Description
ゆえに、外来性DNAは、子孫内に伝達されるために、発生の初期段階に胚の中で安定な形で存在しなければならない。
−第一の解決策は、遊離末端を示すDNAの分解である(例えば細胞が、ウイルス活性を持つことが可能なDNAを除去する場合)。この解決策は、維持および修復のこれらのシステムにおける、遊離末端から消化することによってDNAを分解するエキソヌクレアーゼの存在に基づいている。
−他の解決策は、遊離末端の細胞ゲノムとの組換えである。この組換えは、細胞のゲノムへの外来性分子の組込では特に、異なる形を取ることがある。
−これらの問題のうちで第一の問題は、細胞宿主のDNAへの外来性分子の組込の効率である。効率のこのような欠如は、非常に少ない回数の組込に対して、非常に大量の外来性DNAの注入を意味する。魚、植物、または昆虫などの一部の種においては、エキソヌクレアーゼが、非常に効率的であるため、外来性DNAは染色体中に決して組込まれず、エピソームのままとどまる。例えば外来性DNAに環状形を使用する場合、組込を持つためにはニックが必要である。確かに組込工程は、遊離末端の存在を必要とする。直鎖形を使用する場合、大半の外来性DNAは、遊離末端の存在により、エキソヌクレアーゼによって分解される。
−第二の問題は、組込DNAの完全性である。外来性DNAの直鎖断片は、その遊離末端を、核に到達する前に細胞エキソヌクレアーゼに暴露する。細胞中の末端の長期暴露が、外来性DNAが細胞ゲノムに完全に組込まれる機会を重大な方法で制限する。ゲノムに完全に組込まれる機会を増加させる方法は、例えば同じ制限酵素を用いた消化により作成した、ある凝集性一本鎖DNAオーバーハングを外来性DNAの各末端に添加することに存する。これらの付着末端は、DNA断片が多量体中で会合し、組込のDNAの完全な分解を防止することを可能にする。機会を増加させる別の方法は、長いおよび中性のDNA配列を組込むために、DNA断片を包囲することである。外来性DNAが、スーパーコイルプラスミドに包まれる場合、組込まれた断片は、外来性DNAだけでなく、ベクター全体も含む。
−第三の問題は、組込コピーの数の制御である。故意または故意でない外来性DNAの多量体化は、外来性DNA断片の複数のコピーの挿入を好むという、不都合さを示す。同様に外来性DNAが環状形を持つ場合、プラスミドは、コンカテマーとして組込まれる。この複数の挿入は、結果として、染色体の不安定性を導入し、複数のコピーにおける同一遺伝子の存在による発現の制御の問題を生じる。
原則として、アデノ随伴ウイルスの[Fu, 1998 Nature Biotech, 16, 253-257]またはトランスポゾン[Izsvakら、2000 J Mol Biol 302, 93-102]の隣接反復を用いた技術も、導入遺伝子組込を向上できる。しかしながら、発明者の知るところでは、プラスミド中の反復の潜在的に有害な存在からも害を受けるこれらの技法によって、実際的な結果は報告されていない。更にこれらのベクターは、それらの中へ操作することが可能なDNA配列のサイズについて制限される。

これは、その最も一般的な保存配列モチーフによってクラスター形成されたタンパク質の最大のファミリーである(150配列を超える)。12残基配列の1(5配列)または2のコピー(大多数):ジドデカペプチド。1のドデカペプチド(D)を持つメガヌクレアーゼは、分子量が約20kDaであり、そしてホモダイマーとして作用する。2のコピーを持つこれら(DD)は、各モチーフ間に70〜150の残基を持ち、25kDa(230AA)〜50kDa(HO、545AA)の範囲であり、そしてモノマーとして作用する。

直鎖U3Rβgeo断片は、TAE1Xによる0.8%アガロースゲルでの電気泳動により、Xba IおよびNot IによるptkU3Rβgeoプラスミド10μgの消化の後に分離した。9.7kb断片を分離して、そしてGene−Clean IIのプロトコールを用いてガラスビーズ上で精製した。精製したU3Rβgeoは、形質移入のために、pH7のH2O中で再懸濁した。図4を参照。
形質移入の20時間前に、293T細胞を35mm皿に付き5×104細胞の濃度で、10%不活性化ウシ胎仔血清(SVFi)を含むDMEM(ダルベッコ変法イーグル培地)培地に播種した。細胞は、1気圧、5%CO2、100%湿度、37℃のインキュベータでインキュベートした。形質移入は、DNA2μgの存在下で25℃にて、pH7.12で、125mM CaCl2、NaCl 8.18%(W/V)、HEPES 5.94%、Na2HPO4 0.2%の混合物によるリン酸カルシウム沈殿によって実施した。沈殿物は、細胞上で16時間維持した。次に沈殿物と共に培地を新しいDMEM10%SVFi培地と交換した。
DNA混合物(pVirtkU3Rβgeo+pCMV−I Sce I)2μgを、以下の比を用いて、表1に開示する以下の形質移入対照の存在下で、35mm皿に付き5×104細胞の濃度で形質移入した。

ppSV40NeoIRESegfpプラスミドの作成
ppSV40NeoIRESegfp中間プラスミドは、2工程で得た。第一に、サルウイルス40プロモーター(pSV40)およびネオマイシンホスホトランスフェラーゼをコードするneo耐性遺伝子含有するカセットをpcDNA3.1+プラスミド(Clonetech,パロアルト,カリフォルニア州,米国)から切除して、そしてpIRES2−EGFPプラスミド(Clonetech,パロアルト,カリフォルニア州,米国)のSmaI部位にてクローニングし、IRESバイシストロン要素およびEGFP遺伝子の上流にpSV40−Neoカセットの挿入を生じさせた。第二に、PytknislaczプラスミドからのPyTknlslaczカセット(Henryら、C.R.Acad.Sci.III, 322 (12): 1061-1070 (1999) )を切除し、そしてpSV40−Neo−IRES−EGFP(ppSNIE)カセット全体と置換して、そしてSV40polyA部位の前に挿入した。
形質移入の前日に、5×104のCOS細胞を6ウェルプレートの、10%ウシ胎仔血清、2mM L−グルタミンを添加したDMEM(ダルベッコ変法イーグル培地)培地を播種し、そして37℃にて7%CO2雰囲気中でインキュベートした。形質移入は、Effectene法(Qiagen,チャッツワース,カリフォルニア州,米国)によって実施した。形質移入プロトコールは、メーカーの勧告に従って準備した。簡潔には、プラスミドDNA1μgを、エンハンサー試薬の存在下で(DNA/エンハンサー比は1/8である)DNA濃縮緩衝液100μl中で希釈した。5分間のインキュベーション時間の後、Effectene 10μlをDNAに添加した(DNA/Effectene比は1/10である)。混合物をボルテックスにかけ、そして室温にて10分間インキュベートした。次に完全培地600μlをDNA/Effectene複合体に添加し、混合して、そして細胞に散布した。翌日、細胞を洗浄し、37℃にて7%CO2雰囲気中で、全48時間インキュベートした。
ゲノムDNAは、High Pure DNA Prepキット(Roche,マンハイム,ドイツ)を用いて培養細胞から抽出し、そしてメーカーのプロトコールに従って、Picogreen(商標)dsDNA定量化キット(Molecular Probes,ユージーン,オンタリオ州,米国)を用いて定量した。
ppSNIEプラスミドは、I−SceIエンドヌクレアーゼ発現ベクターpCMV−I−Scel+(Choulikaら、1995 Mol Cel Biol, 15, 1968-1973)によって同時形質移入した。陰性対照として、ppSNIEプラスミドを、エンドヌクレアーゼをコードするORFが逆配向を持つ、そしてしたがってI−SceIを発現させない、pCMV−I−SceI−と同時形質移入した。DNA混合物1μg(ppSNIEプラスミド0.5μg+pCMV−I−SceI−0.5μg)またはppSNIEプラスミド0.5μg+pCMV−I−SceI+0.5μg)を用いて、5×104の細胞を6ウェルプレートで形質移入した。形質移入の48時間後、細胞を収集し、そしてフローサイトメトリーによって分析した。COSの形質移入率は再現的に、GFP+細胞の35〜45%であった。ゆえにppSNIEプラスミドの、I−SceI発現(pCMV−I−SceI+)または対照(pCMV−I−SceI−)ベクターとの同時形質移入は、EGFPレポーターの一過的発現に影響を与えなかった。
魚卵への導入遺伝子組込を上昇させるI−SceIメガヌクレアーゼによるインオボ直鎖化
GFPレポーター遺伝子が続き、そしてI−Sce I認識部位によって隣接された筋肉特異性プロモーターを持つ、環状プラスミド性DNAを1細胞期魚胚に注入した。本構築物を(酵素が、細胞外で不活性を維持するように)マグネシウムを含まない緩衝液中のI−Sce Iメガヌクレアーゼと同時注入した場合、予想GFP筋肉発現は、目覚しく向上する(メガヌクレアーゼなしで注入を実施したときの20%と比較して、胚の80%が体幹筋系において強い蛍光を示した)。更になお顕著なのは、生殖細胞伝達の劇的な上昇であった。古典的な卵注入実験においては、大半の場合で遅いキメラ組込によって2、3パーセントを超えないのに対して、メガヌクレアーゼと同時注入された魚での生殖細胞伝達の頻度は50%に上昇し、単一の挿入が創始魚の1細胞期にて発生することが示唆された。更なるサザン分析は、単一の少ないコピー組込が大半の場合に発生することを確認した。早期組込事象が、メガヌクレアーゼによる核への標的化によってではなく、DNAのインビボ開裂によるか否かをアッセイするために、(I−Sce Iメガヌクレアーゼによって開裂されず、結合された)欠失または突然変異認識部位を持つ対照作成物をメガヌクレアーゼと共に注入した。本発明者らは、メガヌクレアーゼ誘発性インオボ直鎖化が、組込前DNA遊離末端の分解を制限することによって、卵注入による形質転換を向上させるための簡単および有効なプロセスであることを示唆する。
プラスミド構築物
pαact−GFPM2は、α−アクチン筋肉特異性プロモーターによって作動されるEGFPレポーター遺伝子を持つ7.8kbプラスミド内に2のI−SceI認識配列を用いて作成した(pαact−GFP、ヒガシジマ博士より寄贈、[Higashijima, 1997#698])。簡潔には、2つのサブクローニング工程を使用して、Bluescriptポリリンカ中のα−アクチン/GFP/polyAカセットの両端に位置する、Eco RIおよびKpnl部位に<<メガリンカ>>を挿入した。メガリンカは、EcoRlまたはKpnl消化生成物のどちらかと適合性である遊離末端に隣接されるI−SceI認識部位(CCGCTAGGGATAACAGGGTAATATA)を含有する、相補性オリゴヌクレオチドをアニーリングすることによって産生した。メガリンカと直鎖化プラスミドとの連結の後、プラスミドを直鎖化するために使用する酵素によって、DNAを再度消化し、そして熱パルスによってXL1 Blue ultra-competent E.coli (Stratagene)に変換された。クローンは、非パリンドローム性I−SceI部位の配向を決定するために配列決定した。
メダカ胚およびオレンジレッド系統の成魚(日本、東京大学のA.Shimaおよび日本、千葉のY.Ishikawaより寄贈)をすべての実験で使用した。魚は26℃にて20リットル水槽で飼育した。成魚は、繁殖条件(明期14時間/暗期10時間)に置いた。卵は、産卵後、ただちに収集し(明期の開始時)、洗浄し、そしてヤマモトの胚飼育培地に[Yamamoto, 1975]に入れた。注入の場合、最近形成された胚盤を持つ1細胞期胚を選択し[Iwamatsu; 1994]、そして4℃に移して発生を停止させた。1回に約10個の胚を、細胞質盤を上にして、[Westerfield; 1993]で述べられたようにプラスチック型を用いて作成したアガローススロットに配置した。すべての実験において、超微操作装置MM3(Fine Science Tools,ドイツ)および圧力注入器(FemtoJet,エッペンドルフ,ドイツ)を装備したOlympus SZX12立体顕微鏡を使用した。ホウケイ酸ガラス毛細管(外径1mm、GC100T,Clark Electromedical Instruments,英国)は、垂直引張機(PC−10,Narashige,日本)を用いて引いた。毛細管には、滅菌マイクロローダー(Femtotips,エッペンドルフ,ドイツ)を用いて、注入溶液(DNA:10μg/ml;メガヌクレアーゼ緩衝液(Roche Diagnostic,ドイツ):0.5x;メガヌクレアーゼI−SceI:1ユニット/μl;0.1%フェノールレッド)を充填した。注入の直前に、マイクロピペット先端を細い鉗子で直径約1マイクロメートルまで割った。薄い胚盤に直接ピペットを挿入することによって、漿膜を通じてDNAを注入した。1回の圧力パルスが、フェノールレッドによって視覚化された溶滴(見積体積300pl)の注入を引き起こした。次に胚をアガロースから除去し、そして26℃にてペトリ皿に置いた。注入の3日後に、GFPの発現が最初にスクリーニングされた。次に胚を成熟期まで飼育し、魚が導入遺伝子をその子孫に伝達したことを確認するために、対交配を実施した。
サザンブロッティングでは、F1魚からのゲノムDNAをプロテイナーゼKおよびフェノールを用いて抽出した[Sambrook and Russel; 2001]。十分なDNAを得るために、満1ヶ月の全魚を使用した。DNAseを避けるため、より高齢の魚からの筋肉をDNA抽出のために切開した。DNAは、BamHl、EcoRl、Xbal、Xholを用いて完了するまで消化した。DNA標準物質は、対応する酵素を用いてpαact−GFPプラスミドを消化することによって調製した。一倍体ゲノム当たり109bpの見積DNA含有量を使用して、消化されたpαact−GFPプラスミド50pgを装填した。サンプル(レーン当たり100□g)1xTBE中の0.9%アガロースで電気泳動させ、Zetaprobeメーカーが推奨するように毛細管上方移動によって、ナイロンZetaprobe膜 (Biorad)へブロットした。フィルターは65℃にて、αアクチン/GFP/polyAに対応するランダム初回刺激放射標識プローブ(Xhol/EcoRV断片)を用いて、ローラーボトル中で一晩ハイブリダイズした。
胚は、370〜420nm励起フィルターおよび455nm LP発光フィルターを用いて、MZFLIII Leica解剖顕微鏡を使用して観察および評価した。写真は、Nikon DXM1200デジタルカメラを用いて撮影した。写真のために、胚は[Wakamatsu; 1993]で述べられた手順に続いて、孵化酵素によって漿膜除去し[Yasumasu; 1994]、そしてPBS中の80%グリセロールまたはVectashield封入剤に入れた。
注入胚の筋肉におけるGFPの一過的発現は、メガヌクレアーゼは、作成物と同時注入されたときに向上する。
I−SceIメガヌクレアーゼは、あるNLS活性を持つことが既知である。早期組込事象が、メガヌクレアーゼによる核への標的化によってではなく、確かにDNAのインビボ開裂によるものか否かを評価するために、(メガヌクレアーゼによって開裂されず、結合された)欠失(pαact−GFPDM)または突然変異(pαact−GFPMM)認識部位を持つ対照構築物をメガヌクレアーゼと共に注入した。
GFP筋肉発現は、注入実験から生じた一部の成魚において持続した。したがって重要な点は、導入遺伝子が、次世代に伝達されるか否かおよび伝達される方法、および特に、成魚筋肉における発現レベルおよび生殖細胞伝達の頻度との間に何らかの相関があるか否かを調査することである。
本実施例は、複数のレベルでの魚形質転換における目覚しい改良につながるメガヌクレアーゼ仲介技術を報告する。
マウスにおけるI−SceIメガヌクレアーゼによるインオボ直鎖化
形質転換効率を向上させるために、本発明者らはメガヌクレアーゼによるスーパーコイルプラスミドのインオボ直鎖化に基づく新規な方法を開発した。
注入は、B6SJLマウス卵からのオス前核にて、1細胞期に実施した。I−SceI発現ベクターは、B6SJLマウス卵からのオス前核にて1細胞期に、pScel/EGFPプラスミドを微量注入することによってインオボで確認した。EGFP蛍光は、EGFPフィルターを備えた倒立顕微で、注入胚の2細胞期にて既に視覚化された。2のI−SceIおよびEGFP ORFの間にIRESを持つ作成物の性質により、本発明者らは、約95%の場合で、EGFP蛍光が視覚化されたときに、I−SceIは、調和的に生成されることを確信した。Megafluo(図10A)およびpI−SceI/EGFP(図10B)プラスミドは、500コピー/plで同時注入した。注入卵をB6CBA育成メスマウスに移植した。出生を点検し、死産動物を回収し、そして注入導入遺伝子の推定上の組込について、遺伝子型を同定するために、そのDNAを抽出した。新生マウスは、尾の小片を切断することによって3週目に分類した。次にDNA抽出を実施し、続いてPCRおよび/またはサザン実験によって遺伝子型を同定した。本実験より、新生仔5および死産仔2を回収した。DNA抽出の後、DNA回収をアガロースゲル上で点検した(図11A)。抽出されたゲノムDNAの品質を点検するために、β−グロビン遺伝子の494bpサブ断片を増幅するPCRを実施した(図11Bの右パネル)。予想されたPCRβ−グロビン生成物が、試験を行ったすべてのDNAサンプルから視覚化されたため、すべてのDNAサンプルは定性的に正しいと思われる。導入遺伝子の組込について試験を行うために、レポーター遺伝子(DSRed1−E5)の484bpを増幅するPCRを実施した(図11Bの左パネル)。3週齢のマウス子孫3および死産仔2のうち、それらの2(番号1および25)ならびに1(番号A)はそれぞれ遺伝子導入であった。これらの結果はひとまとめにして、これらの結果は、生存動物を考慮した場合に子孫の40%が遺伝子導入であり、そして生存および死産の注入マウスをまとめて考慮した場合には43%が遺伝子導入であることを示した。
プロトコールは、I−SceIが精製タンパク質として1:5 DNA/タンパク質比で供給されたことを除いて、本実験と同じであった。Megafluoスーパーコイルプラスミド750コピー(図10A)をI−SceI精製タンパク質(18μM)と同時注入した。注入卵は、B6CBA育成メスマウスに移植した。新生マウス9匹のうち、Gas5プロモーターまたはプラスミド主鎖配列のどちらかに対応するプローブを使用したサザンブロット分析によって検出されたように、遺伝子導入であった。この結果は、遺伝子導入動物が本方法によって本実験において収率22%で生成されることを示した。
同じプラスミド作成物、Megafluoをマウス形質転換の古典的なプロトコール、すなわち前直鎖化導入遺伝子の注入で使用した。2種類の実験を実施し、一方では1796bpのNdelサブ断片を使用し、もう一方では1824bpのI−SceIサブ断片を使用し、サブ断片は両方ともプロモーターGas5およびレポーター蛍光遺伝子を持っていた。I−SceI断片については、2シリーズの注入を実施した。DNA断片500コピー/plの注入に対応する第一のシリーズは、Red5 PCRによって陰性であるマウス2匹の出生につながった。I−SceI断片750コピー/plの注入に対応する第二のシリーズは、組込について陰性であるマウス8匹の出生につながった。Ndel断片750コピー/plの注入は、8匹の出生につながった。新生仔8匹のうち、1匹のみが遺伝子導入であった(番号18)(図12)。本実験では、遺伝子導入の数は、12.5%であった。3回の実験で、出生動物18匹のうち遺伝子導入1匹が、メガヌクレアーゼ仲介形質転換から得られた数値と比較して、同じDNA配列を使用した場合に得られた遺伝子導入動物の数値を5.5%に低下させる。F1から遺伝子導入オス18への伝達は、このオスをB6SJLメスと交雑させることによって試験した。F1動物25匹のうち、プロモータープローブを使用したサザンブロット分析で検出されたように、得られた6匹のうち1匹および19匹のうちが遺伝子導入であった。これらの結果は、<<古典的な形質転換>>を用いた伝達が約16%であることを示した。
スーパーコイルMegafluoプラスミド750コピー/plの単独での組込も確認した。2つの別個の実験を実施した。マウス10および11がそれぞれ得られた。マウスの遺伝子型を同定するために、PCRおよびサザンブロット分析の両方を使用すると、遺伝子導入動物は検出されなかった。
他のメガヌクレアーゼによるメガヌクレアーゼ仲介形質転換の効率を試験するために、本発明者らは、精製PI−SceIタンパク質を用いて実験を実施した。遺伝子導入構築物、PIFFLagoを図13に示す。レポーター遺伝子LagoZ(CPGなしLacZ配列)の発現につながるGas5プロモーターは、ベクターの主鎖配列を分離する、同じ配向の2のPI−SceI認識/開裂部位と隣接している。プロモーターおよびレポーター遺伝子の間に、単一のI−SceI部位が存在する。
注入用サンプル調製
プラスミドDNA(スーパーコイル)溶液をQiagen Endo-freeカラム内で作成した。沈殿後、次にプラスミドをBrinster緩衝液、10mM TRIS、0.25mM EDTA中で所望の濃度にて再懸濁させた。I−SceI原液は、25mM HEPES、pH8、5%グリセロール中に150μg/ml(10ユニット/μl)を含有していた。表3は、一連のタンパク質:DNA比に対するI−SceIの量を示す。
(プラスミド500コピーは、3.01 10-20molに相当する。I−SceIは、29.3KDaである)
希釈緩衝液:
25mM HEPES、5%グリセロール、25℃にてpH8
同じスーパーコイル作成を発現メガヌクレアーゼベクターまたは精製タンパク質のどちらかと共に注入した。同様に、同じ導入遺伝子の予備直鎖化および精製インビトロ形を使用して、「古典的な形質転換」をメガヌクレアーゼ仲介形質転換と比較した。
プラスミドの生成は、QIAgen Endo-freeキット(QIAGEN)を用いて実施した。回収したDNAを酢酸ナトリウムで沈殿させ、70%エタノールで洗浄し、そして、Brinster緩衝液(10mM TRIS−0.25mM EDTA)中で、注入用の所望の濃度で再懸濁させた。
1.Megafluoを消化して、プラスミドベクター配列から導入遺伝子を遊離した。その目的に2つの異なる酵素を使用した。それぞれ1796bpおよび1824bpにつながる、NdelまたはI−SceI酵素。
2.制限消化生成物を、0.8%TAEを用いてアガロースゲル上で分離した。
3.ゲルをトランスイルミネータ上に置き、所望のバンドを切断し、そして次にQIAquickゲル抽出キット(QIAGEN)を用いて精製した。
4.Tris(pH7.6)中でのDNA溶出後、DNAを沈殿させ、70%エタノールで洗浄し、Brinster緩衝液 Tampon Brinster中で濃度500コピー/plにて再懸濁させた。
5.DNAを−20℃で保存した。
a)過剰***:
−注入3日前に、3週齢B6SJLメスへPMS(Intervetによるfolligon)を5UI/マウスにて腹腔内注入
−注入1日前に、3週齢B6SJLメスへHCG(IntervetによるChorulon)を5UI/マウスにて腹腔内注入、そしてそれらをB6SJLオスと交雑
−B6CBAメスをB6CBA精管切除オスと交差育成。
b)卵母細胞回収:
−B6SJLプラグドB6SJLメスを殺処分し、輸卵管を回収し、そしてそれらをPBI培地に入れた。
−アンプルを鉗子で切断することによって、0.5mg/mlのヒアルロニダーゼ100μl中で卵母細胞を2分間、分離させた。
−卵をPBI中ですすいだ。
−卵をシリコン処理油で覆ったWhittenに写し、それらを37℃、5%CO2で維持した。
c)卵注入
−受精卵を、コンテンションおよび注入毛細管(PhymepによるGC 100−10およびGC100F−10)を用いて、Nikon倒立顕微鏡(TE2000−U)上に微操作装置(transferman NK2 5188)を備えたEppendorf微量注入器(Femtojet5247)を使用してPBI中に微量注入した。
d)育成メスへの注入卵移植:
−Avertin150μg/mlの400μl/マウスでの腹腔内注入によって、B6CBAメスに麻酔をかけた。
−注入卵を切開した輸卵管に移植した。
1)尾部1cm当たり尾部緩衝液750μLおよびプロテイナーゼK40μLまたは胚膜当たり尾部緩衝液500μLおよびプロテイナーゼK30μLを添加した。
2)56℃で一晩インキュベートした。
3)Eppendorfミキサーで5分間混合した。
4)尾部当たり飽和塩化ナトリウム(≒6M)250μLまたは胚膜当たり飽和塩化ナトリウム(≒6M)170μLを添加した。
5)Eppendorfミキサーで5分間混合した。
6)13000rpmで10分間遠心分離した。
7)新しいEppendorf管に、尾部当たり上澄み750μLまたは胚膜当たり上澄み500μLを収集した。
8)尾部当たりイソプロパノール500μLおよび胚膜当たりイソプロパノール350μLを添加した。
9)Eppendorfミキサーで2分間混合した。
10)13000rpmで1分間遠心分離した。
11)室温にて70%のエタノール500μLによって、ペレットを2回洗浄した。
a)RocheによるDig-labellingキットを用いた、精製プラスミドDNA配列 o/n 1μgの標識化。プローブは、G-50 sephadexで精製した。プローブの定量化は、RocheのキットによるDig-controlを用いて、ドットブロット反応について確認した。
b)ゲノムDNA1〜7μgをEcoRI制限酵素(NEB)で消化した o/n。
c)消化したDNAを0.8%アガロースTAEゲルに装填した。
d)ゲルを変性溶液(NaOH(0.5M)+NaCl(1.5M))中で30’変性させ、次に中和溶液(Tris(pH7.4,0.5M)+NaCl(1.5M))中で中和した。
e)DNA転移は、ナイロンHybond−N+膜(Amersham)上で10xSSC o/n 中で毛細管によって行った。
f)DNAは、UV架橋(StratageneによるStratalinker)によって膜に固定して、そして80℃のオーブンで2時間焼成した。
g)膜は、ハイブリダイゼーション緩衝液中で1時間、回転炉内で68℃にて予備ハイブリダイゼーションした。
h)ハイブリダイゼーションは、ハイブリダイゼーション緩衝液中で、回転炉内で68℃にて予備変性Dig標識プローブ(10ng/ml) o/n を用いて実施した。
i)予熱した洗浄溶液によって、回転プラットフォーム上で室温にて5分間、2回洗浄した。
j)緩衝液I(1x)/0.3%tween中で室温にて5分間、1回洗浄した。
k)250rpmで回転させる密閉ポリ袋内のブロッキング溶液中で30分間、膜のブロッキング。
l)ブロッキング溶液中における、抗ジゴキシゲニン−Ap Fab断片(Roche)0.0375U/mlを30分間インキュベーション。
m)緩衝液(1x)/0.3%tween中で2回、それぞれ30分間洗浄。
n)暴露緩衝液(緩衝液III)中で5分間。
o)Rocheによる化学発光サブストレートCDP−Starへの、膜の暴露。
ハイブリダイゼーション緩衝液:0.5M NaPi、7% SDS、1mM EDTA

- ポリヌクレオチドを宿主細胞ゲノム内にランダムに組込むための方法であって、上記方法は、
a)上記宿主細胞内に、遊離5’および3’末端を持たず、そして上記ポリヌクレオチドを含むベクターを導入することであって、上記ベクターは、宿主細胞ゲノム内に見られず、そして上記ポリヌクレオチドに隣接する少なくとも1のエンドヌクレアーゼ開裂部位を含み、

- 個体における状態または障害の処置または予防のための薬剤の調製のための、請求項20記載の細胞の使用。
- 非ヒト遺伝子導入動物を作成する方法であって、胚幹細胞が、請求項1〜19記載の方法によって形質移入され、細胞が、形質移入細胞を組込むことができる期に胚に注入され、次に胚が、代理母に再移植され、そしてキメラ個体が妊娠期間の終わりに得られ、そしてキメラ個体においては生殖細胞系の胚幹細胞によるコロニー形成が観測され、キメラ個体が遺伝子導入動物を得るために交配される、方法。

- 直鎖化される上記導入遺伝子を含む上記ベクターが、更に上記対応するエンドヌクレアーゼをコードする上記核酸を含む、請求項28〜35記載の組成物。
- 上記ポリヌクレオチドが、宿主細胞ゲノムとの相同組換えを受けることができない、請求項28〜36記載の組成物。
