(PBS)、細胞移植緩衝液、フィコール溶液及びマイクロ磁気ビーズへ結合したCD19抗体が挙げられる。
ヒト血液からのB細胞の単離
この実施例では、B細胞の供給源として全血が選択され、全血がネガティブアフィニティー分離により選択される。ヒト自己末梢血B細胞は、密閉且つ滅菌環境で流体移行及び分離プロセスの両方が可能である回収バッグが取り付けられた、特別に設計された分離チャンバー中で単離された。このB細胞選択プロセスで使用される材料は全て滅菌されている。回収された血液は、分離チャンバーへ入れられて、遠心分離チャンバーに誘導される。血液細胞の遠心分離後、血漿を含有する上部層は、廃棄袋に誘導される。中間層であるバフィコート内に含有される富化された白血球は、光学センサにより検出されて、回収バッグに誘導された。細胞をPBSで洗浄し、CNBr活性化セファロースに結合した非B細胞特異的抗原に対する抗体を含有する緩衝液中に再懸濁させた。使用した抗体は、以下のヒト細胞表面抗原:T細胞、NK細胞、顆粒球、単球/マクロファージ及び赤血球上に存在するCD2、CD3、CD14、CD16、CD36、CD43、CD56並びにグリコホリンAに誘導された。細胞−抗体混合物は、温度を制御して20分間インキュベートして、空のカラムへ入れた。溶出緩衝液をカラムに添加して、抗体に結合しない細胞、即ちB細胞が溶出された。B細胞をPBSで洗浄して、緩衝液中に再懸濁させて、患者へ移植し戻す準備が整う。
この実施例で使用される材料の幾つかとして、遠心分離チャンバー、活栓付きのプラスチックカラム、活栓マニホールド、接続ユニット、接続ライン、投入生成物用ポート、洗浄溶液用ポート、再懸濁用ポート、針注入部位を有する排出生成物用ポート、無針ルアー、回収バッグ、廃棄物バッグ、投入ライン離脱装置、最終生成物用のバイアル、リン酸緩衝生理食塩水(PBS)、細胞溶出緩衝液、細胞移植緩衝液、CNBr活性化セファロースに結合した細胞系列特異的抗体(抗CD2、抗CD3、抗CD14、抗CD16、抗CD36、抗CD43、抗CD56及び抗グリコホリンA抗体)が挙げられる。
Claims (11)
- 動物又はヒトの損傷した虚血性組織を治療するための組成物であって、
薬学的に許容可能なキャリア中の少なくとも純度80%の単離された自己のB細胞集団を含み、
前記組成物の量は、損傷した虚血性組織を治療するのに有効な量であり、
前記少なくとも純度80%の単離されたB細胞集団は、
幹細胞及びB細胞を含む不均質の細胞集団から幹細胞を実質的に除去して、少なくとも純度80%のB細胞の集団を得る工程により調製される、組成物。 - 更に、CXCL13を含有する、請求項1に記載の組成物。
- 更に、幹細胞動員因子、成長因子VEGF、FGF、IGF−I、酸化窒素供与体、COX−2阻害剤、利尿薬、血管新生因子、血流促進因子、抗炎症薬、抗高血圧症薬、HMGコレダクターゼ阻害剤、スタチン、アンジオテンシン変換酵素(ACE)阻害剤、創傷治癒促進薬、NSAIDS、ケモカインアンタゴニスト、トロンビン、細胞外分子、ケモカイン、インテグリンリガンド、及びインターロイキン4から選択される1つ以上を含有する、請求項1又は2に記載の組成物。
- 前記B細胞が骨髄、リンパ節、脾臓又は血液に由来するものである、請求項1〜3の何れか1項に記載の組成物。
- 前記B細胞がCD19+細胞、B220+細胞又はB細胞受容体(Igαβ)+細胞あるいはそれらの組合せである、請求項1〜4のいずれか1項に記載の組成物。
- 前記損傷した虚血性組織が心臓組織である、請求項1〜5のいずれか1項に記載の組成物。
- 動物又はヒトの損傷した虚血性組織を治療するための組成物を生産する方法であって、損傷組織を有する動物又はヒトから得られたB細胞を含有するサンプルを精製し、少なくとも純度80%の単離されたB細胞を生産する工程、
前記少なくとも純度80%の単離されたB細胞を薬学的に許容可能なキャリアと組み合わせて、組成物を調製する工程を含み、
前記少なくとも純度80%の単離されたB細胞集団が、幹細胞及びB細胞を含有する不均質の細胞集団から幹細胞を実質的に除去して、少なくとも純度80%のB細胞の集団を得ることを含む方法により調製される、
動物又はヒトの損傷した虚血性組織を治療するための組成物を生産する方法。 - 更に、幹細胞動員因子、成長因子VEGF、FGF、IGF−I、酸化窒素供与体、COX−2阻害剤、利尿薬、血管新生因子、血流促進因子、抗炎症薬、抗高血圧症薬、HMGコレダクターゼ阻害剤、スタチン、アンジオテンシン変換酵素(ACE)阻害剤、創傷治癒促進薬、NSAIDS、ケモカインアンタゴニスト、トロンビン、細胞外分子、ケモカイン、インテグリンリガンド、及びインターロイキン4から選択される1つ以上を加える工程を含む、

- 前記B細胞を含有するサンプルが、CD19+細胞、B220+細胞又はB細胞受容体(Igαβ)+細胞あるいはそれらの組合せである、請求項7〜9のいずれか1項に記載の方法。
- 前記損傷した虚血性組織が心臓組織である、請求項7〜10のいずれか1項に記載の方法。
