Description
The present invention relates to a measuring instrument for measuring a measurement object contained in a sample to be analyzed such as a living body, food, and soil, and in particular, a flow cell type in which the measurement object in a sample can move along a flow path. The present invention relates to an improvement of a measuring instrument including a reaction vessel, a measuring kit using the measuring instrument, a measuring method, and a measuring apparatus.
Quantitative analysis and qualitative analysis using a piezoelectric vibrator such as a quartz vibrator are known. In this method, a complex is generated on a quartz resonator using the interaction between antigen and antibody and the interaction between complementary nucleic acids, and the mass change associated with the complex formation is changed by the change in the frequency of the quartz resonator. It is a method to grasp as. Until now, for example, a method of detecting IgG in a sample using protein A immobilized on a crystal resonator (see, for example, Non-Patent Document 1) or a probe DNA immobilized on a crystal resonator has been used. A method for detecting a target nucleic acid in a sample (see, for example, Patent Document 1) has been reported.
In addition, a flow cell is known in which a quartz vibrator sensor for simply measuring biological reactions such as cell adhesion and blood coagulation in a fluid state is fixed (see, for example, Patent Document 2).
Furthermore, in immunoassay methods other than flow cells, a method is known in which magnetic particles are used as a labeling substance, and the amount of magnetic particles fixed on the crystal resonator is quantified by measuring changes in the oscillation frequency of the crystal resonator. (For example, see Patent Document 3).
Furthermore, a measurement instrument for immunochromatography in which a piezoelectric vibrator is incorporated in a detection site has also been reported (see, for example, Patent Document 4). In this patent document 4, in an immunoassay method in a flow cell, an insoluble carrier is used as a labeling substance, and the amount of insoluble carrier immobilized on a quartz vibrator as a piezoelectric vibrator is measured for a change in the oscillation frequency of the quartz vibrator. A method of quantifying by doing so is disclosed.
However, the immunoassay method for measuring the change in the oscillation frequency of these crystal resonators has a technical problem that the detection sensitivity is not sufficient when measuring a very small amount of antigen or antibody. Therefore, as shown in
For example, in the flow cell type shown in
Furthermore, in the immunoassay method other than the flow cell type shown in Patent Document 3, even if a method using magnetic particles as a labeling substance is adopted, it is still insufficient in terms of reaction efficiency, and the original sensitivity of the sensor cannot be obtained. There are concerns.
Furthermore, in the immunoassay method in the flow cell shown in Patent Document 4, even if an insoluble carrier is used as a labeling substance, there is a concern that the reaction efficiency is still insufficient and the sensor's original sensitivity cannot be obtained.
In recent years, point-of-care testing (POCT), in which sample results are collected in the vicinity of a subject and output of measurement results, has become popular due to the rapid and diversified inspection methods and the flow of inspections in the environmental and food fields. Therefore, it is desired to develop a simple and highly sensitive measuring instrument that can handle POCT, a measuring kit, a measuring method, and a measuring apparatus using the measuring instrument.

This is supported by the examples described later.

According to the present embodiment, in addition to obtaining the same highly sensitive measurement result as in the first embodiment, when the use of the measuring device is finished, the measuring instrument disposable part S1 is discarded and a new one is disposed. The measuring instrument disposable part S1 may be connected to the apparatus instrument analysis processing part S2 to perform the next inspection job. For this reason, the measuring apparatus according to the present embodiment does not require large devices such as the
Embodiment 3
The measuring apparatus according to the first and second embodiments is a model using a single crystal resonator as the
Here, the frequency drift means a frequency fluctuation other than a frequency change based on specific binding of the target substance. For example, when the added sample solution comes into contact with the
In the first and second embodiments, if such a frequency drift exists, the drift component is added to the measurement value, so that there is a concern that an error may occur in the measurement value.
In order to reduce the measurement error due to such a drift phenomenon, a method using a reference crystal resonator can be considered.

The measurement procedure in this embodiment is exactly the same as that in the second embodiment.
According to this embodiment, in addition to non-specific adsorption and temperature change, it is possible to perform measurement in which drift based on an unexpected cause is cancelled. Therefore, improvement in measurement accuracy can be expected.

20A and 20B show an outline of a modified form of the measuring instrument, and FIG. 21 shows an example of the structure of the two-channel crystal resonator 250. FIG.
In the figure, two sets of electrodes are provided on one crystal substrate 255, which are used as a test electrode T (back side) and a reference electrode R (back side), respectively. The front electrode of each electrode is connected to the extraction electrodes TT1 and TR1 by routing the wiring to the back side of the crystal substrate via the end face of the crystal. On the other hand, the back electrode is connected to the lead electrodes TT2 and TR2 on the back surface.
In this way, as in the case where two crystal resonators are used by forming two sets of electrodes T and R on one crystal substrate 255 and pulling out the wirings individually, each resonance frequency is unique. It functions as two vibrators having

Hereinafter, this format is referred to as a reference type two-channel crystal resonator.
FIG. 22 is a diagram showing a
As described above, in the case of using the reference type two-channel crystal resonator 250 instead of using two individual crystal resonators, the drift removal capability is excellent as compared with the case of using two individual crystal resonators. Therefore, it is suitable when higher accuracy and detection limit are required .
Further, a configuration in which a plurality of target substances are detected using two reference type two-channel crystal resonators 250 is also possible. For example, an influenza A virus can be detected by the first reference type 2-channel crystal resonator 250, and an influenza B virus can be detected by the second reference-type 2-channel crystal resonator 250.

As the present embodiment, in addition to the above-described embodiment in which one of the lines is used as a reference crystal resonator, there may be an embodiment in which a test crystal resonator is provided on both lines. In an aspect in which the test crystal resonators are provided on both lines, the same measurement object in two samples can be measured simultaneously, or two types of measurement objects in the same sample can be measured simultaneously.

fo=1/(2・π・(Lx・Cx)1/2) (式3)
Since the admittance Y of the
fo = 1 / (2 · π · (Lx · Cx) 1/2 ) (Formula 3)
The resonance frequency change thus obtained can be converted into the concentration of the measurement object using calibration curve data obtained in advance using a sample solution having a known concentration, and the result can be displayed on the display device.
Further, the size of the crystal resonator was 2.2 mm × 4.5 mm, and a gold electrode having a film thickness of about 150 nm was formed by RF sputtering.
Next, immobilization of the trapper on the crystal resonator will be described.
Prior to immobilization, the gold electrode on the surface of the crystal unit was washed with a sodium hydroxide solution and a hydrochloric acid solution, and then about 15 μL of 100 μg / mL biotinylated BSA aqueous solution was dropped on the surface electrode. It was retained for about 1 hour and immobilized. Next, in order to prevent non-specific adsorption, 15 μL of a 1% BSA (bovine serum albumin) aqueous solution was dropped and kept in a wet box for about 1 hour for blocking.
FIG. 30 is a schematic diagram when the labeling body is reacted with the trapper immobilized on the surface of the
Using the measurement system described above and the sensor substrate, a magnetic particle reaction experiment was performed. The sample solution used in the experiment was prepared by diluting the above-mentioned commercially available magnetic particle suspension with PBS buffer from 40-fold to 5120-fold at 2-fold intervals. The frequency change at that time was measured by sequentially flowing in min. First, the measurement was performed with the concentration magnet removed, and then the measurement was performed with the magnet installed.
FIG. 31 is a diagram showing a typical change in frequency when a sample solution (magnetic particles having streptavidin immobilized on the surface) diluted with the magnet removed is sequentially flowed. The sample solution diluted 5120 times at the timing t1 and the sample solution diluted 2560 times at the time t2, and then the sample solution are supplied at intervals of 5 minutes until the timing t9. Although the change in frequency is not observed at timings t1 to t5 when the concentration of the sample solution is low, the change in frequency is observed from timing t6. Thereafter, the change in frequency increases as the concentration of the sample solution increases. At each timing, the concentration of the sample solution changes and the frequency starts to change, and the concentration dependence was calculated by assuming that the frequency change per minute at that point is proportional to the amount of magnetic particles bound to the trapper. .
FIG. 32 shows the result of concentration dependency.
In the figure, the concentration of magnetic particles is represented by v / v% of a commercially available magnetic particle suspension contained in the sample solution, and 40-fold dilution corresponds to 2.5 v / v%. The vertical axis represents the frequency change rate.
When the magnet is removed, the magnetic particles cannot be detected below 0.3125 v / v% (320 times dilution) and can be measured from 0.625 v / v% (160 times dilution). In this case, it was possible to measure from 0.015 v / v% (640-fold dilution), and a sensitivity difference of about 4 times was recognized.
In the present embodiment, the height of the flow path at the

As the magnetic particles, magnetic particles having a particle size of about 50 nm to 3000 nm are used, and an antibody that specifically binds to the nucleoprotein of influenza A virus is immobilized on the surface thereof. On the other hand, an antibody that specifically binds to the nucleoprotein of influenza A virus is also immobilized on the electrode surface of the
The test is performed by solubilizing the nucleoprotein by adding 0.1% to several percent of surfactant to the nasal rinse or nasal aspirate or gargle collected from the patient. Add to the

The acquisition of the frequency change amount due to the binding of the complex uses the frequency decrease Δfm when a certain time Tm has passed after a certain time Tw after the frequency is lowered by the contact. The relationship between the frequency drop amount Δfm and the amount of nucleoprotein in the virus contained in the sample solution or the amount of virus is determined using calibration curve data stored in the

FIG. 34 is a diagram showing changes in frequency of the

On the other hand, in the reference crystal resonator, the complex is not captured, and only the frequency change due to the drift phenomenon appears.
The frequency change due to the drift phenomenon is considered to occur in the test crystal resonator as well, and by taking the difference between the two, only the frequency change due to the complex containing the target substance can be extracted.
The actual acquisition of the frequency change amount due to the binding of the complex is, for example, by calculating the frequency difference between the
In acquiring the frequency change amount, the difference Δfm is obtained using only the frequency data between two points (T3 to T4), but the frequency drop amount (Δfm2−Δfm1) is continuously obtained to perform curve fitting. The decrease amount Δfm after a certain time may be obtained by estimation using the above method.
According to the present embodiment, in addition to non-specific adsorption and temperature change, it is possible to perform measurement in which drift based on an unexpected cause is canceled, so that improvement in measurement accuracy can be expected.
