A commercially available BSA-free anti-His tag antibody (“carrier antibody”; Qiagen, Germany) is diluted to a final concentration of 2 μg / mL in PBS (pH 7.4).

Dilute 1:20, 1:50, 1: 100, 1: 200, 1: 300, 1: 400, 1: 600, 1: 800 and 1: 1200 in PBS pH 7.4 with BSA. 100 μl is incubated for 3 hours at room temperature in wells containing recombinant vWF-cp. Inhibitory antibody is bound to the surface of the immobilized vWF-cp and unbound antibody is washed by 10 washing steps using PBST pH 7.4. Detection of human antibodies is performed using mouse anti-human IgG Fc specific antibody or mouse anti-human IgM antibody (alkaline phosphatase conjugate). The antibody is diluted 1: 60000 in PBS 2% BSA to the final working solution, incubated for 2 hours at room temperature (100 μl / well), then washed 10 times with PBST pH 7.4. Addition of alkaline phosphatase substrate (pNPP) produces a yellow color, whereby the dye intensity reflects the amount of bound antibody (antibody / vWF-cp). The dye intensity is measured with an ELISA reader and the amount of antibody in the plasma sample is calculated with reference to a standard curve of NPs by serial dilution. As a negative control, normal human plasma (NHP) dilutions are treated accordingly. The results are shown in FIGS. 2A and 2B. This result indicates that human anti-vWF-cp antibody in the patient can be clearly detected at a plasma dilution of at least 1: 600.
